Protective Effects of Geniposide and Extract of Korean Gardeniae Fructus -On Hepatic Injury Induced by Toxic Drugs in Rats- (한국산 치자(梔子) 엑스 및 Geniposide의 약물성(藥物性) 간장해(肝障害)에 대한 보호효과(保護效果))

  • This study was attempted to investigate the effect of Gardeniae Fructus on GOT, GPT, Al.p, LDH activities and level of total cholesterol in serum of $CCl_{4}$ and $_{D}-galactosamine$ intoxicated rats, and bile excretion. The geniposide and extract caused a remarkabel decrease of GPT activities, level of total cholesterol in serum of $CCl_{4}$ intoxicated rats at EtOH Ex. 300, 500 mg/kg p.o., MeOH Ex. and geniposide 100 mg/kg p.o., and GOT, Al.p, LDH activities were significantly decreased compared with control group. It caused a remarkable decrese of GPT, Al.p, LDH activities in serum of $_{D}-galactosamine$ intoxicated rats, and GOT activities was significantly decreased compared with control group. The geniposide and extract caused a remarkable increase of bile excretion, when administration of EtOH extract 500 mg/kg p.o., MeOH extract 100 mg/kg i.d., MeOH extract 50 mg and geniposide 50 mg/kg i. v. compared with normal-control group.

Effect of Dietary Copper Levels on Performance and Carcass Characteristics in Hanwoo Steers (사료중의 Cu 수준이 한우 거세우의 성장 및 도체특성에 미치는 영향)

  • This experiment was conducted to determine the effects of dietary copper levels on performance, carcass characteristics, and economical analysis in Hanwoo steers. Fifteen Hanwoo steers weighing about 300kg were randomly allotted into one of three treatments. The three treatments consisted of Control(7mg Cu/kg feed), TI(12mg Cu/kg feed) and TII(17mg Cu/kg feed). Copper was formulated with concentrate from copper sulfate($CuSO_4$) and animals were fed the diets supplemented with copper from 300kg untill about 630 kg of body weight. Dietary copper levels did not affect daily weight gain, feed intake and feed efficiency during the whole feeding period. Dressing percentage, rib eye area and backfat thickness were not different between animals fed the diets supplemented with copper. Animals fed the diets supplemented with copper received the same B grade in yield grade. Animals fed the diets supplemented with copper received same marbling score. Beef color, fat color, texture and maturity were not affected by Cu supplementation in diet. Animals received the same first grade between treatments in quality grade. Animals fed the diet supplemented with 12mg of copper per kg diet was higher in profit by 13.3% than animals fed the control diet. According to these results, it may be concluded that performance, yield and quality grade were not affected by the Cu levels of 7 to 17mg per kg feed but crude income was enhanced when Hanwoo steers were fed the diet containing 12mg Cu/kg feed.

An Experimental Study on Single-dose and 4 Weeks Repeated-dose Toxicity of Aconitum ciliare Decaisne Pharmacopuncture (초오약침의 단회 및 4주 반복 투여 독성에 관한 실험적 연구)

  • Objectives : This study was performed to examine the toxicity of Aconitum ciliare Decaisne pharmacopuncture(ADP). Methods : The toxicity was evaluated for lethal dose for 50 percent kill(LD50), single dose and repeated dose for 4 weeks. Toxic symptoms, weight measurement, hematological test, blood biochemical test, visual examination and weight measurement of major organs and histopathological test were observed for 4 weeks. Dose of 300, 600, 1,200, 2,400, 3,600, 4,800, 6,000, 7,200 mg/kg in the LD50 experiment, 300, 600, 1,200 mg/kg/day in the single experiment, 150, 300, 600 mg/kg/day in 4 weeks experiments were injected into BALB/c mice. The ADP was injected into ST36 of the right leg. Normal saline solution of same volume was used for control group. In 24 hours after the last treatment, blood samples were taken after anesthesia by inhalation of ethyl ether. After that, the BALB/c mice were euthanized. Their heart, lungs, kidneys, liver and reproductive organs were removed and weighed. Histopathological evaluation was also performed. Results : ADP's LD50 was measured at 6,000 mg/kg. In both single and repeated dose toxicity test, no BALB/c mouse died during the experiments. ADP treatment for 4 weeks did not show any significant changes in toxic symptoms, weight measurement, hematological test, blood biochemical test, visual examination and weight measurement of major organs and histopathological test. Conclusions : As a result, ADP's LD50 was 6,000 mg/kg and repeated dose at a concentration of 600 mg/kg or less is considered to be not harmful for clinical treatment.

