Biological Resources of Pen Shell, Atrina (Servatrina) pectinata japonica in the Coastal Waters of Chungchung-namdo, Korea. 1. Effects of Environmental Factors on Distribution Pattern (충남연안 키조개의 자원생물학절 연구 1. 환경특성이 분포양상에 미치는 영향)

  • The distribution and density of the pen shell, Atrina (Servatrina) pectinata japonica, were investigated to estimate population ecological parameters and biomass. Samples were collected during cruises from April to June 1999 in the coastal waters of Chungchung-namdo, Korea. The oceanographic conditions such as depth, temperature, transparency and suspended solids were measured from sampling stations. Seawater depth ranged from 3.9 m to 75.9 m and the mean was 28.5 m (SE = 0.97). The mean density of the pen shell was higher in the depth of 18.0-30.8 m. The water temperature of the bottom ranged from 7.0-21.5$^{\circ}C$ and the mean was 14.$0^{\circ}C$. The transparency ranged from 1.2-13.5 m and the mean was 4.8 m. The suspended solids ranged from 6.0-93.5 mg/l and the mean was 48.0 mg/l. The sand proportion of the sediment was > 50%. The mean densities of the pen shell by sediment type were 7.3 individuals/40 m$^2$ (SE = 2.16) in the sand, 5.6 individuals/40 m$^2$ (SE = 1.48) in the muddy sand, 3.0 individuals/40 m$^2$(SE = 1.89) in the sandy mud, and none in the mud. The sandy bottom, accounting for 29.8% of the overall habitat area, had a maximum density of 7.3 individuals/40 m$^2$.

Ruminal Degradability of Tropical Feeds and Their Potential Use in Ruminant Diets

  • The objective of this study was to determine the degradability of cassava chip (CC), cassava waste (CW), yellow sweet potato (YP), white sweet potato (WP), purple sweet potato (PP), corn meal (CM), and rice bran (RB) using in situ technique. Two ruminally fistulated steers with an average weight of $303{\pm}10kg$ were used to determine in situ degradabilities of DM and OM. Seven feed sources were weighted in nylon bags ($38{\mu}m$ pore size) and incubated ruminally for 1, 2, 4, 6, 8, 12, 24, and 48 h. The results showed that asymptote (a+b) and effective degradability (ED) of DM of energy sources ranked from the highest to the lowest; CC, YP, WP, PP, RB, CW, and CM (99.3, 92.5; 97.6, 87.9; 97.5, 87.9; 97.2, 87.8; 87.5, 63.6; 78.6, 63.0 and 81.7; 59.3, respectively) and for OM asymptote (a+b) and effective degradability (ED) were similar to those of degradation of DM (99.4, 93.4; 98.8, 89.8; 98.5, 89.4; 98.4, 88.1; 92.4, 65.8; 85.1, 66.9 and 83.6, 63.3, respectively). It was concluded that disappearance characteristic of CC was the highest and it may potentially facilitate the achievement of optimal ruminal availability of energy: protein especially with NPN for microbial protein synthesis.

  • $BaCe_{0.9}Gd_{0.1_O_{2.95}$ powder was synthesized by oxalate coprecipitation method. Precipitate with a stoichimetric ratio of the cations was prepared by adding a mixture of Ba, Ce and Gd nitrate solution to an oxalic acid solution at pH 4. Reaction between the constituent oxides to form a perovskite phase was initiated at $800^{\circ}C$ and a single phase $BaCe_{0.9}Gd_{0.1_O_{2.95}$ powder having good sinterability was obtained after calcination at $1000^{\circ}C$. Sintering green compacts of this powder for 6 h showed a considerable densification to start at $1100^{\circ}C$ and resulted in 93% and 97% relative densities at $1300^{\circ}$ and at $1450^{\circ}C$, respectively. Whereas the power compacts prepared by solid state reaction had lower relative densities, 78% at $1300^{\circ}$and 90% at $1450^{\circ}C$. Fine particles of $CeO_2$ second phase were observed in the surface of the sintered compacts. This was attributed to the evaporation of BaO from the surface that had been exposed during thermal etching.

