Acute Oral Toxicity Test in Japanese Quail (일본 메추리를 이용한 급성 경구독성시험법의 확립)

  • The acute oral LD5O toxicity values of isazofos, pyraclofos, diazinon and methomyl were determined for Japanese quail based on OECD guideline. The $LD_{50}$ of isazofos, pyraclofos and diazinon was 16.26 mg/kg, and 7.11mg/kg body weight In female respectively. And the $LD_{50}$ of each chemical in male was 21.44, 35.64, 8.28 mg/kg body weight respectively. Diazinon was the most susceptible compounds to Japanese quail in both sexes. The $LD_{50}$ of methomyl was 21.24 mg/kg body weights in female, and 28.28 mg/kg body weight in male respectively. Diazinon, isazofos and methomyl were more toxic In the female than male. The symptoms of poisoning were similar in quails administrated with each chemicals. The clinical sign in Japanese quail were ataxia, salivation, diarrhea, ruffled feather and convulsion at dead point. There were severe hemorrhage and catarrhal inflammation from duodenum to ileum In all compounds. In Japanese quail treated with organophosphorus and carbamate compounds, brain acetylcholinesterase was inhibited by 88-96. The recovery was not observed after 5 h in sublethal dose.

Antitumor effect of Ginsenoside Rh2 and $\beta$-glucan in mice

  • In the present study, we investigated the antitumor effects of Ginsenoside Rh2 and $\beta$-glucan using an experimental metastatic mouse model intravenously injected with B 16 melanoma F10 cells. Oral administration to various concentration of $\beta$-glucan (50mg/kg, l00mg/kg and 200mg/kg) dose-dependently reduced the lung-metastatic potential of metastatic BI6 melanoma F10 cells in syngenic mice. At same dose, Ginsenoside Rh2(50mg/kg) has more antitumor effect than $\beta$-glucan(50mg/kg). (omitted)

Effects of dietary copper sources and levels on growth performance, copper digestibility, fecal and serum mineral characteristics in growing pigs

  • This experiment was conducted to investigate the effects of three different copper (Cu) sources (one inorganic and two organics) and levels (0, 50, and 100 mg/kg) on the growth performance, Cu digestibility, fecal mineral excretion, serum mineral concentration, jejunal morphology, and serum biochemical profile of growing pigs. A total of 42 male, growing pigs (31.08 ± 1.82 kg) were randomly assigned to seven treatments consisting of one negative control (0 mg/kg of added Cu level) and treatments with copper sulfate (CuSO4), Cu-amino acid complex (CuAA), and Cu-hydroxy-4-methylthio butanoate chelate complex (CuHMB) at 50 and 100 mg/kg each for 28 d. Pigs fed 50 or 100 mg/kg of Cu showed improved (p < 0.05) average daily gain and feed intake. Although Cu excretion decreased (p < 0.01) in pigs fed 100 mg/kg of organic Cu sources compared to those fed CuSO4, there was no difference between the Cu sources in pigs fed 50 mg/kg. However, the apparent total tract digestibility of Cu increased (p < 0.01) in pigs fed organic Cu sources compared with that in pigs fed CuSO4. The addition of CuHMB increased (p < 0.01) serum phosphorus and sulfur concentrations; however, there were no effects of source and level on jejunal morphology and serum biochemical profile. These results suggest that the inclusion (50 mg/kg) of organic Cu sources (CuAA and CuHMB) in the growing pig diet could be beneficial for growth performance and Cu availability and may reduce environmental pollution.

Inhibitory Effect of Cordyceps Militaris Water Extracts on Sarcoma-180 Cell-Induced Ascities Tumor in ICR Mice (누에번데기 및 누에애벌레 밀리타리스동충하초 (Cordyceps Militaris) 열수추출물이 Sarcoma-180 세포로 유발한 마우스 복수암에 미치는 영향)

