  • Capacitation of mouse spermatozoa in vitro is brought about by epididymal secretions released into the m-Tgrode medium at the time of sperm collection. Epididymal mouse sperm suspension achived by centrifugation were preincubated for a total of 120min with aliquants being removed at 5, 30 and 120min. By gently centrifugation and resuspending in fresh medium, the fertilizing rate of unwashed 5-and 30-min suspensions was increased such that 30-min washed samples did not differ significiantly from full capacitated, highly fertile 120-min unwashed samples. When epididymal suspension was added fertilization of cumulus intact oocyte was markedly inhibited, although fertilization of zona free oocytes was unaffected. Washing sperm suspensions preincubated in the absence of $Ca^{2+}$ with the subsequent introduction of exogenous $Ca^{2+}$ resulted in a significant increase in fertilization rates over equivalent unwashed samples.

Enhanced Germination & Initial Seedling Development by Liquid Phase Ozonation of Plant Seeds (식물종자의 액상오존처리에 의한 종자 발아 및 초기 성장 증진)

  • The effects of liquid phase ozonation on seed dormant alleviation and subsequent seedling growth were studied using two plant seeds of Indian jointvetch (Aeschynomene indica L.) and Indian mallow (Abutilon avicennae Gaertn.). At a constant ozone concentration ($80g/m^3$), contact time varied from 10 to 60 min with 10 min interval. Germination rate, root length, and specific root length were compared after 3-day incubation on gel-medium. The germination rate increased significantly (p<0.05) in the 50 min treatment of Indian mallow by 30% compared to the control. Enhanced root elongation was observed in the seeds of 30 min treatment of Indian jointvetch and 30~50 min treatment of Indian mallow. Specific root length, an indicator of environmental change, did not show significant changes, suggesting the level of ozone treatment has no adverse effect on seedling development. The results indicate that liquid phase seed ozonation can be an effective on-site germination alleviation method in the application of phytoremediation.

Enhancing effect of Multiherb extracts HT008-1 on Memory and Cognitive Function (한약복합물 HT008-1의 인지기능 및 기억력 향상효과)

  • Objectives : Investigation of the memory and cognitive enhancing effect of HT008-1 in scopolamine induced amnesia mice. Methods : At 60 min before acquisition trials, HT008-1 (30, 100, 300 mg/kg p.o.) was administered, and 30 min later, mice were injected with scopolamin (1.0 mg/kg, i.p.). In the passive avoidance test, acquisition trials were carried out 30 min after a single scopolamine treatment. Retention trials were carried out 24h after acquisition trials. Y-maze test was carried out 30 min after a single scopolamine treatment. Spontaneous alternation behavior during an 8-min session was recorded. Inhibitory effects of HT008-1 (0.01, 0.1, 1.0 mg/ml) on AChE activity was measured. Result : HT008-1 ameliorated scopolamine-induced learning impairments and spatial cognitive function in passive avoidance and Y-maze test, respectively. Moreover HT008-1 showed a significant inhibitory effect on AChE activity. Discussion: This study presented that eMultiherb mixture HT008-1 enhanced learning memory and spatial cognitive function in scopolamine-induced amnesia mice. These results suggest that the effect of HT008-1 may be dependent on the inhibition of AChE activity.

Reduction of Suspended Solids in First Flush from a Building Rooftop using Various Media (여과재를 활용한 건물옥상유출 초기빗물의 부유물질 저감)

  • We analyzed the water quality of first flush and rainfall runoff from a building rooftop, and investigated the removal of suspended solids (SS) in first flush using various media to develop a first flush filtration system. Particle size distribution exhibited most of particles in first flush from the rooftop ranged from 10 to 30 ${\mu}m$. SS concentrations maxed in 10~20 min and decreased afterwards. Dissolved organics and inorganic materials in runoff also showed highest levels in first flush (10 min). Filtration tests using anthracite (AC), polyurethane (PU), polypropylene (PP) showed about 50% of SS removal during the first 10 min operation, but the removal rates dramatically decreased after 20 min of filtration. Based upon the results from rinse and run cycle tests, only AC could achieve nice cycles without distinct decease of SS removal. SS removal rates increased with higher depth of media bed and lower flowrate. The system achieved over 50% of SS removal with a media depth of 30 cm and flowrate < 12 L/min.

