팔손이나무(Fatsia japonica) 줄기의 성분연구 (Chemical Constituents of Fatsia japonica Stem)

  • Fatsia japonica is grown wild to Eastern Asia, including Korea, Japan, and Taiwan and it is known as ornamental plant, and it is also known that pharmacological action. In this study, we have selected the stem of F. japonica with consideration about biological activities and amount of yield. In addition, four compounds (1-4) were isolated from the stem of F. japonica. Extensive spectroscopic and chemical studies established the structures of these compounds as maltose (1), begoniifolide A (2), leiyemudanoside B (3), leonticin F (4). All of the compounds were investigated for their anti-inflammatory, anti-neuroinflammatory, and neuro-protective effects on RAW264.7, BV2, and HT22 cells. However, among four compounds, there were no effects by maltose (1), begoniifolide A (2), leiyemudanoside B (3), leonticin F (4) on the anti-inflammatory, anti-neuroinflammatory, and neuro-protective action. This is the first report on the isolation of maltose (1), begoniifolide A (2), leiyemudanoside B (3), leonticin F (4) from the stem of F. japonica. Begoniifolide A (2), leiyemudanoside B (3), leonticin F (4) were isolated for the first time from this plant. It might be necessary to continue the further studies to find the biological active compounds isolated from the stem of F. japonica.

  • Germination ability of a japonica and 15 indica$\times$japonica varieties was tested at day/night tempuratures of 25/2$0^{\circ}C$ and 12/12$^{\circ}C$. Germination ability of a japonica variety, "Jinheung" and two indican$\times$japonica varieties. "Suweon #251" and "Suweon #278" was greater than that of other indica$\times$japonica varieties tested. Germination pattern of varieties was made into 4 groups.ieties was made into 4 groups.

한국산 쑥속의 체세포 염색체수에 의한 분류학적 연구 (A taxonomic study of Korean Artemisia L. using somatic chromosome numbers)

  • 한국산 쑥속 20분류군의 분류를 위하여 체세포 염색체수를 조사하였다. 본 연구에서 취급된 분류군들의 체세포 염색체수는 2n = 16, 18, 34, 36, 50, 52, 54로서 기본 염색체수는 x=8, 9, 10, 13, 17이었으며, 실제비쑥(A. japonica var. angustissima 2n = 36)의 체세포염색체수는 본 연구에서 처음으로 밝혀졌다. 사철쑥 (A. capillaris 2n = 18), 섬쑥(A. japonica var. hallaisanensis 2n = 36), 갯제비쑥(A. japonica subsp. littoricola 2n = 36), 개똥쑥(A. annua 2n = 18), 개사철쑥(A. carvifolia 2n = 18), 큰비쑥(A. fukudo 2n = 16), 맑은대쑥(A. keiskeana 2n = 18), 넓은잎외잎쑥(A. stolonifera 2n = 36), 그늘쑥(A. sylvatica 2n = 16), 물쑥(A. selengensis 2n = 36), 산쑥 (A. montana 2n = 52), 뺑쑥(A. lancea 2n = 16), 산흰쑥(A. sieversiana 2n = 18) 등의 13분류군의 염색체수는 기존 보고와 일치하였으며, 기존 보고와 다르게 파악된 종류는 제비쑥(A. japonica var. japonica 2n = 18, 36 vs 2n = 36), 더위지기(A. sacrorum 2n = 18, 54 vs 2n = 54), 덤불쑥(A. rubripes 2n = 16, 34 vs 2n = 16), 쑥(A. indica 2n = 34, 36 vs 2n = 34), 참쑥(A. codonocephala 2n = 18, 50, 54 vs 2n = 50), 황해쑥(A. argyi 2n = 34, 36, 50 vs 2n = 34)의 6분류군이었다. 한국산 쑥속의 체세포 염색체수는 제비쑥, 더위지기, 참쑥, 황해쑥, 산쑥, 그늘쑥의 분류에 매우 유용한 형질이었다.

