Biomass and Net Primary Productivity of Pinus densiflora Natural Ecosystem in Kangwondo, Korea (강원도산(江原道産) 소나무천연림생태계(天然林生態系)의 Biomass 및 Net Primary Production에 관(關)한 연구(硏究))

  • Dimension analysis was used to estimate biomass and net primary production(NPP) in a 36-year-old Japanese red pine (Pines densiflora S. et Z.) natural forest in Kangwondo. Best estimation was made by the equation model of $Wt=aD^bH^c$ where Wt is weight in kg, D is DBH in cm, and H is total tree height in m. Total aboveground biomass was estimated at 198.82 t/ha. Organic matter was distributed in the Japanese red pine stand as follows ; bolewood 68.8%, live branches 16.5%, bolebark 5.4%, foliage 4.6%, dead branches 3.7%, and cones 0.6%. Net primary production was estimated at 15.87 t/㏊/yr and was distributed: bolewood 44.5%, live branches 30.9%, foliage 14.1%, current twig 7.1%, and bolebark 3.3%. Leaf efficiency was estimated at 1.876. The power equation for biomass and IVPP yielded similar results as the results for Pines densiflora in Japan.

Free Living Amoeba-Bacteria Interactions: Analysis of Escherichia coli Interactions with Nonpathogenic or Pathogenic Free Living Amoeba

  • Free-living amoebae ingest several kinds of bacteria. In other words, the bacteria can survive within free-living amoeba. To determine how Escherichia coli K1 isolate causing neonatal encephalitis and non-pathogenic K12 interact with free-living amoebae, e.g., Acanthamoeba castellanii (T1), A. astronyxis (T7), Naegleria fowleri, association, invasion and survival assays were performed. To understand pathogenicity of free-living amoebae, in vitro cytotoxicity assay were performed using murine macrophages. T1 destroyed macrophages about 64% but T7 did very few target cells. On the other hand, N. fowleri which needed other growth conditions rather than Acanthamoeba destroyed more than T1 as shown by lactate dehydrogenase (LDH) release assay. In association assays for E. coli binding to amoebae, the T7 exhibited significantly higher association with E. coli, compared with the T1 isolates (P<0.01). Interestingly, N. fowleri exhibited similar percentages of association as T1. Once E. coli bacteria attach or associate with free-living amoeba, they can penetrate into the amoebae. In invasion assays, the K1 (0.67%) within T1 was observed compared with K12 (0%). E. coli K1 and K12 exhibited high association with N. fowleri and bacterial CFU. To determine the fate of E. coli in long-term survival within free-living amoebae, intracellular survival assays were performed by incubating E. coli with free-living amoebae in PBS for 24 h. Intracellular E. coli K1 within T1 (2.5%) and T7 (1.8%) were recovered and grown, while K12 were not found. N. fowleri was not invaded and here it was not recovered.

Description and Genomic Characteristics of Weissella fermenti sp. nov., Isolated from Kimchi

  • A Gram-positive, non-motile, and non-spore-forming lactic acid bacterium, designated as BK2T, was isolated from kimchi, a Korean traditional fermented vegetable food, and the taxonomic characteristics of strain BK2T, along with strain LMG 11983, were analyzed. Both strains optimally grew at 30℃, pH 7.0, and 1.0% NaCl. Cells of both strains were heterofermentative and facultatively anaerobic rods, demonstrating negative reactions for catalase and oxidase. Major fatty acids (>10%) identified in both strains were C18:1 ω9c, C16:0, and summed feature 7 (comprising C19:1 ω6c and/or C19:1 ω7c). The genomic DNA G+C contents of both strains were 44.7 mol%. The 16S rRNA gene sequence similarity (99.9%), average nucleotide identity (ANI; 99.9%), and digital DNA-DNA hybridization (dDDH; 99.7%) value between strains BK2T and LMG 11983 indicated that they are different strains of the same species. Strain BK2T was most closely related to Weissella confusa JCM 1093T and Weissella cibaria LMG 17699T, with 100% and 99.4% 16S rRNA gene sequence similarities, respectively. However, based on the ANI and dDDH values (92.3% and 48.1% with W. confusa, and 78.4% and 23.5% with W. cibaria), it was evident that strain BK2T represents a distinct species separate from W. confusa and W. cibaria. Based on phylogenetic, phenotypic, and chemotaxonomic features, strains BK2T and LMG 11983 represent a novel species of the genus Weissella, for which the name Weissella fermenti sp. nov. is proposed. The type of strain is BK2T (=KACC 22833T=JCM 35750T).

