Evaluating Spectral Preprocessing Methods for Visible and Near Infrared Reflectance Spectroscopy to Predict Soil Carbon and Nitrogen in Mountainous Areas (산지토양의 탄소와 질소 예측을 위한 가시 근적외선 분광반사특성 분석의 전처리 방법 비교)

  • The soil prediction can provide quantitative soil information for sustainable mountainous ecosystem management. Visible near infrared spectroscopy, one of soil prediction methods, has been applied to predict several soil properties with effective costs, rapid and nondesctructive analysis, and satisfactory accuracy. Spectral preprocessing is a essential procedure to correct noisy spectra for visible near infrared spectroscopy. However, there are no attempts to evaluate various spectral preprocessing methods. We tested 5 different pretreatments, namely continuum removal, Savitzky-Golay filter, discrete wavelet transform, 1st derivative, and 2nd derivative to predict soil carbon(C) and nitrogen(N). Partial least squares regression was used for the prediction method. The total of 153 soil samples was split into 122 samples for calibration and 31 samples for validation. In the all range, absorption was increased with increasing C contents. Specifically, the visible region (650nm and 700nm) showed high values of the correlation coefficient with soil C and N contents. For spectral preprocessing methods, continuum removal had the highest prediction accuracy(Root Mean Square Error) for C(9.53mg/g) and N(0.79mg/g). Therefore, continuum removal was selected as the best preprocessing method. Additionally, there were no distinct differences between Savitzky-Golay filter and discrete wavelet transform for visual assessment and the methods showed similar validation results. According to the results, we also recommended Savitzky-Golay filter that is a simple pre-treatment with continuum removal.

  • Introduction: Enamel matrix derivative (EMD) is a protein which is secreted by Hertwig root sheath and plays a major role in the formation of cementum and attachment of peridontium. Several studies have shown that EMD promoted the proliferation and differentiation of preosteoblasts, osteoblasts and periodontal ligament cells in vitro: however, reports showing the inhibition of osteogenic differentiation by EMD also existed. This study was designed to simultaneously evaluate the effect of EMD on the two cell lines (human mesenchymal stem cells: hMSC, human periodontal ligament derived fibroblasts: hPDLCs) by means of quantitative analysis of some bone related matrices (Alkaline phosphatase : ALP, osteopontin ; OPN, osteocalcin ; OC). Materials and Methods: hMSCs and hPDLCs were expanded and cells in the 4${\sim}$6 passages were adopted to use. hMSc and hPDLCs were cultured during 1,2,7, and 14 days with 0, 50 and 100 ${\mu}g/ml$ of EMD, respectively. ALP activity was assessed by SensoLyte ALP kit and expressed as values of the relative optical density. Among the matrix proteins of the bony tissue, OC and OPN were assessed and quantification of these proteins was evaluated by means of human OC immunoassay kit and human OPN assay kit, respectively. Results: ALP activity maintained without EMD at $1,2^{nd}$ day. The activity increased at $7^{th}$ day but decreased at $14^{th}$ day. EMD increased the activity at $14^{th}$ day in the hPDLCs culture. In the hMSCs, rapid decrease was noted in $7^{th}$ and $14^{th}$ days without regard to EMD concentrations. Regarding the OPN synthesis in hPDLCs, marked decrease of OPN was noted after EMD application. Gradual decrease tendency of OPN was shown over time. In hMSCs, marked decrease of OPN was also noted after EMD application. Overall concentration of OPN was relatively consistent over time than that in hPDLCs. Regarding the OC synthesis, in both of hPDLCs and hMSCs, inhibition of OC formation was noted after EMD application in the early stages but EMD exerted minimal effect at the later stages. Conclusion: In this experimental condition, EMD seemed to play an inhibitory role during the differentiation of hMSCs and hPDLCs in the context of OC and OPN formation. In the periodontium, there are many kinds of cells contributing to the regeneration of oral tissue. EMD enhanced ALP activity in hPDLCs rather than in hMSCs and this may imply that EMD has a positive effect on the differentiation of cementoblasts compared with the effect on hMSCs. The result of our research was consistent with recent studies in which the authors showed the inhibitory effect of EMD in terms of the differentiation of mineral colony forming cells in vitro. This in vitro study may not stand for all the charateristics of EMD; thus, further studies involving many other bone matrices and cellular attachment will be necessary.

