가역적상호대사과정 모델을 이용한 Prednisolone과 Prednisone의 약동학적 분석 (Pharmacokinetic Modeling of Reversible Interconversion between Prednisolone and Prednisone)

  • Pharmacokinetics of prednisolone and prednisone undergoing reversible interconversion were analyzed from the model including this metabolic process. Blood samples were drawn serially upto 12 hours after I,V. bolus injection of 1 mg/kg prednisolone sodium phosphate and prednisone into 8 dogs as a crossover manner. Plasma concentrations of those two steroids were simultaneously measured with the method of HPLC. After injection, plasma concentrations of administered prednisolone and prednisone were declined with a biexponential pattern and their metabolic partner was rapidly formed. Plasma concentrations of those metaboite were decayed in parallel with their parent steroids throught the elimination phase. Apparent clearances of prednisolone and prednisone were $11.1{\pm}2.0\;ml/min/kg$ and $45.9{\pm}6.4\;ml/min/kg$, and they were underestimated by 29.4% and 33.6% compared to their real clearances$(15.7{\pm}4.4\;and\;69.2{\pm}17.7\;ml/min/kg)$ estimated using reversible interconversion model. Apparent volume of distribution of prednisolone$(1.32{\pm}0.43\;L/kg)$ and prednisone$(4.81{\pm}2.75\;L/kg)$ were overestimated by 53.5 and 52.7% and were compared to the real volumes $(0.86{\pm}0.30\;and\;3.15{\pm}2.13\;L/kg)$. Mean residence time of prednisolone$(2.0{\pm}0.61\;h)$ and prednisone$(1.74{\pm}0.74\;h)$ were much longer than the real sojourn time$(0.93{\pm}0.26\;and\;0.88{\pm}0.54\;h)$. Essential clearances In the reversible interconversion were greater as following orders: $Cl_{21}$(44.3 ml/min/kg) > $Cl_{20}$(24.2 ml/min/kg) > $Cl_{12}$ (7.9 ml/min/kg) > $Cl_{10}$(7.8 ml/min/kg). Estimated mean values of RF, EE, $%X^1_{ss}$ and $RHO^2_1$ were $0.31{\pm}0.10$, $1.49{\pm}0.23$, $69.3{\pm}16.7%$ and $0.65{\pm}0.10$, respectively. These results suggested that true pharmacokinetic parameters estimated from the model including reversible interconversion were significantly different from the apparent parameters estimated from the conventional mamillary model, and disposition of these two steroids seemed to be well explained by the model including reversible interconversion.

  • Oxygen consumption, pulmonary ventilation, heart rate, and breathing frequency were measured on 8 men walking on a treadmill carrying load of 9 kg on hand, back, or head. Besides measurements were made on subjects carrying loads of 2.6 kg each on both feet. The speed of level walking was 4, 5, and 5.5km/hr and a fixed speed off km/hr with grades of 0, 3, 6, and 9%. Comparisons were made between free walking without load and walking with various types of loads. The following results were obtained. 1. In level or uphill walking the changes in oxygen consumption, pulmonary ventilation, breathing frequency and heart rate were smallest in back load walking, and largest in hand load walking. The method of back load was most efficient and hand load was the least efficient. The energy cost in head load walking was smaller than that of in hand load walking. It was assumed that foot load costed more energy than hand load. 2. In level walking the measured parameters increased abruptly at the speed of 5.5 km/hr. Oxygen consumption in a free walking at 4 km/hr was 11.4ml/kg b.wt., and 13.1 ml/kg b.wt. 5.5 km/hr, and in a hand load walking at 4 km/hr was 13.9, and 18.8 ml/kg b. wt. at 5.5 km/hr. 3. In uphill walking oxygen consumption and other parameters increased abruptly at the grade of 6%. Oxygen consumption at 4 km/hr and 0% grade was 11.4 ml/kg b. wt., 13.6 at 6% grade, and 16.21/kg b. wt. at 9% grade in a free walking. In back load walking oxygen consumption at 4km/hr and 0% grade was 12.3 ml/kg b.wt.,14.9 at 6% grade, and 18.7 ml/kg b.wt. In hand load walking the oxygen consumption was the greatest, namely, 13.9 at 0% grade, 17.9 at 6%, and 20.0 ml/kg b. wt. at 9% grade. 4. Both in level and uphill walking the changes in pulmonary ventilation and heart rate paralleled with oxygen consumption. 5. The changes in heart rate and breathing frequency in hand load were characteristic. Both in level and uphill walk breathing frequency increased to 30 per minute when a load was held on hand and showed a small increase as the exercise became severe. In the other method of load carrying the Peak value of breathing frequency was less than 30 Per minute. Heart rate showed 106 beats/minute even at a speed of 4 km/hr when a load was held on hand, whereas, heart rate was between, 53 and 100 beats/minute in the other types of load carriage. 6. Number of strides per minute in level walking increased as the speed increased. At the speed floater than 5 km/hr number of strides per minute of load carrying walk was greater than that of free walking. In uphill walk number of strides per minute decreased as the grade increased. Number of strides in hand load walk was greatest and back load walk showed the same number of strides as the free walk.