  • The Internet of Things has attracted considerable research attention in recent years. In order for the new IoT technology to be widely used, the reliability and protection of information is required. IoT systems are very vulnerable to physical security due to their easy accessibility. Along with the development of SoC technology, many operating systems have been developed and many new operating systems have been introduced. In this paper, we describe the vulnerability analysis results for operating systems running on the ARMv7 Thumb Architecture hardware platform. For the recently introduced "Windows 10 IoT Core" operating system, I implemented the Zero-Day Attack by implanting the penetration code developed through the research into a specific IoT system. The virus detection test for the resulting penetration code was validated by referral to the "virustotal" site.

Comparison of Dietary Values in Seven Species of Marine Diatoms (해산 규조류 7종의 먹이효과 비교)

  • In order to identify some suitable diatoms as a live food source, effects of seven diatom species (Chaetoceros simplex, Navicula incerta, Phaeodactylum tricornutum, Nitzschia closterium, Skeletonema costatum, Thalassiosira pseudonana, Thalassiosira weissflogii) were examined on specific growth rate, mean celt volume, chemical composition and general fatty acid composition. In cell volume. T. pseudonana was the smallest $(125{\mu}m^3)$ and T. weissflogii the largest $(824{\mu}m^3)$. However the specific growth rate of the largest diatom was the lowest (0.5016/day), and that of S. costatum was the highest (0.9928/day). The crude protein content of seven diatom species varied from $18.96\%$ (T. pseudonana) to $24,4\%$ (T, weissflogii). The crude lipid content of C. simplex was the highest $(8.43\%)$, and that of T. pseudonana was the lowest $(3.13\%)$. The total content of polyunsaturate fatty acids which are important for bivalve larvae was higher in C. simplex than those in other species. The total percent of polyunsaturate fatty acids from N. incerta and T. weissflogii was relatively lower than that from other species. Dietary value of seven diatom species were finally examined with Pacific oyster larvae Crassostrea gigas. In this experiment, C. simplex showed the highest survival rate and growth, and the larvae feed on S. costatum and T. weissflogii showed the poorest results. This results C. simplex could be more useful live food for bivalve larvae than other diatom species tested.

  • Infestations of two spotted spider mite (TSSM), Tetranychus urticae Koch, on glasshouse rose (Rosa sp.) grown by an arching method, were determined by counts of the number of TSSM per leaflet in Buyeo, Chungnam Province, for a 2-yr period. Binomial sampling plans were developed based on the relationship between mean density per leaflet (m), and proportion of leaflets infested with ( T mites (PT), according to the empirical model In (m) = a+p In (-ln (1 -PT)). T was defined as tally threshold, and set to 1, 3, 5, 7, and 9 mites per leaflet. Increasing sample size had little effects on the precision of the binomial sampling plan, regardless of tally threshold. However, the precision increased with higher tally thresholds. There was a negligible improvement in precision with T ) 7 mites per leaflet. T= 7 was chosen as the best tally threshold for estimating densities of TSSM based on the precision of the model. Independent data set was used to evaluate the model. The binomial model with T= 7 provided reliable predictions of mean densities of TSSM observed on the commercial glasshouse roses.

Interleukin-7 Enhances the in Vivo Anti-tumor Activity of Tumor-reactive CD8+ T cells with Induction of IFN-gamma in a Murine Breast Cancer Model

  • Interleukin-7 (IL-7) is a potent anti-apoptotic cytokine that enhances immune effector cell functions and is essential for lymphocyte survival. While it known to induce differentiation and proliferation in some haematological malignancies, including certain types of leukaemias and lymphomas, little is known about its role in solid tumours, including breast cancer. In the current study, we investigated whether IL-7 could enhance the in vivo antitumor activity of tumor-reactive $CD8^+$ T cells with induction of IFN-${\gamma}$ in a murine breast cancer model. Human IL-7 cDNA was constructed into the eukaryotic expression plasmid pcDNA3.1, and then the recombinational pcDNA3.1-IL-7 was intratumorally injected in the TM40D BALB/C mouse graft model. Serum and intracellular IFN-${\gamma}$ levels were measured by ELISA and flow cytometry, respectively. $CD8^+$ T cell-mediated cytotoxicity was analyzed using the MTT method. Our results showed that IL-7 administration significantly inhibited tumor growth from day 15 after direct intratumoral injection of pcDNA3.1-IL-7. The anti-tumor effect correlated with a marked increase in the level of IFN-${\gamma}$ and breast cancer cells-specific CTL cytotoxicity. In vitro cytotoxicity assays showed that IL-7-treatment could augment cytolytic activity of $CD8^+$ T cells from tumor bearing mice, while anti-IFN-${\gamma}$ blocked the function of $CD8^+$ T cells, suggesting that IFN-${\gamma}$ mediated the cytolytic activity of $CD8^+$ T cells. Furthermore, in vivo neutralization of $CD8^+$ T lymphocytes by CD8 antibodies reversed the antitumor benefit of IL-7. Thus, we demonstrated that IL-7 exerts anti-tumor activity mainly through activating $CD8^+$ T cells and stimulating them to secrete IFN-${\gamma}$ in a murine breast tumor model. Based on these results, our study points to a potential novel way to treat breast cancer and may have important implications for clinical immunotherapy.

