This experiment was undertaken to investigate proper conditions for protoplast isolation and regeneration from the mycelia of Lyophyllum ulmnrium. Protoplast isolation and regeneration are influenced by a variety of factors such as enzyme, osmotic stabilizer, reaction time and age of mycelia. A combination of Novozyme 234 (10mg/ml) and cellulase Onozuka R-10 (10 mg/ml) with 0.6 M $MgSO_4$ was most effective for isolation of the protoplasts. The optimum reaction time of the mycelia with the lytic enzymes was 3.5~4 hours at $28^{\circ}C$ in shaking condition at 120 strokes per min. High yield of the protoplasts were obtained from its 4~5 days old mycelia on complete agar media. Its protoplasts were regenerated to normal hyphae. Regeneration media with 0.6 M sucrose were proper for regeneration of the protoplasts. Their regeneration frequency on complete agar media was 2.3~2.7%.
Recent study shows that implant design has a great impact on initial stability in bone. The ITI $TE^{(R)}$ implant, designed originally for immediate placement has a tapered/ cylindrical form which fits the anatomical shape of the natural alvelous or tooth root. The increased diameter at the collar region coupled with more threads lead to more bone contact and enhanced stability. The aim of this retrospective study is to evaluate the clinical use and the efficacy of recently introduced ITI TE implant with a new macro-design. The following results are compiled from 139 patients who received ITl TE implant surgery at the periodontal department. of Yonsei University Hospital between July 2002 and September 2005. 1. 139 patients received 173 ITl $TE^{(R)}$ implants in their maxilla and mandible (Mx 82, Mn 91). Posterior area accounted for 84% of the whole implant surgery, 2. In the distribution of bone quality, type III(41,0%) was the most, followed by type IV(41,0%) and type II (27.7%). As for the bone quantity, type B(43.9%) was the most, followed by type C(42.2%), type D(12.2%) and type A(1.7%). 3. 125 implants(83.9%) were treated by single crown, which accounted for the majority. 4, The total implant survival rate was 100% after a mean follow-up period of 21.2 months. This preliminary data with ITl $TE^{(R)}$ implant showed excellent survival rate although the majority of implants evaluated in this study were placed in the posterior region of the jaw and compromised sites.
The purpose of this study was to evaluate the efficacy of the Sonicare $Elite^{(R)}$ power toothbrush in plaque removal and reduction of gingivitis from hard-to-reach sites of the moderate periodontitis compared to regular manual toothbrush in 12 week follow-up. 82 subjects with incipient to moderate periodontitis were randomly assigned to use either the manual or sonic brush, instructed in its use, and asked to brush 2 times a day for 2 minutes. Plaque scores were taken at baseline, 1, 4, 12 weeks using Silness & $L\ddot{o}e$ plaque index and gingival inflammation was assessed by the $L\ddot{o}e$ & Silness gingival index. The results were as follows. 1. The Sonicare $Elite^{(R)}$ power toothbrush showed a significant reduction(p<0.0001) of the plaque(Silness & $L\ddot{o}e$) and gingival inflammation(Loe & Silness). 2. The Sonicare $Elite^{(R)}$ power toothbrush showed a significant better reduction of plaque and gingivitis(p<0.05) than the manual toothbrush after 1, 4, and 12 weeks. 3. The Sonicare $Elite^{(R)}$ power toothbrush demonstrated a significant reduction(p<0.0001) of the plaque in interproximal sites(p<0.0001), buccal sites(p<0.0001) and the lingual sites(p=0.00l8) of the teeth. 4. The Sonicare $Elite^{(R)}$ power toothbrush demonstrated a significant reduction(p<0.0001) of the gingival inflammation in the interproximal sites(p<0.0001), the buccal sites(p<0.0001) and the lingual sites(p<0.0001) of the teeth. The results of this study support the findings that Sonicare $Elite^{(R)}$ power toothbrush has a great potential to remove the plaque and resolve the gingival inflammation during the period of 12 week.
