Distribution and Inheritance of Trypsin Inhibitor Variant in Soybeans Grown in Korea (한국대두에서의 Trypsin Inhibitor Variant의 분포와 유전)

  • Seed proteins of 51 varieties of soybean were separated by disc electrophoresis. Nine varieties had Rf 0.79 electrophoretic band, and 42 varieties had Rf 0.83 band. The frequency for Rf 0.79 was 0.176 and the frequency for Rf 0.83 was 0.824. The F_1 seeds of crossed between Kumgangdaepp (Rf 0.79) and Uidu (Rf 0.83) possessed both bands. Analysis of 96 F_2 seeds showed a ratio of 22 : 53 : 21 (Rf 0.79 : Rf 0.79/Rf 0.83 : Rf 0.83), suggesting single gene control with two co dominant alleles.

QTL Mapping of Agronomic Traits in an Advanced Backcross Population from a Cross between Oryza sativa L. cv. Milyang 23 and O. glaberrima

  • In the previous study, 141 $BC_3F_2$ lines from a cross between the Oryza sativa cv. Milyang 23 and O. glaberrima were used to identify favorable wild QTL alleles for yield component traits. In this study, we carried out QTL analysis of four grain morphology as well as four yield component traits using 141 $BC_3F_5$ lines from the same cross and compared QTLs detected in two different generations. The mean number of O. glaberrima segments in the 141 $BC_3F_5$ lines ranged from 1 to 13 with 2.69 and 5.71 of the average means of homozygous and heterozygous segments, respectively. There was a three-fold difference in the number of QTLs detected for four traits commonly evaluated in two generations (seven QTLs in the $BC_3F_5$ vs 21 in the $BC_3F_2$ population). The percentages of the phenotypic variance explained by QTLs in the BC3F5 population were similar to or less than those in the $BC_3F_2$ population. This is probably due to the difference in the genetic composition of two populations and the environmental effects. The locations of the QTLs commonly detected in both generations were in good agreement except for one QTL for spikelets per panicle. The yield QTL, yd3 was colocalized with the spikelets per panicle, spp3. Yield increase at this locus is due to the increase in spikelets per panicle, because both traits were associated with increase in spikelets per panicle and yield due to the presence of an O. glaberrima allele. Clusters of QTLs for grain morphology traits were observed in two chromosome regions. One cluster harboring five QTLs near SSR markers RM106 and RM263 was detected on chromosome 2. This population would serve as a foundation for development of the introgression line population from a cross between Milyang 23 and O. glaberrima.

Genetic Variation of Korean Fir Sub-Populations in Mt. Jiri for the Restoration of Genetic Diversity (유전다양성 복원을 위한 지리산 구상나무 아집단의 유전변이)

  • To provide a ecological restoration strategy considering genetic diversity of Abies koreana in Mt. Jiri, the genetic diversity and the genetic differentiation among sub-populations such as Banyabong, Byeoksoryeong, and Cheonwangbong were investigated. The average number of alleles (A) was 7.8, the average number of effective alleles ($A_e$) was 4.9, observed heterozygosity ($H_o$) was 0.578, and expected heterozygosity ($H_e$) was 0.672, respectively. The level of genetic diversity within sub-populations ($H_e=0.672$) was lower than those of both population ($H_e=0.778$) and species ($H_e=0.759$) level. However, the level of genetic diversity was high compared those of Genus Abies. Genetic differentiation was 0.014 from F-statistics ($F_{ST}$) and was 0.004 from AMOVA analysis (${\Phi}_{ST}$). There was no almost genetic differentiation among sub-populations in Mt. Jiri from bayesian clustering. Therefore, If the seeds are sampled sufficiently by selecting the parameters from three sub-populations, it is possible that we could obtain genetically appropriate materials for ecological restoration.

