Studies on the Optimum Conditions of Soy Protein Coagulating Enzyme Production from Bacillus sp. IJ-3 Strain and the Action of IJ-3 Strain Enzyme on 75 Globulin (Bacillus sp. IJ-3가 생산하는 대두단백응고효소의 최적생산 조건 및 7S Globulin에 대한 효소적 작용에 관한 연구)

  • A bacterial strain, designated as Bacillu sp. IJ-3 strain, was shown to produce the extracellular soy protein coagulating enzyme and culture conditions for the production of enzyme by this microbial strain was investigated. The culture medium giving a maximum soy protein coagulating activity was consist of 20%(w/v) soymilk, 2.0%(w/v) glucose, 4.0%(w/v) yeast extract, 5.0%(w/v) polypeptone and 1.0%(w/v) potassium phosphate, monobasic. Initial pH was optimal at 6.0 and the enzyme activity in the culture usually reached a maximal level of fermentation at $35^{\circ}C.$ After the culture medium adjustment where required, enzyme activity was reached maximum at 72 hour of cultivation but this enzyme activity was reduced quickly. It can be assumed that Bacillu sp. IJ-3 strain enzyme has a specificity toward the 75 globulin.

Comparison of Protein Electrophoresis Fractions in the Leptospirosis Patient Serum

  • The mechanisms by which leptospires caused disease are not well understood. A number of putative virulence factors have been suggested, but with few exceptions their role in pathogenesis remains unclear. In these days, many cases of leptospirosis are diagnosed by serological immunoassay. Leptospirosis is characterized by the histopathology of liver, kidney, heart, and lung, but the electrophoresis fractions of serum protein are not compared. We analyzed total protein, albumin, aspartic aminotranferase (AST), alanine aminotransferase (ALT), alkaline phosphatase (ALP), creatinine and urea nitrogen (UN) in sera of patients diagnosed leptospirosis. Total protein and albumin were decreased in 18.5% and 31.2% of patients, respectively. AST, ALT, ALP, UN and creatinine were increased in 90.4%, 66.9%, 28.0%, 15.9% and 10.8% of patients, respectively. We performed cellulose acetate membrane electrophoresis (EP) on sera of patients increased both of AST and ALT, and of patients increased both of creatinine and UN. In patients increased both of AST and ALT, and in patients increased both of AST and ALT, the relative percentage of albumin to total protein in patient serum was dcreased in 89.1 % of patients. α₁-globulin, α₂-globulin, β-globulin and γ-globulin were increased in 85.1 %, 75.2%, 33.6% and 98.0% of patients, respectively. In patients increased both of creatinine and UN, the relative percentage of albumin to total protein in patient serum was dcreased in 93.8% of patients. α₁-globulin, α₂-globulin, β-globulin and γ-globulin were increased in 87.5%, 100%, 31.2% and 93.8% of patients, respectively. These data indicate that infection of Leptospira causes severe liver damage to most of leptospirosis patients, but doesn't cause renal damage to most of them. The relative rate of serum protein electrophoresis fractions to total protein are not identical with them of typical hepatitis patient.

  • White and black sesame produced in Korea were defatted with ethyl ether or n-hexane. Defatted sesame meal was extracted with water and salt solution, and protein extraction was precipitated at various pH 1 through 12, with trichloro acetic acid (TCA), tannic acid and ammonium sulfate, respectively. Protein was purified by Sephadex A-25, G-75, G-100 and G-200, and identified its protein fraction by polyacrylamide gel electrophoresis. Amino acids composition of protein in white sesame was analyzed by automatic amino acid analyzer. Protein contents of white sesame, black sesame and sesame meal are 20.5%, 19.2%, and 44.7%, respectively. n-Hexane was the most suitable solvent for extraction of oil from sesame. Crude protein precipitation was better in higher pH. The protein extraction was more effective with the solution containing sodium chloride tinder the pH 8. Globulin in total protein was high and prolamin was less than in other cereal proteins. Glutamic acid contents of white sesame and sesame globulin were 17.1%, and 20%, respectively. Both proteins contained relatively high levels of essential amino acids. 12-13 bands were found in water soluble protein and 2 bands in salt soluble protein were detected by the disc gel electrophoresis, and were identified in both of white and black sesame. The salt soluble protein of white sesame could be purified by Sephadee G-100 and G-200.

