연자육의 6-하이드록시도파민으로 유도된 도파민 세포 독성에 대한 보호효과 (Protective Effects of Nelumbinis Semen Against Neurotoxicity fuduced by 6-Hydroxydopamine in Dopaminergic Cells)

  • Objectives : This study was performed to evaluate the neuroprotective effect of water extracts from Nelumbinis semen (NSW) in dopaminergic cells. Methods : We performed 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging assay, 2,2-azinobis3-ethyl-benzothiazoline-6-sulfonic acid (ABTS) cation scavenging assay, and determination of total polyphenolic content to examine the antioxidant effects of NSW. We also evaluated the neuroprotective effects against 6-hydroxydopamine (6-OHDA)-induced toxicity using 3-(4,5-dimethylthiazol-2-yl)- 2,5-diphenyl-tetrazolium bromide assay (MIT) assay, trypan blue cytotoxicity assay, and nitric oxide assay in SH-SY5Y cells and tyrosine hydroxylase (TH) immunohistochemistry in primary rat dopaminergic neurons. Results : NSW showed $IC_{50}$ values of 184.80 and 92.90 ${\mu}$g/mL in DPPH and in ABTS assays, respectively. NSW showed 1.05% of total polyphenol contents. NSW showed protective effect against 6-0HDA-induced neurotoxicity whereas no influence on cell viability at the concentration of 1${\sim}$50 ${\mu}$g/mL. NSW reduced NO generation while 6-OHDA produced it. Moreover, it protected rat dopaminergic neurons against 6-0HDA at a dose of 1 ${\mu}$g/mL. Conclusions : These results indicated that NSW has neuroprotective effect against 6-0HDA-induced neurotoxicity through antioxidant activity in dopaminergic cell culture.

Caramel 갈색화 반응 생성물의 항산화성에 관한 연구 (A study on the antioxidant activity of products of caramel-type-browning reaction)

  • 10$0^{\circ}C$ 이상에서 당단독, 당과 유기산 또는 유기산염의 caramel 갈색화 반응과 당과 아미노산의 Maillard 갈색 화 반응의 갈색화 물질의 항산화성을 측정하고자 0.1M 의 xylose(XY), glucose(GL), sucrose(SU)를 단독으로, 0.1M glucose에 0.02M의 citric acid를 혼합한 glucose-citric acid(GLCA), 0.02M의 sodium citrate를 혼합한 glucose-sodium citrate(GLSC)와 0.1M의 glycine을 혼합한 glucose-glycine(GLGC)을 80, 120 및 14$0^{\circ}C$에서 12, 24시간 가열하여 얻은 갈색화 반응 생성물의 수소공여능(HDA)에 의한 항산화성을 측정하였으며 이들의 무수 ethanol 추출물의 옥배유 기질에 대한 항산화 효과를 측정하여 기존 항산화제(TBHQ, TOCO)와 비교, 고찰한 결과는 다음과 같았다. 1. Caramel 갈색화 반응 생성물의 수소공여능(HDA)에 의한 환원력은 갈색화 반응온도 및 시간의 경과에 따라 대체적으로 증가되었으며 갈색도가 큰 반응계일수로 환원력이 크게 나타났다. XY, GL, SU, GLCA, GLSC 및 GLGC 갈색화 반응액의 수소공여능은 8$0^{\circ}C$에서 24시간 반응 후 각각 0.290, 0.299, 0.281, 0.301, 0.362 및 0,387로 그 크기는 GLGC > GLSC > GLCA > GL > XY > SU 순이었다. 12$0^{\circ}C$에서 24시간 반응 후의 GLCA와 GLGC에서 갈색도의 증가와는 달리 HDA는 오히려 낮아졌으며, 14$0^{\circ}C$ 에서는 대부분의 갈색화 반응액의 HDA는 온도가 상승함에 따라 높아졌지만 GLCA와 GLGC의 HDA는 낮아져서 각각 0.275 및 0.305로 된 반면에 GLSC의 HDA는 계속 증가하여 0.543으로 가장 높은 것으로 나타났다. 이 때의 HDA는 GLSC > SU > GL > GLGC > XY > GLCA 순이었다. 2. 갈색화 반응 생성물의 무수 ethanol 추출물을 첨가한 옥배유의 항산화 효과는 TBHQ에는 못미치나 TOCO 보다는 우수한 항산화력을 나타내는 것도 있었다. 8$0^{\circ}C$에서 24시간 진행된 갈색화 반응 생성물의 경우는 TBHQ > GLCA > GLGC > TOCO > SU > XY > GL > GLSC > control의 순으로 8$0^{\circ}C$에서는 갈색화 반응 중간생성물은 모두 항산화 효과를 나타내었으며 GLCA의 항산화 효과는 TBHQ 보다는 크게 떨어지나 GLGC, TOCO보다는 높은 항산화력을 보여주었다. 또한 120및 14$0^{\circ}C$에서 24시간 진행된 갈색화 반응생성물의 항산화 효과는 SU, GLCA에서만 보였으며 이들의 항산화 효과는 Maillard형 갈색화 반응액인 GLGC의 항산화 효과 보다 높았다. 그 크기는 TBHQ > SU > TOCO > GLCA > control > GLSC > GLGC > XY > GL의 순으로 80, 120및 14$0^{\circ}C$에서의 갈색화 반응 생성물의 항산화성은 8$0^{\circ}C$에서 가장 좋은 것으로 나타났다. 이상으로 볼 때 각종 갈색화 반응 생성물의 수소공여능에 의한 환원력과 항산화 효과의 관계는 환원력이 증가함에 따라 항산화 효과는 증가하지 않고 감소하여 갈색화 반응물의 항산화 효과와 환원력 사이에는 뚜렷한 상관관계가 없는 것 같았다.

