Synthesis and Structure of Tetrahomodioxa p-phenylcalix(4)arene dihexylether (Tetrahomodioxa p-phenylcalix(4) arene dihexylether의 합성 및 구조에 관한 연구)

  • Tetrahomodioxa p-phenylcalix(4)arene dihexylether(C/sub66/H/sub68/O/sub6/) has been synthesized and structurally characterized by X-ray diffraction. Reaction of tetrahomodioxa p-phenylcalix(4)arene with hexyl halide and NaH in DMF leads to the dihexyl derivatives, 7,13,21,27-tetraphenyl-29,31-dihexyloxy- 2,3,16,17-tetrahomo-3,17-dioxacalit(4)arenes. The crystal is orthorhombic, P2₁2₁2₁, a= 9.764(2), b=16.167(2), c=32.994(3) Å, V=5208(1) Å, Z= 4, Dc = 1.221 gcm/sup -3/. The structure was solved by direct methods and refined by full-matrix least squares. Refinement converged at R = 0.070 for 2009 observed reflections. This molecule has a C-1,2-alternate conformation with pseudo-centrosymmetry and has two pairs of opposite phenyl rings, which are approximately parallel to each other. The benzene rings A and B are up, and the rings C and D rings are down with respect to the plane of the macrocyclic ring.

The Anti-Microbial Activity of Modified Chitosan. (변형 키토산의 항균효과)

  • New type of chitosan derivatives, chitosan-g-MAP, were synthesized by graft copolymerization of mono (2-methacryloyl oxyethyl) acid phosphate (MAP) into chitosan, in order to solubilize chitosan in water. Ceric ammonium nitrate was used as an initiator for graft copolymerization. The optimal conditions for graft copolymerization were determined on the basis of reaction temperature, time, and the concentration of initiator and monomer. The reaction conditions for the highest percentage of grafting were as follows: an initiator concentration, 3.5${\times}$10$\^$-3/ M; monomer concentration, 0.19 M; and reaction temperature, 40$^{\circ}C$ The reaction rate reached the maximum value after 4 hrs of reaction. Antifungal activity was tested against Candida albicans, Trichophyton rubrum and Trichophyton violaceum by using chitosan-g-MAP and two other chitosan samples which have degree of deacetylation of 70% (DA-7) and 90% (DA-90). Their antifungal activities were investigated in weak acidic range. Maximum antifungal activity of them was observed at pH 5.75. Chitosan-g-MAP inhibited thoroughly the growth of Candida albicans and Trichophyton violaceum. Howerver, DA-70 and DA-90 showed higher antifungal activities on Trichophyton rubrum than that of chitosan-g-MAP.

Quantification of Vitamin $D_3$ and Its Derivatives in Marine Animal Foods by HPLC (HPLC에 의한 동물성 수산식품 중의 Vitamin $D_3$와 그 관련물질 정량에 관한 연구)

  • Contents of vitamin $D_3$ and 25-OH-vitamin $D_3$ in marine animal products(20 species) were determined by HPLC. The isomers of vitamin D, $D_2$ and $D_3$, were not clearly separated on a reversed phase, ${\mu}$ Bonda Pak, with 20% methanol-acetonitrile, and on a normal phase, Zorbax SIL. with 0.4% isopropanol-hexane, but 25-OH-vitamin $D_2$ and-$D_3$ were separated on either ${\mu}$ Bonda Pak with 10% methanol-acetonitrile, or on Zorbax SIL with 2.2% isopropanol-hexane, respectively. Although levels of vitamin $D_3$ and 25-OH-vitamin $D_3$ varied remarkably according to species, their average value(fish : $l,l87{sim}36,007$ I.U/sample 100g, mussel : $58{\sim}1,706$ I.U/sample 100g, pickle: $1,208{\sim}79,358$ I.U/sample 100g) was greatly higher than that of meat($80{\sim}100$ I.U/sample 100g) and dairy products($400{\sim}800$ I.U/sample 100g). Fatty tissues of fish and pickled fish intestines contained high level of vitamin $D_3$ and 25-OH-vitamin $D_3$, while the clam and mussel known to have various kinds of sterol including ${\Delta}^7$-sterol showed very low levels of vitamin $D_3$ and its derivative.

