A Combination Strategy for Construction of Peptide-β2m-H-2Kb Single Chain with Overlap Extension PCR and One-Step Cloning

  • The time-consuming and high-cost preparation of soluble peptide-major histocompatibility complexes (pMHC) currently limits their wide uses in monitoring antigen-specific T cells. The single-chain trimer (SCT) of peptide-${\beta}2m$-MHC class I heavy chain was developed as an alternative strategy, but its gene fusion is hindered in many cases owing to the incompatibility between the multiple restriction enzymes and the restriction endonuclease sites of plasmid vectors. In this study, overlap extension PCR and one-step cloning were adopted to overcome this restriction. The SCT gene of the $OVA_{257-264}$ peptide-$(GS_4)_3-{\beta}2m-(GS_4)_4-H-2K^b$ heavy chain was constructed and inserted into plasmid pET28a by overlap extension PCR and one-step cloning, without the requirement of restriction enzymes. The SCT protein was expressed in Escherichia coli, and then purified and refolded. The resulting $H-2K^b/OVA_{257-264}$ complex showed the correct structural conformation and capability to bind with $OVA_{257-264}$-specific T-cell receptor. The overlap extension PCR and one-step cloning ensure the construction of single-chain MHC class I molecules associated with random epitopes, and will facilitate the preparation of soluble pMHC multimers.

비타민 C 보충이 제 2형 당뇨병 환자의 혈당 및 항산화상태에 미치는 효과 (Effect of Vitamin C Supplementation on Blood Sugar and Antioxidative Status in Types II Diabetes Mellitus Patients)

  • Purpose: This study was to determine the effect of oral vitamin C supplements on blood sugar and antioxidative status in Types II diabetes mellitus patients. Method: Data for the study were collected from June 24 to August 31, 2001. Participants(31) took 1g/day vitamin C for 4 weeks, after a 1 - week taking no Vitamin C, followed by Vitamin C 3g/day for 4 weeks. A baseline blood sample was obtained following a 12hour overnight fast and at the end of each 4week Vitamin C administration. Blood samples were taken for plasma vitamin C concentration, fasting blood sugar, HbA1c, superoxide scavenging activity and hydrogen peroxide scavenging activity. The data were analyzed by SPSS for repeated measures ANOVA. Result: Plasma vitamin C concentration was significantly increased over dose(F=3.316, p=.043). Fasting blood sugar and HbA1c was significantly decreased over dose(F=13.192, p=.000; F=11.995, p=.000). Superoxide scavenging activity and hydrogen peroxide scavenging activity was significantly increased over dose(F=486.138, p=.000; F=177.704, p= .000). Conclusion: The results suggest that megadose vitamin C supplementation may have a beneficial effect in diabetes mellitus patients on both glycemic control and antioxidant status. Thus dietary measures to increase plasma vitamin C may be an important health strategy for reducing the compliance of diabetic patients

류마티스 관절염 환자의 통증대처, 통증 및 우울정도의 관계 (The Relationship among the degrees of pain coping strategies, Pain and Depression of patients with Rheumatoid Arthritis)

  • This study has done for the purpose of investigation among the degrees of pain coping strategies, pain, and depression of patients with rheumatoid arthritis. The subjects of this study were 214 patients diagnosed with rheumatoid arthritis. The data were collected from september 4 to 16, 1995 by questionnaires. The data were analysed by the use of frequency, t-test, Pearson Correlation Coefficients, ANOVA, and Duncan test. The results Were as follows ; 1. The mean score of pain was 213.5(range : 4-390) and that of depression was 21.4 (range : 1-50). The mean score of passive pain coping strategies was 30.2(range : 11-48) and that of active pain coping strategies was 18.6(range : 7-33) 2. The passive pain coping degree showed a positive correlation with the pain degree (r=.475, p=.0001) and the active pain coping degree showed a inversive correlation with that(r= -.296, p=.0001). The passive pain coping degree showed a positive correlation with the depression degree (r=.425, p=.0001) and the active pain coping degree showed a inversive correlation with that(r=-.299, p=.0001). The pain degree showed a positive correlation with the depression degree(r=.374, p=.0001).

