Effects of Dietary Apparent Metabolizable Energy and Protein Concentrations on Growth Performance, Nutrient Digestibility, and Quality of Korean Native Ducks (사료내 에너지 및 단백질 함량이 토종오리 생산성, 영양소 이용률 및 도체 특성에 미치는 영향)

  • The objective of this experiment was to investigate the effect of dietary apparent metabolizable energy and protein concentrations on growth performance, carcass characteristics, and nutrient digestibility of Korean native ducks. In $2{\times}2$ factorial arrangement, 1-d-old Korean native ducks were completely randomized to experimental diets with 2,800 and 2,900 kcal of AMEn/kgofdiet, respectively, from 0 to 21 d of age. From 22 to 56 d of age, experimental diets 3,000 and 3,100 kcal of, and contained 16% and 17% CP, respectively. Feed and water were provided ad libitum. And BW gain were higher during 0 to 21 d (P<0.01) in AME 2,800 kcal/kg and CP 21% diet treatment than other treatments. As AME increased from 2,800 to 2,900 kcal/kg, feed intake was decreased (P<0.05) during 0 to 21 d. From 22 to 56 d of age, the, BW gain, feed intake and were not different among the treatments. Carcass ratio, relative weight per 100 of BW (wing, back, neck, breast, and leg), nutrient digestibility, meat color and nutrient composition in breast meat the treatments. In conclusion, diets with 2,800 kcal of AME/kg and 21% CP or with 3,000 kcal of AME/kg and 17% CP at 0 to 21 d and 22 to 56 d, respectively, were more efficient.

Leaf Mineral Contents and Growth Characteristics of Strawberry Grown in Aquaponic System with Different Growing Media in a Plant Factory (식물공장형 아쿠아포닉스 시스템에서 배지 종류에 따른 딸기 잎의 무기이온 함량과 생육 특성)

  • This study was aimed to determine the effects of grow media on the mineral contents of the leaves and growth characteristics of strawberry grown under aquaponics system in a plant factory. For aquaculture, 12 fish (Cyprinus carpio) (total weight, 2.0 kg) were raised in an aquaponics tank (W 0.7 m × L 1.5 m × H 0.45 m, 472.5 L) filled with 367.5 L of water at a density of 5.44 kg·m-3 and total 34 of strawberry seedlings were transplanted in the pots filed with 200 g of orchid stone, hydroball or polyurethane sponge in the growing bed (W 0.7 m × L 1.5 m × H 0.22 m) laid out with holly acrylic sheet (140×60 mm, Ø80) on the top of the system. The pH and EC of the aquaponic solution was ranged from 7.6 to 4.9 and 0.24-0.91 dS·m-1, respectively. The concentration of NO3-N was about 28% lower than that of the hydroponic standard solution, and K, Fe and B were 10, 27 and 3.8 times lower, respectively; however, the mineral contents of strawberry leaves were in the appropriate ranges with lower contents in the leaves grown with sponge media. The organic content (OM), nitrogen (N), phosphorus (P), and potassium (K) of the sludge were 61.5, 5.72, 8.92, and 0.24%, respectively. The leaf area, leaf number, and dry and fresh weights of shoot at 81 DAT were significantly higher in the hydroball, and the average number of fruits per plant was significantly higher in both the orchid stone and hydroball. There was no significant difference in the fresh and dry weights of fruits. Integrated all the results suggest that the orchid stone and hydroball media are more effective to utilize nutrients in solid particles of aquaponic solution, compared to the polyurethane sponge.

