Ginsenoside, Phenolic Acid Composition and Physiological Significances of Fermented Ginseng Leaf (발효처리가 인삼잎의 진세노사이드 및 페놀산 조성 변화와 생리활성에 미치는 영향)

  • This study was carried out to investigate the compositional changes of ginsenosides and phenolic acids of ginseng leaf by fermentation in order to promote the utilization of ginseng leaf. The chief ginsenosides in non-fermented ginseng leaf (NFGL) were ginsenoside-Rg1 (26.0 mg/g), -Re (47.3 mg/g) and -Rd (23.9 mg/g). By fermentation, ginsenoside-Rg1, -Rb1, -Rb2, -Rb3, -Rc and -Re were decreased tremendously and new ginsenoside-Rh2, -Rh1, -Rg2 and -Rg3 appeared. Especially, ginsenoside-Rg3 (3.7 mg/g) on FGL was increased 15-fold compared to that of NFGL (0.2 mg/g). Total phenolic compound content of NFGL and FGL measured by colorimetric analysis was 350.4 and 312.5 mg%, respectively. There were 8 free and 6 ester forms of phenolic acids in NFGL. Among them, content of ferulic acid was the highest, comprised of 12.6 and 50.7 mg%, respectively. In FGL, total content of protocatechuic acid, p-hydroxybenzoic acid, and vanillic acid were increased by 28, 5 and 7.8 fold and ferulic acid was decreased greatly. Tyrosinase inhibitory activity of FGL was stronger than NFGL, while electron donating abilities of FGL were similar to NFGL.

Differences in Phenolic Compounds between Korean Ginseng and Mountain Ginseng (고려인삼과 장뇌삼의 페놀성 성분 비교 연구)

  • Differences in phenolic $\infty$mpounds were observed between cultured and mountain ginsengs. Cinnamic acid and p-hydroxy­b benzoic acid in Korean mountain ginseng and Chinese mountain ginseng were much higher than those in Korean ginseng. C Contents of the esculetin in Korean cultured ginseng and Korean mountain ginseng were higher than that in Chinese m mountain ginseng. The highest contents of esculetin in Korean mountain ginseng was$47.2\mu\textrm{g}/g$. Contents of the ferulic acid a and caffeic acid in red $\infty$lored Korean mountain ginseng were higher than any other ginseng.

  • Bromoxynil is an antidicot herbicide widely used on cereal crops and has a short half life in the soil. A bxn gene, encoding a specific nitrilase that converts bromoxynil to its primary metabolite 3,5-dibromo-4-hydroxybenzoic acid, was inserted in plant binary vector pGA482, and then introduced into tobacco and lettuce plants via Agrobacterium mediated leaf-disc transformation method. Transgenic plants with the bxn gene were selected by kanamycin and regenerated to whole plants. The regenerated transgenic plants were determined level of expression of bxn gene by Northern blot analysis. Leaf-disc analysis and pot-assay confirmed that the transgenic tobacco and lettuce plants were resistant to high doses of bromoxynil.

Bioassay of Allelopathy Substance Related Injury by Successive Cropping in Alfalfa(Medicago sativa L.) (알팔파 연작장해에 관여하는 타감작용 물질의 탐색 및 생물검정)

  • This experiments were conducted to determine the effect of allelopathy and autotoxicity in alfalfa. Among several alfalfa cultivars, the substances exudated from seven cultivars including Medicago sativa, c.v. Luna, Sparta, Magnum, Husky, Milkmaker, Challenger and Anchor inhibited the germination and seedling growth of radish, ranging from 60 to 80%. When allelopathic substances were exudated from the soil of the alfalfa-cultivated field, the increased levels of substance retarded the germination rate and seedling growth of radish and alfalfa. The inhibition rate was about 80%, compared with that of control. Thus, this study indicates that the allelopathy and autotoxicity substances of alfalfa influenced the germination and seedling growth in alfalfa itself and radish. When allelopathic substances were analysed by using gas chromatograph, several kinds of phenol compounds were detected as follows; Salicylic acid, hydroxybenzoic acid, vanillic acid, syringic acid, coumaric acid, and ferulic acid.

  • The effect of butylated hydroxytoluene (BHT) and its major metabolite, 3, 5-di-tert-butyl-4-hydroxybenzoic acid (BHT-acid) on the uptake of taurocholate into hepatocytes was studied using the primary culture of rat hepatocytes. Hepatocyte were isolated by an in situ collagenase perfusion technique and maintained as a monolayer in serum-free meadia for 24 hours before use. The uptake of taurocholate was saturable with an apparent Km of 12.8+2.8 MuM and Vmax of 0.18+0.01 nmol/mg/min. Both BHT and BHT-acid inhibited the hepatocellular uptake of taurocholate when they were added to the culture.

