Characterization and Functional Study of PyrR Orthologues from Genome Sequences of Bacteria (세균 게놈 유래성 PyrR Orthologue의 기능 분석)

  • The regulation of pyrimidine nucleotide synthesis has been proved to be controlled by a regulatory protein PyrR-mediated attenuation in the Gram-positive bacteria. After several bacterial genome sequencing projects, we have discovered the PyrR orthologues in the databases for Haemophilus influenzae and Synechocystis and sp. PCC6803 genome sequences. To investigate whether these PyrR orthologue proteins regulate pyrimidine nucleotide synthesis as well as the cases of Bacillus, the PyrR regions of each strains were amplified by PCR and cloned with pUC19 or T-vector in Escherichia coli and with a shuttle vector pHPS9 for E. coli and B. subtilis. For the regulation test of the PyrR orthologues, the aspartate-transcarbamylase (ATCase) assay was carried out. From the results of the ATCase assay, it was confirmed that Synechocystis sp. PCC6803 could not restore by pyrimidines to a B. subtilis, PyrR but H. influenzae PyrR could. For Purification of PyrR orthologue proteins, PyrR orthologue genes were cloned into the expression vector (pET14b). Over-expressed product of PyrR orthologue genes was purified and analyzed by the SDS-PACE. The purified PyrR orthologue proteins from H. influenzae and Synechocystis sp. PCC6803 turned out to be molecular mass of 18 kDa and 21 kDa, respectively. The result of uracil phosphoribosyl transferase (UPRTase) assay with purified PyrR orthologue proteins showed that H. influenzae PyrR protein only has UPRTase activity. In addition, we could predict several regulatory mechanisms that PyrR orthologue proteins regulate pyrimidine de novo synthesis in bacteria, through phylogenetic analysis for PyrR orthologue protein sequences.

  • Six-coordinate molybdenum(Ⅴ)-oxo complexes, (R4N)[MoO(NCS)2L](R=CH3, C2H5, n-C4, H9) with S-methyl-3-(2-hydroxy-x-phenyl)methylenedithiocarbazate(L1: x=5-H) and its derivatives (L2:x=5-CH3, L3: x=3-CH3O, L4: x=5,6-C4H4 and L5: x=5-NO2) have been synthesized and the structural, spectral and electrochemical properties of the complexes have been characterized by elemental analysis, molar conductivity, UV-Vis, IR, 1H NMR, and CV (cyclic voltammetry).

Food Quality Characterization and Safety of Imported Fish Roe (Japanese flyingfish roe, Capelin roe and Pacific herring roe) (수입 어란(날치 알, 열빙어 알 및 청어 알)의 품질 특성과 안전성)

  • The food quality characterization and safety Japanese flying fish roe (JFF-R), capelin roe (C-R), Pacific herring roe (PH-R) were investigated. The size of JFF-R was 1.48-1.93 mm, which was longer than those of C-R and PH-R. The moisture content of JFF-R imported from Peru (JFF-R-P) was 70.3%, which was higher than that of JFF-R imported from China (JFF-R-C) (67.4%), while was lower than that of JFF-R imported from Indonesia (JFF-R-I) (83.3%). However, the salinity of JFF-R-P was 13.6%, which was lower than that of JFF-R-C, while was higher that of JFF-R-I (1.8%). The moisture contents and salinities of the other fish roes were 80.4% and 3.2%, respectively, for capalin roe, and 65.4% and 20.0%, respectively, for Pacific herring roe. According to the results of pH, volatile basic nitrogen content, heavy metal content, viable cell count and coliform group, the fish roes could be used as sanitary sources for preparation of seasoned JFF-R. The major fatty acids of fish roes were 16:0 (27.8-30.5%), 18:1n-9 (7.2-8.0%), 20:5n-3 (5.6-8.2%) and 22:6n-3 (22.0-25.6%) in JFF-R, and 16:1n-7 (6.7-9.3%) as well as these fatty acids in C-R and PH-R. Total amino acid contents of fish roes ranged from 9.44 g/100 g to 10.39 g/100 g and their major amino acids were aspartic acid, glutamic acid, leucine and lysine. The mineral content of JFF-R were higher than those of the other fish roes expected for zinc of JFF-R-I. According to the results of sensory evaluation, the color and texture of JFF-R-P were superior to those of the other fish roes. No difference was, however, found in flavor, among JFF-R-P and the other fish roes.

