Field Spread of Soy bean Mosaic Virus Strains (콩모자익바이러스 계통의 포장전염)

  • Isolates of soybean mosaic virus (SMV) strains classified based on virulence in silt resistant soybean cultivars caused the same reactions in soybean cultivars used as differentials as those obtained by sap inoculations to the same cultivars. Five species of aphids (Myzus persicae SULZ., Aphis craccivora KOCH, Aphis citricola VAN., Rhopalosiphum maidis FIT., End R. padi L.) were able to transmit each of SMV strains. However, R. maidis and R. padi were inefficient vectors for transmission of SMV strain G3. Spread if four SMV strains (G2, G3, G6, and G7) was monitored in the field from sapinoculated plants in a one meter row of Williams soybeans (source plants) to plants in an adjacent row of Williams 80cm away (test plants). Test plants wert downwind from the source plants. A complete block design was used. Spread of strain G6 was significantly greater than that of other three strains. Two hundred six aphids were collected from June 27, 1979 to August 2, 1979 in the same field. A. citricola was the mist prevalent, comprising $68\%$ of the total aphids. Yields of Williams inoculated with each strain were also compared. Yields were the least from plants inoculated with strain G2 following G6, G3, and G7 in that order.

Variations in Growth Characteristics and Seed Qualities of Korean Soybean Landraces (한국 재래종 콩의 수집지대별 생태적 특성 및 품질 차이)

  • This study was carried out to examine agronomic characteristics and seed qualities of 827 Korean soybean landraces according to the collecting areas. Agronomic characteristics and seed qualities was examined according to the localities by weather characteristics. Days to flowering was the longest (mean 70.9 days) in the northwest seashore, and the most short in the south inland (mean 63.8 days). Growth days was the longest in the Gyeongbuk inland (mean 136.0 days), and the most short in the mountain-high (mean 132.8 days). The stem height was the most short in the north-west seashore (mean 81.3 cm), and in the south seashore (mean 58.8 cm). The seed weight of 100 seeds was the lightest in the north-west seashore (mean 17.1 g), and the heaviest in the south-west seashore (mean 21.5 g). The crude protein contents was the lowest in the north-west seashore (mean 40.1%), the highest in the south seashore (mean 42.8%), and vice versa in the crude oil. The contents of sucrose and oligo-saccharide was the highest 5.8% and 8.6%, respectively, in the mountain-high, and the lowest 4.8% and 7.6% in the Gyeongbuk inland and south seashore, respectively. Among the iso-flavone contests, the contents of genistein and total was the highest, $635.6\;{\mu}g/g$ and $1258.1\;{\mu}g/g$, respectively, in the mountain inland, and the lowest $319.2\;{\mu}g/g$ and $835.3\;{\mu}g/g$ in the south-west seashore. The contents of glycitein was the highest $124.3\;{\mu}g/g$ in the south seashore.

Adsorption Characteristics of Commercial Wood Charcoal in Korea (I) (국내 시판용 목탄의 흡착 특성(I))

  • To evaluate the basic characteristics and adsorption properties of commercial wood charcoal, we investigated the proximate analysis, porosimetry analysis, methylene blue adsorption, removal ratios of formaldehyde, and removal ratio of ethylene gas. Fixed carbon contents of traditional black and white charcoal, and mechanical charcoal were 51.8~76.6%, 72.9~84.6%, and 48.5~80.3%, respectively. Refining degrees of the most traditional black charcoal were 9, and those of white charcoal and mechanical charcoals were zero. Specific surface area of traditional black charcoal was 0.1~13.7 $m^2/g$, which was quite lower than that of white charcoal (53.2~372.6 $m^2/g$) and mechanical charcoals (224.3~464.6 $m^2/g$). Also, amounts of methylene blue adsorption were quite lower in black charcoal (0.53~1.97 mg/g) compared with white charcoal (2.68~7.68 mg/g) and mechanical charcoal (11.63~26.10 mg/g). Removal ratios of formaldehyde of the black charcoal were 11.4~26.7%, which is quite similar to white charcoal (17.9~34.9%) and mechanical charcoal (5.5~25.8%). Removal ratios of ethylene gas for traditional black charcoal, traditional white charcoal, and mechanical charcoal were 2.2~43.5%, 21.7~39.1%, 21.7~39.1%, respectively. There was no significant difference in the removal ratios of formaldehyde and ethylene gas among traditional black charcoal, traditional white charcoal, and mechanical charcoal.

