Histopathological study of acute toxicity of ammonia to the eel, Anguilla japonica in high temperature and pH levels (고수온(高水溫) 고(高)pH에서 뱀장어에 미치는 암모니아 급성독성(急性毒性)의 병리조직학적(病理組織學的) 연구(硏究))

  • This study was carried out to examine the acute toxic effects of ammonia to the eel, Anguilla japonica in high temperature and pH levels by histopathological observations. The eels of 40 g average body weight were exposed to 4 different concentrations of total ammonia (0, 10, 20, 30 mg/$\ell$) for 24~120 hours. Each concentration was treated under 4 different levels of pH (7.5, 8.0, 8.5, 9.0) and each of these treatments was tested at 2 different temperatures ($27^{\circ}C$, $32^{\circ}C$). Histopathological changes in gill tissues were observed by hematoxylin-eosin stain. As increasing of pH (from 7.5 to 9.0), water temperature (from $27^{\circ}C$ to $32^{\circ}C$), total ammonia concentration (from 0 mg/$\ell$ to 30 mg/$\ell$) and exposure time (from 24 hours to 120 hours), gill discolorated to dark brown with the naked eye and gill tissues showed hypertrophy of gill lamellae, winding of the secondary gill lamellae, epitherial separation and necrosis histopathologically. When gill lamellae epithelium was separated from the blood vessels, gill discolorated to dark brown with the naked eye.

Expression of a Human Histone H1.5 in Transgenic Tobacco Cultured Cells (담배 배양세포에서 인간 히스톤 단백질 H1.5의 발현)

  • Transgenic tobacco (Nicotiana tabacum L. cv Bright Yellow-2) cell lines expressing a human histone H1.5 (referred to as hH1.5), which suppress collagen-induced rheumatoid arthritis, were developed under the oxidative stress-inducible peroxidase (SWPA2) promoter. Tobacco BY-2 cells were transformed by Agrobacterium-mediated method. The kanamycin-resistant calli were selected on the modified MS medium containing 150mg/L kanamycin and 300mg/L claforan. Transgenic cell lines were confirmed by PCR and northern blot analysis. Recombinant hH1.5 (rhH1.5) protein (42 kDa) was also detected by Western blot analysis, showing a different molecular weight of human hH1.5 (32 kDa). These results suggested that a hH1.5 gene was properly introduced in tobacco cultured cells under the control of SWPA2 promoter. The further characterization of rhH1.5 protein remains to be studied.

Fermentation for Liquid-type Yogurt with Lactobacillus casei 911LC

  • This study was carried out to find the attributes for liquid-type yogurt with Lactobacillus casei 911LC during 72 h fermentation at $37^{\circ}C$. The pH decreased up to 32 h and plauteaued thereafter, and the titratable acidity increased up to 40 h. The growth of lactic acid bacteria sharply increased with $3.5{\times}10^7$ cfu/ml up to 40 h of fermentation and slowly decreased thereafter. The free amino acids produced during fermentation reached the maximum value at 44 h and gradually decreased thereafter. Bitterness sensory scores were the highest at 44 h of fermentation. In the result of electrophoresis, the band mostly disappeared at 72 h fermentation. The present data showed that the range of optimum fermentation time for liquid-type yogurt using Lactobacillus casei 911LC was from 40 to 44 h.

  • This experiment was carried out to develope a technique for labelling pine gall midge with ·radioactive phosphorus and calcium. This technique evolved would help to obtain basic information on the appropriate concentration of $^{32}P\; and\; ^{45}Ca$ in tagging and determination of adult flight ranges which considered as one of the factors in dissemination. 1. Labelling at the larval stage of pine gall midge by injection of $^{45}Ca$ into pine tree trunk appeared not to be effective for tagging the adults because radioactivity was gradually decreased to background by emergence. 2. The radioactivities of larvae in the galls gradually increased in September but from October 1 the activity was gradually decreased for 10 days then equilibrium was maintained thereafter. The results seem to indicate that larval diet termination occured early October. 3. A maximum level of radiation was detected at 3 weeks after $^{32}P$ injection and thereafter it was gradually reduced. This result implies that $^{32}P$ is not appropriate radioisotope in labelling pine gall midge through trunk injection method. 4. After washing in running water for 10 minutes, the optimum$^{32}P$ concentration for treated overwintered larvae and pupae was found to be 0.5 micro curie per ml. for 30 minutes dipping.