Processing of Intermediate Product(Krill Paste) Derived from Krill (크릴을 원료로 한 식품가공용 중간소재(크릴페이스트) 가공에 관한 연구)

  • As a part of investigation to use the Anatrctic krill, Euphausia superba, more effectively as a food source, processing conditions, utilizations and storage stability of krill paste (intermediate product of krill) were examined and also chemical compositions of krill paste were analyzed. Frozen raw krill was chopped, agitated with $25\%$ of water to the minced krill and then centrifuged to separate the liquid fraction from the residue. This liquid fraction was heated at $98^{\circ}C$ for 20 min. to coagulate the proteins of krill, and it was filtered to separate the protein fraction. Krill paste was prepared with grinding the protein fraction, adding $0.2\%$ of polyphosphate and $0.3\%$ of sodium erythorbate to the krill paste for enhancing of functional properties and quality stability. The krill paste was packed in a carton box, and then stored at $-30^{\circ}C$. Chemical compositions of krill paste were as follows : moisture $78\%$, crude protein $12.9\%$, crude lipid $5.9\%$, and the contents of hazardous elements of krill paste as Hg 0.001 ppm, Cd 1.15 ppm, Zn 9.1 ppm, Pb 0.63 ppm and Cu 11.38ppm were safe for food. The amino acid compositions of krill paste showed relatively high amount of taurine, glutamic acid, aspartic acid, leucine, lysine and arginine, which occupied $55\%$ of total amino acid and also taurine, lysine, glycine, arginine and proline were occupied $65\%$ of total free amino acid. Fatty acid compositions of krill paste consist of $32.4\%$ of saturated fatty acid, $29.6\%$ of monoenoic acid and $38.0\%$ of polyenoic acid, and major fatty acids of product were eicosapentaenoic acid ($17.8\%$), oleic acid ($16.9\%$), palmitic acid ($15.3\%$), myristic acid ($8.7\%$) and docosahexaenoic acid ($8.4\%$). In case of procssing of fish sausage as one of experiment for krill paste use, Alaska pollack fish meat paste could be substituted with the krill paste up to $30\%$ without any significant defect in taste and texture of fish sausage, and the color of fish sausage could be maintained by the color of krill paste. Judging from the results of chemical and microbial experiments during frozen storage, the quality of krill paste could be preserved in good condition for 100 days at $-39^{\circ}C$.

Effect of VA Mycorrhizal Fungi on Alleviation of Salt Injury in Hot Pepper (Capsicum annuum L.) (VA 균근균(菌根菌) 접종(接種)에 의한 고추의 염류장해(鹽類障害) 경감효과(輕減效果))

  • Vesicular arbuscular mycorrhizal (VAM) fungi are known to increase plant growth as well as to enhance salt tolerance of plants where plant roots are colonized by VAM. In pot experiment, pepper was grown in soil containing 0, 200, 400, and $600P\;kg\;ha^{-1}$ with and without mycorrhizal inoculum. Pots were irrigated with saline water containing 0.5, 2.0, and $6.0dS\;m^{-1}$. At 0, 200, and $400P\;kg\;ha^{-1}$ of three EC treatments, plant hight in mycorrhizal treatments was significantly different compared to nonmycorrhizal treatments. However, plant hight at $600P\;kg\;ha^{-1}$ was not different between mycorrhizal and nomycorrhizal treatments. Leaf area at $0P\;kg\;ha^{-1}$ of three EC treatments, and $200P\;kg\;ha^{-1}$ of $6.0dS\;m^{-1}$ in mycorrhizal treatments significantly increased compared to nonmycorrhizal treatments. However, these increase were not discovered in high salinity and P level. Level of EC affected dry weight, and especially, interection of P and EC, or P and VA inoculation highly affected root dry weight. R/S ratio generally decreased in mycorrhizal treatments. Significantly decreased R/S ratio was shown at 0, 400, and $600P\;kg\;ha^{-1}$ of $6.0dS\;m^{-1}$. Chlorophyll content generally increased with decreased salinity and P level where mycorrhizal treatments showed higher chlorophyll content compared to nonmycorrhizal treatments. The benefits of VAM inoculation on fruit production was discovered at only low P level and salinity. Mycorrhizal dependency on dry weight basis was generally shown in $0P\;kg\;ha^{-1}$ of three EC treatments and 0.5, $2.0dS\;m^{-1}$ of $200P\;kg\;ha^{-1}$ level. Colonization rate ranged 3.3 to 43.3% and number of spores was 47.7 to 198.3 $100g^{-1}$ soil. Colonization rate and number of spores increased with decreased P level and salinity where there was high correlation ($r=0.858^{**}$) between both. Also improved uptake of mineral nutrients was discovered at mycorrhizal treatments in decreased P level and salinity.