Perceptions held by Investigators, IRB Members and IRB Administrators on the Bioethical Oversight System of National R&D Projects (국가연구개발과제 생명의학윤리 감독체계에 대한 인식조사 및 제언)

  • Purpose: Aim of this study was to gather and evaluate perceptions of investigators, IRB members, and IRB administrators on the appropriateness of IRB review process and bioethical oversight system of national R&D (NR&D) projects. Method: Investigators, IRB members, and IRB administrators at 17 different institutions were surveyed using convenience sampling and survey questionnaires were partially group-specialized to consider any differences between the groups. Results: Participants included 29 investigators, 37 IRB members, and 17 administrators with response rate of 100% (83 of 83). According to the responses obtained, insufficient preparation time for constructing protocol and gaining IRB approval was one of the main problems in the IRB review process (investigator 79.3%, IRB administrator 88.2%). Also, discrepancy between NR&D and IRB's protocol formats was another major issue (IRB members 96.4%, IRB administrator 100%) and most investigators (89.7%) had to modify the original NR&D protocol to obtain IRB approval. Moreover, it was reported that 13.8% of investigators and 31.3% of IRB administrators did not submit midyear reports to IRB and for bioethical issues of NR&D projects, 17.2% of investigators did not include information on project status and safety issues in the annual reports. Conclusion: In conclusion, for successful and ethical completion of R&D projects, revision of both IRB review process and NR&D project protocol formats as well as implementation of appropriate bioethical oversights are necessary.

Study on Oil Production from Pyrolysis of Mixed Plastic Waste Using Multidimensional Chromatography (Multidimensional Chromatography/Mass Spectrometry를 이용한 혼합 폐플라스틱의 열분해 오일 특성 평가에 관한 연구)

  • The total hydrocarbon distribution of oil products obtained from the pyrolysis of four kinds of mixtures of polyethylene-polystyrene waste has been studied by multidimensional chromatography(high performance liquid chromatography followed by capillary gas chromatography)/mass spectrometry. Saturated, unsaturated and aromatic hydrocarbons in oil products were selectively pre-separated according to structural groups by HPLC and the weight fraction of each group was estimated by analysis of each component using GC-FID response factors. The hydrocarbon distribution of aliphatic fraction consists of $C_{5}$ to $C_{25}$ saturated and unsaturated hydrocarbons. And that of aromatics fraction consists of benzene, toluene, xylene, styrene, propenyl benzene, naphthalene, and some of derivatives. Pyrolysis temperature did not affect the ratio of total weight fraction of aliphatic over aromatic hydrocarbon distribution in case of PS only and PE-PS mixtures (1:1 and 1:4 wt. ratio) as a feed while affected the ratio of total wt. fraction in case of PE only. The optimal temperature for the maximum oil production was $600^{\circ}C$ for pyrolysis of PS and 1:1 and 1:4 mixtures of PE and PS. The optimal condition for aromatic recovery was $600^{\circ}C$ with 1:1 mixture of PE and PS. In this condition, aromatic was produced up to 90% of total oil product. The maximum yield of toluene, xylene, styrene, and propenyl benzene were 8.6, 8.9, 51.0 and 7.4% of feed for pyrolysis PS at $700^{\circ}C$, respectively. However, only 1.3% naphthalene was recovered at $700^{\circ}C$ with 1:1 PE:PS(by wt.).