  • In order to reduce energy input in direct ethanol production from raw starch by co-immobilized Aspergillus awamori(A) and Zymomonas mobilis(Z), A-Z 36 culture system which was changed to anaerobic after 36 h of aerobic fermentation without sterilization was investigated. This immobilized cell system can not be carried out under unsterile conditions because of growth of microbial contaminants from original medium. Among some food additives such as sorbic acid, benzoic acid, dehydroacetic acid, p-hydroxybenzoic acid, Vantocil IB and Neupectin-L, Vantocil IB and Neupectin-L were a potent antibacterial agent in A-Z 36 culture cell system and were not affected in hydrolysis of substrate as compared with the case of control. Ethanol yield(6.9 g/l) in system of addition of 0.1% Neupectin-L was slightly higher than that in control(6.4 g/l). When 2% starch was fed five times in fed-batch culture with 0.1% Neupectin-L, ethanol yield and productivity were 34 g/l and 2.0 g/l/day, respectively.

  • The 12 compounds were isolated from MeOH extract of Magnolia biondii and their structures were identified as seven lignans, two phenolics, one coumarin, and two flavonoid compounds, respectively. Among these constituents, tiliroside (3), kaempferol-7-methyl ether (4), 4-hydroxybenzoic acid (5), vanilic acid (6), and scopoletin (9) were isolated from Magnolia biondii for the first time.

  • A phytochemical investigation of Spergularia marina (L.) Griseb. growing in Egypt, has been carried out, which resulted in the isolation of seven compounds from the different extracts of the plant namely; ${\beta}$-sitosterol glucoside, tricin (1) dihydroferulic acid (2), vanillic acid (3), 4-hydroxybenzoic acid (4), uracil (5) and 8-hydroxy cuminoic acid (6) Structure elucidation of the isolated compounds was carried out using different spectroscopic techniques. This is the first report for the isolation of these compounds from genus Spergularia. Furthermore, 8-Hydroxy cuminoic acid and uracil were isolated for the first time from family Caryophyllaceae. The chemical composition of the volatile components present in the petroleum ether extract of Spergularia marina (L.) Griseb. using combined gas chromatography-mass spectrometry (GC-MS) is reported here for the first time. Of the 97 components present, 59 were identified including three sulfur containing compounds which represented about 1.8% of the volatiles of the total petroleum ether extract. This prompted us to study and report its possible antimicrobial activity. In addition, the antibacterial and antifungal screening of different extracts of Spergularia marina (L.) Griseb. as well as some isolates have been performed using agar diffusion method.

Phenolic Constituents of Boehmeria pannosa and α-Glucosidase Inhibitory Activity (왕모시풀의 페놀성 성분 및 항당뇨 활성)

  • Boehmeria pannosa (Urticaceae) is a perennial herb widely distributed in Korea. In this study, investigation on the constituents of B. pannosa was conducted by chromatographic techniques and spectroscopic analysis. As a result, nine compounds including eight phenolic compounds, 3-hydroxy-1-(4-hydroxyphenyl)propan-1-one (1), β-hydroxypropiovanillone (2), C-veratroylglycol (3), 3-hydroxy-1-(4-hydroxy-3,5-dimethoxyphenyl)-propan-1-one (4) 2,3-dihydroxy-1-(4-hydroxy-3,5-dimethoxyphenyl) -1-propanone (5), p-coumaric acid (6), 4-hydroxybenzoic acid (7), vanillic acid (8) and one lignan, (-)-(7R,8S)-dihydrodehydrodiconiferyl alcohol (9) were isolated from the EtOAc-soluble fraction of B. pannosa. Among them, compounds 5, 6 and 9 inhibited α-glucosidase inhibitory activity more than 50% at the concentration of 100 μM.

In vitro Free Radical Scavenging and Hepatoprotective Compound from Sanguisorbae Radix

  • In the course of searching for hepatoprotective agents from natural products, four compounds were isolated from the MeOH extract of Sanguisorbae Radix, as guided by their DPPH free radical scavenging activity. The structures were determined as 4,5-dimethoxy-3-hydroxybenzoic acid methyl ester (1), (+)-gallocatechin (2), methyl $6-O-galloyl-{\beta}-D-glucopyranoside$ (3), and pomolic acid $3-O-[{\alpha}-L-arabinopyranoside]-28-O-[{\beta}-D-glucopyranosyl]$ ester (ziyu-glycoside I) (4). Compounds 2 and 3 showed significant DPPH free radical scavenging effects, exhibiting $IC_{50}$ values of 11.4 and $13.0\;{\mu}M$, respectively. L-Ascorbic acid was used as a positive control and exhibited the $IC_{50}$ value of $50.3\;{\mu}M$. In evaluation of the hepatoprotective activity of the isolated compounds on drug-induced cytotoxicity, compound 2 showed the significant hepatoprotective effect with the $EC_{50}$ value of $91.84\;{\pm}\;11.0\;{\mu}M$ on tacrine-induced cytotoxicity in Hep G2 cells, while silybin, a positive control, exhibited $EC_{50}$ value of $122.4\;{\pm}\;12.5\;{\mu}M$.
