Caspase-11 Promoter-GFP Construct as a Dual Reporter of Cytotoxicity and Inflammation

  • Caspase-11 has been known as a dual regulator of apoptosis and inflammatory response. An unusual feature of caspase-11 is that its expression is induced by apoptotic or proinflammatory stimuli. Utilizing these unusual features of caspase-11, we have developed a simple and sensitive assay method to screen pro- or anti-apoptotic/inflammatory molecules. To develop this assay method, we generated a reporter construct where GFP expression is regulated by caspase-11 promoter. When several types of cultured cells were transfected with this reporter construct and subsequently treated with various apoptotic or proinflammatory molecules, expression of GFP by the activation of caspase-11 promoter was easily detected by fluorescence microscopy or spectrofluorometry. In addition, a reduction of the GFP fluorescence was detected when an agent reported to suppress caspase-11 induction was applied. These results suggest that our reporter system can be used to screen pro- or anti-apoptotic/inflammatory molecules.

레이저 형광법을 이용한 우식유발 예측모형 (CARIES PREDICTION MODEL USING LASER FLUORESCENCE)

  • 레이저 형광법을 이용하여 각 개인의 우식의 활성도를 측정할 수 있는지를 규명하기 위해 $7\sim9$세의 아동 50명을 대상으로 치아의 순면과 협면에 아르곤 레이저를 조사하고 특수 필터를 사용하여 초기 치아우식증이 관찰되는 치아의 수를 측정하고, 이와 같은 초기 치아우식증의 수를 측정하는 우식활성법과 기존의 우식 활성도 측정 방법인 dDfFtT rate, Streptococcus mutans colony count와 상관성을 비교, 평가하고, 레이저 형광법을 이용한 우식활성검사의 특이도, 민감도, 진단력을 평가한 결과, 다음과 같은 결론을 얻었다. 1. 아르곤 레이저 형광법을 이용한 우식활성도 측정법은 기존의 Streptococcus mutans colony count 검사법과 비교적 높은 상관관계를 보였다($\gamma=0.48$, P<0.01). 2. 아르곤 레이저 형광법을 이용한 우식활성도 측정법은 dDfFtT rate 검사와 상관관계가 있었다($\gamma=0.39$, P<0.01). 3. Streptococcus mutans colony count와 dDfFtT rate와는 낮은 상관관계가 있었다($\gamma=0.27$, P<0.05). 4. dDfFtT rate를 기준검사 방법으로 하였을 때, 레이저 형광법을 이용한 우식활성 검사법은 특이도 44.4%, 민감도 85.7%, 진단력이 87.8%였다. 5. dDfFtT rate를 기준검사방법으로 하였을 때, Streptococcus mutans colony count는 특이도 77.8%, 민감도 92.9%, 진단력이 84.8%였다. 6. Streptococcus mutans colony count 검사법을 기준검사 방법으로 하였을 때 레이저 형광법을 이용한 우식활성 검사법은 특이도 40.0%, 민감도 84.8%, 진단력이 95.1%였다. 이를 종합해 볼 때 레이저 형광법을 이용하여 관찰한 우식활성 검사법은 기존의 우식활성검사 및 구강환경 검사와 비교적 높은 상관관계, 우수한 진단학적 지표를 보여 주므로써 향후 임상적으로 신뢰도와 타당성이 높은 우식활성 검사법으로서 그 활용 가능성이 높을 것으로 사료된다.

1,2-비스피라질 에틸렌의 합성과 분광학적 성질에 대한 연구 (Synthesis and Spectral Properties of 1,2-Bispyrazyl Ethylene)

  • 메틸 피라진과 피라진산으로 부터 새로운 화합물 1,2-비스피라진 에틸렌을 합성하여 기기분석과 원소분석으로 이 화합물의 구조를 확인하였다. UV-VIS 스펙트럼에서는 예상했던 $(n,\;{\pi}^*)^1$ 흡수띠가 흡광도가 강한 $({\pi},\;{\pi}^*)^1$ 흡수띠에 묻혀 볼 수가 없었으나 형광에 미치는 염의 효과로 부터 $(n,\;{\pi}^*)^1$ 상태가 $({\pi},\;{\pi}^*)^1$ 상태와 거의 같은 에너지를 갖고 있음을 알았다. $77^{\circ}K$에서 형광의 양자 수득률은 0.025로서 같은 조건에서 형광 양자 수득륙이 1인 스틸벤과 좋은 대조를 이루며 이는 $(n,\;{\pi}^*)$$({\pi},\;{\pi}^*)$의 혼합으로 단일 상태에서 삼중 상태로의 intersystem crossing이 효율적으로 잘 일어난다는 것을 말해 준다.

  • Phosphor particles ((Sr,Mg)2 SiO4:Eu2+ were suspended in deionized water in quartz cuvette and used for measuring liquid temperature field by using two-color-ratio method. In the temperature range of 23~77℃, it showed the relative error from 2.4% to 4% and the temperature sensitivity of 0.65 %/℃ at 30℃ and 0.95 %/℃ at 77 ℃. This performance is comparable to measurement techniques using thermographic liquid crystal or laser induced fluorescence or other thermographic phosphor particle. Among investigated potential error sources, the particle number density affected the intensity ratio and the temperature, but the effect of laser fluence was not evident.

Identification of Novel Genes with Proapoptotic Activity

  • In order to identify novel proapoptotic genes, we screened approximately 1,000 hypothetical genes whose functions are completely unknown. After these genes were transiently expressed in HeLa cells, their nuclei images were captured using automated high-speed fluorescence microscope, through which the ratio of apoptotic nuclei was estimated. We selected genes that induce greater than 3-fold increase in apoptotic nuclei compared to that of the vector control. The candidate proapoptotic genes were sequenced and their effects on cell death were further confirmed by the additional assay, DNA fragmentation ELISA. Finally, we were able to identify 4 full-length hypo-thetical genes with proapoptotic activity.

