Low Ruminal pH Reduces Dietary Fiber Digestion via Reduced Microbial Attachment

  • In vitro rumen incubation studies were conducted to determine effects of initial pH on bacterial attachment and fiber digestion. Ruminal fluid pH was adjusted to 5.7, 6.2 and 6.7, and three major fibrolytic bacteria attached to rice straw in the mixed culture were quantified with real-time PCR. The numbers of attached and unattached Fibrobacter succinogenes, Ruminococcus flavefaciens and Ruminocococcus albus were lower (p<0.05) at initial pH of 5.7 without significant difference between those at higher initial pH. Lowering incubation media pH to 5.7 also increased bacterial numbers detached from substrate regardless of bacterial species. Dry matter digestibility, gas accumulation and total VFA production were pH-dependent. Unlike bacterial attachment, maintaining an initial pH of 6.7 increased digestion over initial pH of 6.2. After 48 h in vitro rumen fermentation, average increases in DM digestion, gas accumulation, and total VFA production at initial pH of 6.2 and 6.7 were 2.8 and 4.4, 2.0 and 3.0, and 1.2 and 1.6 times those at initial pH of 5.7, respectively. The lag time to reach above 2% DM digestibility at low initial pH was taken more times (8 h) than at high and middle initial pH (4 h). Current data clearly indicate that ruminal pH is one of the important determinants of fiber digestion, which is modulated via the effect on bacterial attachment to fiber substrates.

Effect of an Acid pH Shock on Physiological Changes of Chlamydomonas acidophila (Chlorophyta), UTCC 122 (pH 충격에 의한 Chlamydomonas acidophila (Chlorophyta), UTCC 122의 생리적 변화에 관한 연구)

  • The effect of low pH on physiological changes was studied with the acidophilic green alga, Clamydomonas acidophila, UTCC 122. The growthrates (${\mu}$) were identical, $0.5{\sim}0.7\;day^{-1}$, at pH 3.7${\sim}$6.7 and no significantly different (ANOVA, p =0.134), showing cell volume reduced gradually as they were growing, whereas that at pH 2.7 was falling to zero and cell volume increased dramatically. Chlorophyll a concentration of the cultures incubated for one day was $191{\sim}255\;pg\;cell^{-1}$, after then it declined from $60{\sim}103\;pg\;cell^{-1}$ at pH 3.7${\sim}$6.7 except $210\;pg\;cell^{-1}$ at pH 2.7, which was directly related with cell volume. External carbonic anhydrase (CA) activity was varied from1.1 to$3.7{\times}10^{-4}\;E.U.\;mm^{-2}$, showing the gradualincrease during culture, except at 2.7 and pH 5.7. However there was not found any relationship among the pH gradient cultures. CA molecular mass of C. acidophila was 29 kBa, and concentration of that was identical in all cultures. The proteins of 41 kDa and 63 were not or very faintly expressed in low pH cultures, in contrast that of 17 kDa more expressed. In this work, we found that C. acidophila could live optimally within a wide range of acidic pH, and 17 kDa of unidentified protein might be concerned with tolerating in low acid environment.

  • The stability difference of metmyoglobin in $H_2O$ and $D_2O$ at pH 5.7 and pH 7.0 toward pressure denaturation is studied. Metmyoglobin is denatured in $D_2O$ at smaller pressure than in $H_2O$. The stability difference in $H_2O$ and $D_2O$ is more pronounced at pH 5.7 than at pH 7. The main reasons for the stability difference in $H_2O$ and $D_2O$are the difference in positive charge due to $H^+$and $D^+$ binding to the protein in $H_2O$ and $D_2O$, and the structural change that accompany deuteration.

