Development and Prey Consumption of Phytoseiid Mites, Amblyseius womersleyi, A. fallacis, and Typhlodromus occidentalis under controlled Environments (점박이응애 천적인 3종 이리응애의 발육 및 포식량 비교)

  • Comparative studies on some ecological characteristics of three phytoseiid mitespecies (one native; Arnblyseius womersleyi Schica, and two introduced species; A. fallacis Garmanand Typhlodromus occidentalis Nesbit) were carried out. The two-spotted spider mite (Tetranychusurticae Koch) was supplied as prey. Under four constant temperatures of 20, 23, 25 and 30f OS$^{\circ}$C,duration of growth from egg to adult for A. womer.vleyi was 11.5, 7.7, 6.7 and 5.6 days. While twoother species needed slightly shorter time but not significantly different. Critical temperature andeffective degree-days (DD) of A. womerslevi females were 83$^{\circ}$C and 1 1 1.6 DD, whereas those of A..fallacis were 10.7"C and 86.0 DD, and those of 7: occidentalis were 10.7"C and 94.1 DD. Also, thoseof males were similar to their females. Average longevity of females of A. womersleyi, A. fallucisand 7: occidentalis were 18.2 k 8.67, 19.6 3~7.18 and 13.0f5.66 days, total fecundity were 34.3 $-11.93, 39.8k 12.64 and 23.6k8.86, respectively. Under four constant temperatures of 20, 23, 25 and30-t0.S$^{\circ}$C, A. womersleyi consumed 9.1 f2.49, 9.7 k2.00, 9.7 f 2.61, and 10.3 k2.33 eggs of 7:urticae throughout their development. A. ,fizlluc~i.sc onsumed 10.2 k 2.52, 9.7 f2.29, 10.7 f 2.37 and10.1 k2.62 eggs, while, 7: occidentalis consumed 1 1.9 k3.43, 14.2 f4.50, 14.8 k 3.2 1 and 12.7 f2.95 eggs, respectively. Gravid females of A. womersleyi, A. f~zllacis and 7: occidentalis consumed11.4f1.59, 12.5k1.43 and 11.7k3.07 eggs, or consumed 11.9f 2.63, 12.4k3.82, and 12.5f 3.73protonymphs of 7: urticae in a day at 25-30$^{\circ}$C.e in a day at 25-30$^{\circ}$C.

Effects of Immersion Temperatures and Times on Chestnut Fruit and Mortality of the Chestnut Weevil, Curculio sikkimensis Heller (밤바구미 방제를 위한 온도 및 시간별 침지처리 효과와 종실 변화)

  • To control populations of the chestnut weevil, Curculio sikkimensis Heller, the effects of various immersion temperatures and times on the chestnut weevil were tested. The effects of immersion treatments on chestnut fruits were also analyzed. The mortality levels of C. sikkimensis larvae were evaluated at various temperatures ($30^{\circ}C$, $35^{\circ}C$, $40^{\circ}C$, $45^{\circ}C$, and $50^{\circ}C$) and time intervals (1, 2, 3, 5, 7, 10, 15, 22, and 24 h). Changes in the chestnut fruit due to the immersion treatment were measured in terms of color (lightness, redness, and yellowness), physiological effects (germination rate and decay rate), hardness, and change in constituent properties (moisture content, free sugar content, and tannin content). Mortality levels of C. sikkimensis larvae after immersion treatment at $30^{\circ}C$ were over 70% and 100% after 2 h and 7 h, respectively. Color (lightness, redness, and yellowness) of the chestnut fruits was not significantly different at $30^{\circ}C$ and $35^{\circ}C;$ however, the color was affected at temperatures over $40^{\circ}C$. The germination rate of the fruit was not affected by the immersion treatments, but the decay rate at $40^{\circ}C$ increased with an increase in immersion time. The hardness of the fruits decreased with an increase in immersion times and temperatures. These results can be used in the future for developing methods for the control of chestnut weevil populations.