난소적출 rat에 녹용, 홍삼, 홍화, ipriflavone 및 estrogen을 투여 했을 때 호르몬, Ca, P 및 ALP수준에 미치는 영향에 관한 연구 (Effects of antler, red ginseng, safflower, ipriflavone and estrogen on hormones, Ca, P and ALP levels in ovariectomized rats)

  • This study was performed to elucidate the effects of antler, red ginseng, safflower seed, ipriflavone and estrogen on ovariectomized rats. The rats were fed with Ca and P deficient diet for five weeks to induce osteoporosis. After this period, these animals were fed with normal feed and treated every other day with antler(600 mg/kg, p.o), red ginseng(200 mg/kg, p.o), safflower(200 mg/kg, p.o), ipriflavone(80 mg/kg, p.o) and estrogene(400$\mu\textrm{g}$/kg, i.m) for 5 weeks. During the treatment, the rats were examined for serum concentrations of estradiol, calcitonin, Ca, p and alkaline phosphatase(ALP) activities. The results are summarized as follows 1. The levels of serum 17 $\beta$-estradiol after five weeks of treatment were showed 39.6$\pm$3.0 pg/ml by antler, 33.2$\pm$2.5pg/ml by red ginseng, 34.9$\pm$2.4pg/ml by safflower, 28.1$\pm$3.1 pg/ml by ipriflavone and 40.6$\pm$3.0pg/ml by estrogen-treated group. They were lower than 50.8$\pm$3.1pg/ml of normal control group which had not received ovariectomy. They, however, were significantly higher than 26.8$\pm$1.8pg/ml of ovariectomized non-treatment group(p<0.05). 2. The levels of serum calcitonin after five weeks of treatment were showed 0.60$\pm$0.02 ng/ml by antler, 0.55$\pm$0.04ng/ml by red ginseng, 0.59$\pm$0.02ng/ml by safflower, 0.56$\pm$0.04ng/ml by ipriflavone and 0.62$\pm$0.02ng/ml by estrogen-treated group. They were lower than 0.67$\pm$0.03pg/ml of normal control group. However, they were significantly higher than 0.45$\pm$0.05ng/ml of ovariectomized non-treatment group(p<0.05). 3. The levels of serum Ca of the rats after five weeks of treatment with antler, red ginseng, safflower, ipriflavone and estrogen were 23.51$\pm$2.19$\mu\textrm{g}$/$m\ell$, 25.22$\pm$3.44$\mu\textrm{g}$/$m\ell$, 23.20$\pm$0.02$\mu\textrm{g}$/$m\ell$, 24.76$\pm$3.57$\mu\textrm{g}$/$m\ell$, 23.07$\pm$3.66$\mu\textrm{g}$/$m\ell$, respectively. They were a bit higher than 21.43$\pm$2.22$\mu\textrm{g}$/$m\ell$ of normal control group. And non-treatment group showed 26.12$\pm$0.29$\mu\textrm{g}$/$m\ell$ which was significantly higher than that of control group(P<0.05). 4. The serum P concentraions after five weeks of treatment were showed 12.11$\pm$2.14$\mu\textrm{g}$/$m\ell$ by antler, 13.18$\pm$1.64u91m4 by red ginseng, 12.67 $\pm$2.31$\mu\textrm{g}$/$m\ell$ by safflower, 12.38$\pm$2.07$\mu\textrm{g}$/$m\ell$ by ipriflavone, 11.86$\pm$1.93$\mu\textrm{g}$/$m\ell$ by estrogen-treated group. They were a bit higher than 11.29$\pm$1.23$\mu\textrm{g}$/$m\ell$ of normal control group. And non-treatment group showed 13.42$\pm$1.87$\mu\textrm{g}$/$m\ell$ which was higher than that of control group but not significant. 5. The levels of serum ALP after five weeks of treatment were showed 164.8$\pm$3.8IU/ml by antler, 277.7$\pm$4.8IU/ml by red ginseng, 288.5$\pm$4.5IU/ml by safflower, 214.7$\pm$5.7IU/ml by ipriflavone and 159.4$\pm$5.4IU/ml by estrogen-treated group. They were significantly higher than 144.1$\pm$3.5IU/ml of normal control group(p<0.05). However, they were significantly lower than 336.9 $\pm$12.7IU/ml of ovariectomized non-treatment group(p<0.05). Antler and safflower elevated serum estradiol and calcitonin, and decreased serum ALP significantly. Therefore they were thought to have therapeutic effect on osteoposis by making inhibitory effect on osteoclasts rather than activating osteoblasts.