Characterization of a Vibrio parahaemolyticus Phage Isolated from Marine (해양에서 분리한 Vibrio parahaemolyticus Phage의 특성)

  • A novel bacteriophage, designated as VPP97, that infects the strains of Vibiro parahaemolyticus (hallophilic, Gram-negative bacterium) isolated most commonly from marine environments, has been discovered, and several of its properties have been determined. The plaques were clear and sized $0.6{\sim}1.0\;mm$ in diameter. The virion forms a single band on 70% sucrose gradient and ${\rho}1.50$ CsCl gradient by sucrose gradient centrifugation and CsCl gradient centrifugation respectively. It has a hexagonal head and a relatively long tail, as shown by electron microscopy. Vibrio alginolyticus, Vibrio fluvialis and Vibrio furnissii were also sensitive to this phage. It was almost totally inactivated at $70^{\circ}C$ and at pH below 5 or over 10. The nucleic acid of VPP97 is composed of DNA. The VPP97 had 9 specific structural proteins sized between 21.5 kDa and 97.4 kDa on SDS-PAGE. When V. parahaemolyticus cultures were treated with either phage VPP97 or one of the several antibiotics for 2 hours, the viable number of V. parahaemolyticus treated with the phage VPP97 is lower than that treated with chloramphenicol, erythromycin or penicillin, but not lower than that treated with tetracycline. Mice that have responded to the phage treatment revealed the lower numbers of V. parahaemolyticus in small intestine and less damage on small intestine compared to the untreated mice. Therefore, we suggest that the phage treatment appears effective to the infection by V. parahaemolyticus.

  • The purpose of this study is to investigate the optimal condition for the hydrogen-rich gas production and the CO removal by reforming of gliding arc plasma reforming system using biogas. The parametric screening studies were carried out according to changes of steam feed amount, catalyst bed temperature in water gas reactor and catalyst bed temperature, input air flow rate in preferential oxidation reactor. The standard condition is as follows. The steam/carbon ratio, catalyst bed temperature, total gas flow rate, input electric power and biogas composition rate ($CH_4$ : $CO_2$) were fixed 3, $700^{\circ}C$, 16 L/min, 2.4 kW and 6 : 4, respectively. The results are as follow, HTS optimum operating conditions were S/C ratio of 3 and reactor temperature of $500^{\circ}C$. LTS were S/C ratio of 2.9 and temperature of $300^{\circ}C$. Also, PROX I optimum conditions were input air flow rate of 300 mL/min and reactor temperature of $190^{\circ}C$. PROX II were 200 mL/min and $190^{\circ}C$ respectively. After having passed through each reactor, the results were as follows: 55% of $H_{2}$ yield, 0% of CO selectivity, 99% of $CH_4$ conversion rate, 27% of $CO_2$ conversion rate, respectively.

  • The performance of adiabatic capillary tubes are measured to provide the database for a generalized correlation. Test conditions and capillary tube geometries are selected to cover a wide range typically observed in air-conditioning and heat pump applications. Based on extensive experimental data for R22, R290, and R407C measured in this study, a generalized correlation for refrigerant flow rate in adiabatic capillary tubes is developed by implementing dimensionless parameters for tube inlet conditions, capillary tube geometry, and refrigerant properties. The correlation yields good agreement with the present data for R22, R290, and R407C with average and standard deviations of 0.9% and 5.0%, respectively. In addition, approximately 97% of the data for Rl2, R134a, R152a, R410A, and R600a obtained in the open literature are correlated within a relative deviation of $\pm$ 15%.

  • To answer the question whether last clonogenic cell should be eradicated for the tumor to be controlled, radiation tumor control study was performed using microscopic tumors of variable sizes ranging from 101 to 10s tumor cells. $TCD_{50}'s$ estimated from experimental data were 14.8, 27. 1, 42.4, 49.9 and 65.5 Gy for $10^1,\;10^2,\;10^3,\;10^4\;and\;10^4$ tumor cells, respectively. Theoretical calculations, assuming that all the clonogenic cells should be inactivated, were 15.65, 8.50, 40.97, 53.41 and 65.85 Gy. From this well matched data, it can be concluded that all the clonogenic cells should be eradicated for tumor control, at least in this tumor model.