Synergistic Anticancer Effect of the Cinnamomi Cortex Ethanol Extract (CcEE) and Hyperthermia in AGS Human Gastric Cancer Cells (AGS 인체 위암세포에서 육계 에탄올 추출물(CcEE)과 온열치료의 항암 시너지 효과)

  • Objectives : In this study, we investigated the combination effects of Cinnamomi cortex Ethanol Extract (CcEE) and hyperthermia in the human AGS gastric cancer cell line. Methods : AGS cells were treated with the indicated concentrations of CcEE (0, 50 or $60{\mu}g/mL$) for 1h prior to hyperthermia. And then incubated for a further 30 min at the indicated temperatures (37, 42 or $43^{\circ}C$) in a humidified incubator containing 5% $CO_2$ or a thermostatically controlled water bath for hyperthermia. The cell viability was measured by MTT assay, Morphology assay and Trypan blue assay. To investigate the possible molecular signaling pathways, the activation of mitogen-activated protein kinase (MAPK) proteins (ERK, p38 and JNK) and expression of various anti-apoptotic proteins such as Caspase-3, Caspase-9, p53, Cyclin D1 and MMP-2 were assessed by Western blot analysis. In addition, Annexin V and 7-amino-actinomycin D (7-AAD) staining was performed to examine the apoptotic mechanism. Results : Combination of CcEE with hyperthermia effectively suppressed the cell viability and changed cellmorphology compared with CcEE or hyperthermia treatment alone. Combined treatment also abated the expression of Caspase-3, Caspase-9, Cyclin D1 and MMP-2. Whereas, the expression level of p53 was up-regulated by co-treatment. Moreover, combination treatment enhanced phosphorylation of ERK, p38 and JNK. In addition, this combination increased anti-cancer effect by inducing cell death through the apoptosis. Conclusions : Taken together, all these findings suggest that the combination treatment with CcEE and hyperthermia may have therapeutic potential as a promising approach to patients with stomach cancer.

OGLE-2017-BLG-1049: ANOTHER GIANT PLANET MICROLENSING EVENT

  • We report the discovery of a giant exoplanet in the microlensing event OGLE-2017-BLG-1049, with a planet-host star mass ratio of q = 9.53 ± 0.39 × 10-3 and a caustic crossing feature in Korea Microlensing Telescope Network (KMTNet) observations. The caustic crossing feature yields an angular Einstein radius of θE = 0.52 ± 0.11 mas. However, the microlens parallax is not measured because the time scale of the event, tE ≃ 29 days, is too short. Thus, we perform a Bayesian analysis to estimate physical quantities of the lens system. We find that the lens system has a star with mass Mh = 0.55+0.36-0.29 M⊙ hosting a giant planet with Mp = 5.53+3.62-2.87 MJup, at a distance of DL = 5.67+1.11-1.52 kpc. The projected star-planet separation is a⊥ = 3.92+1.10-1.32 au. This means that the planet is located beyond the snow line of the host. The relative lens-source proper motion is μrel ~ 7 mas yr-1, thus the lens and source will be separated from each other within 10 years. After this, it will be possible to measure the flux of the host star with 30 meter class telescopes and to determine its mass.

Effect of the Physical Parameters and Alkalinity in the Ammonia Stripping (반응조의 물리적 인자와 알칼리도가 암모니아 탈기에 미치는 영향에 관한 연구)

