Continuous Foam Separation of Yeast Cells (효모의 연속 포말 분리)

  • Cell separation by means of continuous foam separation of Saccharomyes formosensis without additive was investigated. The yeast separation ratio was improved at low feed rates, high nitrogen rates, optimum pH and temperature for ethanol production, dilution of cultivation medium and addition of 0.5g/$\ell$ $CaCl_2$. Percentage of yeast removal and yeast separation ratio were more than 85 when continuous foam separation was operated in optimum condition..

Effects of Antioxidant Activity and Changes in Vitamin C during Storage of Lycii folium Extracts Prepared by Different Cooking Methods (조리방법을 달리한 구기엽의 저장기간별 항산화 효과 및 비타민 C 함량의 변화)

  • Contents in polyphenol and vitamin C and their antioxidant activities during storage of Lycii folium prepared by different treatment methods were investigated. The antioxidant activity of extracts prepared from 80% MeOH was evaluated using DPPH radical scavenging and SOD-liked activities. Total phenolic acid contents were 1.34 mg/mL when dry, but extracts during 15-day storage after NaCl 0% treatment decreased. DPPH radical scavenging activity of extracts was 65.48% when dry and decreased during 15-day storage after dry and NaCl 0% treatment. SOD-liked activity was 98.63% in NaCl 0% during 15-day storage after dry. Contents of vitamin C decreased during 15-day storage after dry and NaCl 0% treatment. Consequently, our results indicate that Lycii folium prepared from dry treatment could be a promising biomaterial for the production of functional food for total phenolic acid and vitamin C.

Electrochemical Reduction of Perchlorate Using Mercury Film Electrode (수은 막전극을 이용한 수용액 중 과염소산이온의 전기화학적 환원)

  • A method for electrochemical degradation of the perchlorate anion ($ClO_4{^-}$) using mercury film electrode has been studied. Electrochemical method has relatively simple pre-treatment. However, electrochemical method should avoid interference from hydrogen evolution at the applied potential to degradation of perchlorate ion, and thus applied electrode should have large hydrogen overvoltage which suppressed the hydrogen evolution at the working reduction potential to prevent hydrogen evolution. In this study, we used mercury film electrode as a working electrode which has a large overvoltage. Ag / AgCl (sat. NaCl) was used as a reference electrode, and platinum was used as a counter electrode. Mercury film electrode was made by cyclic voltammetry (CV) method. The deposition time was decided as 10 minute, and the stability of the mercury electrode in perchlorate solution was confirmed by CV. The reduction potential of perchlorate was checked by using CV method, and decomposition of perchlorate was performed by using chronoamperometric (CA) method. Also, ion chromatography (IC) was used to confirm the degradation rates of perchlorate.

The Characteristics of silicon nitride thin films prepared by atomic layer deposition method using $SiH_2Cl_2 and NH_3$ ($SiH_2Cl_2와 NH_3$를 이용하여 원자층 증착법으로 형성된 실리콘 질화막의 특성)

  • Silicon Nitride thin films were deposited on p-type Si (100) substrates by atomic layer deposition (ALD) method at $550^{\circ}C$ using alternating exposures of $SiH_2Cl_2$ and $NH_3$, and the physical and electrical propeties of the deposited films were characterized. The thickness of the films was linearly increased with the number of deposition cycles, and the growth rate of the films was 0.13 nm/cycle with the reactant exposures of $3.0\times10^{9}$ L. The silicon nitride thin films deposited by Alf exhibited similar physical properties with the silicon nitride thin films deposited by low-pressure chemical vapor deposition (LPCVD) method in terms of refractive index and wet etch rate, lowering deposition temperature by more than 200 $^{\circ}C$. The ALD films showed the leakage current density of 0.79 nA/$\textrm{cm}^2$ at 3 MV/cm, which is lower than 6.95 nA/$\textrm{cm}^2$ of the LPCVD films under the same condition.