The Predictable Factors of the Postoperative Kyphotic Change of Sagittal Alignment of the Cervical Spine after the Laminoplasty

  • Objective : Laminoplasty is an effective surgical method for treating cervical degenerative disease. However, postoperative complications such as kyphosis, restriction of neck motion, and instability are often reported. Despite sufficient preoperative lordosis, this procedure often aggravates the lordotic curve of the cervical spine and straightens cervical alignment. Hence, it is important to examine preoperative risk factors associated with postoperative kyphotic alignment changes. Our study aimed to investigate preoperative radiologic parameters associated with kyphotic deformity post laminoplasty. Methods : We retrospectively reviewed the medical records of 49 patients who underwent open door laminoplasty for cervical spondylotic myelopathy (CSM) or ossification of the posterior longitudinal ligament (OPLL) at Pusan National University Yangsan Hospital between January 2011 and December 2015. Inclusion criteria were as follows : 1) preoperative diagnosis of OPLL or CSM, 2) no previous history of cervical spinal surgery, cervical trauma, tumor, or infection, 3) minimum of one-year follow-up post laminoplasty with proper radiologic examinations performed in outpatient clinics, and 4) cases showing C7 and T1 vertebral body in the preoperative cervical sagittal plane. The radiologic parameters examined included C2-C7 Cobb angles, T1 slope, C2-C7 sagittal vertical axis (SVA), range of motion (ROM) from C2-C7, segmental instability, and T2 signal change observed on magnetic resonance imaging (MRI). Clinical factors examined included preoperative modified Japanese Orthopedic Association scores, disease classification, duration of symptoms, and the range of operation levels. Results : Mean preoperative sagittal alignment was $13.01^{\circ}$ lordotic; $6.94^{\circ}$ lordotic postoperatively. Percentage of postoperative kyphosis was 80%. Patients were subdivided into two groups according to postoperative Cobb angle change; a control group (n=22) and kyphotic group (n=27). The kyphotic group consisted of patients with more than $5^{\circ}$ kyphotic angle change postoperatively. There were no differences in age, sex, C2-C7 Cobb angle, T1 slope, C2-C7 SVA, ROM from C2-C7, segmental instability, or T2 signal change. Multiple regression analysis revealed T1 slope had a strong relationship with postoperative cervical kyphosis. Likewise, correlation analysis revealed there was a statistical significance between T1 slope and postoperative Cobb angle change (p=0.035), and that there was a statistically significant relationship between T1 slope and C2-C7 SVA (p=0.001). Patients with higher preoperative T1 slope demonstrated loss of lordotic curvature postoperatively. Conclusion : Laminoplasty has a high probability of aggravating sagittal balance of the cervical spine. T1 slope is a good predictor of postoperative kyphotic changes of the cervical spine. Similarly, T1 slope is strongly correlated with C2-C7 SVA.

Twist2 Regulates CD7 Expression and Galectin-1-Induced Apoptosis in Mature T-Cells

  • In the periphery, a galectin-1 receptor, CD7, plays crucial roles in galectin-1-mediated apoptosis of activated T-cells as well as progression of T-lymphoma. Previously, we demonstrated that $NF-{\kappa}B$ downregulated CD7 gene expression through the p38 MAPK pathway in developing immature thymocytes. However, its regulatory pathway is not well understood in functional mature T-cells. Here, we show that CD7 expression was downregulated by Twist2 in Jurkat cells, a human acute T-cell lymphoma cell line, and in EL4 cells, a mature murine T-cell lymphoma cell line. Furthermore, ectopic expression of Twist2 in Jurkat cells reduced galectin-1-induced apoptosis. While full-length Twist2 decreased CD7 promoter activity, a C-terminal deletion form of Twist2 reversed its inhibition, suggesting an important role of the C-terminus in CD7 regulation. In addition, CD7 expression was enhanced by histone deacetylase inhibitors such as trichostatin A and sodium butyrate, which indicates that Twist2 might be one of candidate factors involved in histone deacetylation. Based on these results, we conclude that upregulation of Twist2 increases the resistance to galectin-1-mediated-apoptosis, which may have significant implications for the progression of some T-cells into tumors such as Sezary cells.