Protective Efficacy of Alpha-lipoic Acid against AflatoxinB1-induced Oxidative Damage in the Liver

  • Alpha-lipoic acid (${\alpha}$-LA) is not only involved in energy metabolism, but is also a powerful antioxidant that can protect against hepatic oxidative stress induced by some drugs, toxins, or under various physiological and pathophysiological conditions. Here, we investigated the effect of ${\alpha}$-LA against liver oxidative damage in broilers exposed to aflatoxin $B_1$ ($AFB_1$). Birds were randomly divided into four groups and assigned different diets: basal diet, 300 mg/kg ${\alpha}$-LA supplementation in basal diet, diet containing 74 ${\mu}g/kg$ $AFB_1$, and 300 mg/kg ${\alpha}$-LA supplementation in diet containing 74 ${\mu}g/kg$ $AFB_1$, for 3 weeks. The results revealed that the addition of 300 mg/kg ${\alpha}$-LA protected against the liver function damage of broilers induced by chronic low dose of $AFB_1$ as estimated by a significant (p<0.05) change in levels of plasma total protein, albumin, alkaline phosphatase and the activities of liver glutamic-oxalacetic transaminase and glutamic-pyruvic transaminase. The histopathological analysis also showed that liver tissues were injured in the $AFB_1$ diet, but this effect was alleviated by the addition of 300 mg/kg ${\alpha}$-LA. Additionally, $AFB_1$ induced a profound elevation of oxidative stress in birds, as indicated by an increase in malondialdehyde level, a decrease in glutathione peroxidase activity and a depletion of the glutathione content in the liver. All of these negative effects were inhibited by treatment with ${\alpha}$-LA. Our results suggest that the inhibition of $AFB_1$-induced excess production of lipid peroxides and the maintenance of intracellular antioxidant status may play important roles in the protective effects of ${\alpha}$-LA against $AFB_1$-induced oxidative damage in the liver.

The Effects of Gobonyangjeonbang Administration on Reproductive Toxicity of 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) induced Rats. (고본양정방 투여가 TCDD 유도 랫드의 생식독성에 미치는 영향)

  • Sperm formation disorders and sperm quality degradation comprises the largest proportion of male infertility caused by TCDD. To solve this problem, this study examined the effects of Gobonyangjeonbang oriental medicine prescription on the endocrine function and reproductive toxicity-related indicators in rat-induced TCDD-induced reproductive. Male SD rats were divided into five groups of seven animals and tested. The normal control group was administered the vehicle and saline, the TCDD alone group was administered intraperitoneally with TCDD (2 ㎍/kg, weeks) and physiological saline, and the test group was administered orally by dividing GYB (75, 150, and 300 mg/kg) into three concentrations for six weeks. Weight loss was observed in all groups administered TCDD. Regarding the hormonal changes, a significant decrease in free testosterone was observed in the GYB 300 mg/kg group (p<0.01). In addition, some of the germ cell destruction, seminiferous tubular atrophy, and decrease in sperm count was improved in a concentration-dependent manner in the testicular tissue of the GYB-treated group. In addition, Johnsen's score and serotoli cell index (SCI) were improved in a concentration-dependent manner (p<0.05). Overall, GYB can be used in drug therapy rather than medical procedures to solve male infertility in the future.