Preparation of Barley Leaf Powder Tea and Its Quality Characteristics (보리잎 분말차의 제조와 그 품질특성)

  • This study was carried out to establish the manufacturing process of barley leaf powder tea. The optimal manufacturing process among many trials was determined with sensory evaluation. Finally established process and operation conditions were as follows: pretreatment (cutting and washing), steaming ($100^{\circ}C$, 30 sec), primary drying and roasting ($130^{\circ}C$, 40 min), rolling (RT, 25 min), middle drying and roasting ($60^{\circ}C$, 30 min), final drying and roasting ($55^{\circ}C$, 25 min), drying ($60^{\circ}C$, 20 min), roasting ($85^{\circ}C$, 20 min), and powdering (120 mesh). The barley leaf powder tea produced by this process mainly consisted of dietary fiber (33.8%), amino acids (12.9%), minerals (4.7%) and ${\beta}-carotene$ (6.9 mg%).

Prevention of Ischemic Damage in Working Rat Hearts by Calcium Channel Blocker and Calmodulin Inhibitors (흰쥐심장의 허혈손상에 대한 Calcium 통로봉쇄제와 Calmodulin 억제제의 예방효과에 대한 연구)

  • This study was investigated under the postulation that activation of intracellular calcium- calmodulin complex during ischemia-reperfusion leads to myocardial injury. The protective effects of calcium channel blocker, diltiazem and calmodulin inhibitors, trifluoperazine, flunarizine and calmidazolium from ischemic injury in rat hearts were observed by using Langendorff apparatus when the antagonists were infused for 3 min in the beginning of ischemia. Thereby, an increase in resting tension developed during 30-min ischemia was analyzed with regard to [1] the degree of cardiac functional recovery following 60-min reperfusion, [2] changes in biochemical variables evoked during 30-min ischemia. The results obtained were as follows: l. In the ischemic group, the resting tension was increased by 4.1*0.2 g at 30-min ischemia. However, the increase in resting tension was markedly reduced not only by pretreatment with diltiazem [3.3 p M] but also with calmodulin inhibitors, trifluoperazine [3.3 p M], flunarizine [0.5 p M] and calmidazolium [0.5 p M], respectively. 2. Recovery of myocardial contractility, +dF /dt and coronary flow were much reduced when evoked by reperfusion in the ischemic group. These variables were significantly improved either by pretreatment with diltiazem or with calmodulin inhibitors. 3. The resting tension increment evoked during ischemia was significantly inversely correlated with the degree of cardiac function recovered during reperfusion. 4. Following 30-min ischemia, the production of malondialdehyde and release of lysosomal enzyme were much increased in association with a decrease in creatine kinase activity. 5. The increases in malondialdehyde production and release of free lysosomal enzyme were suppressed by pretreatment with calmodulin inhibitors as well as diltiazem. Likewise, the decrease of creatine kinase activities was prevented by these calcium antagonists. With these results, it is indicated that a increase in resting tension observed during ischemia has an inverse relationship to the cardiac function recovered following reperfusion, and further, the later may be significantly dependent on the degree of biochemical alterations occurred during ischemia such as decrease in creatine kinase activity, increased production of malondialdehyde and increased release of free lysosomal enzyme. Thus it is concluded that calmodulin plays a pivotal role in the process of ischemic injury.

Gradual Reperfusion Lowers the Incidence of Reperfusion-Induced Ventricular Fibrillation in a Cat Model of Regional Ischemia

  • Blood flow restoration to ischemic zone of the heart is essential to salvage of ischemic tissue. However, there is a large body of evidence documenting that the reperfusion can induce reperfusion injury like reperfusion-induced malignant arrhythmias. In the present study, employing a cat model of regional cardiac ischemia, we examined if reperfusion rendered in a gradual fashion could lower the incidence of reperfusion-induced ventricular fibrillation (VF), which usually precipitated within a few to several tens of seconds after abrupt reperfusion. The experiments were conducted with male mongrel cats (n=46, 2.5-5 kg). The animals in the control and 30 MIN groups were subjected to an episode of 20- and 30-min left anterior descending coronary artery occlusion, respectively, followed by abrupt reperfusion. The animals in 5 G and 10 G groups received gradual reperfusion over a 5- and 10-min period, respectively, following a 20-min occlusion. The proportion of animals that exhibited VF during the reperfusion phase was 11/15 in the control, 7/10 in the 30 MIN, 5/10 in the 5 G and 2/11 in the 10 G groups. The incidence of VF in the 10 G group was significantly lower than that in the control or 30 MIN group subjected to abrupt reperfusion. These results suggest that the gradual reperfusion is a useful procedure against reperfusion-induced VF.