High Concentrated Spore Production of Bacillus thuringliensis by Fed-Batch Processes (유가식 배양공정에 의한 Bacillus thurngiensis의 고농도 포자생산)

  • Both the production of high spore concentration and high insecticidal activity are required in the production of Bacillus thuringiensis to be used for the bacterial insecticide. In the production of high cell and spore concentrations of B. thuringiensis the continuous fed-batch culture(CFBC) and intermittent fed-batch culture(IFBC) were investigated at $28^{\circ}C$ by maintaining 40% dissolved oxygen concentration. When the final glucose concentration was 50 g/L the maximum viable cell number obtained using the CFBC with linear gradient feeding was $9.37{\times}109$ cells/mL and maximum spore concentration was $8.33{\times}109$ spores/mL which was approximately 84.4% yield of spore formation. When the final glucose concentration was 100 g/L the aximum viable cell and spore concentrations obtained using the IFBC with pH-statb were $1.38{\times}$1010 cells/mL and $1.35{\times}1010$ spores/mL respectively and the yield of spore formation was approximately 97.8%.

Optimization of Lactic Acid Fermentation of Prickly Pear Extract

  • Lactic acid fermentation of prickly pear extract (PPE) was performed by Lactobacillus rhamnosus LS, Lactobacillus bulgaricus, and Lactobacillus brevis. The PPE was pasteurized to eliminate indigenous microorganisms as well as to dissolve the partially insoluble pulp. The PPE fermented without yeast extract by L. rhamnosus LS exhibited 0.57% acidity and 3.5${\times}$10$^{8}$ CFU/mL bacteria count. With the addition of 0.2% edible yeast extract the PPE fermented by L. rhamnosus LS exhibited 1.15% acidity,2.7${\times}$10$^{9}$ CFU/mL bacteria count and 95.0% retention of red color. When 5% fructose syrup was added, the PPE fermented by L. rhamnosus LS had 1.09% acidity, 6.5${\times}$10$^{8}$ CFU/mL, and 97.7% retention of red color. With 1∼3% (w/v) concentrations of starter, the PPE fermented by L. bulgaricus and L. brevis showed 0.97% and 0.65% acidities, respectively. The viable cell counts from L. rhamnosus LS fermentation were higher compared with those of other LAB. During cold storage at 4$^{\circ}C$, the viable cell count was well maintained for 3 weeks, but then rapidly decreased. The red pigment was highly stable during cold storage for 4 weeks. The pasteurized PPE fortified with 5% fructose syrup, 0.2% yeast extract, and 0.05% CaCO$_3$ was successfully fermented by inoculating with 3% LAB and incubating at 3$0^{\circ}C$ for 2 days. Both viable cell counts and the red color of the fermented PPE were well maintained during cold storage for 3 weeks.

Study on the Microwave Dielectric Properties and Dielectric Resonator Performance of the $Mg_4Ta_2O_9$ Ceramics with $TiO_2$ Addition ($TiO_2$ 첨가에 따른 $Mg_4Ta_2O_9$ 세라믹스의 마이크로파 유전특성과 유전체 공진기 거동에 관한 연구)

  • The $(1-x)Mg_4Ta_2O_9-xwt%TiO_2\;(x=5\sim20)$ microwave dielectric ceramics were prepared by solid-state reaction method and sintered at $1450^{\circ}C$. According to the X-ray diffraction data, the $Mg_4Ta_2O_9-xwt%TiO_2$ ceramics had main phase of $Mg_4Ta_2O_9$ and $MgTi_2O_5$ peaks were added by increasing of $TiO_2$ addition. Microwave dielectric properties of the $Mg_4Ta_2O_9-xwt%TiO_2$ ceramics were influenced by $MgTi_2O_5$ phase and properties of $TiO_2$. There was a little decrement of the quality factor from 116,800GHz of pure $Mg_4Ta_2O_9$ to 100,100GHz of 15wt% $TiO_2$ added one. But there was excellent improvement in temperature coefficient of the resonant frequency (TCRF) by addition of 15wt% $TiO_2$. The dielectric constant quality factor and TCRF of the $Mg_4Ta_2O_9-xwt%TiO_2$ ceramics sintered at $1450^{\circ}C$ were $13.08\sim16.41,\;45,000\sim165,410GHz,\;-24.82\sim+3.88ppm/^{\circ}C$, respectively, depending on the value of x. Simulated dielectric resonator (DR) with $Mg_4Ta_2O_9-15wt%TiO_2$ ceramics had the operating frequency of 11.97GHz and $S_{2,1}$ of -35.034dB.