The aim of this study was to achieve the healing of peri-implantitis defects and the hard tissue regeneration using the augmentation of a xenograft on defect site. Two patients were treated with the surgical approach. With a full muco-periosteal flap elevation, the implant surfaces were exposed and taken the debridement of granulation tissue around the abutment. Each surface of the abutments was prepared with the air-abrasive device (PerioFlow$^{(R)}$) for decontamination. Bovine-derived bone mineral (Bio-Oss collagen$^{(R)}$) was then used to fill the defects, and no membrane was placed on the grafting site. Radiographs and clinical photo was taken to compare from baseline status. Within the limits of the present case, this case shows the significance of the surgical treatment of peri-implantitis. And this also verifies the stability of bovine-derived bone mineral and effectiveness of Air-abrasive device (PerioFlow$^{(R)}$).
The purpose of this study was to evaluate 6 years cumulative survival rate (CSR, %) of mandibular posterior single tooth implants replaced with $Br{\aa}nemark$$TiUnite^{(R)}$ implant system. The 55 (55 persons) single implants that were placed in the mandibular first molar region were successful except 2 cases and showed 96.36% survival rate. And, among the 57 (56 persons) single implants replacing the mandibular second molar. 2. failed showing 96.49% survival rate. There was no significant statistical difference. 3. Among the total 112 implants, 5.0mm wide diameter implants were placed in 96 cases(85.7%) showing 96.9% survival rate. 4.0mm standard diameter implants were placed in 16 cases showing 93.8% survival rate. There was no significant statistical difference. 4. Long implants above 10.0mm length were placed 103 cases(91.0%) and showed 96.1% survival rate. Short implants within 8.5mm length were placed 9 cases and showed 100% survival rate. There was no significant statistical difference. 5. 37 implants placed in type I, II bone quality were showed higher survival rate(100%) than that of 52 implants placed in type III, IV bone quality(92.3%). But, there was no significant statistical difference. In conclusion, $Br{\aa}nemark$$TiUnite^{(R)}$ implant showed successful results when replacing manbibular single molar.
Recently, it was reported that enamel matrix derivative may be beneficial in periodontal regeneration procedures in expectation of promoting new bone and cementum formation. The aim of present study was to evaluate the effect of enamel matrix derivative($Emdogain^?$)and Caso4 sulfate paste in 1-wall intrabony defects in beagle dogs. Surgically created 1-wall intrabony defects were randomly assigned to receive root debridement alone or $Emdogain^{(R)}$ or $Emdogain^{(R)}$ and Caso4. Clinical defect size was 4 X 4mm. The control group was treated with root debridement alone, and Experimental group I was treated with enamel matrix derivative application, and Experimental group II was treated with enamel matrix derivative and Caso4 sulfate paste application,. The healing processes were histologically and histometrically observed after 8 weeks and the results were as follows: 1. The length of junctional epithelium was $0.41{\pm}0.01mm$ in the control group, $0.42{\pm}0.08mm$in the experimental group I and $0.50{\pm}0.13mm$in the experimental group II. 2. The connective tissue adhesion was $0.28{\pm}0.02mm$ in the control group, $0.13{\pm}0.08mm$ in the experimental group I and $0.19{\pm}0.02mm$ in the experimental group II. 3. The new cementum formation was $3.80{\pm}0.06mm$ in the control group, $4.12{\pm}0.43mm$ in the experimental group I and $4.34{\pm}0.71mm$ in the experimental group II. 4. The new bone formation was $1.43{\pm}0.03mm$ in the control group, $1.53{\pm}0.47mm$ in the experimental group I and $2.25{\pm}1.35mm$ in the experimental group II. Although there was limitation to present study, the use of enamel matrix derivative in the treatment of periodontal 1-wall intrabony defect enhanced new cementum and bone formation. Caso4 sulfate paste will be the candidate for carriers to deliver enamel matrix derivative, and so enhance the regenerative potency of enamel matrix derivative.
