상처치유과정중 관상어류 피부 색소체계의 재분화경로를 규명하기 위하여 외부 색채가 화려한 담수산 금붕어 (Carassius auratus L.)를 실험재료로 하여 피부의 일정부위에 인위적인 상처를 유도한 후, 시간 경과에 따른 조직과 색소 체계의 치유과정을 고배율의 투과 전자현미경으로 관찰하였다. 금붕어 피부 색소세포는 정상조직에서 황색소세포, 백색소세포 그리고 혹색소세포 등 세 종류의 진피성 색소세포로 이루어져 있었다. 황색소세포에는 pterinosome과 carotenoid vesicle 등 두 종류의 색소과립이 분포되어 있었고, 백색소세포와 혹색소세포에는 무정형 색소결정인 leucosome과 전자밀도가 높은 구형의 색소과립인 melamosome이 각각 함유되어 있었다. 초기 상처치유반응은 상처 유도직 후에 표면 손상부위로 전이되는 상피세포와 혈구세포에 의하여 수행되었다. 상처 유도후 $5\sim7$일이 경과된 조직의 표본에서는 조면소포체가 특이하게 발달되어 진피성 색소세포의 공통 원기로 추정되는 세포의 출현이 확인되었다. 또한 재생된 조직내에서 재분화된 색소세포는 상처유도 후 3주가 경과된 표본에서 처음 관찰되었다. 색소과립의 재분화 경로는 세포 내에서의 색소과립 형성 과정과 마찬가지로 색소세포내에 잘 발달된 조면소포체와 골지체를 경유한 후, 분비소포의 형태로 생성림이 확인되었다. 그리고 재분화된 색소세포로 유입되는 색소 원기물질은 음세포과립을 통하여 수송되었는데, 특히 조면소포체가 풍부한 원기세포와 연접된 색소세포의 원형질막에서 매우 활발한 물질의 수송이 관찰되었다. 한편, 각 색소세포의 일차적인 재분화과정은 상처 유도후 4주가 경과되어 피부 상처가 치유되는 시점을 전후하여 완료되었으나, 재분화된 색소세포의 수나 분포밀도는 충분한 체색발현을 위한 상태에 비해 크게 미달되는 것으로 분석되었다. 따라서 특별한 환경의 변화가 없는 한, 상처유도 이전의 색소체계와 동일한 상태로의 복구에는 적어도 3개월 이상이 소요되는 것으로 확인되었다.
태반유래 중간엽줄기세포(PD-MSCs)는 재생의학에서 세포기반치료제로 잘 알려진 세포군이다. PD-MSCs의 손상된 부위로의 이동과 호밍 기능은 MSC 생착의 중요한 특성이다. miRNA는 최근 MSC의 증식, 생존 이동과 같은 중요한 기능을 조절하는 것으로 알려져 있다. 본 연구의 목적은 담관결찰(BDL) 쥐 모델에서 PD-MSCs 호밍에 관련된 miRNA 및 표적 유전자를 동정하는 것으로, 마이크로어레이 분석을 이용하여 PD-MSCs 호밍에 관여하는 유전자 표적 miRNA를 선별하였다. BDL 쥐모델에 PD-MSCs을 이식한 일주일 후 간 조직에서 PD-MSCs 생착여 부는 면역형광분석법과 qRT-PCR에 의한 인간 Alu유전자 발현으로 확인되었다. 저산소 및 정상조건(Hyp/Nor)에서 이동한 PD-MSC에 비하여, PD-MSCs 이식한 BDL군 간 조직에서 miRNAs 발현의 차이가 크게 나타났으며, PD-MSCs 호밍 관련 miRNA와 표적유전자를 검증하였다. miR199a-5p 및 miR-148a-3p에 대한 표적 유전자 인테그린 α4 (ITGA4)와 α5 (ITGA5)의 발현은 이식(Tx)그룹에서(p<0.05) 유의하게 상향 조절되었다. 또한 인테그린 β1 (ITGB1)과 β8 (ITGB8)의 발현은 miR-183-5p 및 miR-145-5p억제에 의하여 크게 증가되었다. 따라서 이러한 결과는 BDL에 의해 손상된 쥐간에서 PD-MSCs가 호밍효과을 위해 인테그린 그룹과 관련된 miRNA 발현 조절에 관여함을 나타내었다. 본 연구결과는 miRNA에 의한 인테그린 그룹 조절기능이 BDL에 의해 유도된 간섬유증 쥐모델에서 PD-MSCs의 치료효과에 기여할 수 있음을 시사한다.