Extraction and Electrophoretic Characterization of Rice Proteins

  • Rice proteins were extracted from brown and milled rice of five varieties: Kwanganbyeo, Daeanbyeo, Daejinbyeo, Surabyeo, Hwaseongbyeo; and their electrophoretic patterns were analyzed by SDS-PAGE. Albumin was extracted with water, globulin with 5% NaCl, prolamin with 70% ethanol, and glutelin with 0.2 M sodium borate buffer (pH 10.0) containing 0.5% SDS, 0.6% $\beta$-mercaptoethanol. The ratios of albumin : globulin : prolamin : glutelin in the brown rice were 10.8~14.1 : 12.4~16.4 : 3.6~5.3 : 68.6~72.8, and in milled rice were 4.4~5.6 : 10.6~12.0 : 3.9~5.4 : 75.7~79.8. In albumin seven major bands were observed with molecular weights ranging from 14.g~96.8 kDa, in globulin four bands with molecular weights in the range of 14.4~56.9 kDa, prolamin had only one band with a molecular weight of 14.4 kDa, and glutelin had four bands with molecular weights of 14.4 ~ 57.4 kDa. There were no differences in electrophoretic patterns between rice varieties or between brown and milled rice.

Genetic Correlation of Carcass and Meat Production Traits with Hormones and Metabolic Components in Hawoo (가축의 혈청 호르몬 및 대사물질 농도와 도체 및 산육형질에 대한 유전상관에 관한 연구)

  • This study was aimed to investigate genetic relationships, variables, and correlations between economic traits and metabolic materials in serum components according to bleeding periods and breeding locations for the castrated and not castrated Hanwoo cattle at National Livestock Research Institute. Analysis of variance for serum hormones and metabolic materials showed significant differences by breeding locations except for testosterone and globulin. Statistical differences for serum components were detected by birth year except for cortisol, total protein, globulin and creatinine, and by castration except for total protein and BUN. All the serum components were tended to have sire effects except for testosterone resulting in some degree of additive gene actions. Breeding locations showed statistical significances for carcass weight and back fat thickness, but not in carcass rate, KPH, live weight and transportation weight loss. Effects of breeding locations and castration were significant for all weight measurement periods except for 9 month and 6 month, respectively. A significant sire effect was observed in all weight measurements. Least squared means for concentration of serum components by breeding year, season and castration were not significant. High concentration of cortisol, creatinine and triglyceride and low concentration of IGF-1 and glucose were detected in castrated cattle. Concentration of testosterone with castrated cattle was $5.2\%$ corresponding to non castrated cattle. Estimation of heritabilities of serum components using a sire model with restricted maximum likelihood were ranged 0.07 to 0.58. High heritabilities were estimated for total protein, albumin, globulin, cortisol, creatinine and BUN were 0.53, 0.54, 0.42, 0.45, 0.58 and 0.54, respectively. Low heritabilities were estimated fur calcium, testosterone and IGF-1 for 0.07, 0.15 and 0.12, respectively. Heritabilities for carcass weight, back fat thickness, meat yield index, KPH, and IMF were estimated as 0.39, 0.45, 0.30 0.13, and 0.93. Heritabilities of weights on 18, 12, 9, 6, and 24 month were estimated as 0.78, 0.76, 0.62, 0.58 and 0.58. Estimated heritabilities for average daily gain on 6${\~}$2, 12${\~}$18, and 18${\~}$24 month were 0.80, 0.75 and 0.19, respectively.