Immune-enhancing effect of hydrolyzed and fermented Platycodon grandiflorum extract in cyclophosphamide-induced immunosuppressed BALB/c mice

  • BACKGROUND/OBJECTIVES: The immunomodulatory effect of Platycodon grandiflorum (PG) has been reported, but studies on its mechanism are still lacking. This study was undertaken to confirm whether the hydrolyzed and fermented PG extract (HFPGE) obtained by adding hydrolysis and fermentation to the extraction process has an immune-enhancing effect in the in vivo system. MATERIALS/METHODS: Five-week-old BALB/c mice were divided into 4 groups: normal control group (NOR), control group (CON), 150 mg/kg body weight (BW)/day HFPGE-treated group (T150), and 300 mg/kg BW/day HFPGE-treated group (T300). The mice were administered HFPGE for 4 weeks and intraperitoneally injected with cyclophosphamide (CPA, 80 mg/kg BW/day) on day 6, 7, and 8, respectively, to induce immunosuppression. The levels of immunoglobulins (Igs) and cytokines were measured in the serum. In splenocytes, proliferation and cytokine levels were measured. RESULTS: Serum IgA, IgG, and IgM levels were observed to decrease after CPA treatment, which was recovered by HFPGE administration. The levels of serum interleukin (IL)-12, tumor necrosis factor (TNF)-α, IL-8, and transforming growth factor (TGF)-β were also decreased after exposure to CPA but increased after HFPGE administration. Decreased splenocyte proliferation was seen in CPA-treated mice, but was observed to increase in the T150 and T300 groups as compared to the NOR group. Compared to the CON group, splenocyte proliferation stimulated with concanavalin A (ConA) or lipopolysaccharide (LPS) in the HFPGE-treated groups was significantly increased. The cytokines secreted by ConA-stimulated splenocytes (IL-2, IL-12, interferon-γ, TNF-α) were increased in the T150 and T300 groups, and cytokines secreted by LPS-stimulated splenocytes (IL-4, IL-8, TGF-β) were also increased by HFPGE administration. CONCLUSION: These results suggest that HFPGE stimulates the immunity in immunosuppressed conditions, thereby enhancing the immune response. Therefore, it is expected that HFPGE has the potential to be used as functional food and medicine for immune recovery in various immunocompromised situations.

  • Aspirin과 고려 홍삼의 항산화 활성 성분으로 분리된 maltol을 축합하여 신물질, AFRS를 합성하였다. Aspirin율 저용량 (100-300 mg/day)으로 복용하면 말초순환 개선 효과가 있으며, 이는 aspirin의 혈소판 cyclooxygenase에 대한 특이적, 비가역적 저해 작용에 기인한다. 그러나 aspirin 복용은 위궤양을 유발한다고 알려져 있어 그 사용이 제한되어왔다. AFRS는 aspirin과 비교하여 우수한 지혈 시간 연장 효과를 보이며, aspirin보다 4배의 고용량에서 위궤양을 유발하지 않았다. 또한, AFRS는 in vivo와 in vitro에서 maltol기에 기인하는 항산화 활성을 가지며, 그 효능은 mole 비로 비교할 때 maltol과 유사하였다. AFRS는 aspirin보다 약 5배의 고용량에서 aspirin과 유사한 해열 작용을 나타내었으며, 300-450 mg/kg의 용량에서 소염작용을 나타내지 않았다.

A 26-Week Repeated Oral Dose Toxicity Test and a 4-Week Recovery Test of Cassia tora L. Water Extract in Sprague-Dawley Rats (Sprague-Dawley Rats을 이용한 결명자 물 추출물의 26주 반복 경구투여 독성시험 및 4주 회복시험)