  • The effect of the physical parameters in the reactor (aeration depth, bubble size, and surface area) and the alkalinity of the solution on the ammonia stripping by bubbling were evaluated. When an airflow of 30 L/min was bubbled below the solution surface in the range 6-53 cm, the ammonia removal rate were observed to be the same regardless of the bubbling depths. At pH 10.0 and a temperature of $30^{\circ}C$, the average rate constant and the standard deviation were $0.178h^{-1}$ and 0.004. No appreciable changes in the ammonia removal rate were also observed with varying the bubble size and the air-contacting surface area. Alkalinity of the solution was found to affect the ammonia removal rate indirectly. This is expected because the pH of the solution would vary with dissolution of gaseous $CO_2$ by air bubbling. The real wastewaters from landfill site and domestic wastewater treatment plant were tested. In the case of domestic wastewater (pH = 7.1, alkalinity = 75 mg/L), the ammonia removal rate was poor even with the control of pH to 9.3. The raw landfill leachate (pH = 8.0, alkalinity = 6,525 mg/L), however, showed the appreciable removal rate with increasing pH during aeration. When the initial pH of the leachate was adjusted 9.4, the removal rate was significantly increased without changing the pH during aeration.

Experimental life history of Echinostoma hortense (호르린스극구흡충의 실험실 내 생활사에 대한 연구)

  • The complete life cycle of Echinostoma hertense has been maintained in the laboratory, using Lymnaea persia snails and Rana nigromaculata tadpoles as the first and second intermediate hosts. ICR mice was used as the definitive host. Within the egg of 5. hotense, the miracidium was fully matured in 13 days of incubation at $29~30^{\circ}C$. The miracidium was $93.8{\times}53.6{\;}{\mu\textrm{m}}$ in average size, covered with numerous cilia of $7~11{\;}{\mu\textrm{m}}$ length. The epidermal plates were arranged in 6-8-4-2 formula. The first generation rediae ($1.19{\times}0.27{\;}mm$ in average size) were observed in 14 days after miracidial challenge to the snails, and the second generation rediae ($1.40{\times}0.26{\;}mm$ in average size) in 30 days. The average sixte of the cercaria was $295.5{\times}145.0{\;}{\mu\textrm{m}}$. Their head crown was poorly developed, and collar spines were not yet observed. After a cercarial challenge to the tadpoles, all of the tadpoles became infected and the average worm recovery rate was 88.5%. The majority of the metacercariae (75.5%) were recovered from the muscle of the tadpole's posterior body and the rest (24.3%) from their gills. The metacercariae from the tadpoles were elliptical, and $167.7{\times}129.9{\;}{\mu\textrm{m}}$ in average size. The recovery rate of adults from the mice was difFerent by the age of the metacercariae grown in the tadpoles. The metacercariae younger than 5 hrs could not infect mice whereas those older than 6 hrs could infect mice. The recovery rate became higher as the metacercaria matured, with the peak recovery rate of 90.0 % at the metacercarial age of 9 days. Thereafter the recovery rate decreased to 55.0% at the age of 50 days. As shown by the above results, the whole life cycle of E. hcrtense has been completed in the laboratory. At least 55~58 days were required to maintain one egg-to-egg cycle of E. hortense.

  • boehmite which is the by-product in alcohol process contains anions such as $Cl^-,{NO_3}^-.or\; CH_3COO^-$ Influe-nce of these anions was studied on properties of $\alpha$-alumina powders prepared by treating the transformed ${\gamma}$-alumina with the alumina sol and $\alpha$-alumina seeds (d_{50}=0.36\mu\textrm{m}$) Disperal 20/1 and 10/1 containing $Cl^-$ produced spherical powder Disperal 20/2 with ${NO_3}^-$ produced equiaxed powder and Disperal 10/3 with $CH_3$ $COO^-$ irregular shaped and sized powder. All of these $\alpha$-alumina powders were submicron. Although the green density of the alumina powder derived from Disperal 10/1 by calcination at $1200^{\circ}C$/1h. was 53% it did not sinter even at $1550^{\circ}C$ for 2h. which implies that its d50 is greater than $3\mu\textrm{m}$. The others gave green densities in the range of 44~47% but they sintered as well as the AKP-30 at $1500^{\circ}C$/2 h or $1550^{\circ}C$/2 h.