개에서 실험적으로 유발한 복수증의 초음파 진단 (Ultrasonographic Evaluation of Experimentally Induced Ascites in Dogs)

  • In this study, ultrasonographic evaluations were made after experimentally induced ascites in dogs. Ultrasonographic evaluation allowed to identify the earliest fluid accumulation between the spleen and the left kidney after the administration of 0.9% normal saline at 8 ml/kg into the abdominal cavity in dogs. Ultrasonographic finding were observed in order between the spleen and the left kidney, the liver, the pelvic cavity. It is considered that the most adequate pare f3r the early detection of ascites is between the spleen and the left kidney, and the least amount of detection fluid is 8 ml/kg in dogs.

바륨 현탁액과 메틸셀룰로스(Methylcellulose)를 사용한 변형 상부위장관조영술 :전통적 바륨조영술과의 비교 (Modified Upper Gastrointestinal Study Using Methylcellulose After Administration of Barium Suspension : Comparison with Conventional Series)

  • For comparison with conventional barium-gastrointestinal study, modified method using barium and methylcellulose was performed in 16 normal dogs (4-8 kg) divided into two groups. The group 1 received 8ml/kg of 40% w/v barium suspension only, and group 2 had taken 15 ml/kg of 0.6% w/v methylcellulose after administration 4 ml/kg of 40% w/v barium suspension by feeding tube. The barium suspension was prepared mixing full strength-barium suspension with water and methylcellulose solution was produced by blending methylcellulose sterilized water Sequential radiographs, lateral and ventrodorsal projections were obtained at 5 minute, 20 minute,40 minute. 60 minute and every 30 minutes thereafter, until the contrast is seen in the large intestine Inage qual- ity was rated by three veterinary radiologists as " poor" " fair ". "good", or "excellent" We analyzed the relationship between image quality,, transit time. Between two techniques, the modified method with 4ml of 40% w/v barium suspension and 15 ml of 0.6% w/v methylcellulose showed much better image quality ["excellent" result in 7 of the 8 subjects (88%)] and shorter transit time (107 minutes) toe the cecum. In addition, the best image quality was obtained at 20 and 40 minutes after admin-istration of contrast agent. It call be concluded the modified gastrointestinal study using methylcel-lulose after administration of barium suspension was a simple technique and easily improved the image quality and diagnostic accuracy of gstrointestinal disorders in small animal.racy of gstrointestinal disorders in small animal.