카라멜화 반응 생성물의 갈색도와 항산화 효과와의 관계 (A study on the relations between the color intensity and the antioxidant activity of caramelization products)

  • The study was carried out to compare the relation between the color intensity and antioxidant activity of caramelization products using xylose(XY), glucose(GL). sucrose(SU), glucose+citric acid(GLCA), glucose+sodium citrate(GLSC), heated at 80, 120 and 140$\^{C}$ for 24hrs, respectively. The color intensity(absorbance at 490nm) of the browning mixtures increased as the browning temperature and time increased. But the degrees of color intensity of SU and GLCA changed very little. The hydrogen donating ability(HDA) of browning reaction products was generally enhanced as the browning temperature and time increased. When browning mixtures were heated at 80$\^{C}$, the HDA of GLGC was the highest, but the HDA of GLSC was the highest when heated at 120 and 140$\^{C}$. The antioxidant activities for the corn oil substrate containing the anhydrous ethanol extracts from the browning mixtures was inferior to that of SU, but was superior to that of GLCA. The relations among the color intensity, the antioxidant activity, and the hydrogen donating ability(HDA, reducing power) of the browning reaction mixtures were as follows: As the color intensity increased, the antioxidant activity decreased. The correlation coefficient of the color intensity and the antioxidant activity by regression equation was -0.73 ∼ -0.82. As the reducing power increased, the antioxidant activity decreased. The correlation coefficient between the reducing power and the antioxidant activity by regression equation was -0.98 ∼ -0.99. Therefore, the antioxidant activity of browning reaction mixtures seemed not correlated with the color intensity and the reducing power.

청혈단(淸血丹)의 6-hydroxydopamine에 의해 유발된 독성에 대한 신경세포보호효과 (The Protective Effect of Chunghyul-dan(Qingxuedan) Against 6-hydroxydopamine Induced Neurotoxicity.)

  • Objectives : This Study was performed to assess the antioxidant and neuroprotective effect of Chunghyul-dan(Qingxuedan) in PC12 cells and primary rat mesencephalic dopaminergic neurons. Methods : The anioxidant effect was investigated using the DPPH radical and ABTS cation scavenging assays and total polyphenol amout of Chunghyul-dan(Qingxuedan). The neuroprotective effect of Chunghyul-dan(Qingxuedan) in PC12 cells was evaluated using MTT assay. The scavenging activity of Chunghyul-dan(Qingxuedan) on ROS production induced by 6-OHDA(6-hydroxydopamine) in PC12 cells was evaluated, as well as the attenuating effect on GSH reduction. Finally, we examined the neuroprotective effect of Chunghyul-dan(Qingxuedan) against 6-0HDA-induced toxicity in the primary culture of rat mesencephalic doperminergic neurons. Results : Chunghyul-dan(Qingxuedan) showed concentration-dependent scavenging activities in DPPH radical and ABTS cation scavenging assays and it was not cytotoxic to PC12 cells. In postand co-treatment, Chunghyul-dan(Qingxuedan) protected PC12 cells from the 6-OHDA induced toxicity at 50 and 100 ${\mu}$g/mL significantly. And Chunghyu!-dan(Qingxuedan) decreased the 6-OHDA induced ROS production at a dose dependent manner, while increaing the 6-OHDA induced GSH reduction at 50 and 100 ${\mu}$g/mL significantly. Finally, Chunghyul-dan(Qingxuedan) showed signicant protection of rat mescencephalic dopaminergic neurons from 6-OHDA at 1 ${\mu}$g/mL. Conclusions : These results demonstrate that Chunghyul-dan(Qingxuedan) has the antioxidant and neuroprotective effect against 6-0HDA induced cytotoxicity through decreasing ROS production and increasing GSH reduction.

