The Protein Kinase A Pathway Regulates Zearalenone Production by Modulating Alternative ZEB2 Transcription

  • Zearalenone (ZEA) is an estrogenic mycotoxin that is produced by several Fusarium species, including Fusarium graminearum. One of the ZEA biosynthetic genes, ZEB2, encodes two isoforms of Zeb2 by alternative transcription, forming an activator (Zeb2L-Zeb2L homooligomer) and an inhibitor (Zeb2L-Zeb2S heterodimer) that directly regulate the ZEA biosynthetic genes in F. graminearum. Cyclic AMP-dependent protein kinase A (PKA) signaling regulates secondary metabolic processes in several filamentous fungi. In this study, we investigated the effects of the PKA signaling pathway on ZEA biosynthesis. Through functional analyses of PKA catalytic and regulatory subunits (CPKs and PKR), we found that the PKA pathway negatively regulates ZEA production. Genetic and biochemical evidence further demonstrated that the PKA pathway specifically represses ZEB2L transcription and also takes part in posttranscriptional regulation of ZEB2L during ZEA production. Our findings reveal the intriguing mechanism that the PKA pathway regulates secondary metabolite production by reprograming alternative transcription.

Visual Perception of Garment Surface Appearance

  • This paper concerns with the relationship between the visual perception of the degree of pucker or wrinkles of garment surfaces and the geometrical parameters of surfaces. In this study, four potentially relevant parameters of the surface profile are considered, namely, the variance ($\sigma$$^2$), the cutting frequency (F$\_$c/), the effective disparity curvature (D$\_$ce/) (Defined as the average disparity curvature of the wrinkled surface over the eyeball distance of the observer) and the frequency component of the disparity curvature ( D$\_$cf/). Based on the experiments using garment seams having varying degree of pucker (i.e. the wrinkles along a seam line), it was found that, while the logarithm of each of these four parameters has a strong linear relationship with the visually perceived degree of wrinkles, following the Web-Fetchner Law, the effective disparity curvature ( D$\_$ce/) and the frequency component of the disparity curvature (D$\_$cf/) appeared to have stronger relationships with the visual perception. This finding is in agreement with the suggestion by Rogers '||'&'||' Cagenello that human visual system may compute the disparity curvature in discriminating curved surfaces. It also suggested an objective method of measuring the degree of surface wrinkles.

Parameterization for Longwave Scattering Properties of Ice Clouds with Various Habits and Size Distribution for Use in Atmospheric Models

  • A parameterization for the scattering of longwave radiation by ice clouds has been developed based on spectral scattering property calculations with shapes and sizes of ice crystals. For this parameterization, the size distribution data by Fu (1996) and by Michell and Arnott (1994) are used. The shapes of ice crystal considered in this study are plate, solid column, hollow column, bullet-rosette, droxtal, aggregate, and spheroid. The properties of longwave scattering by ice crystals are presented as a function of the extinction coefficient, single-scattering albedo, and asymmetry factor. The heating rate and flux by the radiative parameterization model are calculated for wide range of ice crystal sizes, shapes, and optical thickness. The results are compared with the calculated results using a six-stream discrete ordinate scattering algorithm and Chou's method. The new method (with various habits and size distributions) provides a good simulation of the scattering properties and cooling rate in optically thin clouds (optical thickness < 5). Depending on the inclusion of scattering by ice clouds, the errors in the calculation of the cooling rates are significantly different.

Parametric study on dynamic behavior of rectangular concrete storage tanks

  • Tanks are used to store a wide variety of liquids such as oil, gasoline and water. It is reported that, a large number of tanks have been damaged during severe earthquakes. Therefore, understanding their behavior under earthquake is an important subject for structural engineers. In this paper, a comprehensive study is presented on dynamic response of tanks. A parametric study has been completed on the rectangular storage tanks with aid of finite element method (FEM). Various parameters are investigated, such as; liquid height, density and earthquake with different peak ground acceleration (PGA). When investigating these parameters, modal and time history method is used. Six different earthquake records are used for time history analysis. The analysis results show that when the PGA increases by 10.7 times, the maximum displacements, stress, sloshing and base shear increase by 11.4, 22.6, 5.46 and 17.8 times, respectively and when the liquid height increases by two times, the absolute maximum values of stress, displacements, base shear and sloshing increase 1.65, 2.04, 2.05 and 1.34. Furthermore, values of sloshing increase with decrease in density.

The Effects of Chicken Leg Bone Extract on Antioxidative Properties under Different Heating Condition

  • The aim of this study was to extract chicken leg bone, which is a by-product of industrial poultry processing, using different heating temperatures (80, 90 and $100^{\circ}C$) and durations (5, 10 and 15 min). The pH value, soluble protein content, peptide content and antioxidative properties, including superoxide anion scavenging ability, 1, 1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging ability, reducing capacity and inhibitory activity of linoleic acid peroxidation, were measured. The results showed no significant differences (p>0.05) in pH value among all treatments. Decreased soluble protein content and peptide content were observed in chicken leg bone extract obtained under higher heating temperatures (90 or $100^{\circ}C$) and longer heating durations (10 or 15 min). In antioxidative properties, the extracts which were heated at 90 or $100^{\circ}C$ for 15 min exhibited significantly higher superoxide anion scavenging ability, DPPH free radical scavenging ability, reducing capacity and inhibitory activity of linoleic acid peroxidation (p<0.05).