새로운 항HIV-1제, KR-V series의 개발을 위한 약물동태연구 (Pharmacokinetic analysis for the development of new potent anti-HIV-1 agents, the KR-V series)

  • 새로운 항HIV 후보물질인 19개의 KR-V 경구제제의 생체이용률을 평가하기 위해 랫드에서 정맥 및 경구투여후 약물동태를 연구하였다. 혈장내 KR-V 화합물들의 검출은 HPLC-UVD 법을 이용하여 분석하였다. 19개 KR-V series 중 KR-V 3, 10, 14, 16 및 18-1만이 랫드에서 경구로 흡수되어 생체이용성을 나타내었다. 10mg/kg의 정맥투여후 약물 통태 연구에 있어서는 5개물질, KR-V3, 10, 14, 16 및 18-1의 소실 반감기는 서로 비슷하였으나 KR-V 3, 10, 14 및 16의 총청소율($CL_{total}$, >4L/hr/kg)은 KR-V 18-1(1.1L/hr/kg) 보다 유의성있게 높았다. KR-V 3, 10, 14 및 16에 비해 KR-V3 18-1이 혈중곡선하면적(AUC, $8.97{\mu}g{\cdot}hr/ml$)은 크고 겉보기분포용적(Vd, 0.58L/kg)은 적었다. 50mg/kg의 경구투여후 약물동태 연구에 있어서는 KR-V 18-1의 반감기가 다른 4개의 물질에 비해 비록 짧았지만 경구 AUC($3.659{\mu}g{\cdot}hr/ml$), 최고혈중농도($C_{max}$, $1.891{\mu}g/ml$) 는 현저히 높았다. 또한 별도의 in virus 실험결과 생체이용률을 나타낸 이들 5개의 물질들 중 KR-V 18-1만이 HIV-1 돌연변이종(mutants)에 대한 억제효과를 나타내었다. 따라서 KR-V 18-1이 항에이즈(AIDS)제의 새로운 후보물질 혹은 선도물질로서 가능성이 기대되었다.

Immunomodulating activities of water extract from xanthium strumarium 2

  • One of water and/or methanol extracts from 14 herbal deugs which were screened using murine splenocytes showed immunosuppressive activities previously. After water extract from Xanthium strumarium was treated with chloroform. $100 \mu{g/ml}$ of water layer (XS-WCI) has very strong immunosimulating activities tested by $^3H$-thmidine incorporation (control as $100 \mu{g/ml}$, 26345 cpm was 69515 cpm). MLR also appears to be simulated strongly (control vs $100 \mu{g/ml}$, 4962 cpm vs 78688 cpm). When $100 \mu{g/ml}$ of XS-WCI and $0.8 \mu{g/ml}$ of concanavalin a (ConA) were added. more $^3H$-thymidine were incorporated significantly, compared with $0.8 \mu{g/ml}$ of ConA only. In contrast with ConA. results from $5 \mu{/ml}$ of lipopolysaccharide (LPS) and $100 \mu{g/ml}$ of XS-WCI were not different. compared with $5\mu{/ml}$of LPS only. These results indicated the responses of XS-WCI to B cell and T cell may be different. XS-WCI was injected intraperitoneally (10 mg/kg. 50mg/kg/ 100 mg/kg) for 4 days or 10 days and tested secretion of IgM or IgG by direct and indirect hemolytic plaque-forming cell assays, respectively. Numbers of hemolytic plaques for both IgM and IgG were increased significantly. Especially, secretion of IgGs was increased more than 10 times. After administration of XS-WCI for 7 days (50 mg/kg. 100 mg/kg) splenomegaly deu to graft vs host reaction was observed. Human lymhocytes separated from whole blood by Ficoll-Hypaque method were also proliferated after treatment of $10 \mu{g/ml}$ and $50 \mu{g/ml}$ of XS-WCI. As seen in murine lymphocytes, human lymphocyte proliferation was increased synergistically after treatment with both of XS-WCI and phytohemagglutinin (PHA). It appears that XS-WCI may have potential immunosimulating activities and that it remains to be purified further for isolation of active components.