Many tooth cleansing instruments and agents have been developed for removal of plaque, inhibition of plaque formation and reduction of gingival inflammation. The aim of this study was to evaluate the plaque control effect and the therapeutic effect of newly developed tooth cleansing instrument(Belloblanco(R)). 40 healthy subjects with gingivitis or early periodontitis were divided into two groups. Subjects in control group only used manual tooth brushing and in experimental group used manual tooth brush and additive tooth cleansing instrument(Belloblanco(R)). Additive tooth cleansing instrument was used once a week. At baseline scaling and tooth brushing instruction was performed. Probing depth, bleeding on probing, plaque index, gingival index were scored at baseline, 2weeks, 4weeks. Probing depth of control and experimental group were significantly reduced at 2 weeks, 4weeks, hut there were no differences between two groups(P<(0.05). Bleeding on probing, plaque index and gingival index of control and experimental group were significantly reduced at 2weeks and 4weeks and there was significantly more reduction in the experimental group than the experimental group than the control group(p<(0.05). From these finding. it can be conclude that newly developed tooth cleansing instrument(Belloblanco(R)) are effective on the removal of plaque and the reduction of gingival inflammation
The ultimate goal of periodontal disease therapy is to promote the regeneration of lost periodontal tissue, there have been many attempts to develop a method to achieve this goal, but none of them was completely successful. The purpose of this study was to compare the effects of treatment using BBP(R) with control treated by only modified Widman flap. 22 intrabony defects from 12 patients with chronic periodontitis were used for this study, 10 sites of them were treated with BBP(R) as experimental group and 12 site were treated by only modified Widman flap as control group. Clinical parameters including probing depth, gingival recession, bone probing depth and loss of attachment were recorded at 6 months later, and the significance of the changes was statistically analyzed. The results are as follows : 1. Probing depth of control(${\triangle}2.7{\pm}1.3mm$) and experimental group(${\triangle}3.6{\pm}1.8mm$) weres reduced with statistically significance(P<0.05), but this changes were not different between the two experiment, control group with statistically significance. 2. Gingival recession showed statistically significant increase in control group(${\triangle}2.1{\pm}1.2mm$)(P<0.05), but not in experimental group(${\triangle}0.5{\pm}0.7mm$), and this changes were different between the two experiment, control group with statistically significance(P<0.05). 3. Bone probing depth showed statistically significant decrease in experimental group(${\triangle}2.9{\pm}1.0mm$)(P<0.05), but not in control group(${\triangle}1.1{\pm}1.4mm$), and this changes were different between the two experiment, control group with statistically significance(P<0.05). 4. Loss of attachment showed statistically significant decrease in experimental group(${\triangle}3.1{\pm}1.7mm$), but not in control group(${\triangle}0.6{\pm}1.2mm$), and this changes were different between the two experiment, control group with statistically significance(P<0.05) On the basis of these results, treatment using BBP(R) improves the probing depth, bone probing depth and loss of attachment in intrabony defects.
This paper reports an improved protocol for isolation, culture and regeneration of Lotus corniculatus protoplasts. A range of parameters which influence the isolation of L. corniculatus protoplasts were investigated, i.e., enzyme combination, tissue type, incubation period and osmolarity level. Of three enzyme combinations tested, the highest yield of viable protoplasts was achieved with the combination of 2% Cellulase Onozuka RS, 1% Macerozyme R-10, 0.5% Driselase and 0.2% Pectolyase. The use of etiolated cotyledon tissue as a source for protoplast isolation proved vital in obtaining substantially higher protoplast yields than previously reported. Culture of the protoplasts on a nitrocellulose membrane with a Lolium perenne feeder-layer on the sequential series of PEL medium was highly successful in the formation of microcolonies with plating efficiencies 3-10 times greater than previous studies. Shoot regeneration and intact plants were achieved from 46% of protoplast-derived cell colonies.