  • This study was undertaken to evaluate the antitumor activities of Cordyceps militaris of silkworm pupa (CMP) and silkworm larva (CML), as compared with the effect of cordycepin, an active compound found in Cordyceps militaris. Antiproliferation effect of the test materials were evaluated in the sarcoma-180 cells using the MTT test. For the in vivo study, ICR mice were inoculated i.p. with 1.0 ${\times}$ 10$^{6}$ sarcoma-180 cells/mouse on Day 0, and were again i.p. injected with one of the following substances from Day 1 to Day 10 : saline (control group), 50 mg/kg (CMP50, CML50) ,100 ma/kg (CMP100, CML100), or 200 mg/kg (CMP200, CML200) of Cordyceps militaris water extracts, or 1 mg/kg (C1), 2 mg/kg (C2), or 4 mg/kg (C4) of cordycepin. Pretreatment of the sarcoma-180 cells with 100 mg/ml, 500 mg/ml, and 1000 mg/ml of CML (60.1$\pm$2.5%, 49.8$\pm$3.7%, and 45.4$\pm$0.1% of the value for untreated control cells, respectively) or CMP (68.3$\pm$2.1%, 55.1$\pm$0.9%, and 51.4$\pm$3.5% of the value for control cells, respectively) for 48 hrs significantly decreased the survival rate (proliferation) of tumor cells (p<0.05). Body weight of the control mice bearing ascites tumor and injected with saline was 1.4 times of the value for normal animals at day 18. Mice bearing ascites tumor and injected with cordycepin (1, 2, or 4 mg/kg) exhibited a significantly lighter body weight compared with the control mice, while animals injected with CMP or CML (50, 100, or 200 mg/kg) showed a significantly lighter body weight compared with the mice injected with cordycepin. Mice injected with CMP50, CMP100, or CMP200 mg/kg (or CML50, CML100, or CML200 mg/kg) showed a 133% (or 90%), 80% (or 62%), and 68% (or 52%) longer mean survival time, and those treated with C1, C2, or C4 exhibited a 54%, 91% and 80% longer survival time compared to the value for control mice injected with saline. These results indicate that the hot-water extracts of Cordyceps militaris of both silkworm pupa and silkworm larva have an anti-proliferation effect of tumor cells as well as the life prolongation effect in mice bearing ascites tumor, which are superior to the activities of cordycepin.

In vivo Antitumor Activity and Acute, Subacute Toxicity of Keumsa (Phellinus linteus) Extracts (금사목질진흙버섯 자실체 추출물의 in vivo 항암활성 및 급성, 아 급성 독성 시험)

  • This study used an S-180 cell-injected mouse model to evaluate the antitumor effects of the acute and subacute toxicity of Keumsa (Phellinus linteus) extract intravenously administrated in ICR mice. When administered intravenously (31.3-250 mg/kg body weight), Keumsa (Phellinus linteus) extract significantly inhibited the growth of the solid tumor cell. The antitumor activity of Keumsa (Phellinus linteus) extract increased in a dose-dependent manner. The highest dose (250 mg/kg body weight) was highly effective, reducing tumor formation by 42.7% compared with the control group. In the acute toxicity test, $LD_{50}$ of the Keumsa (Phellinus linteus) extract showed 632.84 mg/kg (♂) and 814.48 mg/kg (♀) after intravenous administration. In addition, liver and spleen weight were increased in a dose-dependent manner. In the subacute toxicity test, the mice were intravenously administered over the course of 28 days. The $LD_{50}$ of the Keumsa (Phellinus linteus) extract showed 355.41 mg/kg (♂) and 383.53 mg/kg (♀) after intravenous administration. The liver and spleen weight also increased in a dose-dependent manner. In the case of the group that received more than 125 mg/kg of intravenous administration, exercise capacity, such as jumping ability and agility, were significantly increased. These results suggest that Keumsa (Phellinus linteus) extract can be regarded as a potent enhancer of the innate immune response, and it can be considered as a new natural product with low toxicity that may be used as a candidate for antitumor action.

Optimum Level of Nitrogen Fertilizer Based on Content of Nitrate Nitrogen for Growing Chinese Cabbage in Green House (시설조건(施設條件)의 배추 재배(栽培) 토양(土壤)에서 질산태질소(窒酸態窒素) 검정(檢定)에 의한 질소실비량(窒素施肥量) 결정(決定))