만성 폐쇄성 폐질환 환자의 기능적 용량 설명모형 (An Explanatory Model on Functional Capacity in Patients with Chronic Obstructive Pulmonary Disease)

  • Purpose: This study was conducted to develop and test an explanatory model on functional capacity in patients with chronic obstructive pulmonary disease using path analysis. Methods: Data were collected from 149 chronic obstructive pulmonary disease patients using 6-minute walk test, measurement of oxygen saturation, pulmonary function test, and self-reported questionnaires from June to October, 2005. The collected data were analyzed using SPSS/WIN 12.0 program and AMOS/WIN 4.0 program. Results: The overall fitness indices of modified model were good($x^2$ = 14.324, p = .281 GFI = .981, RMSEA = .006, AGFI = .944, NFI = .927, NNFI = .999, CFI = .999, PNFI = .613, $x^2$/df = 1.194). Functional capacity was influenced directly by age(${\beta}$ = -.304, p = .000), dyspnea(${\beta}$ = -.278, p = .000), self-efficacy(${\beta}$ = .240, p = .000), social support(${\beta}$ = .175, p = .004), pulmonary function(${\beta}$ = .169, p = .008), and oxygen saturation(${\beta}$ = .099, p = .048). These variables explained 39.3% in functional capacity. Conclusion: The findings of this study suggest that comprehensive nursing interventions should focus on decreasing dyspnea and increasing self-efficacy, social support, and oxygen saturation. In this perspective, pulmonary rehabilitation would be an effective strategy for improving functional capacity in patients with chronic obstructive pulmonary disease.

P2X Receptor 3D Structure Prediction Using Homology Modelling

  • P2X receptors are ATP-activated ion channels in the plasma membrane. P2X receptors have a role in a diverse range of disorders, making them a valuable therapeutic target. Hence, the present investigation employed homology modelling of the P2X receptor based on the crystal structure of 5SVJ, 6AH4, 5YVE and 5SVL. Twenty models, using both single- and multiple template-based methods, were developed, and the best model was chosen based on the validation result. We observed that a strategy based on multiple templates provided greater accuracy. Future studies involving binding site and docking analysis can make use of the produced structures.

Abiotic Degradation Degradation of the Herbicide Oxadiazon in Water

  • The performance of abiotic degradation of oxadiazon was investigated by applying zerovalent iron(ZVI), potassium permanganate($KMnO_4$) and titanium dioxide($TiO_2$) in the contaminated water. Experimental conditions allowed the disappearance of oxadiazon in the abiotic system. The degradation of this herbicide was monitored in buffer solutions having pH 3, 5 and 7 in the presence of iron powder in which the maximum degradation rate was achieved at acidic condition(pH 3) by 2% of ZVI treatment. The oxidative degradation of oxadiazon was observed in aqueous solution by $KMnO_4$ at pH 3, 7 and 10 in which the highest disappearance rate was found at neutral pH when treated with 2% of $KMnO_4$. The catalytic degradation of oxadiazon in $TiO_2$ suspension was obtained under dark and UV irradiation conditions. UV irradiation enhanced the degradation of oxadiazon in aquatic system in the presence of $TiO_2$. Conclusively, the remediation strategy using these abiotic reagents could be applied to remove oxadiazon from the contaminated water.

백목련의 가지 생장 유형

  • 백목련의 가지 생장유형은 두 가지의 생장전략에 의해 이루어지는 것으로 보인가. 하나는 환경 극복 전략이고 다른 하나는 생명 유지 전략으로 구분할 수 있는데, 이러한 전략은 한 그루의 나무 안에서도 같이 일어나고 있었다. 가지의 생장율은 조도에 밀접한 관련이 있었고(P<0.001) 물리적인 접촉의 유무와도 상당히 관련(P<0.01)되어 있는 반면에 방위별로는 우의차가 없었다. 상대 조도 1%미만의 경우는 주변 가지들과 거의 물리적인 접촉으로 대부분 생장이 제한되어 있었고, 10% 초과의 경우는 접촉에 의해 생장 제한이 거의 일어나지 않았다. 가지간의 물리적인 접촉의 유무는 분지율에 영향을 미치고 있었으나(P<0.05), 분지율은 조도에 무관하고, 방위별로도 유의차가 없었다. 생장 초기에는 주가지에서 부가지로의 물질 분배비가 크게 나타났으나 나무가 자라면서 환겨의 제한을 받게 됨에 따라 분배비는 다양하고 때로는 아주 적은 값을 보이기도 하였다. 이러한 다양한 생장율, 즉 물질의분배비의 다양(0.16~0.98)은 생장과 더불어 빚어지는 제약을 극복하여 태양 에너지를 최대한 효율적으로 흡수하기 위한 가지의 배치로 해석되며, 주어진 환경하에서 백목련이 택하는 최적의 생장 전략인 것으로 생각되다.