  • This study, examines the performance and the heat pump cycle systematizing characteristics for non-azeotropic refrigerant systems. In order to conduct such an examination, the cycle characteristics of heat pumps for pure R-22, R-114, and their mixtures were experimentally investigated. The results show that cooling/heating capacities for the mixtures was more suited at the evaporating temperature of $5^{\circ}C$ than that of $0^{\circ}C$, $-5^{\circ}C$, and $-10^{\circ}C$. The C.O.P of the 50 wt% mixtures was considerably higher than for pure R-22, and the compression power of the 25 wt% was as much as 60% lower than that of pure R-22. Even small fractional mixture variations can lead to significant changes in the characteristics of the heat pump cycle. This experiment verified the importance of accurate weight fractions of refrigerant mixtures.

  • In this paper, a class of more general delayed viral infection model with lytic immune response is proposed by Song et al.[1] ([Journal of Mathematical Analysis Application 373 (2011), 345-355). We derive the basic reproduction numbers $R_0$ and $R_0^*$ 0 for the viral infection, and establish that the global dynamics are completely determined by the values of $R_0$ and $R_0^*$. If $R_0{\leq}1$, the viral-free equilibrium $E_0$ is globally asymptotically stable; if $R_0^*{\leq}1$ < $R_0$, the immune-free equilibrium $E_1$ is globally asymptotically stable; if $R_0^*$ > 1, the chronic-infection equilibrium $E_2$ is globally asymptotically stable by using the method of Lyapunov function.

Let n be a fixed positive integer, R be a noncommutative 2n!-torsion free prime ring with the center $C_R$ and d, g be a pair of derivations on R. If < $d^2(x)+g(x)$, $x^n$ > $\in$ $C_R$ for all x $\in$ R, then d = g = 0. Then we apply these purely algebraic techniques to obtain several range inclusion results of pair of (generalized-)derivations on a Banach algebra.

Performance of Alternative Refrigerants in Low Temperature Chillers

  • This paper is about the performance of alternative refrigerants for low temperature and transport refrigeration chillers. A breadboard refrigeration chiller was constructed with counterflow heat exchangers. R502, its transitional alternatives of R402A and R402B, and long-term alternatives of R404A and R507 were tested in an attempt to compare the performance of each refrigerant against R502. Measurements were conducted at two condensing temperatures of 43.3$^{\circ}C$ and 52.$0^{\circ}C$ and the evaporating temperature was varied over a range from -$25^{\circ}C$ to -5$^{\circ}C$. The evaporator superheat and condenser subcooling were maintained constant at about 5$^{\circ}C$ for all tests. Test results showed that all alternative fluids tested in this work can be used as 'drop-in fluids' to replace R502 without any major problem. It is also found that in the long run HFC alternatives are to be used due to their favorable environmental characteristics and better performance.

Evaporation Heat Transfer Characteristics of R-22, R-134a in Small Diameter Tubes (세관내 R-22, R-134a의 증발 전열 특성에 관한 연구)

  • Evaporating heat transfer coefficients of R-22 and R-134a were measured in smooth horizontal copper tubes with inner diameters of 1.77, 3.36 and 5.35mm, respectively. The experiments were conducted in a closed loop, which was driven by a magnetic gear pump. Experiments were performed for the following range of variables: mass velocity (200 to 400 kg/$m^2$.s), saturation temperature($0^{circ}C,; 5^{\circ}C$) and quality(0 to 1.0). Main results obtained are as follows: evaporating heat transfer coefficients in the small diameter tubes (ID<7mm) were observed to be strongly affected by various diameters and to differ from those in the large diameter tubers. The heat transfer coefficients of the small diameter tubes were higher than those of the large diameter tubs. And it was very difficult to apply some well-known previous predictions (Shah`s, Gungor-Winterton`s and Kandlikar`s correlation) to small diameter tubes.