동백나무 추출물을 향장소재로 사용하기 위한 항균, 항산화 활성 탐색 (Antimicrobial and Antioxidant Activity of Camellia japonica Extracts for Cosmetic Applications)

  • This study was carried out to investigate the antimicrobial and antioxidative activities of Camellia japonica extracts for cosmetic applications. Antimicrobial effects of C. japonica were determined against Bacillus cereus by methanol extract of new leaf, stem and stem-leaf; Malassezia pachydermatis, by methanol extract of new leaf and stem-leaf. A methanol extract of new leaf of C. japonica showed strong antimicrobial effect using paper disc method against most species especially in Staphylococcus aureus. Antioxidative activities of C. japonica seed oils were determined by DPPH and ABTS radical scavenging activities. The value of $EC_{50}$ of DPPH scavenging activity was 500 mg/mL and that of ABTS scavenging activity was 96.10 mg/mL. C. japonica oil extracts showed lower antioxidative activities than those of gallic acid and ${\alpha}$-tocopherol. Electron microscopic observation of damaged virgin hairs of different ages gave a stabilizing effects after C. japonica seed oil treatment. These results indicated that the extracts of stem, leaf and seed of C. japonica could be used as cosmetic ingredient combined with appropriate formula.

Feeding Ecology of Sillago japonica in an Eelgrass (Zostera marina) Bed

  • Feeding habits of Sillago japonica collected from in an eelgrass (Zostera marina) bed in Jindong Bay, Korea were studied. S. japonica was a carnivore which consumed mainly gammarid amphipods, polychaetes, bivalves, caridean shrimps and crabs. Its diets also included a small amount of fishes, copepods and caprellid amphipods. The diet of S. japonica underwent significant size-related changes; small individuals (<5cm SL) fed mainly on gammarid amphipods and crab larvae, while proportion of polychaetes and bivalves increased with increasing fish size and gammarid amphipods were also important prey for medium size individuals (5.1-9.9cm SL). The large individuals (>10cm SL) ate polychaetes, caridean shrimps and crabs. The dietary breadth of S. japonica were varied with size. The diet of S. japonica also underwent seasonal changes that could be related to differences in prey availability; gammarid amphipods were mainly consumed in spring and polychaetes in summer.

비파 분말을 첨가한 매작과의 품질 특성 (Quality Characteristics of Maejakgwa Containing Various Levels of Eriobotrya japonica Leaf Powder)

  • The purpose of this study was to investigate the quality characteristics of Maejakgwa prepared with Eriobotrya japonica leaf powder (0, 1, 3, 5, 7%) substituted for flour. The pH and density of Maejakgwa dough significantly decreased in response to addition of all levels of E. japonica leaf powder. Furthermore, when spread factor values were compared among the groups, they were found to be inversely proportional to the E. japonica leaf powder concentration. With increasing amounts of E. japonica leaf powder, L and a values decreased, whereas b value increased. In terms of textural characteristics, addition of E. japonica leaf powder increased hardness, cohesiveness, springiness, chewiness, and brittleness compared to control. Finally, the results of an acceptance test showed that Maejakgwa containing 5% E. japonica leaf powder had the highest scores.

Phylogenetic analysis of Neottia japonica (Orchidaceae) based on ITS and matK regions

  • To elucidate the molecular phylogeny of Neottia japonica, which is a terrestrial orchid distributed in East Asia, the internal transcribed spacer (ITS) of nuclear DNA and the matK of chloroplast DNA were used. A total 22 species of 69 accessions for ITS and 21 species of 114 accessions for matK phylogeny were analyzed with the maximum parsimony and Bayesian methods. In addition, we sought to establish a correlation between the distribution, morphology of the auricles and genetic association of N. japonica with phylogenetic data. The phylogenetic results suggest that N. japonica is monophyletic and a sister to N. suzukii in terms of the ITS phylogeny, while it is paraphyletic with N. suzukii in terms of the matK phylogeny. N. japonica and N. suzukii show similar morphologies of the lip and column, they both flower in April, and they are both distributed sympatrically in Taiwan. Therefore, it appears to be clear that N. japonica and N. suzukii are close taxa within Neottia, although there is incongruence between the nrDNA and cpDNA phylogenies of N. japonica. The incongruence between the two datasets may have various causes, meaning that further studies are needed to confirm the evolutionary process of N. japonica. The phylogenetic status of N. kiusiana, which was not included in previous studies, was as a sister to N. nidus-avis. Meanwhile, the ITS and matK phylogenies are unsuitable for identifying genetic associations with the characteristic of auricles. The phylogenetic topologies of Korean, Taiwanese and mainland Chinese individuals suggest that the populations of N. japonica in Korea originated from China's mainland and island areas. The characterization of regional gene differences could provide useful preliminary data for future studies.