양파껍질에서 분리된 용매 추출물의 항산화효과 (Antioxidant Activity of Solvent Extract from Onion Skin)

  • 양파껍질중에 존재하는 항산화물질을 여러 용매로 분획하여 그 항산화효과를 비교하고, butanol fraction의 TLC에 의한 재분획 및 총페놀함량, 수소공여능 및 UV spectral 분석을 행하였다. 메타놀추출물의 항산화 효과는 첨가농도가 증가함에 따라 증가되었다. 메타놀추출물의 용매 분획물의 항산화효과는 buthanol> ethyl acetate> ethyl ether>water fraction의 순으로 나타났다. 이들의 항산화효과는 총페놀함량 및 수소공여능과 밀접한 관계를 보였다. TLC에 의한 butanol fraction의 재분획 결과 Rf치 0.20, 0.33, 0.49, 0.60 및 0.94을 나타내는 형광띠를 나타내었다. Rf치 0.94의 형광 band는 다른 형광 band에 비해 현저히 강한 것으로 보여 주었으며 flavonol의 존재를 나타내는 에서 나타나는 255 nm와 317 nm에서 강한 흡수가 보였다. Spectral analyses 결과 양파껍질 중에 함유되어 있는 주요 항산화성 물질은 quercetin aglycone (3,3',4',5,7-pentahydroxyflavone)인 것으로 확인되었다.

서울, 경기 일부지역(一部地域)의 단체급식소(團體給食所)에 대(對)한 영양실태조사(營養實態調査) (A Nutritional Survey on the Actual Condition of Group Feeding in Seoul City and Kyongki Province)

  • The purpose of this nutritional survey on the actual conditions of group feeding is to be of value for promoting the health of the blue collar workers through consice understanding of food intake trends in businesses as well as realizing our desirable target. The results of the survey are summarized as follow. 1, Status of food economics 1) The average feeding cost amounts to 453 won per day per person. 2) The average school years of food buyers were 12.3 years, cookers were 12.0 years and dieticians were 14.5 years. 3) Kerosene(57.6%), gas(24.2%), electric(12. 1%) and cool briguette(6.1%) were used for fuel. 2. Status of food intake The average food intake per person per day in surveyed businesses was 1193.4g. The total intake of food was consisted of 42% grains, 40.4% vegetables, 4.7% legumes, 4.5% fish and shellfish, 4.0% potatoes, 1.6% meats, 1.0% seasonings. 0.8% eggs, 0.5% sea weeds, 0.3% oil and fats, 0.1% sugar. These findings led us to the conclusion that workers in the surveyed businesses retied heavily on plant foods. 3. Status of nutrient intakes 1) The average intake of calorie was 2752.6 kcal per day, which was slightly higher than 2,700 kcal of RDA. Though the quantity was above the level of RDA, it was largely from plant foods. 2) The average intake of protein was 84.7g per day, which was slightly higher than 80g of RDA. 3) The average intake of fat was 26g per day, which was much lower than 36g of RDA. 4) The averse intake of calcium was 532.3mg per day, which was lower than 600mg of RDA. 5) The average iron intake was 9.8mg per day, which was much lower than 10mg of RDA and 18mg of HDA for women. 6) Intakes of vitamin group were high. Especially, intake of vitamin A was 2-3 times higher than RDA and intake of vitamin C and niacin were 2 times higher than RDA.

Effect of Acupuncture on 6-Hydroxydopamine-induced Nigrostriatal Dopaminergic Neuronal Cell Death in Rats

  • Objectives: Acupuncture treatment has been clinically used for functional recovery in Parkinson's disease. In the present study, we investigated the effect of acupuncture at Zusanli (ST36) on nigrostriatal dopaminergic neuronal cell death in rats. Methods: A Parkinson's disease model was induced by the unilateral injection of 6-hydroxydopamine (6-OHDA) into the striatum. Acupuncture treatment was performed at Zusanli (ST36) and at the hip, as a non-acupoint, once a day for 14 days. Two weeks after 6-0HDA injection, an apomorphine-induced rotational behavior test showed significant rotational asymmetry in rats with Parkinson's disease. Immunostaining for tyrosine hydroxylase demonstrated a dopaminergic neuronal loss in the substantia nigra and dopaminergic fiber loss in the striatum. Results: Acupuncture at the ST36 acupoint significantly inhibited rotational asymmetry in rats with Parkinson's disease, and also protected against 6-OHDA-induced nigrostriatal dopaminergic neuronal loss. These effects of acupuncture were not observed for non-acupoint acupuncture. Conclusions: The present study shows that acupuncture treatment, especially at the ST36 acupoint, can be used as a useful strategy for the treatment of Parkinson's disease.