  • Background: Cassia tora L., an annual or perennial plant of the Fabaceae family, is traditional medicine with various biological activities, including anti-constipation and, anti-inflammation. Chemical compounds such as anthraquinone glycoside and naphthalene derivatives have been isolated from this plant. Cassia tora L. is a common contaminant of agricultural commodities, but is toxic to cattle and poultry. Methods and Results: To investigate the potential toxicity, Cassia tora L. aqueous extract (CO) was administered orally to rats for 26 weeks at 0 (control), 300, 1,500 and 3,000 mg/kg/day (n = 10 for male rats for each dose). The positive control comprised animals orally administered anthraquinone 100 mg/kg/day. There was no treatment-related mortality. An increase in the kidney weight was observed at 3,000 mg/kg/day of CO and anthraquinone 100 mg/kg/day. Macrophage infiltration in the colon was observed at CO 1,500 and 3,000 mg/kg/day and anthraquinone 100 mg/kg/day, but there were no significant toxicological changes in the incidence and severity of the finding. Conclusions: The oral no-observed-adverse-effect level (NOAEL) of CO was 3,000 mg/kg/day in male rats and no target organs were identified. In addition, 300 mg/kg was found to be the no-observed-effect level (NOEL) for systemic toxicity under the conditions of the study.

Effects of Unripe Rubus coreanus Miquel Extract on Improvement of Lipid Metabolism in C57BL/6 Mice Fed a High-Cholesterol Diet (미숙과 복분자 열매 추출물이 고콜레스테롤 식이를 섭취시킨 C57BL/6 Mice의 지질대사 개선 효과)

  • Hypercholesterolemia is the presence of high levels of cholesterol in the blood, and it is regarded as a risk factor for cardiovascular disease due to the effects of cholesterol. The objective of this study was to investigate the effects of unripe Rubus coreanus Miquel (uRC) extract on lipid metabolism in hypercholesterolemic mice fed a high cholesterol diet (HC). uRC 50 (unripe R. coreanus 5% ethanol extract 50 mg/kg/day), uRC 100 (unripe R. coreanus 5% ethanol extract 100 mg/kg/day), uRC 300 (unripe R. coreanus 5% ethanol extract 300 mg/kg/day), and BO (borage seed oil containing minimum of 20% ${\gamma}$-linolenic acid 30 mg/kg/day) were orally administered for 60 days after HC. Oral administration of uRC 50, uRC 100, uRC 300, and BO significantly reduced serum total-cholesterol, triglyceride, LDL-cholesterol, glutamic oxaloacetic transaminase, glutamic pyruvic transaminase, atherogenic index, and cardiac risk factor levels. Similarly, uRC treatment elevated serum HDL-cholesterol levels. These results suggest that unripe R. coreanus extract could be established as a functional food for the improvement of lipid metabolism.

Tar Colors in Foods Distributed throughout the Gyeong-In Region;Monitoring Favorite Food Items of Children Near Elementary Schools (경인지역 유통식품 중 타르색소 실태 조사;학교주변 어린이 기호 식품을 중심으로)

  • In Korean, nine tar colors are permitted in foods. This study assessed these compounds in the favorite food items of children found near elementary schools. A total of 439 items categorized under six food types were included in the analysis. The most frequently detected tar colors were tartrazine (Y4), Brilliant Blue FCF (Y5), Allura Red, and Sunset Yellow FCF, respectively. One or a mixture of two tar colors were commonly found in products such as gums, ice bars, soft drinks, and cereals. However, most often, combinations of two or three tar colors were detected. The levels of tar colors in candies, chocolates, gums, ice bars, cereals, and soft drinks were 0.11-1169.58 mg/kg, 0.73-468.02 mg/kg, 0.10-602.46 mg/kg, 0.25-162.32 mg/kg, 0.11-753.68 mg/kg, and 0.21-69.45 mg/kg, respectively. Tar color levels were higher in chocolates and gums than in soft drinks and ice bars. And Y4 and Y5 were detected at the highest levels. For ages 7-12, the total estimated daily intake (${\sum}EDI$) of each tar color ranged from 0.004 to 1.017 mg/day/person. These values were 0.02-5.98% of the FAO/WHO's acceptable daily intake (ADI).