Purification and Characterization of a Keratinase from a Feather-Degrading Fungus, Aspergillus flavus Strain K-03

  • A keratinolytic enzyme secreted by Aspergillus flavus K-03 cultured in feather meal basal medium (FMBM) containing 2% (w/v) chicken feather was purified and characterized. Keratinolytic enzyme secretion was the maximal at day 16 of the incubation period at pH 8 and $28^{\circ}C$. No relationship was detected between enzyme yield and increase of fungal biomass. The fraction obtained at 80% ammonium sulfate saturation showed 2.39-fold purification and was further purified by gel filtration in Sephadex G-100 followed by ion exchange chromatography on DEAE-Sephadex A-50, yielding an active protein peak showing 11.53-fold purification. Sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) and zymograms indicated that the purified keratinase is a monomeric enzyme with 31 kDa molecular weight. The extracellular keratinase of A. flavus was active in a board range of pH ($7{\sim}10$) and temperature ($30^{\circ}C{\sim}70^{\circ}C$) profiles with the optimal for keratinase activity at pH 8 and $45^{\circ}C$. The keratinase activity was totally inhibited by protease inhibitors such as phenylmethylsulfonyl fluoride (PMSF), iodoacetic acid, and ethylenediaminetetraacetate (EDTA) while no reduction of activity by the addition of dithiothreitol (DTT) was observed. N-terminal amino acid sequences were up to 80% homologous with the fungal subtilisins produced by Fusarium culmorum. Therefore, on the basis of these characteristics, the keratinase of A. flavus K-03 is determined to be subtilisins-like.

Immobilization on Chitosan of a Thermophilic Trehalose Synthase from Thermus thermophilus HJ6

  • A thermostable trehalose synthase (TtTSase) from Thermus thermophilus HJ6 was immobilized on chitosan activated with glutaraldehyde. The yield of immobilization was evaluated as 39.68%. The optimum pH of the immobilized enzyme was similar to that of the free enzyme. However, the optimal temperature ranges were shifted by about $4^{\circ}C$ owing to better thermal stability after immobilization. The half-life of heat inactivation for free and immobilized enzymes was 5.7 and 6.3 days at $70^{\circ}C$, respectively, thus showing a lager thermostability of the immobilized enzyme. When tested in batch reaction, the immobilized enzyme retained its relative activity of 53% after 30 reuses of reaction within 12 days, and still retained 82% of its initial activity even after 150 days at $4^{\circ}C$. A packed-bed bioreactor with immobilized enzyme showed a maximum yield of 56% trehalose from 100 mM maltose in a continuous recycling system (bed volume: 10 ml) under conditions of pH 7.0 and $70^{\circ}C$.

Purification and Characterization of Pretense from Sarcodon aspratus (Berk.) S. Ito (능이버섯에서 분리한 Protease의 정제와 특성)

  • 능이버섯 〔Sarcodon aspratus(Berk.) S.Ito〕으로부터 단백질 가수분해 효소를 추출하여 75%(NH$_4$)$_2$SO$_4$ 염석과 DE52 anion exchange column chromatography 와 sepharyl-S 200 column 및 Mono s column chromatography 에 의해 정제하였는데 조 효소의 특이 활성은 55.2U/mg protein으로 조효소액에 비하여 11.26배 증가하였고 수율은 49.5%로 나타났다. 정제된 효소는 전기영동을 행한 결과 단일 band를 나타내었으며 분자량은 29,300으로 추정되었다. pH의 안정성은 4$^{\circ}C$에서 48시간 보존하였을 때 pH 8.5에서 가장 안정하였고, pH 5.5~10.5까지 높은 활성을 유지하였다. 온도에 대한 안정성은 30분간 보존 한 후 활성을 검토한 결과 단백분해능은 5$0^{\circ}C$까지 비교적 처음활성을 유지하다가 6$0^{\circ}C$에서는 53%정도의 활성이 유지되었으나 그 이상의 온도에서는 급격히 실화하여 7$0^{\circ}C$에서는 완전히 실화하였다. 금속 이온에 의해서는 크게 저해를 받지 않았으나 PMSF 저해제에 대하여 저해되어 본 효소가 serine protease임을 시사하였다.