Quantitation of Plasma Apolipoprotein A-I with a Sandwich Type Enzyme-Linked Immunosorbent Assay Using Monoclonal Antibodies

  • A sandwich-type enzyme-linked immunosorbent assay (ELISA) for the quantification of human apolipoprotein A-I (apoA-I) was developed using monoclonal antibodies. For this assay, we used three monoclonal antibodies to trap and detect apo A-I. HDAI16 and HDA15 monoclonal antibodies were used for trapping apoA-I and HDAI8 monoclonal antibody was for detecting apoA-I. These three monoclonal antibodies were produced by immunizing mice with high density lipoprotein (HDL) isolated from human plasma. By immunoblot analysis, these three monoclonal antibodies were specific to apoA-I and showed no cross-reactivities with other plasma proteins. The results of competition assays for epitope cross-reactivity test also verified that these monoclonal antibodies identified separate and distinct epitopes on HDL and apoA-I. Affinity constants of monoclonal antibodies were measured by ELISA. Their association constants ranged from $10^7$ to $10^8$ $M^{-1}$. For this assay, pure apoA-I was isolated by affinity chromatography using monoclonal antibodies. In this sandwich assay, the amount of HRP-labeled HDAI8 bound to apoA-I trapped by HDAI16 and HDAI5 was proportional to apoA-I concentration in the range of 0 to 500ng/ml. ApoA-I concentration in plasma was calculated from the linear regression equation of standard curve. The precision and reliability of the assays are reflected in the low intra-and interassay coefficients of variation that averaged 3.25% and 4.30%, respectively. This assay is sensitive, simple, reproducible, convenient in incubation interval, and does not use radioisotope: thus it can be widely applied in clinical laboratories.

Yaw방향의 보정이 가능한 Tomotherapy couch device의 개발 (Development of Tomotherapy couch device capable of yaw-directional correction)

  • 목 적 : "Yaw방향의 보정이 가능한 Tomotherapy couch device"를 자체 제작하였고, 그 유용성을 평가하였다. 대상 및 방법 : 휨 강도가 $200kg/cm^2$의 경질 섬유판 재질로 Yaw방향의 회전 가능한 Tomotherapy couch device를 제작하고, 제작된 device의 물리적 정확성을 판단하기 위해 Varian사의 Novalis Tx의 CBCT Image와 MED-TEC사의 Iso-Align Phantom을 사용하여 device의 Yaw방향의 보정각도의 정확성을 측정하였고, Tomotherapy HDA에서의 치료를 위해 치료계획은 Accuray PrecisionTM를 이용하여 In House Head and Phantom 위에 치료계획을 설계하였고 제작한 device의 보정각도마다 치료 전 후의 MVCT의 adaptive plan을 Accuray PrecisionARTTM and PrecisionTM을 사용하여 선량을 평가하였다. 결 과 : Yaw방향의 보정이 가능한 Tomotherapy couch device를 자체 제작하였으며, 물리적 정확성의 측면에서 모든 각도별 vertical, lateral, longitudinal의 보정값은 약 1 mm 내외였고, 최대 보정값이 1.5 mm를 넘지 않았으며 Yaw방향의 보정은 최대 0.12도 오차 이내의 범위 안에 들어왔다. 치료후의 MVCT촬영 영상을 이용한 adaptive plan을 이용한 선량 평가에서도 모든 각도실험에서 target은 D95=Prescription dose를 만족하고, target내 선량이 95 %~107 % 이내에 들어왔으며, OAR 또한 본원에서 평가한 SMC Head and Neck tolerance dose 영역 내에 모두 들어왔다. 결 론 : 자체 제작한 "Yaw방향의 보정이 가능한 Tomotherapy couch device는 Tomotherapy의 치료에서 Yaw방향의 set-up 오차를 정확히 보정해준다. 비용적인 측면에서도 효과대비 저비용으로 장치를 만들 수 있을 뿐만 아니라 재촬영에 대한 환자가 받는 MVCT image dose를 방지하고 시간적으로도 환자의 회전율을 상승시킬 수 있으며, Yaw방향의 set up 오차를 보정하므로 더 정확한 방사선치료를 시행할 수 있다고 판단된다.