Combination Effects of Benzoate, Sorbate and pH for Control of Escherichia coli O157:H7 (Escherichia coli O157:H7의 제어를 위한 Benzoate, Sorbate 및 pH의 병용처리 효과)

  • Effects of benzoate (0~0.6 g/$\ell$ ) and sorbate (0~2.0 g/$\ell$) on the growth of Escherichia coli O157:H7 in tryptic soy broth at various pH levels (4~8) and temperatures (4$^{\circ}C$, 37$^{\circ}C$) were investigated. Benzoate and sorbate were inhibited the growth of E. coli O157:H7 up to 12 hours cultivation at 4$^{\circ}C$, and 2.0 g/$\ell$ sorbate was only inhibited during 48 hours cultivation at 37$^{\circ}C$. Among the pH levels tested, pH 4 showed significant inhibitory effect against the E. coli O157:H7 on 4$^{\circ}C$ and at 37$^{\circ}C$, respectively. When used in combination 0.2 g/$\ell$ benzoate and sorbate were completely inhibited the growth of E. coli O157:H7 on pH 4 and at 37$^{\circ}C$. While on pH 5 at 4$^{\circ}C$, all of the concentration tested did not exert any inhibitory effect. The combined effects were retarded more than single treatment of E. coli O157:H7.

The Comorbidity of Attention Deficit Hyperactivity Disorder and the Effect of Methylphenidate on it (주의력 결핍 과잉 운동 장애 환자들의 약물치료 효과 및 Comorbidity에 관한 연구)

  • The objectives of this study were to evaluate the effect of methylphenidate on attention deficit hyperactivity disorder (ADHD) and the comorbidity of the disorder, using child attention problem checklist to 56 (male: 38, female: 18) patients from, March 992 to February 1993. The results were as follows: Among 56 subjects, ADHD alone were 20 (335.71%) subjects, and with one additional diagnosis were 31 (55.35%) subjects and with two additional diagnosis were 5 (8.93%) subjects. There was significant improvement on symptoms in the scores rated by teachers in 7th and 28th day after menthlphenidate administration compared to baseline score (P<0.05) and 28th day score showed significant improvement compared to 7th day score (P<0.05) and 28th day score showed significant improvement compared to 7th day score (P<0.05). There was significant improvement on symptoms in the scores rated by parents in 28th day after methylphenidate administration improvement compared with 7th day score. In single administration of methylphenidate in Sunday morning score compared to afternoon score (P<0.05). In the administration of significant improvement on symptoms compared with the Sunday morning rating score of parents (P<0.05) and the 28th day comparison was also showed significant improvement on symptoms in the scores rated by teachers compared with the scores rated by parents on symptoms (P<0.05). Among group comparison, all groups showed significant improvements (P<0.05) except conduct disorder & oppositional defiant group.

Changes of multi-purpose solutions for soft contact lens depending on using period or keeping temperature (사용기간 및 온도에 따른 소프트콘택트렌즈 다목적용액의 변화)

  • To investigate changes of multi-purpose solutions for soft contact lens(MPS) depending on using period or keeping temperature, we evaluate four brands of MPS. No significant difference was seen in protein deposit removing efficacy after samples had used for 24 weeks and kept at $4^{\circ}C$, $20^{\circ}C$ or $30^{\circ}C$. The pH values of the samples of 4 brands measured weekly over the 24 week testing period. The initial average pH value of samples were 7.0, 7.5, 7.6 or 8.2. One brand of MPS was in the range of the threshold for ocular awareness, which is outside the zone of 6.6 ~ 7.8. During the testing period, the pH value were decreased in using period-dependent manner. At the 24th week, the average pH values of samples turned to 6.6, 7.2, 7.2 or 7.7. However, the difference of keeping temperature was not associated with decreased levels of pH values. After 24 weeks, one of total 36 samples was contaminated by bacteria. Furthermore, the change of components was shown after 24 weeks in the analysis using thin layer chromatography and the analysis of UV absorption pattern. The results of our study provides that the keeping temperature of MPS is not the important factor of changes of MPS, but the using period of MPS can cause contact lens wearers discomfort.