방풍갈근탕(防風葛根湯)의 항알레르기효과에 관한 연구 (Study on Anti-allergic Effect and Safety of Bangpung-galgeun-tang)

  • The purpose of this study was to examine the anti allergic effect in vivo and in vitro, and to observe single and four weeks repeated toxicity in mice of Bangpung-galgeun-tang (BGT). We investigated anti DNP IgE-mediated passive cutaneous anaphylaxis in rodents and compound 48/80-induced active systemic anaphylatic shock in mice after oral administration with BGT of 0.4 g/kg and 0.8 g/kg for 8 days, and also examined MTT assay, ${\beta}-hexosaminidase$ activity, IL-4 and $TNF-{\alpha}$ from RBL-2H3 and $TNF-{\alpha}$ from Raw264.7 after pre-treatment with BGT of 0.25 mg/ml, 0.5 mg/ml, 1 mg/ml and 2 mg/ml. To ascertain safety and toxicity of BGT, we divided into single and four weeks repeated administration test. In single test, three groups were administrated different dosages and routes (2 g/kg/i.p., 4 g/kg/i.p. and 15 g/kg /p.o.) of BGT, and in four weeks repeated test, 0.8 g/kg BGT was administrated. Control groups were administrated with only saline according to on Korean Food and Drug Administration, respectively. We observed attentively motality, abnormal clinical sign, body weight change, organ weight, AST and ALT of mice after BGT administration. BGT inhibited passive cutaneous anaphylaxis and active systemic anaphylatic shock by oral administration. All the concentrations of BGT from 0.25 to 2 mg/ml didn't have an effect on cell viability and cytotoxicity. In RBL-2H3, ${\beta}-hexosaminidase$ release, IL-4 and $TNF-{\alpha}$, and in Raw264.7, $TNF-{\alpha}$ were significantly reduced by treated all concentrations of BGT. During toxicity experiment period, there was no difference in body weight change, organ weight, AST and ALT among different dose groups. Death were found 3 mice from day 2 to day 3 in single test i.p. group. (2 g/kg, 4 g/kg). Several individuals of single test i.p. group were observed that decreased locomotor activity, exophthalmos, bloodshot eyes, loss of eyesight and so on in early period after administration. But there was no difference in clinical signs among p.o. group. These results indicate that BGT have inhibition effects on allergy and suggest that no observable effect level of the test orally administration was considered to be more than 2 g/kg in mice under the conditions employed in this study.

담수조건에서 토양의 표면에 시용된 요소태 질소의 행동에 미치는 Methanol의 영향 (Effect of Methanol on the Behavior of Urea-N Applied to the Soil Surface under Submerged Condition)