Studies on Serum Chemical Values of Holstein Cows (Holstein 암소 혈청(血淸)의 화학성분(化學成分)에 관(關)하여)

  • Thirty of healthy non-pregnant Holstein cows being raised in the vicinity of Daejeon, which had been delivered of calves 4 to 6 times since they were imported to Korea, were selected with the purpose of serum chemical analysis. Total protein, albumin, globulin, total cholesterol, urea nitrogen and glucose and A/G ratio were investigated. The results of present investigation were summarized as follows: 1. Ranges and mean values (with standard error) of total protein, albumin and globulin were 6.87~8.58g/100ml, $7.75{\pm}0.60g/100ml$; 2.6~3.5g/100ml(37.1~45.0%), $3.26{\pm}0.27g/100ml$(40.1%); and 3.78~5.48g/100ml(55.1~64.6%), $4.51{\pm}0.43g/100ml$(58.1%), respectively. 2. Range and mean value (with standard error) of A/G ratio were 0.52~0.86 and 0.73~0.093. 3. Range and mean value (with standard error) of glucose were 35~50mg/100ml and $45.2{\pm}9.1mg/100ml$. 4. Range and mean value(with standard error) of total cholesterol were 122~228mg/100ml and $194.6{\pm}11.1mg/100ml$. 5. Serum urea nitrogen showed that the range of 10.5~15.1mg/100ml and the mean value(with standard error) of $12.7{\pm}1.5mg/100ml$.

Effect of Rutin on Major Lipid-Related Parameters in Sera of Rats Fed High Fat Diet (루틴 첨가가 고지방 식이 랫드의 혈청 지질 농도의 변화에 미치는 영향)

  • This study was performed to investigate the changes of the serological lipid-related parameters of the rats when they were fed with the high fat diets supplemented with or without rutin for five weeks. Twenty-four Sprague-Dawley male rats ($272.2{\pm}7.2 g$ of body weight) were randomly divided into three groups: control (C) group and two treatment groups. Rats in the C group were fed with the high-fat diet containing 20% lard, 1% cholesterol and 0.5% sodium cholate (w/w) which was modified from the formula of the American Institute of Nutrition-76 (AIN-76) diet. Rats in treatment groups were fed with above diet supplemented with 0.75% rutin (R-0.75) or 1.5% rutin (R-1.5) on the weight to weight basis, respectively. The supplementation of rutin did not induce any significant difference on the final body weight, gain of body weight, the amount of feed intake and the feed efficiency of rats in both control and treatment groups. In addition the values of glucose concentration, total protein, albumin, globulin and albumin/globulin (A/G) ratio showed no significant differences among groups. The values of total cholesterol (TC) and low density lipoprotein-cholesterol (LDL-C) in sera of rats in both R-0.75 and R-1.5 groups were lower than those in C group but the significances were showed in only between R-0.75 and C group (p < 0.05 and p < 0.01, respectively). The values of high density lipoprotein-cholesterol (HDL-C) in sera of rats in both R-0.75 and R-1.5 groups were higher than those in C group but the significances were showed in only between R-1.5 and C group (p < 0.01). The values of atherogenic index(AI) of rats in both R-0.75 and R-1.5 groups were the lower than those in C group (p < 0.01 and p < 0.05, respectively). The values of triglyceride in sera of rats showed no significant differences among groups. The values of AST and ALT in sera of rats showed no significant differences among groups. Therefore the supplementation of rutin to high fat diet in rats reduced effectively the serum lipid levels such as TC and LDL-C which were regarded as to cause the cardiovascular diseases, and moreover it elevated effectively HDL-C value which was regarded to protect cardiovascular diseases.

A study on the epidemiology of caprine anaplasmosis in Korea III. Seasonal variation in hematologic profiles (산양의 anaplasmosis에 대한 역학적 조사 III. 혈액치의 계절적 변화)