Genetic Diversity of Thread-sail Filefish Stephanolepis cirrhifer Populations in Korean Coastal Waters Inferred from Mitochondrial DNA Sequence Analysis

  • The genetic diversity and population genetic structure of thread-sail filefish, Stephanolepis cirrhifer (Temminck & Schlegel), were examined with a nucleotide sequence analysis of a 495bp fragment of the 5'-end of the cytochrome b gene in 113 fish collected from five populations from the south and east coasts of the Korean Peninsula. Seventeen variable nucleotide sites and 16 haplotypes were defined. The observed haplotypes had a shallow haplotype genealogy and no geographical association. Most of the populations had high haplotype diversity and low nucleotide diversity, and significant negative values for Fu's $F_S$, suggesting rapid, recent population growth from an ancestral population and sudden population expansion. The estimated pairwise fixation indices ($F_{ST}$) indicate that substantial gene flow occurs among these populations. Thread-sail filefish in the South Sea of Korea and East Sea Korean populations forms a single panmictic population. Thus, thread-sail filefish in these areas should be treated as one management unit.

  • Epigenetic alterations such as DNA methylation, histone acetylation, and chromatin remodeling can control gene expression by regulating gene transcription. DNA methylation is one of the frequent epigenetic events that play important roles in cancer development. Cancer cells can gain significant resistance to anticancer drugs and escape programmed cell death through major epigenetic changes, including DNA methylation. To date, several research groups have identified instances of both (i) hypermethylation of tumor suppressor genes, and (ii) global hypomethylation of oncogenes. These changes in DNA methylation status could be used as biomarkers for the diagnosis and prognosis of cancer patients undergoing chemotherapies or other clinical therapies. Herein, we describe genes for which methylation is dependent upon anticancer drug resistance in patients with gastric cancer; we then suggest a significant epigenetic target to focus on for overcoming anticancer drug resistance.

Properties of Plasma Sprayed $Al_2O_3/SS316$ Graded Coatings (플라즈마 용사용 $Al_2O_3/SS316$ 복합 분말 제조 및 경사 코팅충의 제조에 관한 연구)

  • In the case of using high temperature by coating ceramic/metal, large stress was produced due to difference of thermal expansion coefficient between those. And then lead to delamination. In order to relaxation of the stress A1$_2$O$_3$/SS316 composite powders with $10wt.%Al_2O_3$ compositional gradient and $10wt.%Al_2O_3$ agglomerated powder were made by spray drying method. These powders were sintered to improve the strength and to be plasma sprayed in order to fabricate the FGC(functionally graded coating). The influence of gun power, working distance and Ar pressure on the microstructure of the coating layer was studied in order to optimize the plasma spray conditions. It was proven that the optimum conditions were 40kw gun power, 5cm working distance and $100ft^3/h$ Ar flow for both powders. FGC with 10 compositional steps was fabricated and the total thickness was 1.3mm. FGC was heat treated at $1100^{\circ}C$for 10hours to evaluate the heat resisting characteristics.

Superior Vena Cava Syndrome -2 Bypass Graft Cases- (상공정맥 증후군 -Dacron & Nylon 환치수술 2예-)

  • Superior Vena Cava Syndrome: Dacron and Nylon graft between the left innominate vein and the right atrial appendage. Two cases with typical superior vena cave syndrome treated by by-pass graft between the left innominate vein and the right atrial apepndage were presented. One of them was a 58 year old farmer who suffered from marked swelling of the neck and upper half of body, the other was a 50 years old government employee who had acutely progressive symptoms of superior vena cave obstruction. Both of cases revealed that [1] cubitel venous pressure was markedly increased. [2] tumors were noted in the posterior mediastinum by laminography. [3] preoperative cavogram showed the occlusion of superior vena cava and marked collaterals. Dacron and Nylon graft were inserted between the left innominate vein and the right atrial appendage. Postoperatively, the symptoms were relieved markedly, showing edema free face and decreased cubital venous pressure. Postoperative cavogram showed patent graft. Histologically the first case was diagnosed as squamous cell carcinoma and the second as undifferentiated carcinoma, originated probably from bronchus. Total doses of 3150 r X-ray irradiation and 5000 mg of 5-FU were administered in each cases. The first case expired 11 months postoperatively without recurrence of superior vena cave obstruction symptom and the second case is living now without obstruction signs, 4 months after by-pass operation.

Development of a position sensitive CsI(Tl) crystal array

  • A position-sensitive CsI(Tl) crystal array coupled with the multi-anode position sensitive photomultiplier tube (PS-PMT), Hamamatsu H8500C, has been developed at the Institute of Modern Physics. An effective, fast, and economical readout circuit based on discretized positioning circuit (DPC) bridge was designed for the 64-channel multi-anode flat panel PSPMT. The horizontal and vertical position resolutions are 0.58 mm and 0.63 mm respectively for the 1.0 × 1.0 × 5.0 ㎣ CsI(Tl) array, and the horizontal and vertical position resolutions are 0.86 mm and 0.80 mm respectively for the 2.0 × 2.0 × 10.0 ㎣ CsI(Tl) array. These results show that the CsI(Tl) crystal array with low cost could be applied in the fields of medical imaging and high-resolution gamma camera.