  • 담수토양에서 이분해성 유기물 (methanol)이 표면에 시용된 요소로 부터 유래하는 요소태 질소 (Urea-N)와 암모늄태 질소 ($NH_4-N$) 및 질산태 ($NO_3-N$) 질소의 행동에 미치는 영향을 조사하였다. 토양에 요소를 $150kg\;N\;ha^{-1}$ (관행구)와 $300kg\;N\;ha^{-1}$ (배비구)의 수준으로 처리하고 이분해성 유기물인 메탄올 ($CH_3OH$)을 2, 4 및 8 ml 처리한 후 30일 동안 담수 항온시키면서 표면수와 토양의 pH, Urea-N, $NH_4-N$$NO_3-N$의 농도를 시간별로 조사한 결과는 다음과 같다. 담수 10일째에 관행구와 배비구의 표면수에서는 Urea-N이 검출되지 않았다. 그러나 토양 중 Urea-N은 관행구에서는 검출되지 않았으나 배비구에서는 메탄올을 8 ml 처리한 경우에 담수 10일째에 $4.7{\mu}g\;g^{-1}$의 농도를 나타내었으며 그 이후에는 검출되지 않았다. 표면수의 $NH_4-N$ 농도는 무처리 0.5-3.5, 관행구 4.1-12.1 및 배비구 $7.2-20.5{\mu}g\;ml^{-1}$의 범위로서 배비구에서 가장 높았다. 무처리, 관행구 및 배비구 모두 메탄올 처리량이 증가할 수록, 담수시간이 경과할 수록 약간 증가하는 경향이었다. 토양의 $NO_3-N$ 농도는 무처리 1.7-6.7, 관행구 2.2-7.3 및 배비구 $2.8-7.8{\mu}g\;g^{-1}$로서 거의 비슷한 농도를 나타내었다. 무처리, 관행구 및 배비구 모두 메탄을 처리량에 따른 토양의 $NO_3-N$ 농도의 큰 차이가 없었으며, 담수시간이 경과할 수록 감소하는 경향이었다. 표면수와 토양의 $NH_4-N$ 총량은 배비구에서 가장 많았으며, 무처리, 관행구 및 배비구 모두 메탄올 처리량이 증가할 수록 $NH_4-N$ 총량은 감소하였다. 담수시간 별로는 무처리 관행구 및 배비구 모두 담수 20일까지는 증가하고 그 이후에는 감소하는 경향으로서 토양의 $NH_4-N$ 농도변화와 관계가 있었다. 표면수와 토양의 $NO_3-N$ 총량은 무처리, 관행구 및 배비구 모두 메탄올을 처리하지 않은 경우와 메탄올을 2 ml 처리한 경우에는 차이가 없었으나 메탄올을 4와 8 ml 처리한 경우에는 약간 증가하는 경향이었다. 무처리, 관행구 및 배비구 모두 담수시간이 경과함에 따라 $NO_3-N$ 총량은 감소하는 경향이었다.

  • 최근 줄기세포를 이용한 재생의학분야의 세포치료 연구가 활발히 진행되고 있으나 상대적으로 복잡한 구조를 갖는 신장에서는 난치성 질환인 급성신부전에 대한 줄기세포 치료의 효과는 아직도 논란의 여지가 많다. 이는 연구자마다 다양한 동물모델을 사용하는 것이 한 원인으로 지적되고 있다. 본 연구에서는 줄기세포의 치료효과 분석을 위한 최적의 동물모델 조건을 확립하고자 한다. 쥐를 사용하여 세 종류의 급성신장손상모델을 다음과 같이 다양한 농도와 조건에서 분석하였다: glycerol (대조군, 8 ml/kg, 10 ml/kg), cisplatin (대조군, 5 mg/kg, 10 mg/kg)과 허혈/재관류 (대조군, 35 분 허혈, 45 분 허혈). 줄기세포의 재생능력 평가를 위한 모델의 적합성 정도를 신장손상과 회복의 형태적, 기능적 패턴과 생존률 분석을 통하여 실시하였다. 독성모델인 glycerol과 cisplatin은 각각 10 ml/kg과 7.5 mg/kg 농도에서, 그리고 허혈모델은 45 분 모델에서 대조군 및 다른 비교모델에 비해 장기간의 관찰 기간 동안 신기능의 유의적 차이를 보였다. 또한 45 분 허혈 모델에서 50%의 사망률을 보인 것을 제외하고는 모든 군에서 관찰기간 동안 100% 생존율을 보였다. 조직소견에서 허혈과 glycerol 모델에서는 2일째, cisplatin 모델에서는 5일째 각각 유의적인 조직변화가 관찰되었다. 따라서, 관찰기간 동안 급격한 생존율 감소 없이 안전하고, 조직학적 평가와 신기능에서 대조군과 유의적인 차이를 보이는 cisplatin 7.5 mg/kg, glycerol 10 ml/kg, 35 분 허혈 모델이 치료효과를 평가하기에 적합한 모델로 사료되며, 줄기세포의 세포치료효과 분석에 적합한 급성신장손상 모델로 추천된다.