Evaluation of Korean Native Ducks on Production Efficiency Factor, Carcass Yield, Partial Meat Ratio and Meat Quality with Weeks (토종오리 대형종의 주령별 생산지수, 도체 수율, 부분육 비율 및 육질 평가)

  • This work was carried to evaluate production efficiency factor, carcass yield and meat quality with weeks of Large-type Korean native ducks. Korean native ducks (n = 90) from National Institute of Animal Science (RDA, Korea) were used in this work. Ninety ducks were divided into 6 groups (15 birds/group) and were fed with meat-type duck diets for 8 wk old. When ducks grew at specific wk (6, 7 and 8 weeks), 2 ducks per group were slaughtered at 6, 7 and 8 wk old. Production efficiency factor, carcass yield, partial meat and meat quality were researched in this work. There was no significant difference on livability with weeks, but body weight at 7 and 8 wk old was higher than that at 6 wk old (P<0.05). Feed conversion ratio at 6, 7 and 8 wk old were 2.25, 2.69 and 3.21, respectively, so there was significant difference with weeks (P<0.05). Production efficiency factor at 6, 7 and 8 wk old were 256.6, 199.8 and 153.0, respectively, so there was significant difference with weeks (P<0.05). Carcass yield at 8 wk old was higher than that at 6 and 7 wk old as 73.5% (P<0.05). Lightness at 6, 7 and 8 wk were 41.8, 39.0 and 38.1, respectively, and that at 6 wk old was the higher than other weeks (P<0.05). There was no significant difference on redness at 6, 7, and 8 wk old (P>0.05) and yellowness at 8 wk old was higher compared to other weeks (P<0.05). Cooking loss was the highest at 6 wk old as 31.6%, but water holding capacity was the highest at 8 wk old (P<0.05). There was no significant difference on shear force among weeks. pH at 6 wk old was the lower than that of other weeks as 5.84. Moisture content significantly decreased with weeks (P<0.05) and fat content at 8 wk was the highest as 1.88% (P<0.05). Protein content significantly increased with weeks until 20.9% at the age of 8 wk (P<0.05). Ash content at 7 and 8 wk old was the higher than that at 6 wk old (P<0.05). There was no significant difference on juiciness, tenderness, and flavor with weeks. Finally, these results may provide that shipping time at 7 wk old preferred to that at 6 and 7 wk old, but further research was needed because of deficiency of data.

Modification of Papain with $IO_4-Oxidized$ Soluble Starch ($IO_4$-산화 가용성 전분에 의한 파파인의 변형)

  • Periodate-oxidized soluble starch was reacted with papain at pH 4.0, pH 7.0, and pH 9.7, and an oxidized soluble starch-papain conjugate was produced. When compared with native papain, the specific activity decreased to 60%, in both the modified papain reacted with 0.4% $NaBH_4$ and in the modified papain not reacted with $NaBH_4$. The specific activity decreased to 70% in the modified papains reacted with 1.5% $NaBH_4$ and 4.0% $NaBH_4$, respectively. The reduction by $NaBH_4$ did not have an effect in the thermal stability of either the modified or nonmodified papain. An activity of 54.7% remained in the papain modified at pH 4.0, which was incubated at $80^{\circ}C$ for 40 min. The papains modified at pH 7.0 and pH 9.7 and incubated for 40 min at two different temperatures, respectively, were stable to $60^{\circ}C$, and at $80^{\circ}C$ their activities at 56.3% and 44.1 %, respectively. The modified papain's thermal stability pattern was similar to that of native papain, with no increase in its statbility. In the range of pH $2.0{\sim}13.0$, the stability of the papain modified at pH 4.0 decreased greatly between pH $3.0{\sim}5.0$, but it was similar to the native papain at other pH values. The stability of the papain modified at pH 7.0 showed a similar pattern to the native papain at pH $2.0{\sim}6.0$, while its stability increased when moving into the alkali pH range. The papain modified at pH 9.7 also had increased stability, when moving into the alkali range. The results of Hammerstein milk casein, which was reacted with the papains modified at pH 4.0, pH 7.0, and pH 9.7, respectively, and analyzed by FPLC, showed different peaks according to the different modification pHs, and the greatest peak differences were observed with the modification at pH 9.7.