Development and Validation of the Analytical Method for Oxytetracycline in Agricultural Products using QuEChERS and LC-MS/MS (QuEChERS법 및 LC-MS/MS를 이용한 농산물 중 Oxytetracycline의 잔류시험법 개발 및 검증)

  • An analytical method was developed for the determination of oxytetracycline in agricultural products using the QuEChERS (Quick, Easy, Cheap, Effective, Rugged and Safe) method by liquid chromatography-tandem mass spectrometry (LC-MS/MS). After the samples were extracted with methanol, the extracts were adjusted to pH 4 by formic acid and sodium chloride was added to remove water. Dispersive solid phase extraction (d-SPE) cleanup was carried out using $MgSO_4$ (anhydrous magnesium sulfate), PSA (primary secondary amine), $C_{18}$ (octadecyl) and GCB (graphitized carbon black). The analytes were quantified and confirmed with LC-MS/MS using ESI (electrospray ionization) in positive ion MRM (multiple reaction monitoring) mode. The matrix-matched calibration curves were constructed using six levels ($0.001{\sim}0.25{\mu}g/mL$) and coefficient of determination ($r^2$) was above 0.99. Recovery results at three concentrations (LOQ, $10{\times}LOQ$, and $50{\times}LOQ$, n=5) were from 80.0 to 108.2% with relative standard deviations (RSDs) less than of 11.4%. For inter-laboratory validation, the average recovery was in the range of 83.5~103.2% and the coefficient of variation (CV) was below 14.1%. All results satisfied the criteria ranges requested in the Codex guidelines (CAC/GL 40-1993, 2003) and the Food Safety Evaluation Department guidelines (2016). The proposed analytical method was accurate, effective and sensitive for oxytetracycline determination in agricultural commodities. This study could be useful for safety management of oxytetracycline residues in agricultural products.

Effects of Vitamin E on the Changes of Mineral Contents in Chronic Cadmium-Poisoned Rats

  • This study was carried out to investigate the effects of vitamin E on the cadmium contents of bone and on the calcium and phosphorous contents of the blood, urine and feces. Male Sprague-Dawley rats weighing 100$\pm$10g were randomly assigned to one normal group and three cadmium poisoned groups. The cadmium poisoned groups consisted of a vitamin E free diet (Cd-0E) group; a 40 mg vitamin E /kg diet (Cd-40E) group; and a 400 mg/kg diet (Cd-400E) group. Experimental animals were maintained on their respective diets for 20 weeks and were simultaneously administered 50 ppm $Cd^{2+}$ dissolved in the drinking water. At the end of the trial, the average hematocrit value in the Cd-0E group was 28.13% lower than in the normal group. However, the average hematocrit value in the Cd-400E group was significantly higher than in the Cd-0E and Cd-40E groups. WBC levels in the cadmium-poisoned groups were lower than in the normal group, but Cd-400E group levels were significantly higher than in the Cd-0E and Cd-40E groups. The contents of calcium of tibia has no significant difference between normal group and cadmium exposed group at $10^{th}$ week After 20 weeks, the calcium contents of the tibia in the Cd-0E and Cd-40E groups were lower than in the normal group by 25.5% and 22.1 %, respectively, although the calcium contents of the tibia in the Cd-400E group were higher than in the normal group. After 10 weeks, the calcium contents of the femur in the Cd-0E and Cd-40E groups were 19.25% and 15.45% lower than in the normal group, respectively, but the calcium contents of the femur in the Cd-400E group were at the same levels as in the normal group. The levels of calcium in the femur after 20 weeks were similar to the 10-week levels. Calcium levels of the urine in the Cd-0E and Cd-40E groups were 3.92 fold and 2.92 fold higher, respectively, than in the normal group, but levels in the Cd-400E group were significantly lower than in either the Cd-0E group or the Cd-40E group. Calcium levels of the feces in cadmium-poisoned groups were significantly higher than in the normal group, although levels in the Cd-400E group were significantly lower than in the Cd-0E and Cd-40E groups. Phosphorous levels of the blood in the Cd-0E group were 17% lower than in the normal group, although levels in the Cd-400E group were significantly higher than in the Cd-0E group. Phosphorous levels of the urine in the Cd-0E and Cd-40E groups were significantly higher than in the normal group, while Cd-400E group levels were found to be at the same level as in the normal group. Cadmium contents of the tibia in the Cd-40E and Cd-400E groups were 13% and 17% lower, respectively, than in the Cd-0E group. Regarding cadmium levels in the femur, only the Cd-400E group achieved lower levels (10% lower) than the Cd-0E group. In conclusion, vitamin E supplementation resulted in a suppression of the release of calcium from bone, and a reduction in the excretion of calcium via the urine and feces, thus having a normalizing effect on calcium metabolism in rats with chronic cadmium poisoning.