Analysis of the Genotype Distribution in Cattle Breeds Using a Double Mismatched Primer Set that Discriminates the MC1R Dominant Black Allele (소 MC1R 우성흑모색 대립인자를 구분하는 변형 프라이머를 이용한 소 품종들의 유전자형 분포 분석)

  • With a double mismatch primer set designed for amplifying the modified DNA sequence fragments, bovine melanocortin-1 receptor(MC1R) gene encoded in Extension locus which plays a critical role in coat color development was analyzed using polymerase chain reaction mediated restriction fragment length polymorphism(PCR-RFLP). Amplified PCR fragments were successfully discriminated with combining the MspI- and AluI-RFLP into three major alleles(ED, E+, and e), directly related to bovine coat color phenotypes. The genotyping results showed that Jeju black cattle contained three MC1R alleles, but yellowish-red colored Hanwoo and bridle colored Korean Brindle cattle did not contained the dominant black allele ED. However, two dominant black-colored cattle breeds, Holstein and Angus, contained the ED allele over 96% in frequency. Hanwoo×Holstein F1 and Hanwoo×Angus F1 crossbred calves showed ED/e MC1R genotypes, and uniformly black coat color. the results suggested that this MC1R genotyping method be useful in allele discrimination for bovine MC1R gene which used for breed classification and characterization, as one of the important genetic markers, using combination of MspI- and AluI-RFLP for modified PCR product amplified with a newly designed double mismatch primer set.

Diallel Analysis for Cyanidin-3-glucoside Content in Pigmented Rice (이면교배에 의한 흑자색미 안토시아닌 함량의 유전분석)

  • A complete diallel cross was performed to determine the inheritance of 1,000 grain weight of brown rice and cyanidin-3-glucoside (C3G) content and combining ability of five pigmented rice that showed different level of C3G content. Parents and F1 of 20 crosses were evaluated for 1,000 grain weight of brown rice and C3G content which extracted from pigmented rice grain with 0.1% trifluoroacteic acid (TFA) in 95% EtOH. For 1,000 grain weight of brown rice and C3G content, both additive and dominant gene effects were detected and the dominance was partial and there were more dominant alleles than recessive ones. Genetic parameters pointed out that there are significant additive and dominant effects and the additive effect is bigger than the dominant one for both grain weight and C3G content. The frequency of negative alleles were higher than positive alleles. Narrow sense heritability estimates were 0.66 for grain weight of 1,000 brown rice and 0.70 for C3G contents. General combining ability (GCA) effects and specific combining ability (SCA) effects were highly significant and GCA effects were drastically higher than SCA ones. It indicates that the inheritance of the C3G contents is controlled mainly by additive genetic factor.

Identification of QTLs controlling somatic embryogenesis using RI population of cultivar ${\times}$ weedy soybean

  • Quantitative trait loci (QTLs) controlling ability of somatic embryogenesis were identified in soybean. A frame map with 204-point markers was developed using an RI population consisting of 117 $F_{11}$ lines derived from a cross between cultivar 'Keburi' and a weedy soybean 'Masshokutou Kou 502'. The parents differed greatly in their abilities of somatic embryogenesis using immature cotyledons as explants. The ability of somatic embryogenesis was evaluated in five different experiments: the $F_{11}$ (evaluated in 1998) and $F_{15}$ (2002) generations cultured on basal media supplemented with $40\;mg\;l^{-1}$ 2,4-D (2,4-D1998 and 2,4-D2002), $F_{14}$ (2001) generation on medium with $40\;mg\;l^{-1}$ 2,4-D and high sucrose concentration [2,4-D2001 ($30\;g\;l^{-1}$ sucrose)], and the $F_{11}$ (1998) and $F_{12}$ (1999) generations on medium with $10\;mg\;l^{-1}$ NAA (NAA1998 and NAA1999). The RILs showed wide and continuous variations in each of the five experiments. In the composite interval mapping analysis, 2 QTLs were found in group 8 (D1b + W, LOD = 5.42, $r^2$ = 37.5) in the experiment of 2,4-D1998 and in group 6 (C2, LOD = 6.03, $r^2$ = 26.0) in the experiment of 2,4-D2001 (high concentration sucrose). In both QTLs, alleles of 'Masshokutou Kou 502' with high ability of somatic embryogenesis contributed to the QTLs. For the other three experiments, no QTL was detected in the criteria of LOD >3.0, suggesting the presence of minor genes.