영양사의 기호도에 따른 학교급식에서 전통음식의 이용실태와 활용증대 방안 (Utilization Status and Efforts to Increase Usage of Traditional Foods in School Lunch according to Dietitians' Preference)

  • The study was conducted to assess school dietitians' preferences for traditional foods, to determine the association with their perception and usage status, and to develop the strategies to increase its utilization of traditional foods in school lunches. The information was obtained by the self-administered questionnaire from the subjects, which were 198 dietitians in schools located in Seoul and Gyeonggi-do. According to the preference for traditional foods, subjects were distributed into three groups (High = 54, Medium = 128, Low = 16). The significances of differences between groups were determined using SPSS 12.0 at p < 0.05. Compared to the dietitians with lower (medium and low) preferences, those with high preferences answered that they had better knowledge (p < 0.001) for traditional foods, and served them more in school lunches (p < 0.01). Also, more dieticians in the high preference group not only felt that they need to train school administrators (p < 0.01) as well as themselves (p < 0.05), but also were currently providing students with the nutrition education on traditional foods (p < 0.05). The results presented the evidence suggesting that having dietitians with higher preference for traditional foods can offer an effective strategy to increase the opportunities for schoolchildren to be exposed to traditional foods. Development of the standard recipes for use in school lunches by the government or the association was answered as the best strategy to increase the use of traditional foods, regardless of the dietitians' preferences.

학교 정온시설 앞 식물군락 조성지역에서 도로교통소음 특성에 대한 사례연구 (A Case Study on the Characteristics of the Road Traffic Noise in Plant Communities)

  • 본 연구에서는 식물군락이 조성된 일부 학교 정온시설을 선택하여 도로교통소음 특성을 파악하고 식물군락 유무에 따른 소음의 효과를 분석하였다. 차량을 4가지로 분류할 때 전체 교통량에 가장 기여하는 차량은 소형차로 나타났고 전체 교통량 대비 도로소음에 가장 영향을 미치는 차량은 오토바이로 나타났다. 교통량(Q)과 자롱차 속력(V) 관계에서 자동차 속력이 증가할수록 소음도는 감소하였다. 하지만 자동차를 4가지로 분류한 후 각 자동차 속력의 분포별로 분석한 결과, 소형차의 경우 교통량은 $2016{\leq}Q{\leq}2388$ 및 속력 67 km/h에서 소음도가 감소한 결과를 제외한 모든 구간의 교통량에 따른 속력에서 소음도가 증가하였다. 또한 다른 차종(중형차, 대형차, 오토바이 등)에서는 교통량에 따른 차량속력에 의해 모든 구간에서 소음도가 증가하였다. 식물군락 유무에 따른 두 집단 차이를 쌍체 t 검정을 수행한 결과 두 집단간 차이(p<0.05)가 있었고 식물군락이 존재하지 않는 지역에서 주간과 야간의 차이를 이표본 t 검정을 수행한 결과 두 집단 간 차이(p<0.05)가 있었다. 하지만 식물군락이 존재할 경우 주간과 야간의 차이를 비교시 두 집단 차이(p>0.05)가 없었다.

환경 일기 쓰기를 통한 환경 소양 함양 (Cultivating Environmental Literacy through Journal Writing on the Environment)

  • This study examined the educational effects of journal writing on environmental literacy of elementary students. Thirty-one 3rd grade students were involved in the journal writing which is considered as one of the most frequently performed writing activities in elementary students. Topics for the journal writing were chosen from a textbook on the environment. Before and after the journal writing, an instrument was used to measure the environmental literacy of the students. The environmental literacy instrument was originally developed by Jin (2004) then were modified by Kim (2005) for elementary students. The results of this study are as follows. The environmental literacy of the students, on average, was improved from 140.84 (74.5%) to 161.84 (85.6%) after the journal writing and the difference was statistically significant(t=7.46, p<.001). The environmental literacy scores were significantly improved in every variable after the journal writing: ecological knowledge(t=3.60, p<.01), environmental sensitivity(t=2.54, p<.05), knowledge of environmental issues(t=3.11, p<.01), environmental attitude(t=5.92, p<.001), environmental concern(t=5.27, p<.001), sense of environmental crisis(t=2.97, p<.01), environmental issue investigation and action strategy knowledge (t=2.34, p<.05), environmental skill(t=4.86, p<.001), locus of control(t=5.63, p<.001), and responsible environmental behavior(t=4.64, p<.001). This study also shows that improvement of the students was statistically significant in each of the four levels of goals in environmental education: EE goal level I (t=4.22, p<.001), level II (t=7.53, p<.001), level III(t=5.01, p<.001), and level IV(t=5.46, p<.001). The results imply that the journal writing on the environment is effective in cultivating elementary students' environmental literacy and in achieving goals in environmental education.