Heat shock protein 70 (HSP70)은 다양한 질병상태와 치명적인 열 손상에서 세포 및 조직을 보호하는데 중요한 역할을 하며, 또한 외부의 stress로부터 세포내 단백질의 파괴와 변화를 감소시키는 단백질로 알려져 있다. 본 연구에서는 오랜 기간 동안 조혈기관의 치료제로 콩팥에서도 세포보호효과가 있는 것으로 알려진 Erythropoietin(EPO)을 투여하여 콩팥내의 HSP70의 발현변화를 세포수준에서 관찰하고자 하였다. Sprague-Dawley계 흰쥐를 사용하여 전자현미경적 면역조직화학법으로 rHuEPO투여군과 대조군에서 HSP70의 발현변화를 관찰하였다. 대조군에서 HSP70은 콩팥의 바깥수질과 속수질에서 관찰되었으며, 특히 속수질에서 강하게 발현되었고 그 부위는 속수질집합관세포와 헨레고리의 내림가는 부분이었다. EPO 투여군에서는 속수질과 바깥수질의 내림가는 부분에서는 발현변화가 관찰되지 않았으나, 바깥수질의 집합관 세포에서 발현이 급격히 증가함을 관찰되었다. 특히 대조군에서의 핵주변부위뿐 아니라 세포내 핵상부분을 비롯한 세포막주변부위에도 강한 면역 염색성을 나타내었다. 이러한 결과는 콩팥의 바깥수질에서 stress성 단백질인 HSP70의 조절기전이 EPO에 의해 매개됨을 보여주면, 세포 stress및 질병상태에서도 이러한 기전이 작용할 것으로 생각된다.
흡수성 차페막을 이용한 조직 유도 재생술시 차폐막의 견고성으로 미루어 보아 재생을 위한 공간의 유지가 어려울 수 있다. 조직 유도 재생술과 함께 골이식술을 시행함으로써 공간 확보와 함께 적절한 혈병의 유지를 도모할 수 있고 이식된 골은 선생골 형성을 위한 핵으로 작용할 수도 있다. 최근에 사람의 치아회분말과 연석고를 혼합한 골이식재가 여러 연구를 통해 좋은 골이식재로 평가되었다. 본 연구에서는 성견 하악 소구치 2급 치근이개부위에 외과적으로 형성하여 흡수성 차폐막과 치아회분말-연석고 혼합 이식재를 이용한 조직유도재생 술을 시행하여 치주 조직 재생의 양상을 조직학적으로 관찰하고자 한다. 생후 12개월에서 16개윌 된 체중 15 Kg 내외의 성견 4마리를 이용하였다. 실험 재료로 생체흡수성 차폐막 (Biogide(R), Swiss) 를 사용하였고, 골이식재로 치아회분말-연석고를 혼합매식 하였다. 양측 상악 소구치 부위에 변연 치조골하방에 4 mm ${\times}$ 4 mm ${\times}$ 4 mm, (깊이 ${\times}$ 근원심 ${\times}$ 협설폭경) 깊이로 골내낭을 형성하였다. 형성된 골내낭의 기저부위 치근 표면에 1/4 round bur로 notch를 형성하여 참고점으로 하였다. 무작위로 선택된 한 쪽의 결손부를 대조군으로 오직 생체 흡수성 차폐막을 사용하였고, 실험군으로 치아회분말-연석고와 생체 흡수성 차폐막을 결손부로부터 2 mm 이상 덮을 수 있도록 다듬어 결손부 위에 위치시킨 후 협측 판막을 덮고 봉합하였다. 4주 후 2마리 ,8주 후 2마리를 희생시키고 통상의 방법으로 고정, 탈회, 포매의 과정을 거쳐 광학 현미경으로 검경하였다. 그 결과, 1. 4주 대조군에서 Bio-gide(R)는 완전한 흡수를 보였고, 치근이개부내에는 큰 공간이 존재하였다. 2. 4주 실험군에서 역시 Bio-gide(R)는 완전한 흡수를 보였고, 골 결손부내에 더 많은 신생골 관찰되었다. 그러나 아직까진 기존골과 신생골간에 명확한 차이가 있어서 쉽게 구분할 수 있었다. 또한 골이식재 주변으로 파골세포가 다수 관찰되며 이로 미루어 보아 활발한 골흡수가 일어남을 알 수 있었다. 3. 8주 대조군에서 결손부내에서는 기존골에 인접하여 신생골 형성이 부분적으로 일어났으나 연조직 침입이 관찰되었다. 4. 8주 실험군은 신생골이 기존골과 매우 유사한 형태로 관찰되었고, 신생골 형성 부위에 신생 혈관 증식이 관찰되었다. 또한 골내낭 기저부위에서는 백악질과, 치주인대가 재생됨이 관찰되었다. 이상의 결과에서 치아회분말-연석고 혼합매식은 골재생을 위한 골전도성이 있는 재료로 사료되며, 이를 이용히여 치주조직재생술시 흡수성 차폐막과 병행하여 사용한다면 더 많은 골재생이 있을 것으로 기대된다.