Effects of a Pre-Exercise Meal on Plasma Growth Hormone Response and Fat Oxidation during Walking

  • The purpose of this study was to determine the effects of a pre-exercise meal on the plasma human growth hormone (hGH) response and fat oxidation during walking. Subjects (n=8) were randomly provided with either 1 g/kg body weight of glucose in 200 mL water (CHO) or 200 mL water alone (CON) 30 min prior to exercise and subsequently walked on a treadmill at 50% of VO2max for 60 min. Plasma hGH concentrations were significantly higher in subjects who received CHO compared to those who received CON at 15 and 30 min. The fat oxidation rate in the CHO was significantly lower than the CON while walking for 5~15, 25~35 and 45~55 min. Plasma FFA levels were also significantly lower in the CHO compared to the CON at 30, 45 and 60 min. Plasma glucose levels in the CHO were significantly lower while plasma insulin levels were significantly higher than in the CON at 15 and 30 min. Therefore, the results of this study suggest that the elevation of plasma hGH levels due to the intake of a pre-exercise meal may not be strongly related to fat oxidation and plasma free fatty acid (FFA) levels during low-intensity exercise.

Quality Characteristics of Sasambeong according to Processing Conditions (사삼병의 제조조건에 따른 품질특성)

  • This study was performed in order to determine the processing conditions (pre-drying at room temperature for 0 or 30 min, frying at $160^{\circ}C$ for 1 min or at $180^{\circ}C$ for 40 sec, and soaking for 0~24 h) of Sasambeong recorded in the "Sumunsasul". The Sasambeong was evaluated for crude lipid content, hardness, Hunter's color values, and sensory characteristics. After pre-drying at room temperature for 30 min, the crude lipid contents and hardness of Sasambeong were significantly increased. In addition, the crude lipid content and hardness of Sasambeong did not differ significantly according to the frying conditions. After pre-drying, the sensory characteristics of Sasambeong showed more improvement. The crude lipid content, hardness, Hunter's color values, and sensory characteristics of Sasambeong, which were prepared according to different soaking times (0, 6, 12, or 24 hr) were investigated. As soaking time increased, the crude lipid content and hardness of Sasambeong decreased with a range of 25.43~24.31% and 525.90~388.98 g, respectively. The sensory characteristics of Sasambeong showed no significant difference according to the soaking time. Overall, we think that the best processing conditions of Sasambeong were pre-drying at room temperature for 30 min and then frying at $160^{\circ}C$ for 1 min.

Effect of Microcurrent Stimulation at Hand Somatic Acupuncture Points on Experimental Pain Threshold (미세전류가 수부체성경혈점의 실험적 통증 역치에 미치는 영향)

  • The purposes of this study were 1) to examine the effects of microcurrent electrical neuromuscular stimulation 2) to compare surface electrode with needle electrode at somatic acupuncture points on experimental pain threshold measured at the distal end of the radius 3) to determine the changes in effect over time. A total of sixty healthy adult male and female subjects were assigned randomly to one of two experimental group or to a control group. Group 1(n=20) received MENS(Microcurrent Electrical Neuromuscular Stimulation) with surface electrode. Group 2(n=20) received HENS with needle electrode. Group 3(n=20) received no MENS. It measured experimental pain threshold at the wrist on pretreatment, 0 min after treatment. 15 min after treatment, 30 min after treatment in two experimental group. The results were as follows: 1. Experimental pain thresholds were higher in males than females(P<0.01). 2. Only the experimental group exhibited a significant increased in pain threshold after MENS treatment(P<0.05). 3. Surface electrode group increased significant pain threshold 0 min, 15 min after treatment, but greatly decrease 30 min after treatment. 4. Needle electrode group increased significant pain threshold 0 min, 15 min after treatment, but decrease 30 min after treatment. The results suggest that MENS applied to appropriate somatic acupuncture point can increase pain threshold. Further research is needed to assess the effects of greatly variety intensity of MENS of pain sufferes.