Physicochemical Quality Properties of Mudflat Solar Salt and Roasted Salt (갯벌천일염과 구운 소금의 이화학적 품질 특성)

  • The purpose of this study was carried out to investigate the cause of sulfur dioxide occurrence, general element composition, sulfur compounds, heavy metals, macro- & micro-minerals, and oxidation-reduction potential (ORP) following baking time course of RS (RS1, RS2, RS3, and RS4) and mudflat solar salts (MSS). Sulfur dioxide ($SO_2$) and sulfite ($SO{_3}^{2-}$) were not detected in MSS or RS. However, sulfate ($SO{_4}^{2-}$) content significantly decreased in RS (29,878.15~36,097.45 ppm) compared to that in MSS (35,601.65 ppm). ORP was 181.15 mV in MSS, and 58.55 mV in RS1. Moisture content was 9.34% in MSS and 0.00% in RS with increased NaCl (94.77~95.77%). Moisture and NaCl contents showed no significant difference in RS. Insoluble and sandy residues were higher in RS than in MSS, whereas Ca and K showed no significant difference. Mg and Cl contents were higher in RS than in MSS. Br level was higher in MSS (628.1 ppm) than in RS (512.72~586.62 ppm), but there was no significant difference in $NO_3$. Heavy metals (Pb, As, and Hg) were more abundant in RS than in MSS, but levels were still safe. These results suggest that MSS and RS may increase protection against from $SO_2$ and $SO{_3}^{2-}$.

Fundamental Study on a Distillation Separation of a LiCl-KCl Eutectic Salt from Rare Earth Precipitates (희토류 침전물로부터 LiCl-KCl 공융염의 증류 분리에 관한 기초연구)

  • The distillation rate on LiCl-KCl eutectic salt under different vacuums from 0.5-50 mmHg was first investigated by using both a non-isothermal and a isothermal thermogravimetric (TG) analysis. Based on the non-isothermal TG data, distillation rate equations as a function of the temperature could be derived. Calculated flux by these model flux equations was in agreement with the distillation rate obtained from isothermal TG analysis. A distillation rate of $10^{-4}-10^{-5}$ mole $cm^{-2}sec^{-1}$ is obtainable at temperatures less than 1300K and vacuums of 0.5-50 mmHg. About a 99% salt distillation efficiency was obtained after an hour at a temperature above 1150 K under 50 mmHg in a small scale distillation test system. An increase in the vaporizing surface area is relatively effective for removing residual salt in the remaining particles, when compared to that for the vaporizing time. Over 99.95% of total distillation efficiency was obtained for a 1-h distillation operation by increasing the inner surface area from $4.52cm^2$ to $12.56cm^2$.

  • Spoiled mushroom cans were collected from several canneries and examined the spore bearing bacteria in them. Thirty four isolates of anaerobic mesophiles were isolated. The one isolate(Cl-5) which was the most heat stable among the above isolates, the other isolate (D-29) which was examined in the previous experiment (Korea J. Food Sci. Technol., 10(2), 224 (1978) and PA 3679 were compared their heat resistancy in neutral phosphate buffer. They were confirmed the spoilage of mushroom products. Z value and $D_{250}$ value of the Cl-5(isolated from mushroom casing soil at Gimjae) possessing the highest resistancy were $21.3^{\circ}F$ and 1.8 min in mushroom can (4 oz. piece and stem). It's F value was 8.95 when Z value was 21.3.

Discrimination of Geographical Origin for Scutellaria baicalensis Using Energy Dispersive X-ray Fluorescence Spectrometer (에너지 분산형 X-선 형광분석기를 이용한 황금의 원산지 판별)

  • A discrimination technique for domestic and imported Scutellaria baicalensis was developed using an energy dispersive X-ray fluorescence spectrometer (ED-XRF). Mineral content ratios, of a total of 43 species, including P, S, Cl, K, Ca, Mn, Fe, Cu, and Zn, were measured among 204 samples. Macro element content ratios and trace element content ratios were determined using the standardless fundamental parameters (SLFP) analysis. Inorganic element ratios of P, S, K, Ca, Cl, Mn, and Fe were significantly different between domestic and imported samples. The result from the canonical discriminant analysis showed that the accuracy of geographical origin discrimination was 95.15%; the correlation coefficient was 0.888. It was concluded that this technique could be used as a useful method in discriminating the geographical origins between domestic and imported Scutellaria baicalensis.