Cultural Conditions for the Mycelial Growth of Phellinus spp. (진흙버섯속의 배양적 특성)

  • This study was carried out to determine the optimum culture conditions for Phellinus spp. known as white rot fungi showing anti-cancer activity. The optimum solid medium for mycelial growth at $25^{\circ}C$ was potato dextrose agar medium and optimum pH range was $6.0{\sim}8.0$, while all species showed reduced or no growth at pH 4.0. Most species showed good growth at $25{\sim}30^{\circ}C$. Out of 10 species of Phellinus examined, P. biscuspidatus was the best growing fungus in the range of pH $6.0{\sim}7.0$ based on mycelial density. Three species such as P. biscuspidatus, P. johnsonianus and P. lloydii could be grouped in mesophile fungi, showing $30{\sim}35^{\circ}C$ optimum temperature.

  • The photoreaction of 2, 3-benzo-1, 1-diphenyl (or dimethyl)-1-sila-2-cyclobutene (9 or 10) with an aldehyde or ketone results in 1:1 cycloadduct of [4 + 2] type. In the reactions of 2, 3-benzo-1, 1-dimethyl-1-sila-2-cyclobutene (10) with acetone and butanone, another 1:1 adducts (13) were also formed, respectively. The following facts indicate that the formation of adduct involves an attack of a triplet carbonyl compound on the silicon of the benzosilacyclobutene, an $S_H2$ process. (1) Even when the reaction of 9 with acetophenone was carried out under conditions such that more than 99% of incident light was absorbed only by acetophenone using the filter solution of aq. cupric sulfate, the same adduct was still formed. (2) When the reaction of 9 with acetone was carried out under oxygen atmosphere, only trace amount of adduct was formed.

  • We studied the interaction of 3,3',3'',3'''-ethylenetetrakis-4-hydroxycoumarin (EHC) with bovine serum albumin (BSA) in acetate buffer and phosphate buffer with different pH values by UV-vis absorption spectrometry and fluorescence spectrometry respectively. It was found that the pH values of the buffer solutions had an effect on the interaction process. In acetate buffer of pH 4.70, the carbonyl groups in EHC bound to the amino groups in BSA by means of hydrogen bond and van der Waals force, which made the extent of peptide chain in BSA changed. By contrast, in phosphate buffer of pH 7.40, hydrophobic force played a major role in the interaction between EHC and BSA, while the hydrogen bond and van der Waals force were also involved in the interaction. The results of spectrometry indicated that BSA could enhance the fluorescence intensity of EHC by forming a 1:1 EHC-BSA fluorescent complex through static mechanism at pH 4.70 and 7.40 respectively. Furthermore, EHC bound on site 1 in BSA.

The influence of saliva pH value on the retention and durability of bar-clip attachments

  • PURPOSE. The aim of this study was to compare the durability and retention of 4 types of attachments placed over computer aided design/computer aided manufacturing (CAD/CAM) titanium bars when subjected to different pH conditions. MATERIALS AND METHODS. Four commercially available attachments were investigated: Hader Yellow, Hader Red, Ackerman Gold and Ackerman Stainless Steel. These attachments and Ackerman CAD/CAM titanium bars were placed in 2 vessels containing different artificial saliva solutions (pH 7/pH 4) at $37^{\circ}C$ for one month to simulate corrosion conditions, and they were then subjected to mechanical testing (5400 cycles of insertion and removal). RESULTS. The results revealed that there were significant differences in the average values of insertion/removal force due to the pH (F (1, 24)=9.207, P<.05) and the type of attachment (F (3, 24)=11.742, P<.05). CONCLUSION. More acidic pH values were found to have a negative influence on the retention capacity of the attachments.