Protective Effects of Phellinus linteus and Curry-Added Cooked Mixed Grain Rice Extracts on Oxidative Stress-Induced LLC-PK1 Cell Damage (상황버섯 및 카레를 첨가한 잡곡밥 추출물의 LLC-PK1 세포에서의 산화적 스트레스 보호 효과)

  • The aim of this study was to investigate the protective effects of methanolic extracts of cooked mixed grain rice samples, including grain rice (sorghum, black bean, proso millet, and Job's tears) mixed with fermented brown rice (GR), GR added with 0.5% water extract of Sanghwang mushroom (GRS) or 0.1% curry (GRK), and traditional five grain mixed rice (TMR, Ohgokbap), on $H_2O_2$-induced oxidative injury in LLC-PK1 pig renal epithelial cells. White rice (WR) was used as a positive control. Cells were first exposed to $H_2O_2$ ($250{\mu}M$) for 4 hr, followed by treatment with $100{\mu}g/mL$ of different GR extracts for 24 hr. $H_2O_2$ significantly induced cell damage (P<0.05). Cellular levels of reactive oxygen species (ROS), lipid peroxidation, and antioxidant enzymes, including catalase (CAT), superoxide dismutase (SOD), and glutathione peroxidase (GSH-px), were measured. In addition, mRNA levels of antioxidant enzymes were determined by RT-PCR assay. Mixed grain rice, particularly GRS and GRK, were able to reduce cellular levels of ROS, decrease lipid peroxidation, and also increase mRNA expression of antioxidant enzymes compared to other samples. These results suggest that mixed grain rice, specifically GRS and GRK, have strong protective effects against $H_2O_2$-induced oxidative injury in LLC-PK1 cells through inhibition of lipid peroxidation, reduction of ROS levels, and elevation of antioxidant enzyme activities.

Human Impact on Sedimentary Environment of Estuarine Coastal Salt Marches, Southern Coastal Region of Korea Peninsula (인위적 환경변화에 따른 해안지역 퇴적환경의 변화)

  • An estuary is semi-inclosed inlets, located between terrestrial and marine environment. Since many estuaries along south-western coasts of Korean peninsula were affected by human settlements and activities, significant changes in sedimentation environments have been observed. The research area is divided into three distinct morpho-stratigraphic units: fluvial dominated area(Area1), mixed area(Area 2), tide-dominated area(Area3). The landform of this area has been changed by reclamation and river channel change. Temporal variations affected by dam construction, periodic freshet was iterrupted. Sediments began to continuously accmulate on estuary banks by tide. Meanwhile, because of the continuous but reduced discharge of fresh water, the salinity of estuarine sediments was declined. That processes made vegetated area( Phregmites lonivalvis and Suaeda japonica) to be expanded. It indicates that the magnitude and frequency of geomorphic processes has been significantly changed.