  • To assess the genetic diversity of Aconitum coreanum (Ranunculaceae) populations in Korea, we have amplified and sequenced eight organellar marker regions, and developed and analyzed microsatellite markers. No sequence variation was detected from the eight organellar markers. Ten microsatellites were developed using Next Generation Sequencing and two microsatellite markers, AK_CA03 and AK_CT07, were identified polymorphic and applied for 143 individuals of twelve A. coreanum populations. Four and five alleles were detected for the two microsatellite loci, respectively, and number of migrants ($N_m$) was estimated as 1.12586. Two microsatellite marker loci showed $F_{ST}$ of 0.205 and 0.275, respectively. The heterozygosity deficit, low level of among-population differentiation, small size of gene flow, and lack of sequence variation of the organellar markers suggest that A. coreanum is reproductively isolated from other Aconitum species and there has been continuous gene flow among the populations of A. coreanum or it has dispersed relatively recently after speciation. Though population pairwise $F_{ST}$'s presented significant geographic structure, further sampling and study will be necessary to confirm this.

Genetic Variation in Wild and Cultured Populations of the Sea Squirt Halocynthia roretzi Inferred from Microsatellite DNA Analysis

  • We used nine microsatellite DNA markers to estimate genetic variation among wild and cultured populations of the sea squirt Halocynthia roretzi. The loci were polymorphic, with 6-32 alleles, and allelic richness ranged from 6.0 to 26.1 in each population. The wild and the cultured populations had similar mean heterozygosities ($H_O$ and $H_E$), allele numbers, and allelic richness. One cultured population with softness syndrome had a lower mean in the observed heterozygosity ($H_O$ = 0.57) and higher mean inbreeding coefficient ($F_{IS}$ = 0.261) than any other populations. This suggests that the loss of genetic variation in the diseased population might be due to increased inbreeding. A neighbor-joining tree and pairwise population estimates of $F_{ST}$ showed moderate genetic differentiation between the wild and the cultured populations. Additionally, the softness syndrome population was genetically divergent from wild populations, but it was genetically close to the cultured populations.

Genetic Diversity of Goats from Korea and China Using Microsatellite Analysis

  • Nine microsatellite loci were analyzed in 84 random individuals to characterize the genetic variability of three domestic goat breeds found in Korea and China: Korean goat, Chinese goat and Saanen. Allele diversity, heterozygosity, polymorphism information content, F-statistics, indirect estimates of gene flow (Nm) and Nei's standard distances were calculated. Based on the expected mean heterozygosity, the lowest genetic diversity was exhibited in Korean goat ($H_E$=0.381), and the highest in Chinese goat ($H_E$=0.669). After corrections for multiple significance tests, deviations from Hardy-Weinberg equilibrium were statistically significant over all populations and loci, reflecting the deficiencies of heterozygotes (global $F_{IS}$=0.053). Based on pairwise FST and Nm between different breeds, there was a great genetic differentiation between Korean goat and the other two breeds, indicating that these breeds have been genetically subdivided. Similarly, individual clustering based on the proportion of shared alleles showed that Korean goat individuals formed a single cluster separated from the other two goat breeds.

Electrophoretic Analysis of Haemolymph Proteins during Silkworm (Bombyx mori L.) Ontogenesis

  • A study was made of the haemolymph protein spectrum of mulberry silkworm (Bomhyx mori L.) from the first larval instar to imago. Horizontal starch gel electrophoresis was used. Sixteen races and eight F1 interracial hybrids, raised in Bulgaria, were analyzed. During the ontogenesis, a total of 17 protein bands (15 cathodic and 2 anodic) were detected. Distinct dynamics in the haemolymph protein spectrum was observed, in result of different expression during the individual development associated with the processes of growth, histolysis and histogenesis. Based on the ontogenetic dynamics found, a correspondence was assumed between some proteins detected by us using the starch gel electrophoresis and major haemolymph proteins (SP1, SP2, MHPs and Vg) detected by other authors using the polyacrilamide gel electrophoresis. Intraracial and interracial polymorphism was observed in four protein zones. The effect of four polymorphic loci with codominant and null alleles was suggested.