마가루 첨가에 따른 스폰지케익의 품질 및 관능적특성 (Quality Properties and Sensory Characteristics of Sponge Cakes as Affected by additions of Dioscorea japonica flour)

  • In the fabrication of sponge cakes, wheat flour was substituted by Dioscorea Japonica flour at 3%, 6%, and 9% (C1, C2, and C3, respectively), and the sensory, physicochemical, texture properties, viable cell counts and chromaticity of the samples were investigated and compared with the control cake (C0). The addition of Dioscorea Japonica flour increased the density of the sponge cake, but decreased the size and moisture content of the cakes (C0〉C1〉C2〉C3). The pH value increased with increasing Dioscorea Japonica flour in the sponge cakes, and the microbial growth was remarkably suppressed by Dioscorea Japonica flour compared with the control. There was an increase in the chromaticity of the sponge cakes with increased Dioscorea Japonica flour in the cakes. Also, the hardness, gumminess, and chewiness were enhanced approximately two times higher than those of control by increasing Dioscorea Japonica flour. In the sensory evaluation, the addition of Dioscorea Japonica flour into sponge cakes showed a significant increment in the internal color, size/uniformity of the voids, and the sweetness, while the adhesiveness and moistness were the highest in the control. The overall preference was in the order C0〉C2〉C3〉Cl, and therefore, the substitution of 6% of wheat flour by Dioscorea Japonica was recommended in the fabrication of sponge cakes.

새삼, 실새삼 및 갯실새삼 추출물이 Clone M-3 세포주의 Melanin 생합성 및 Tyrosinase 활성에 미치는 영향과 세포독성 및 항산화효과 (Inhibitory Effects on Melanin Biosynthesis and Tyrosinase Activity; Cytotoxicity in Clone M-3 and Antioxidant Activity by Cuscuta japonica, C. australis, and C, chinensis Extracts)

  • Water extracts, ethanol extracts, and juice of Cusuta japonica, C, australis, and C. chinensis were prepared, and their cytotoxicity, antioxidant activity and inhibitory effects on tyrosinase activitiy and melanin biosynthesis were estimated by using melanoma Clone M-3. From this study; the following conclusions were attained. Extracts of Cuscuta japonica, C. australis, and C. chinensis showed noticeable cytotoxicity except ethanol extracts from the stem of C. australis. A maximual cytotoxicity was observed with tile ethanol extract from the seed of C, australis (87.39%). While the ethanol extract from the seed of C. japonica (91.88%) showed the most pronounced inhibitory effect on melanin biosynthesis, the water extract from the stem of C. japonica (1.05%) possessed very little inhibitory effect. The most inhibitory effect on tyrosinase activity was observed with the water extract from the stem of C. australis (76.67%). Howeverr the water extract from the stem of C. japonica showed a very poor effect on the inhibition of tyrosinase activity All the preparations, except extracts from the seed of C. australis were able to remove reactive oxygen species (ROS) in a dose-dependent manner. The juice of C. japonica demonstrated the strongest activity (59.02%).

해부형태적 특징과 SCAR Marker를 이용한 백출의 기원식물 판별 (Discrimination of Atractylodes Rhizome White Using Anatomical Characteristics and SCAR Markers)

  • 국내 한약재 시장에서 유통되고 있는 백출 건재약재의 기원을 판별하고자 해부형태적 특징과 종 특이적인 SCAR 마커 (SAjR2, SAmR1)를 적용한 실험 결과는 다음과 같다. 해부형태적 특징으로 A. japonica에서는 주피에 코르크층이 매우 발달하여 있었고, 코르크층 내부 또는 아래에 석세포대가 연속적으로 발달하고 있었다. 유실은 A. japonica에서 뿌리횡단면에 전체적으로 다수 분포하여 잘 발달하고 있었으며 피충부분과 잘 발달된 방사조직 내에서도 목섬유다발이 다수 관찰됐다. 또한 유세포 내의 침상 결정물도 A. macrocephala보다 많이 함유하고 있었다. 이러한 특징들을 종합한 결과, 국내 유통 국산 및 수입 백출은 모두 A. japonica의 해부형태적 특징을 따르고 있어 A. japonica로 동정되었다. 한편 RAPD clone으로부터 개발된 A. japonica 특이적인 SAjR2를 이용하여 PCR 한 결과 모든 샘플에서 600bp 크기의 한국백출 특이적인 밴드가 형성되었으며, A. macrocephala 특이적인 SAmR1을 이 용한 결과에서는 작물시험장에서 재배중인 A. macrocephala에서만 1200 bp크기의 특이적인 밴드가 형성되어, 시중에서 유통되고 있는 한국백출과 중국백출은 모두 A. japonica로 동정되었다. 결론적으로 해부형태적 특성과 SCAR 마커는 A. japonica와 A. macrocephala 기원식물의 동정과 유통 건재 약재를 판별하기에 유용한 방법이었다.