  • 4',5'-디히드로소랄렌(DHP)을 합성하여 상온 및 77K에서 이 화합물의 형광 양자수율은 0.08, 인광양자수율은 0.013, 형광수명은 0.95ns, 인광수명은 0.039s 임을 알았다. 이 결과로부터 형광수명은 5,7-디메톡시쿠마린보다 작은반면 소랄렌보다 크고 반면 인광수명은 DMC 보다 크고 소랄렌보다 작음을 알았다. 소랄렌 유도체들의 광독성을 분자적 차원에서 규명하고 DNA에의 광부가 반응을 알아내기 위하여 DHP와 TME의 광부가 반응을 연구하였다. 주 생성물은 TLC로 분석하고 Preparative TLC와 용매추출로 분리해냈다. 분리된 생성물의 구조는 질량분석기, UV, IR, NMR 그리고 원소분석결과에 의해 DHP와 TME외 광고리화첨가반응에 의한 생성물 임을 알았다. DHP는 형광의 자체소광 효과를 볼 수 없으며 TME, 에틸푸마레이트, 푸마로니트릴에 의해 형광이 효과적으로 소광된다. 광부가 반응의 양자수율과 소광실험에서 DHP의 광첨가반응은 단일상태와 삼중상태에서 다 일어나는 것으로 추측된다.

Physiological Responses of Porphyra yezoensis Ueda (Bangiales, Rhodophyta) Exposed to High Ammonium Effluent in a Seaweed-based Integrated Aquaculture System

  • Porphyra yezoensis is known to act as a biofilter against nutrient-rich effluent in seaweed-based integrated aquaculture systems. However, few studies have examined its physiological status under such conditions. In this study, we estimated the photosynthetic activity of P. yezoensis by chlorophyll fluorescence of PSII (${\Delta}F/F'm$ and relative $ETR_{max}$) using the Diving-PAM fluorometer (Walz, Germany). In addition, bioremediation capacity, tissue nutrients, and C:N ratio of P. yezoensis were investigated. The ammonium concentration in seawater of seaweed tank 4 decreased from $72.1{\pm}2.2$ to $33.8{\pm}0.4{\mu}M$ after 24 hours. This indicates the potential role of P. yezoensis in removing around 43% of ammonium from the effluents. Tissue carbon contents in P. yezoensis were constant during the experimental period, while nitrogen contents had increased slightly by 24 hours. In comparison with the initial values, the ${\Delta}F/F'm$ and $rETR_{max}$ of P. yezoensis had increased by about 20 and 40%, respectively, after 24 hours. This indicates that P. yezoensis condition improved or remained constant. These results suggest that chlorophyll fluorescence is a powerful tool in evaluating the physiological status of seaweeds in a seaweed-based integrated aquaculture system.

Protein Binding Characteristics of Brazilin and Hematoxylin

  • 천연색소 Brazilin 및 Hematoxylin의 BSA에 대한 결합 특성을 fluorescence probe 법을 이용하여 측정하였다. Brazilin 및 Hematoxylin은 BSA에 대해 강한 결합 친화력을 보였으며 Hematoxylin은 Brazilin 보다 더 강한 결합력을 보였다. Brazilin 및 Hematoxylin의 농도 증가에 따라 결합상수는 감소하였으며, 이는 probe-단백 결합체와 양화합물간의 상호작용 또는 Probe와 양화합물간의 결합체 형성에 기인하는 것으로 추정되었다. 양화합물과 BSA의 결합은 pH 및 이온강도에 의존적이었으며, 이 결합에는 electrostatic force 및 hydrophobic force 가 관여하는 것으로 추정되었다.

Tracking Intravenous Adipose-Derived Mesenchymal Stem Cells in a Model of Elastase-Induced Emphysema

  • Background: Mesenchymal stem cells (MSCs) obtained from bone marrow or adipose tissue can successfully repair emphysematous animal lungs, which is a characteristic of chronic obstructive pulmonary disease. Here, we describe the cellular distribution of MSCs that were intravenously injected into mice with elastase-induced emphysema. The distributions were also compared to the distributions in control mice without emphysema. Methods: We used fluorescence optical imaging with quantum dots (QDs) to track intravenously injected MSCs. In addition, we used a human Alu sequence-based real-time polymerase chain reaction method to assess the lungs, liver, kidney, and spleen in mice with elastase-induced emphysema and control mice at 1, 4, 24, 72, and 168 hours after MSCs injection. Results: The injected MSCs were detected with QD fluorescence at 1- and 4-hour postinjection, and the human Alu sequence was detected at 1-, 4- and 24-hour postinjection in control mice (lungs only). Injected MSCs remained more in mice with elastase-induced emphysema at 1, 4, and 24 hours after MSCs injection than the control lungs without emphysema. Conclusion: In conclusion, our results show that injected MSCs were observed at 1 and 4 hours post injection and more MSCs remain in lungs with emphysema.

Highly efficient deep-blue electroluminescence using doped PCVtPh with a new host material

  • Novel blue host material, 4,4'-(dinaphthalen-2-yl)-1,1'-binaphthyl (DNBN), was designed and synthesized for OLEDs. In order to test the electroluminescent properties of DNBN, DNBN was used as the host materials for a blue emitter, PCVtPh. The device exhibited deep-blue emission with the CIEx,y coordinates (x=0.15, y=0.08) at 8.0 V, a luminous efficiency of 1.66 cd/A, a power efficiency of 0.77 lm/W and an external quantum efficiency of 2.30 % at 20 mA/$cm^2$, respectively.