  • Anaplasmosis is a tick-borne disease of large and small ruminants, causing losses through mortality, abortion, weight loss and reduced milk production. In one dairy farm, for example, 250 of a total of 800 imported goats were diagnosed with a mysterious type of anemia during the summer and autumn of 1992. The etiologic agent was identified as Anaplasma spp by acridine orange and ultrastructure by electron microscopy. In order to monitor variations in blood biochemical and hematological parameters associated with the disease, blood samples were collected by jugular venipuncture from 50 goats at 3 month intervals between the period of February and October, 1993. The levels of RBCs, HB and HCT decreased from $18.48{\pm}1.96$ to $13.47{\pm}2.48X10^6/mm^3$, $12.25{\pm}1.41$ to $9.54{\pm}1.77g/dl$, and $43.09{\pm}4.75$ to $30.93{\pm}5.78%$, respectively. The values of MCH(Mean corpuscular hemoglobin), MCHC(Mean corpuscular hemoglobin concentration) and PLT(Platelet) were elevated from $6.58{\pm}0.30$ to $7.05{\pm}0.47pg$, $28.40{\pm}1.20$ to $30.82{\pm}1.85g/dl$ and $1688.34{\pm}750$ to $2046.82{\pm}783X10^3/mm^3$, respectively. Percent parasitized erythrocytes(PPE) increased from $0.61{\pm}0.5$ to $2.22{\pm}1.9%$, clinical biochemical parameters aspartate aminotransferase and alanine aminotransferase were $66.64{\pm}23.1K.U$ and $14.90{\pm}6.59K.U$, respectively and persisted at high levels throughout the observation period. The level of albumin(2.46)0.52 g/dl) was decreased corresponding to an elevated globulin and a reduced albumin/globulin ratio in October as compared with the values in February. It is concluded that caprine anaplasmosis may be an important cause of anemia and hepatic malfunction in goats.

The Effect of Supplementary Feeding of Tannic acid on Growth and Hematological Changes in Rats Fed High Fat Diet (탄닌산(tannic acid) 첨가가 고지방 식이 흰쥐의 성장과 혈액학치 및 혈액화학치의 변화에 미치는 영향)

  • This study was performed to investigate the growth rate, hematological and serological changes of the rats when they were fed with the high fat diets supplemented with or without the tannic acid for five weeks. Thirty-two Sprague-Dawley male rats(235.7\pm10.7g\;of\;body\;weight)$ were randomly divided into four groups, control group and three treatment groups(T1, T2 and T3). Rats in control group were fed with the high fat diet containing $15\%\;lard,\;1\%$ cholesterol and $0.5\%$ sodium cholate(wt/wt) which was modified from the formula of American Institute of Nutrition (AIN)-76 diet and rats in treatment groups were fed with above diet supplemented with $0.25\%(T1),\;0.5\%(T2)$ or $0.75\%(T3)$ of tannic acid(wt/wt), respectively. The supplementation of tannic acid(TA) did not affect the final body weight, gain of body weight and feed intake of rats in both control and treatment groups. The numbers of red blood cells, hemoglobin concentrations and hematocrit values in blood of rats showed no significant differences between control group and treatment groups. The glucose concentration and albumin/globulin(A/G) ratio of rats in treatment groups were slightly lower than that of control group without significance. The values of total protein, albumin and globulin showed no significant differences between control group and treatment groups. The values of total cholesterol, low density lipoprotein­cholesterol and atherogenic index in sera of rats in treatment groups were much lower than that of control group without significance. The values of triglycerides in sera of rats in T3 group were significantly lower than that of control group (p<0.05). The values of AST and ALT in sera of rats in T3 group were significantly lower than that of control group (p<0.05). Thus supplementation of tannic acid to high fat diet could be effective to reduce the serum lipid levels such as total cholesterol, high density lipoprotein-cholesterol and triglycerides which were regarded as to cause the cardiovascular diseases.

Treatment of Damp-heat Type Liver Disease With Yugan-tang (임상관찰관우유간탕치료습열형간병(臨床觀察關于愈肝湯治療濕熱型肝病))

  • The aim of the present study is to determine clinical efficacy and adverse reaction of yugan-tang. We compared symptoms and liver function test results of 160 patients with damp-heat type liver disease in before and after taking yugan-tang. The patients took 200ml yugan-tang two times daily for three months. Then we analyzed these datas statistically. The overall therapeutic effect was "markedly improved" in 70 of 160 patients (43.75%) and "improved" in 75 of 160 patients (46.88%). yugan-tang markedly improved damp-heat type liver disease, especially jaundice, abdominal distention, hypochondriac pain, nause, vomiting and anorexia. Alanine aminotransferase (ALT) level and total bilirubin (TBIL) level were within normal limits at baseline. But albumin-globulin ratio (A/G) level was not changed markedly. I think that yugan-tang is worth applying widely to clinic.