Purpose : This research evaluates the effect of the use of absorbable membrane barrier with deproteinized bovine bone (Bio-$Oss^{(R)}$, Switzerland) on bone healing in surgically created critical-sized defects in rat calvaria. Materials and Methods : Two standardized transosseous circular calvarial defects (5 mm in diameter) are made in each calvarium of 30 rats. These rats are divided into negative control group(n=15), positive control group(n=15) and two experimental groups(n=15). In the negative control group, defects are only filled with blood clots. In the positive control group, defects are filled with autogenous bone obtained from calvarium; in the experimental group 1, defects are filled with deproteinized bovine bone; and in the experimental group 2, defects are filled with deproteinized bovine bone with absorbable membrane. At the postoperative 1 week, 3 weeks. and 6 weeks, clinical. histologic and histomorphometric evaluations of the defects are performed. Results : 1. The grafted bone without membrane in the calvarial bone defect was scattered but, the grafted bone with membrane was stable. 2. $BioMesh^{(R)}$ membrane was absorbed beginning at 3 weeks, and was absorbed considerably at 6 weeks while maintaining the structural form of the membrane. 3. The use of membrane blocked soft tissue invasion. 4. In histomorphometric analysis. it showed the greatest amount of new bone formation in the positive control group. The amount of new bone formation was greater in the experimental group 2 than experimental group 1. At 6 weeks. the amount of new bone formation was greater in the positive control group than experimental group l(p<0.005). Conclusion : These results suggest that membrane increase the stability of grafted bone and protects from soft tissue invasion, and the use of the membrane may promote new bone formation in deproteinized bovine bone graft area.

We identified the main impacts, drivers, and restoration projects for Atlantic Forest in Northwest of the Rio Grande do Sul State, Brazil. The objective was to analyze the quantity, distribution, and causes of the environmental crimes in 2000-2014. To verify differences between degraded and restored areas, we performed a t-test; ANOVA for the municipalities with more quantity of crimes, simple linear regression analysis for the relationship between sizes of degraded areas and quantity of seedlings planted, and Principal Component Analysis (PCA) for environmental damages categories and population of the municipalities. The main environmental damages found were deforestation outside permanent preservation area (20%) and those related to Permanent Preservation Area (37%). Environmental crimes in these areas fall into two categories: native and exotic vegetation removal (17%), and impediment to natural regeneration (20%). The average size of the degraded areas was $5,359{\pm}526m^2$, while for restored areas was $3,337{\pm}255m^2$. The sizes of the degraded fragments were similar among the five municipalities with the higher number of environmental crimes (ANOVA: p>0.05, F=1.24; df=241). The number of seedlings planted was positively related to the sizes of the degraded fragments (p<0.001, $R^2=0.53$). Segregation between the less and the most populous municipalities was found with the PCA analysis along PC1 (51.7%), while PC2 represented 19.2% of the total variation. The most populous municipalities showed the highest number of environmental crimes, and the majority of degraded areas were recovered by planting native seedlings. Atlantic Forest fragments need to be recognized and preserved as an ecosystem with a unique ecological function by the population and public administration.

Brassinosteroids are steroidal plant hormones and are known to modulate physiological and cellular response in a wide range of plant species. Considerable insights has been achieved of the physiological role of brassinosteroid in Brassica species in the past few years, but their effect on direct organogenesis has not been extensively studied. In this sense, under optimal basal media and growth conditions we tested the cellular and organogenic response of 24-epibrassinolide (EBL) in a variable concentration (0.1 to $5.0\;{\mu}M$) and Zeatin (Z) (1.0 to $100\;{\mu}M$) and their synergic effect on hypocotyl explants of cauliflower and broccoli. The isolated EBL accelerated cell elongation and promotes direct organogenesis. One micromolar EBL + $10\;{\mu}M$ of Z was the most efficient combination for cell elongation, cell differentiation as well as for organogenesis. A suppressing effect on root induction was confirmed for all the tested hormone levels. The general results indicate a synergic effect of EBL-Z and EBL potentates Zeatin activity, at least in certain tissues. Besides de genetic factors, we can speculate that the natural hormone concentration in the explants might affect the responses by application of exogenous growth regulators. Experiments with new plant growth regulators, like brassinolide, are important aiming to maximize or accelerate plant regeneration for in vitro multiplication or for genetic transformation.