Distribution of Human Rotavirus Genotypes in a Tertiary Hospital, Seoul, Korea During 2009-2013 (2009년부터 2013년까지 서울의 일개 대학병원에서 동정된 로타바이러스 유전형의 분포)

  • Purpose: Group A rotavirus (RV) is most common etiologic agent of acute gastroenteritis (AGE) in children worldwide. Recently, vaccination has been introduced in several countries to reduce the disease burden caused by RV infections, but continuous surveillance of RV strains is necessary to detect the emergence of potential variants induced by vaccine-immune pressure. This study aimed to investigate the changing pattern of RV genotypes in children with AGE, following the introduction of vaccination in Korea. Methods: Genotyping of RVs by RT-PCR on the basis of VP7 and VP4 gene segment sequence was carried out on 201 rotavirus-positive stool samples, from children hospitalized with AGE between August 2009 and June 2013. We have directly sequenced PCR products and analyzed the phylogenetic tree. Results: The most prevalent G genotype was G9 (33.3%), followed by G1 (22.4%), G3 (15.9%), G2 (6.0%), G4 (3.0%), G10 (1.5%), and mixed G-type (15.4%), with some nontypeable cases (2.5%). The detected P genotypes were P[4] (45.3%), P[8] (43.8%), mixed P-type (10.4 %), and P[2] (0.5%). The G9P[4] genotype was predominantly observed in hospitalized cases in Seoul in 2010/2011, however G1P[8] has been re-emerged as the predominant genotype in the following season (P =0.004). Conclusions: It seems that the periodic fluctuation in predominance of the G1, G3, and G9 strains occurred in Korea during 2009-2013, following the introduction of RV vaccination.

Oral Concentrated Grape Juice Suppresses Expression of NF-kappa B, TNF-α and iNOS in Experimentally Induced Colorectal Carcinogenesis in Wistar Rats

  • The aim of this study was to evaluate the effects of grape juice on colon carcinogenesis induced by azoxymethane (AOM) and expression of NF-kB, iNOS and TNF-${\alpha}$. Methods: Forty male Wistar rats were divided into 7 groups: G1, control; G2, 15 mg/kg AOM; G3, 1% grape juice 2 weeks before AOM; G4, 2% grape juice 2 weeks before AOM; G5, 1% grape juice 4 weeks after AOM; G6, 2% grape juice 4 weeks after AOM; G7, 2% grape juice without AOM. Histological changes and aberrant crypt foci (ACF) were studied, while RNA expression of NF-kB, TNF- and iNOS was evaluated by qPCR. Results: The number of ACF was higher in G2, and G4 presented a smaller number of crypts per focus than G5 (p=0.009) and G6. Small ACF (1-3) were more frequent in G4 compared to G2, G5 and G6 (p=0.009, p=0.009 and p=0.041, respectively). RNA expression of NF-kB was lower in G3 and G4 compared to G2 (p=0.004 and p=0.002, respectively). A positive correlation was observed between TNF-${\alpha}$ and NF-kB gene expression (p=0.002). In conclusion, the administration of 2% grape juice before AOM reduced the crypt multiplicity, attenuating carcinogenesis. Lower expression of NF-kB was observed in animals exposed to grape juice for a longer period of time, regardless of concentration.

Efficacy of antibacterial treatments of fresh ginseng (Panax ginseng C.A. Meyer) (항균제 처리에 따른 수삼의 미생물 저감화 효과)

  • Fresh, washed ginseng can be contaminated with microorganism loads as high as 6.5 log CFU/g for total bacteria and 4.3 log CFU/g for mold. The goal of this study was to test eight antibacterial agents on ginseng. Immersing fresh ginseng washed in 1% (w/w) sodium citrate, sodium diacetate, sodium acetate, citric acid, and sodium lactate solution for 1 h resulted in a bactericidal effect of 31.0-97.5% for total bacteria. Among the organic acids, sodium citrate had the best antibacterial effect, with total bacteria reduced from 6.5 log to 4.9 log CFU/g. A 1% (w/w) vitamin B1 lauryl sulfate solution with surfactant function by hydrophilic and hydrophobic sites can reduce 2.7 log CFU/g (99.8% inactivation) on total bacteria. In the 1% (w/w) calcium oxide solution, total bacteria were reduced by 3 log, showing an excellent inactivation effect of 99.9%. Calcium oxide is a highly useful material for inactivation of microorganisms in fresh ginseng.

Optimization of biomass production of Acetobacter pasteurianus SRCM101388 (Acetobacter pasteurianus SRCM101388 바이오매스 생산 최적화)

  • In this study, culture conditions were optimized to confirm the feasibility of Acetobacter pasteurianus as a starter for fermentation vinegar. Acetobacter pasteurianus strain can be used as a food ingredient. The optimal temperature and pH conditions of the selected Acetobacter pasteurianus SRCM101388 were 28℃ and pH 6.00, respectively. The response surface methodology (RSM) was used to optimize the composition of the medium, and Plackett-Burman design (PBD) was used to obtain the effective selection of culture medium, resulting in that glucose, sucrose, and yeast extract had the highest effect on increasing biomass. The optimal concentration, which was performed by central composite design (CCD), were determined to be 10.73 g/L of glucose, 3.98 g/L of sucrose, and 18.73 g/L of yeast extract, respectively. The optimal concentrations of trace elements for the production of biomass were found to be 1 g/L of ammonium sulfate, 0.5 g/L of magnesium sulfate, 2 g/L of sodium phosphate monobasic, 2 g/L of sodium phosphate dibasic, and the final optimized medium was pH 6.10. When incubated in a 5 L jar fermenter, the SRCM101388 strain showed a faster-dissolved oxygen (DO) reduction at a lower agitation rate (rpm), and it was able to grow even at reduced DO level when aeration was maintained. The amount of final biomass produced was 2.53±0.12×109 CFU/mL (9.40±0.02 log CFU/mL) when incubated for 18 hours at 150 rpm, 0.5 vvm, pH 6.0, and 28℃.

