Studies on Basidiomycetes(I) -On the Mycelial Growth of Agaricus bitorquis and Pleurotus ostreatus- (담자균(擔子菌)에 관한 연구(硏究)( I ) -고온성(高溫性) 양송이와 느타리의 균사(菌絲) 생육(生育)에 관하여-)

  • The mycelial growth of Agaricus bitorquis and Pleurotus ostreatus in synthetic media were carried out by ordinary methods. The optimum pH and temperature for mycelial growth were from pH 6.0 to 6.5 and 25 to $30^{\circ}C$, and from pH 5.0 to 6.5 and $25^{\circ}C$ in A. bitorquis and P. ostreatus, respectively. Among the carbon and nitrogen sources, glucose, starch, and peptone showed the good result for the mycelial growth of A. bitorquis, and glucose, fructose, starch and peptone were good for the mycelial growth of P. ostreatus. The yield of mycelium decreased under lower or higher C/N ratio. Also, at the same C/N ratio, the higher the concentration of glucose and peptone, the more the yield was increased. Among various vitamins thiamine, Ca-pantothenate and folic acid were suitable for the mycelial growth of A. bitorquis, and thiamine, folic acid and ino­sitol for the mycelial growth of P. ostreatus. Although pH, total nitrogen and glucose contents of media decreased gradually during culture period the yield of mycelium increased.

Selection of Agaricus bitorquis hybrid strains based on RAPD analysis (RAPD분석을 통한 여름양송이(Agaricus bitorquis)의 교잡주 선발)

  • Agaricus bitorquis is an edible white mushroom of the genus that is cultivated at high temperature($25{\pm}1^{\circ}C$) unlike A. bisporus is cultivate at $16{\pm}2^{\circ}C$. Unlike Agaricus bisporus, an edible white A. bitorquis mushroom is cultivated at high temperature ($25{\pm}1^{\circ}C$). Most farmers cultivate this mushroom for a long cultivation period in Korea. For this reason, we made heterokayons to develop a new cultivar that generate fruitbodies for short cultivation period. Over one hundred SSIs(single spores isolates) were collected from selected A. bitorquis ASI1151 and ASI1349 strains. Seventy-three SSIs were germinated on CDA(compost dextrose agar) media after 20 days (minimum) or 83 days (maximum) incubation under different media condition. The mycelial growth rate of germinated SSIs was different. 9 homokaryons in ASI 1151 and 11 homokaryons in ASI 1349 from SSIs were selected by OPN-02 primer in RAPD analaysis. Also this primer was used to select heterokaryon that cross among each homokaryon with compatible locus. Therefore 44 compatible matings were confirmed of 99 crossed lines.

Ribosomal Intergenic Spacer 1 Based Characterization of Button Mushroom (Agaricus bisporus) Strains

  • Breeding the button mushroom requires genetic information about its strains. This study was undertaken to genetically characterize four domestically bred button mushroom strains (Saea, Saejung, Saedo, Saeyeon cultivars) and to assess the possibility of using the intergenic spacer 1 (IGS1) region of rDNA as a genetically variable region in the genetic characterization. For the experiment, 34 strains of Agaricus bisporus, two strains of A. bitorquis, and one strain of A. silvaticus, from 17 countries were used. Nucleotide sequence analysis of IGS1 rDNA in these 37 Agaricus strains confirmed that genetic variations exist, not only among the four domestic strains, but also between the four domestic strains and foreign strains. Crossing two different haploid strains of A. bisporus seems to generate genetic variation in the IGS1 region in their off-spring haploid strains. Phylogenetic analysis based on the IGS1 sequence revealed all A. bisporus strains could be differentiated from A. silvaticus and A. bitorquis strains. Five genetic groups were resolved among A. bisporus strains. Saejung and Saeyeon cultivars formed a separate genetic group. Our results suggest that IGS1 could be complementarily applied in the polymorphism analysis of button mushroom.

Characteristic evaluation of collected strains of Agaricus spp. based on ITS rDNA sequence (ITS 계통분석을 이용한 주름버섯류 수집균주의 특성평가)

  • Phylogenic relationships and morphological characteristics were classified and investigated among the 233 collected strains of Agaricus spp. The 38 strains were differently identified as different characteristic group using analysis of ITS regions in rDNA. A. bitorquis was showed close relationship in groupA whereas A. campestris was in groupC as different characteristic group among with A. bisporus. There was no phylogentic difference with strains collected by country and different pileus colored Agaricus bisporus. Also the strains were cultivated twice to investigate morphological characteristics of fruiting bodies. The characteristics and yield of collected strains were compared with molecular varieties and seasons by the cultivation. In this result, A. campestirs showed good yield and quality in terms of hardness off-white mushroom was more harder than other number of A. bisporus. Also earliness and color of pileus was influenced by external environment all conditions.