Bioequivalence of Burophil Capsule to Surfolase Capsule (Acebrophylline 100 mg) (설포라제 캡슐(아세브로필린 100 mg)에 대한 부로필 캡슐의 생물학적 동등성)

  • Acebrophylline is a compound produced by salifying ambroxol with theophylline-7 -acetic acid. After acebrophylline administration, the salt splits into these two components which feature a peculiar pharmacokinetic behavior, an adequate ambroxol and a low theophylline-7-acetic acid serum levels. The purpose of the present study was to evaluate the bioequivalence of two acebrophylline capsules, Surfolase (Hyundai Pharm. lnd. Co., Ltd.) and Burophil (Kuhnil Pharm. Co., Ltd.), according to the guidelines of the Korea Food and Drug Administration (KFDA). The release of ambroxol from the two acebrophylline formulations in vitro was tested using KP VIII Apparatus II method with various dissolution media (pH 1.2, 4.0, 6.8 buffer solution and water). Twenty eight healthy male subjects, $23.25{\pm}1.43$ years in age and $64.82{\pm}6.77$ kg in body weight, were divided into two groups and a randomized $2{\times}2$ cross-over study was employed. After two capsules containing 100 mg as acebrophylline were orally administered, blood was taken at predetermined time intervals and the concentrations of ambroxol in serum were determined using HPLC with electrochemical detector (ECD). The dissolution profiles of two formulations were similar at all dissolution media. In addition, the pharmacokinetic parameters such as $AUC_t$, $C_{max}$ and $T_{max}$ were calculated and ANOVA test was utilized for the statistical analysis of the parameters using logarithmically transformed $AUC_t$, $C_{max}$ and untransformed $T_{max}$. The results showed that the differences between two formulations based on the reference drug Surfolase, were -1.64, -3.33 and -0.92% for $AUC_t$, $C_{max}$ and $T_{max}$, respectively. There were no sequence effects between two formulations in these parameters. The 90% confidence intervals using logarithmically transformed data were within the acceptance range of log 0.8 to log 1.25 $(e.g., \;log\;0.93{\sim}log\;1.05\;and\;log\;0.88{\sim}log\;1.05$ for $AUC_t$, and $C_{max}$, respectively). Thus, the criteria of the KFDA bioequivalence guideline were satisfied, indicating Burophil capsule was bioequivalent to Surfolase capsule.

Effects of Liquid Culture of Coriolus Versicolor on Lipid Metabolism and Enzyme Activities in Rats Fed High Fat Diet (운지버섯 균사체 배양액이 고지방 식이를 급여한 흰쥐의 지질대사 및 효소활성에 미치는 영향)

  • The effects of liquid culture of Coriolus versicolor on weight gain, food intakes, food efficiency ratios, serum and hepatic lipid concentrations, serum protein levels and serum enzyme activities, were studied in growing male rats. Sprague-Dawley rats were given four different types of diets for a succeeding period of 5 weeks, respectively: a normal diet group (7% corn oil), a high fat diet group (7% corn oil+15% lard), a 20% or 30% C. versicolor diet groups (high fat diet+20% or 30% C. versicolor in water) according to the levels of C. versicolor supplementation. The body weight gains of the rats fed the 30% C. versicolor diets were lower than those in the rats fed high fat diet. The epididymal fat pad weight of the rats fed high fat diet and 20% or 30% C. versicolor diets were significantly higher than that of the rats fed normal diet. The concentrations of triglyceride in the serum and the liver of the rats fed the 30% C. versicolor diets were more significantly decreased compared to rats on the high fat diet. The concentrations of total cholesterol in the serum and the liver of rats fed the high fat diet, 20% and 30% C. versicolor diets were similar to those of rats fed the normal diet. The HDL-cholesterol concentration and the HDL-cholesterol/total-cholesterol ratio of the rats fed 20% and 30% C. versicolor diets were significantly lower than those of the rats fed high fat diet. But the antherogenic index of the rats fed 20% or 30% C. versicolor diets were significantly higher than those of the rats fed high fat diet. There were no differences in the activities of glutamic oxaloacetic transaminase, glutamic pyruvic transaminase and alkaline phosphatase in the serum among the experimental groups. These results showed that the 30% C. versicolor diet feeding decreased the triglyceride in serum and liver of the rats.
