Comparative Simulation of 3-zone SMB (Simulated Moving Bed) and 4-zone SMB for IgY (Immunoglobulin Yolk) Purification (IgY (Immunoglobulin Yolk) 분리를 위한 3-영역 SMB (Simulated Moving Bed)와 4-영역 SMB 비교전산모사)

  • IgY (Immunoglobulin Yolk) is a specific antibody in egg yolk, and it protects human body from virus and antigen. There are a lot of egg yolk components such as lipoprotein and protein. To separate IgY, HPLC (High Performance Liquid Chromatography) and precipitation were used in a batch mode and SMB (Simulated Moving Bed) was adopted for continuous purification of yolk proteins. IgY and other proteins in yolk were separated by using three-zone and four-zone SMB chromatography. Before performing SMB experiments, batch chromatography simulation parameters and adsorption isotherms were obtained. The parameters of batch chromatography were used to simulate SMB using Aspen chromatography. To compare three-zone and four-zone SMB chromatography, simulations in $m_2-m_3$ plane on the triangle theory were carried out. In terms of concentration and purity of both IgY and other lipoproteins, 3-zone SMB process is considered as ideal at the vertex of triangle ($m_2$, $m_3$=0.1, 1.1). 4-zone SMB yields the highest IgY purity at the coordinate ($m_2$, $m_3$=0.06, 0.5), which is the pure raffinate region. In 3-zone SMB without recycle, other lipoproteins in extract are largely affected in purity by small shift from the vertex of triangle ($m_2$, $m_3$=0.1, 1.1).

  • A new one-column chromatography process, analogous to a four-zone simulated moving bed (SMB), was presented. The basic principle of the process was identical to that of a four-zone SMB. The process consisted of one chromatographic column and four tanks, instead of the four columns in the four-zone SMB (1-1-1-1), and has been used for the separation of two amino acids, phenylalanine and tryptophan, using an ion exchange resin. The operating parameters for the one-column process and four-zone SMB were obtained from equilibrium theory. Computer simulations were used to compare the performances of the new one column process to that of the general four-zone SMB, using Aspen $Chromatography^{TM}$ v 11.1. The differences between the one-column and SMB processes in terms of the purities and yields of phenylalanine and tryptophan were less than 4 and about $6\%$, respectively. The lower purities of the one-column process were due to the loss of the developed concentration profiles in the column when the liquid was stored in tanks. The one-column process gave great flexibility, and would be useful for reconstructing an existing conventional chromatography process to one of a SMB.

  • SMB (simulated moving bed) is a continuous chromatographic process by shifting periodically port position. Binary of mixture, Blue dextran and Orange G, was separated by SMB. These components have unique color individually, that is, Blue dextran is blue and Orange G is orange. It is easy to understand SMB process by observing the shift of color changes in SMB. These components was not adsorbed to stationary phase and isolated by difference of size exclusion factor. Mass transfer coefficient was determined by single pulse test under several flow rate conditions. Operation condition was obtained by standing wave theory and optimized for high purities in extract and raffinate streams. Experiment was performed in open loop 4 zone (2-2-2-2) SMB. There are several advantages in open loop SMB, where extract is product for high purity. It is also easy to control flow rate and monitor experimental state during operation. Experimental, extract and raffinate history is well fitted with simulation results, however, column concentration profile is a little different from simulation results. Purities were 99.5% for extract and 98.9% for raffinate and extract and raffinate yields were obtained as 98.9% and 99.4% respectively.

  • SMB (simulated moving bed) chromatography is a very useful utility for the separation of binary system. We simulated the separation of L-arabinose and L-ribose from the mixture by using lab-scale 4(1-1-1-1)-zone SMB chromatography. Preliminary experiments of PIM (pulse input method) were performed to measure adsorption isotherms of L-ribose and L-arabinose in $NH_2$ HPLC column, and experimental and simulated results from ASPEN chromatography were compared. To find the most suitable separation condition in SMB, we carried out a simulation in $m_2-m_3$ plane base on the triangle theory and calculated operating parameters (flow rate of four zone, switching time and feed concentration and so on) using ASPEN chromatography under the conditions of linear isotherms obtained from PIM.

  • Conventional SMB process is operated using 4-zone having several chromatography columns in each zone. Unlike batch chromatography, SMB process can continuously separate binary materials. Both high productivity and purity are obtainable by using SMB process. In this study, the simulation on Thermal Simulated Moving Bed Concentrator (TSMBC) which is a SMB process with thermal swing adsorption was carried out. The advantage of TSMBC is that adsorption isotherm can be easily controlled by thermal wave or direct heating. Recovery of pure water and concentration of phenol was studied in simulation. To verify environmental-friendly potential of TSMBC, DOWEX $1{\times}4$ was chosen as an adsorbent and phenol was selected as a target material. When 3 columns were used in this study, concentration of phenol is 2.29, 2.28 and 1.31 times higher than injected sample. However, a contamination of phenol in solvent port was found, probably due to the restriction of adsorption isotherm of phenol on DOWEX $1{\times}4$.