Structure-Activity Relationships of 13- and 14-Membered Cyclic Partial Retro-Inverso Pentapeptides Related to Enkephalin

  • A series of 13- and 14-membered cyclic enkephalin analogs based on the moderately $\mu$ selective prototype compound Tyr-C[D-$A_2bu$-Gly-Phe-Leu] 8a were synthesized to investigate the structure-activity relationship. The modifications of sequence were mainly focused on two positions 3 and 5, critical for the selective recognition for $\mu$ and $\delta$ opioid receptors. The substitution of hydrophobic $Leu^5$ with hydrophilic $Asp^5$ derivatives led to Tyr-C[D-$A_2bu$-Gly-Phe-Asp(N-Me)] 7 and Tyr-C[D-Glu-Phe-gPhe-rAsp(O-Me)] 5, the peptides with a large affinity losses at both $\mu$ and $\delta$ receptors. The substitution of $Phe^3$ with $Gly^3$ led to Tyr-C[D-Glu-Gly-gPhe-rLeu] 3 and Tyr-C[D-Glu-Gly-gPhe-D-rLeu] 4, the peptides with large affinity losses at $\mu$ receptors, indicating the critical role of phenyl ring of $Phe^3$ for $\mu$ receptor affinities. One atom reduction of the ring size from 14-membered analogs Tyr-C[D-Glu-Phe-gPhe-(L and D)-rLeu] 6a, 6b to 13-membered analogs Tyr-C[D-Asp-Phe-gPhe-(L and D)-rLeu] 1, 2 reduced the affinity at both $\mu$ and $\delta$ receptors, but increased the potency in the nociceptive assay, indicating the ring constrain is attributed to high nociceptive potency of the analogs. For the influence of D- or L-chirality of $Leu^5$ on the receptor selectivity, regardless of chirality and ring size, all cyclic diastereomers displayed marked $\mu$ selectivity with low potencies at the $\delta$ receptor. The retro-inverso analogs display similar or more active at $\mu$ receptor, but less active at $\delta$ receptor than the parent analogs.

Study on the Binding of New Anti-ulcer Agent(IY-80843 and IY-80845) to Histamine $H_2$-Receptor in Isolated Guinea Pig Gastric Glands (새로운 항궤양제(IY-80843과 IY-80845)의 기니픽 위점막세포 $H_2$-수용체에 대한 결합반응연구)

  • Receptor binding study was carried out as an in vitro assay to test the anti-ulcer effect for newly synthesized test compounds(IY-80843 and IY-80845) which were reported to have a strong anti-secretory effect in Shay-ligated rats. The histamine H$_{2}$-receptor fraction was prepared from the membranes of the isolated gastric glands in guinea pigs and $^{3}$H-cimetidine was used as a radioligand. The binding of $^{3}$H-cimetidine to the membranes of the isolated gastric glands was found to be time dependent, saturable and confined to a single population of binding sites with $K_{D}$ value of 0.13$\pm$0.03 $\mu{M}$ and B$_{max}$ value of 52.5$\pm$1.5 pmol/mg. From the competition experiments, both IY-80843 and IY-80845 were shown to have a strong blocking effect against binding of $^{3}$H-cimetidine to the histamine H$_{2}$-receptor. The IC$_{50}$, Ki, and Hill coefficient(nH) values for IY-80843 were 0.18$\pm$0.02 $\mu{M}$, 0.16$\pm$0.02 $\mu{M}$, and 0.97$\pm$0.15, respectively and those values for IY-80845 were 0.27$\pm$0.02 $\mu{M}$, 0.24$\pm$0.02 $\mu{M}$, and 0.82$\pm$0.13, respectively. The results demonstrated that the blocking effects of IY-80843 and IY-80845 were 7 and 5 times stronger than that of cimetidine, respectively. Therefore, the newly synthesized compounds, IY-80843 and IY-80845, appeared to be the highly potent competitive inhibitors of histamine on the H$_{2}$-receptor.