α-Glucosidase Inhibition Activity of the Extracts of Katsura Tree (Cercidiphyllum japonicum Sieb. Et Zucc) Leaves

  • Katsura tree (Cercidiphyllum japonicum Sieb. Et Zucc) leaves were collected, air-dried and extracted with 70% aqueous acetone, then concentrated and sequentially fractionated using n-hexane, $CH_2Cl_2$, EtOAc, and $H_2O$ to be freeze dried for antioxidant and ${\alpha}$-glucosidase inhibition activity tests. The antioxidant activity of the extracts was evaluated using DPPH (1,1-diphenyl 2-picrylhydrazyl) free radical scavenging assay. The test concentrations were adjusted to 500, 250, 125, 62.5, 31.25, 15.62, 7.81, 3.9, 1.95 and 0.97 ppm. The $H_2O$ and EtOAc fractions showed higher activities compared with the control, ${\alpha}$-tocopherol, at all concentrations. The crude fraction also gave better activity at the concentrations lower than 62.5 ppm. However, the nonpolar n-hexane and $CH_2Cl_2$ fractions gave prominently lower activities compared with the control at all concentrations. The $IC_{50}$ values of the crude, EtOAc, and $H_2O$ fractions exhibited 11.78, 4.29 and $9.80{\mu}g/m{\ell}$, respectively, compared with $12.08{\mu}g/m{\ell}$ of the control. But the n-hexane and $CH_2Cl_2$ fractions indicated 300 and $91.85{\mu}g/m{\ell}$ of $IC_{50}$, respectively. ${\alpha}$-Glucosidase inhibition activity was evaluated at the concentrations of 50, 25, 12.5, 6.3, 3.1, 1.6 and 0.8 ppm. The inhibition activities were increased according to as the increase of sample concentrations. However, the nonpolar n-hexane and $CH_2Cl_2$ fractions indicated very low inhibition activities compared with acarbose, a positive control. The EtOAc fraction showed very good capability as almost 100% compared with the control at the higher concentrations than 12.5 ppm and the crude fraction also indicated good potential as 95% and 100% at 25 and 50 ppm, respectively. The $H_2O$ fraction gave good inhibition value as 90% at 50 ppm although the value was lower than the control. These results showed that the polar fractions had better ${\alpha}$-glucosidase inhibition activities. The $IC_{50}$ values of the nonpolar fractions, n-hexane and $CH_2Cl_2$, showed very lower values as 468 and $103.6{\mu}g/m{\ell}$, respectively, than the control. ${\alpha}$-Glucosidase Inhibition Activity of the Extracts of Katsura Tree (Cercidiphyllum japonicum Sieb. Et Zucc) Leaves However, the polar fractions, crude, EtOAc and $H_2O$, showed 7.1, 3.7 and $13{\mu}g/m{\ell}$, respectively, indicating that these fractions can be used as natural bioresources for treating diabetes mellitus. Also ${\alpha}$-glucosidase inhibition activity had a positive correlation with antioxidant activity of the extracts.

  • A 1.95Kb Sau3Al fragment coding for $\alpha$-amylase from Bacillus amyloliquefaciens was isolated by the shotgun method using Escherichia coli as a host. The genome of Bacillus amyloliquefaciens was partially digested with the restriction endonuclease Sau3Al and joined to plasmid YRp7 cleaved with the restriction endonuclease BamHI. The $\alpha$-amylase gene present in a 1.95Kb insert was stably maintained and expressed in Escherichia coli. The amount of $\alpha$-amylase activity produced by Escherichia coli containing the hybrid plasmid pEA24 was about 65% of the activity produced by the donor Bacillus amyloliquefaciens strain. The properties of $\alpha$-amylase produced by Escherichia coli were very similar to those produced by Bacillus amyloliquefaciens as based on optimum temperature, pH, and effect of CaCl$_2$ concentration. About 70% of the $\alpha$-amylase produced by Escherichia coli was localized in the periplasmic space, whereas the remaining enzyme was localized in the inner part of the cell.