Effect of xylooligosaccharide-sugar mixture on defecation frequency and symptoms in young women with constipation: a randomized, double-blind, placebo-controlled trial (자일로올리고당을 함유한 설탕이 20대 여성의 변비 개선에 미치는 효과: 이중맹검 연구)

  • Purpose: To investigate the effects of the intake of xylooligosaccharide-sugar mixture (XOS) on defecation frequency and symptoms in 56 young women (mean age of 22.1 years old) with constipation. Methods: Two experiments were conducted. In experiment 1, a randomized double-blind study was performed to evaluate the effect of 6 weeks' intake of 10 g sucrose containing 7% xylooligosaccharide or 10 g sucrose on constipation. In experiment 2, 24 g coffee mixture containing 12.8 g plant cream and 11.2 g xylooligosaccharide-sugar mixture was consumed by the subjects. During the study, the clinical efficacy was assessed by using a daily diary. The subjects indicated the number of frequencies they defecated in a day and the clinical symptom scores. Results: In experiment 1, the mean frequency of defecations was 2.07 in the pretreatment week and increased significantly to 4.05, 4.42, 4.84, 4.84, and 4.05 in weeks 2 to 6 of XOS intake, in comparison with the 3-3.67 with sucrose intake (sucrose, SUC). In experiment 2, the mean frequency of defecations significantly increased from 2.47 in the pretreatment week to 4.11-5.67 in weeks 1-6 of XOS intake. The occurrence of very loose or loose stools in the XOS group was significantly increased in weeks 5 and 6, compared with the pretreatment week and SUC group. XOS intake significantly alleviated the abdominal displeasure and feeling of residual stool leftness in weeks 2, 3, 5, and 6, while SUC did so in weeks 4 and 6 (p < 0.05). The coffee mixture containing xylooligosaccharide-sugar mixture reduced the abdominal displeasure and feeling of residual stool leftness from week 3 until the end of the experiment (p < 0.05). Conclusion: Our results suggest that xylooligosaccharide-sugar mixture intake was effective, without adverse effects, for the alleviation of constipation in the young women in this study.

Changes of Phytic acid and Minerals by Heat Treatment in Korean Soybeans (한국산 대두의 열처리에 의한 피트산과 무기성분의 함량변화)

  • In this study, effect of heat treatment on phytic acid, phosphorus compounds, and minerals in Korean soybean varieties was investigated. Results were summarized as follwo: 1. In the soybeans tested, protein content ranged from 34.6 to 44.6%, lipid content from 15.4 to 20.2%, fiber content from 4.8 to 6.1% and ash content from 4.5 to 5.9%. 2. Content range for phytic acid in soybean varieties was 1,300 to 1,542mg/100g and its mean was measured 1,392mg/100g. With increasing of the temperature, the phytic acid tends to be destroyed, especially at 6$0^{\circ}C$ the loss was averaged about 20%. 3. Total phosphorus content in soybean ranged from 607 to 681mg/100g and the decending order of phosphors content in soybean varieties was Millyang > Hwangkeum > Kwangkyo > Danyoup > Hill > Jangyoup. It was also destroyed with increasing temperature. 4. Phytate phosphorus content range in soybean was from 315.6 to 318.0mg/100g and decreased with increasing temperature. 5. Inorganic phosphorus content ranged from 95.5 to 110.0mg/100g and it was increased by temperature rising among soybean varieties. 6. Phytate phosphorus to total phosphorus ratio ranged from 5.2 to 5.7 and decreased by temperature rising. 7. The content of nonphytate phosphorus varied widely between soybean varieties and decreased with temperature increase. 8. The content of calcium, iron and magnesium ranged from 15.7 to 25.7mg/100g from 8.8 to 16.8mg/100g and from 121.9 to 143.6mg/100g respectively. The content of Mg showed small difference among soybean varieties. The change of mineral content with heat treatment in soybean did not give any meaningful change mineral content.

A Study on the Actual Condition for Portion Control of Meal Served by Elementary School Foodservice Operation (초등학교 급식의 적정분량에 관한 연구)

  • This study was focused on setting up a proper portion by elementary school foodservice operation. The results were summarized as follows : Individual average consumption size and consumption rate from elementary school foodservice : cooked rice 238g(97.84%), soup 213g(93.8%), pot stew 277g(91.2%), stir fry 68g(83.9%), stew 67g(81.3%), fresh salad and seasoned vegetable 54g(81.0%), kimchi 49g(95.6%), one course dish 429g(96.3%). Proper portion of meal for the elementary school foodservice based on a statistical data was as follows : cooked rice 230g, soup 205g, pot stew 262g, stir fry 40∼55g, stew 70g, fresh salad and seasoned vegetable 45∼50g, kimchi 50g, one course dish 360∼400g.