In vitro Protective Effects of Glehnia Littoralis on Alpha-amanitin Induced Hepatotoxicity (알파 아마니틴에 의한 간독성에 대한 갯방풍의 보호 효과)

  • Purpose: Glehnia littoralis has been used to treat ischemic stroke, phlegm, cough, systemic paralysis, antipyretics and neuralgia. The pharmacological mechanisms of Glehnia littoralis include calcium channel block, coumarin derivatives, anticoagulation, anti-convulsive effect, as well as anti-oxidant and anti-inflammatory effects. Alpha-amanitin (${\alpha}$-amanitin) is a major toxin from extremely poisonous Amanita fungi. Oxidative stress, which may contribute to severe hepatotoxicity was induced by ${\alpha}$-amanitin. The aim of this study was to investigate whether Glehnia littoralis ethyl acetate extract (GLEA) has the protective antioxidant effects on ${\alpha}$-amanitin -induced hepatotoxicity. Methods: Human hepatoma cell line HepG2 cells were pretreated in the presence or absence of GLEA (50, 100 and $200{\mu}g/ml$) for 4 hours, then exposed to $60{\mu}mol/L$ of${\alpha}$-amanitin for an additional 4 hours. Cell viability was evaluated using the MTT method. AST, ALT, and LDH production in a culture medium and intracellular MDA, GSH, and SOD levels were determined. Results: GLEA (50, 100 and $200{\mu}g/ml$) significantly increased the relative cell viability by 7.11, 9.87, and 14.39%, respectively, and reduced the level of ALT by 10.39%, 34.27%, and 52.14%, AST by 9.89%, 15.16%, and 32.84%, as well as LDH by 15.86%, 22.98%, and 24.32% in culture medium, respectively. GLEA could also remarkably decrease the level of MDA and increase the content of GSH and SOD in the HepG2 cells. Conclusion: In the in vitro model, Glehnia littoralis was effective in limiting hepatic injury after ${\alpha}$-amanitin poisoning. Its antioxidant effect is attenuated by antidotal therapy.

Synthesis and Characterization of Magnetic Nanoparticles and Its Application in Lipase Immobilization

  • We demonstrate herein the synthesis and modification of magnetic nanoparticles and its use in the immobilization of the lipase. Magnetic $Fe_3O_4$ nanoparticles (MNPs) were prepared by simple co-precipitation method in aqueous medium and then subsequently modified with tetraethyl orthosilicate (TEOS) and 3-aminopropyl triethylenesilane (APTES). Silanization magnetic nanoparticles (SMNP) and amino magnetic nanomicrosphere (AMNP) were synthesized successfully. The morphology, structure, magnetic property and chemical composition of the synthetic MNP and its derivatives were characterized using transmission electron microscopy (TEM), Fourier transform infrared spectroscopy (FT-IR) analysis, X-ray diffraction, superconducting quantum interference device (SQUID) and thermogravimetric analyses (TGA). All of these three nanoparticles exhibited good crystallization performance, apparent superparamagnetism, and the saturation magnetization of MNP, SMNP, AMNP were 47.9 emu/g, 33.0 emu/g and 19.5 emu/g, respectively. The amino content was 5.66%. The AMNP was used to immobilize lipase, and the maximum adsorption capacity of the protein was 26.3 mg/g. The maximum maintained activity (88 percent) was achieved while the amount of immobilized lipase was 23.7 mg $g^{-1}$. Immobilization of enzyme on the magnetic nanoparticles can facilitate the isolation of reaction products from reaction mixture and thus lowers the cost of enzyme application.

Preliminary Study on the Crystal Structure of Cholesteryl Isopropyl Carbonate (Cholesteryl Isopropyl Carbonate의 결정구조에 관한 예비연구)

  • Cholesteryl isopropyl carbonate(C31O3H52) is orthorhombic, space group P212121, a=6.266(4), b=10.836(5), c=47.364(20)Å, Z=4, Dc=0.98 g/cm3 and Dm=1.01 g/cm3. The intensity data were collected on a Nonius CAD-4 diffractometer with a graphite-monochromatized MoKα radiation to a maximum 2θ value of 40°. The structure are solved by direct methods and refined by Fourier and full matrix least-squares methods. The present R factor was 0.22 for 1513 observed reflections. The further refinements are in progress. Compared with other cholesterol derivatives, the cholesteryl ring and tail region of the molecule are normal. The molecular long axes are parellel to the c-axis. There are close packings of cholesterol groups and loose packings of isopropyl carbonate chains forming monolayers.