  • To establish N fertilizer recommended scheme for the Chinese cabbage cultivation in green house based on the soil test of nitrate nitrogen, relationship among the content of soil nitrate and fertilizer effects and fertilizer N use efficiency were investigated from nine soils which differed amount of nitrate nitrogen from $14mg\;kg^{-1}$ to$226mg\;kg^{-1}$. The amount of nitrate nitrogen in soil showed a positive correlation with the dry weight of chinese cabbage in the plot of no fertilization. When the fertilizer effects were calculated by difference between the plots of fertilization and no fertilization in the dry weight and the amount of N uptake, a negative correlation was obtained between the amount of nitrate nitrogen in soils and the fertilizer effects. There was also a negative correlation between the amount of nitrate nitrogen in soils and fertilizer use efficiency. Recommendation of application rate of nitrogen fertilizer based on content of $NO_3-N$ in soils was evaluated by the regression equation among the content of soil nitrate, fertilizer effects and fertilizer N use efficiency. Incase the content of $NO_3-N$ nitrogen in soil is more than $200mg\;kg^{-1}$, No N fertilization is recommended; However, The standard N fertilization($320kg\;ha^{-1}$) is recommended for the soils with less than $50mg\;kg^{-1}$. For the soils ranged from $50mg\;kg^{-1}$ to $200mg\;kg^{-1}$ in the amount of nitrate nitrogen, an equation has been developed in order to calculate the recommended amount of fertilizer N.

The Effect of Red Ginseng Ethanol Extract on the Immunotoxicity of Diethylstilbestrol in ICR Mice (마우스에 있어서 Diethylstilbestrol의 면역독성에 미치는 홍삼 Ethanol 유출물의 영향)

  • The effect of red ginseng ethanol extract on the immunotoxicity of diethylstilbestrol (DES) was studied in ICR mice. ICR male mice were divided into S groups (10 mice/group), and red ginseng ethanol extract (50, 100 and 200 mg/kg body wt., respectively) and DES (1 mg/kg body wt.) were injected intraperitoneally (i.p.) to ICR mice once a day for 2 weeks. Mice were sensitized and challenged with sheep red blood cells (S-RBC). Immune response were evaluated by humoral immunity, cell-mediated immunity, non-specific immunity, and circulating leukocyte counts. The results of this study were summarized as followings: 1. The DES-treated control group as compared with normal group showed the tendency to decrease body weight rate and relative liver weight, decreased both humoral and cellular immune responses, phagocyte activity, and circulating leukocyte counts, but increased the natural killer (NK) cell activity. 2. Compared with the DES-treated control group, DES plus red ginseng ethanol extract-treated groups significantly decreased the body weight rate (P<0.01). Relative liver weight was significantly decreased in DES plus red ginseng ethanol extract (50mg/kg)-treated group (P<0.01), but significantly increased in DES plus red ginseng ethanol extract (100mg/kg)-treated group (P<0.01). Relative spleen and thymus weights were significantly enhanced in DES plus red ginseng ethanol extract (100 mg/kg)-treated group (P<0.01), but significantly decreased in DES plus red ginseng ethanol extract (200 mg/kg)-treated group (P<0.01). 3. Both humoral and cellular immune responses were significantly decreased in DES plus red ginseng ethanol extract-treated groups rather than in the DES-treated control group (P<0.01). Especially, it weakened the decrease in DES plus red ginseng ethanol extract (100 mg/kg)-treated group. 4. Phagocyte activity and circulating leukocyte counts were significantly decreased in DES plus red ginseng ethanol extract-treated groups rather than in the DES-treated control group (P<0.01). Especially, it weakened the decrease in DES plus red ginseng ethanol extract (100 mg/kg)-treated group. NK cell activity was significantly enhanced in DES plus red ginseng ethanol extract (100 mg/kg)-treated group (P<0.01), but significantly decreased in DES plus red ginseng ethanol extract (50 and 200 mg/kg)-treated groups (P<0.01).

Effect of Supplemental Selenomethionine on Growth Performance and Serum Antioxidant Status in Taihang Black Goats