Disappearance of Organic Phosphate Insecticides Residue on Vegetables and Fruit Crops (과실 및 채소류에 대한 유기 인계 농약의 잔류소장)

  • Sumithion and EPN residues on grapes, EPN and Diazinon on chinese cabbage, Parathion on peaches, Dimethoate on tomatoes, and EPN and Malathion on cucumber were analyzed in terms of 0, 3, 7, 14, 21 and 30 days after last application for the pesticides safty use. From the disappearance rate for various organo-phosphate insecticides on vegetables and fruit crops, following results are obtained. 1. On Chinese cabbage, Diazinon residues were 0.25~0.38p.p.m three weeks after one application, and EPN were 1.39~2.69p.p.m seven days after one application and 0.96~2.34p.p.m two weeks after twice application. 2. EPN residues on grapes were 1.09~1.80p.p.m seven days after one application and Sumithion were 0.17~0.53p.p.m fourteen days after one application. 3. On peaches, Parathion residues were 0.40~0.61p.p.m two weeks after last application. 4. Dimethoate residues on tomatoes were 0.141p.p.m seven days after four times application. 5. On cucumber, EPN residues were 2.11~2.14p.p.m three days after twice application, and Malathion were 0.46p.p.m 3 day after four times application but 0.062~0.025p.p.m three days after last application. 6. Rate of degradation of organo-phosphate chemicals is inversely related to half-life of its. 7. Minimum intervals between last treatment and harvest to prevent unsafty residues are as follows. 7 days for EPN with one application and 14 days with twice application on chinese cabbage, 3 days on cucumber and 7 days on grape, 14 days for parathion, 7 days for dimethoate on tomatoes, 0 to 3 days for Malathion on cucumber, 21 days for Sumithion on grape, 21 days on chinese cabbage for Diazinon.

Relationship between Nursing Organizational Climate and Job Satisfaction of Nurses in general hospitals (병원 간호조직풍토와 간호사 직무만족도의 관계)

  • The purpose of this study was to identify the relationship between nursing organizational climate and job satisfaction of nurses in general hospitals and also the factors which had influences in the nursing organizational climate and job satisfaction of nurses. Data were collected from 200 nursing managers and 800 nurses with structured questionnaires at 11 general hospitals in Taegu and Kyungbuk-area, from June 1 to June 30, 1999. Data were analyed with SPSS 7.5 using program such as t-test, ANOVA and stepwise multiple regression. The results were as follows: 1) In the nursing organizational climate there were significant differences by age(F=9.246, p=.000), religion(f=5.658, p=.001), educational level(F=4.660, p=.010), position(F=27.016, p=.000), and the total length of service(F=7.274, p=.000). Also there were significant differences by subsidiary school(F=11.224, p=.000), the number of beds(F=9.893, p=.000), the number of nurses(F=6.365, p=.000), and kind of medical agency(F=5.251, p=.000) in the hospitals. 2) In the nurses' job satisfaction there were significant differences by age(f=11.528, p=.000), religion(F=3.003, p=.000), position(F=22.485, p=.000), career the department of the present service(F=5.157, p=.000), total career of service(F=9.243, p=.000), and salary(F=5.507, p=.000). Also there were significant differences by religious background(F=4.779, p=.009), subsidiary school(F=7.039, p=.000), the number of beds(F=7.039, p=.000), and kind of medical agency(F=2.778, p=.006) in the hospitals 3) There was significant correlation between nursing organizational climate and job satisfaction of nurses(r=.686). 4) The nursing organizational climate was explained 21.8% by salary 9.5%, position 7.4%, religious background of hospital 4.1%, and subsidiary school of hospital 0.8%. 5) The nurses' job satisfaction was explained 70.9% by nursing organizational climate 46.7%, salary 21.9%, kind of medical agency 1.4%, position 0.3%, religious background of hospital 0.3%, religion 0.3%.