  • Currently, researches on recycling and reuse of waste energetic materials have recently gained a great attention from advanced countries due to ever tightening environmental regulations. In this study, as a part of a recycling technology, the experiments and dynamic simulation of simulated moving bed (SMB) process were performed to efficiently separate TNT and RDX from their mixture, which are main components of ammunition. In order to determine the operation zone of SMB process, the retention times of TNT and RDX were measured using HPLC at different flow rates and the adsorption equilibrium of each component was obtained by using a moment method. According to the adsorption equilibrium and the triangle theory of SMB process, four operation points were determined and separation experiments were carried out by the SMB process using the solvent consisting of acetonitrile and water. Two different mixing ratios (6:4 and 1:1) of acetonitrile and water were chosen for the experiment due to the great impact of mixing ratio of the solvent on separation. The performance of SMB process was evaluated by purity, recovery, productivity and solvent consumption. Pure TNT and RDX were successfully obtained from the SMB process and the dynamic simulation for the SMB process agreed well with the experimental results. Therefore, the dynamic model could be applied for predicting the dynamic behavior of the SMB process and designing a large scale SMB process.

  • Simulated moving bed (SMB) process is a continuous chromatographic technology used to separate a large amount of petrochemicals, fine chemicals, pharmaceuticals, and so on, drawing a great attraction of related industries. With the recent development of biotechnology, the SMB process has been adopted for the separation of various useful bio-products. Attempts to understand the separation mechanism of the SMB process in many aspects are reported in many publications. These researches have dealt with the improvement of SMB for easier operation and solving problem in process. The feed mixture fed into the SMB process may be of different concentration batch by batch rather than in uniform concentration. Retention behaviors of feed (psicose (A) and fructose (B) mixture) existing in the SMB unit in dynamic steady-state and feed (psicose (C) and fructose (D) mixture) newly injected into the SMB were analyzed. It was observed that the existing components, (A) and (B), were eluted relatively faster as the injection time of new feed was earlier during the port-switching period. In addition, the components (C) and (D) were eluted earlier as fresh feed was injected earlier in a port-switching time.

  • Using strong cation exchange simulated moving bed (SMB) chromatography, a nitrogen-phosphate-potassium (NPK) fertilizer is produced in a cost-effective manner. The SMB process operated in a non-traditional way is divided into production and regeneration sections for exclusion of undesirable ions, and composed of six zones including two wash-water zones. This paper addresses modeling and simulation studies on the ion-exchange SMB process, comparing simulation results with experimental data obtained both from a pilot plant and an industrial plant. The simulation results show a good agreement with in situ experimental data obtained in the two plants. The model equation validated by the experiments will be applicable for optimization problems to obtain optimum operating conditions of the process.

  • Nanostructured materials have received attention due to their unique electronic, optical, optoelectrical, and magnetic properties as a results of their large surface-to-volume ratio and quantum confinement effects. Thermal chemical vapor deposition process has attracted much attention due to the synthesis capability of various structured nanomaterials during the growth of nanostructures. In this study, silicon oxide nanowires were grown on Si\$SiO_2$(300 nm)\Pt(5~40 nm) substrates by two-zone thermal chemical vapor deposition with the source material $TiO_2$ powder via vapor-liquid-solid process. The morphology and crystallographic properties of the grown silicon oxide nanowires were characterized by field-emission scanning electron microscope and transmission electron microscope. As results of analysis, the morphology, diameter and length, of the grown silicon oxide nanowires are depend on the thickness of the catalyst films. The grown silicon oxide nanowires exhibit amorphous phase.

  • Countries such as the United States, Russia, and Europe are developing and operating UAVs for various purposes, including stealth UAVs. North Korea is also operating unmanned aerial vehicles, and it is presumed that it is continuously flying south of the demarcation line to obtain information on the national security zone, but it is difficult to detect and neutralize it effectively. Therefore, this paper analyzed the military UAV development trends and military radar specifications in Western countries such as the United States and communist countries such as Russia, China, and North Korea through literature research. In addition, based on the investigation of the AESA radar-based UAV response system in the country, not only general responses but also countermeasures such as direct strike and electromagnetic pulses to North Korean UAVs that cannot be jammed were suggested.