  • An experiment was conducted to evaluate the effect of different levels of supplemental selenomethionine (Se-Met) on growth performance and serum antioxidant status in Taihang Black goats. Fifty 16-week-old goats with an average body weight of 12.5${\pm}$0.5 kg were randomly assigned to five treatments fed a basal diet (0.049 mg Se/kg DM) supplemented with 0 (control), 0.10, 0.30, 0.50 and 1.00 mg of Se/kg DM (form Se-Met) for 80 days. Average daily gain and feed efficiency were higher (p<0.05) in the groups supplemented with 0.30 to 0.50 mg Se/kg DM compared with the control group. However, Se-Met supplementation had no influence on average daily feed intake (p>0.05). Se-Met supplementation significantly increased (p<0.01) the activity of glutathione peroxidase enzymes (GSH-Px) and superoxide dismutase (SOD) in serum. The group supplemented with 0.50 mg Se/kg DM had the highest activity of GSH-Px compared with other groups (p<0.05). Serum SOD activity was higher (p<0.05) in goats supplemented with both 0.30 and 0.50 mg Se/kg DM than in control goats and goats supplemented with 1.00 mg Se/kg DM. Serum glutathione-S-transferase (GST) activity and malondialdehyde (MDA) concentration were significantly decreased (p<0.05) in goats supplemented with 0.30, 0.50 and 1.00 mg Se/kg DM compared with control values. These results indicated that Se-Met supplementation markedly improved the antioxidant status in goats. Blood Se concentration increased linearly (p<0.001) and quadratically (p<0.001) as the level of supplemental Se-Met increased. The concentration of Se in the control diet (0.049 mg Se/kg DM) did not satisfy the Se requirement in goats as indicated by reduced growth rate, feed efficiency, activities of GSH-Px and SOD in serum, and blood Se concentrations. In conclusion, it is recommended that 0.30 to 0.50 mg of Se/kg DM from Se-Met (total diet Se of 0.349 to 0.549 mg/kg DM) be supplied in the diet of Taihang Black goats to enhance growth performance and improve antioxidant status.

Antihypertensive Effect of $\alpha-Methyldopa$ Administered Concurrent with Timolol or Labetalol in SHR. (SHR에 $\alpha-Methyldopa$를 Timolol 또는 Labetalol과 병용투여시 혈압강하효과에 관한 연구)

  • The antihypertensive effect of $\alpha-Methyldopa$ administered concurrently with timolol or labetalol were studied with spontaneously hypertensive rats (SHR). Every drugs were administered orally once a day after prechecking the systolic blood pressure and heart rate of SHR. The blood pressure and heart rate of SHR were significantly decreased in groups of combination while those of non-combination groups were fallen slightly. Compared with control group, the significant changes of blood pressure was obseved in group of $\alpha-methyldopa$ with timolol(100mg+2mg/kg)and $\alpha-methyldopa$ with labetalol (100mg+12.5mg/kg). The group of $\alpha-methyldopa$ with timolo 1 (100mg + 1.0mg/kg) reduced blood Pressure in similar degree as manifested in group of $\alpha-methyldopa$ with labetalol (100mg + 50mg/kg). In the group that $\alpha-methyldopa$ was administered concurrently with timolol, the maximum antihypertensive effect and heart rate decreasing effect were appeared after 3hr and 1hr of administration respectively and those effects in group of $\alpha-methyldopa$ with labetalol were appeared after 6hr of administration. The acute oral toxicity test was performed using albino mice with $\alpha-methyldopa$ alone, $\alpha-methyldopa$ with timolol (100:1) and $\alpha-methyldopa$ with labetatol (4:1), and was found that the $LD_{50}$ of $\alpha-methyldopa$ alone was 1104 mg/kg, $\alpha-methyldopa$ with timolol (100:1) was 1115 mg/kg and $\alpha-methyldopa$ with labetalol (4:1) was 354mg/kg.

Acute Toxicity Test of KH-502 (Flupyrazofos) in Rats and Mice (KH-502의 랫트 및 마우스를 이용한 급성독성시험)

  • KH-502 (Flupyrazofos), a new organophosphorus insecticide synthesized by Korea Re-search Institute of Chemical Technology, was found to be effective against diamond-back moth(Plutella xylostella). This study was carried out to determine the acute toxicity of KH-502 in Sprague-Dawley rats and ICR mice. The test article was orally or dermally administered to the animals. Death, tremors, salivation, lacrimation, abnormal gait and corneal opacity were observed. Decrease in body weight gain was observed in all treatment groups. At necropsy, dark red coloration of lung, enlargement of adrenal glands and atrophy of spleen were observed. The oral $LD_{50}$ value was 372 mg/kg in male rats, 605 mg/kg in female rats, 186 mg/kg in male mice, and 115 mg/kg in female mice. And the dermal $LD_{50}$ was 4086 mg/kg in male and 3881 mg/kg in female rats.