One of the limitations to conducting maize Agrobacterium-mediated transformation using explants of immature zygotic embryos routinely is the availability of the explants. To produce immature embryos routinely and continuously requires a well-equipped greenhouse and laborious artificial pollination. To overcome this limitation, an Agrobacterium-mediated transformation system using explants of type II embryogenic calli was developed. Once the type II embryogenic calli are produced, they can be subcultured and/or proliferated conveniently. The objectives of this study were to demonstrate a stable Agrobacterium-mediated transformation of maize using explants of type II embryonic calli and to evaluate the efficiency of the protocol in order to develop herbicide-resistant maize. The type II embryogenic calli were inoculated with Agrobacterium tumefaciens strain C58C1 carrying binary vector pTF102, and then were subsequently cultured on the following media: co-cultivation medium for 1 day, delay medium for 7 days, selection medium for $4{\times}14$ days, regeneration medium, and finally on germination medium. The T-DNA of the vector carried two cassettes (Ubi promoter-EPSPs ORF-nos and 35S promoter-bar ORF-nos). The EPSPs conferred resistance to glyphosate and bar conferred resistance to phosphinothricin. The confirmation of stable transformation and the efficiency of transformation was based on the resistance to phosphinothricin indicated by the growth of putative transgenic calli on selection medium amended with $4mg\;1^{-1}$ phosphinothricin, northern blot analysis of bar gene, and leaf painting assay for detection of bar gene-based herbicide resistance. Northern blot analysis and leaf painting assay confirmed the expression of bar transgenes in the $R_1$ generation. The average transformation efficiency was 0.60%. Based on northern blot analysis and leaf painting assay, line 31 was selected as an elite line of maize resistant to herbicide.
Etifoxine (etafenoxine, $Stresam^{(R)}$) is a non-benzodiazepine anxiolytic with an anticonvulsant effect. It was developed in the 1960s for anxiety disorders and is currently being studied for its ability to promote peripheral nerve healing and to treat chemotherapy-induced pain. In addition to being mediated by $GABA_A{\alpha}2$ receptors like benzodiazepines, etifoxine appears to produce anxiolytic effects directly by binding to ${\beta}2$ or ${\beta}3$ subunits of the $GABA_A$ receptor complex. It also modulates $GABA_A$ receptors indirectly via stimulation of neurosteroid production after etifoxine binds to the 18 kDa translocator protein (TSPO) of the outer mitochondrial membrane in the central and peripheral nervous systems, previously known as the peripheral benzodiazepine receptor (PBR). Therefore, the effects of etifoxine are not completely reversed by the benzodiazepine antagonist flumazenil. Etifoxine is used for various emotional and bodily reactions followed by anxiety. It is contraindicated in situations such as shock, severely impaired liver or kidney function, and severe respiratory failure. The average dosage is 150 mg per day for no more than 12 weeks. The most common adverse effect is drowsiness at the initial stage. It does not usually cause any withdrawal syndromes. In conclusion, etifoxine shows less adverse effects of anterograde amnesia, sedation, impaired psychomotor performance, and withdrawal syndromes than those of benzodiazepines. It potentiates $GABA_A$ receptor-function by a direct allosteric effect and by an indirect mechanism involving the activation of TSPO. It seems promising that non-benzodiazepine anxiolytics including etifoxine will replenish shortcomings of benzodiazepines and selective serotonin reuptake inhibitors according to animated studies related to TSPO.

③ NDSL에서 제공하는 콘텐츠를 보도, 비평, 교육, 연구 등을 위하여 정당한 범위 안에서 공정한 관행에
합치되게 인용할 수 있습니다.
④ NDSL에서 제공하는 콘텐츠를 무단 복제, 전송, 배포 기타 저작권법에 위반되는 방법으로 이용할 경우
저작권법 제136조에 따라 5년 이하의 징역 또는 5천만 원 이하의 벌금에 처해질 수 있습니다.
제 14 조 (유료서비스)
① 당 사이트 및 협력기관이 정한 유료서비스(원문복사 등)는 별도로 정해진 바에 따르며, 변경사항은 시행 전에
당 사이트 홈페이지를 통하여 회원에게 공지합니다.
② 유료서비스를 이용하려는 회원은 정해진 요금체계에 따라 요금을 납부해야 합니다.
제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)
회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