Discriminant analysis of grain flours for rice paper using fluorescence hyperspectral imaging system and chemometric methods

  • Rice paper is an element of Vietnamese cuisine that can be used to wrap vegetables and meat. Rice and starch are the main ingredients of rice paper and their mixing ratio is important for quality control. In a commercial factory, assessment of food safety and quantitative supply is a challenging issue. A rapid and non-destructive monitoring system is therefore necessary in commercial production systems to ensure the food safety of rice and starch flour for the rice paper wrap. In this study, fluorescence hyperspectral imaging technology was applied to classify grain flours. Using the 3D hyper cube of fluorescence hyperspectral imaging (fHSI, 420 - 730 nm), spectral and spatial data and chemometric methods were applied to detect and classify flours. Eight flours (rice: 4, starch: 4) were prepared and hyperspectral images were acquired in a 5 (L) × 5 (W) × 1.5 (H) cm container. Linear discriminant analysis (LDA), partial least square discriminant analysis (PLSDA), support vector machine (SVM), classification and regression tree (CART), and random forest (RF) with a few preprocessing methods (multivariate scatter correction [MSC], 1st and 2nd derivative and moving average) were applied to classify grain flours and the accuracy was compared using a confusion matrix (accuracy and kappa coefficient). LDA with moving average showed the highest accuracy at A = 0.9362 (K = 0.9270). 1D convolutional neural network (CNN) demonstrated a classification result of A = 0.94 and showed improved classification results between mimyeon flour (MF)1 and MF2 of 0.72 and 0.87, respectively. In this study, the potential of non-destructive detection and classification of grain flours using fHSI technology and machine learning methods was demonstrated.

Suppression of Load Pendulation Using Tagline Control System for Floating Crane (해상 크레인에 의해 인양되는 중량물의 거동 감쇠를 위한 Tagline 제어 시스템)

  • This paper describes the control system to suppress the load pendulation using tagline for the floating crane. Dynamic equation of motion of the floating crane and the load is derived using Newton's 2nd law and free body model. The floating crane and the load are assumed that they move in center plane. Each rigid body has 3 DOF (surge, heave, pitch), because it moves in two directions and rotates. Then, this system, which is composed of two rigid bodies, has 6 DOF. The gravitational force, the hydrostatic force, the hydrodynamic force and the tension of the wire rope are considered as external forces, which affect to the floating crane. To suppress the pendulation of the load, the tagline, which connects between the load and the float crane, is applied to the system. The tagline is composed of the spring and the wire rope. Proportional and Derivative control is used as a linear control algorithm. The results of the numerical analysis of the 3,600 ton floating crane show that the tagline system is effective to suppress the load pendulation.

Non-Destructive Sorting Techniques for Viable Pepper (Capsicum annuum L.) Seeds Using Fourier Transform Near-Infrared and Raman Spectroscopy

  • Purpose: This study examined the performance of two spectroscopy methods and multivariate classification methods to discriminate viable pepper seeds from their non-viable counterparts. Methods: A classification model for viable seeds was developed using partial least square discrimination analysis (PLS-DA) with Fourier transform near-infrared (FT-NIR) and Raman spectroscopic data in the range of $9080-4150cm^{-1}$ (1400-2400 nm) and $1800-970cm^{-1}$, respectively. The datasets were divided into 70% to calibration and 30% to validation. To reduce noise from the spectra and compare the classification results, preprocessing methods, such as mean, maximum, and range normalization, multivariate scattering correction, standard normal variate, and $1^{st}$ and $2^{nd}$ derivatives with the Savitzky-Golay algorithm were used. Results: The classification accuracies for calibration using FT-NIR and Raman spectroscopy were both 99% with first derivative, whereas the validation accuracies were 90.5% with both multivariate scattering correction and standard normal variate, and 96.4% with the raw data (non-preprocessed data). Conclusions: These results indicate that FT-NIR and Raman spectroscopy are valuable tools for a feasible classification and evaluation of viable pepper seeds by providing useful information based on PLS-DA and the threshold value.

Rancidity Prediction of Soybean Oil by Using Near-Infrared Spectroscopy Techniques

  • Purpose: This study evaluated the feasibility of a near-infrared spectroscopy technique for the rancidity prediction of soybean oil. Methods: A near-infrared spectroscopy technique was used to evaluate the rancidity of soybean oils which were artificially deteriorated. A soybean oil sample was collected, and the acid values were measured using titrimetric analysis. In addition, the transmission spectra of the samples were obtained for whole test periods. The prediction model for the acid value was constructed by using a partial least-squares regression (PLSR) technique and the appropriate spectrum preprocessing methods. Furthermore, optimal wavelength selection methods such as variable importance in projection (VIP) and bootstrap of beta coefficients were applied to select the most appropriate variables from the preprocessed spectra. Results: There were significantly different increases in the acid values from the sixth days onwards during the 14-day test period. In addition, it was observed that the NIR spectra that exhibited intense absorption at 1,195 nm and 1,410 nm could indicate the degradation of soybean oil. The PLSR model developed using the Savitzky-Golay $2^{nd}$ order derivative method for preprocessing exhibited the highest performance in predicting the acid value of soybean oil samples. onclusions: The study helped establish the feasibility of predicting the rancidity of the soybean oil (using its acid value) by means of a NIR spectroscopy together with optimal variable selection methods successfully. The experimental results suggested that the wavelengths of 1,150 nm and 1,450 nm, which were highly correlated with the largest absorption by the second and first overtone of the C-H, O-H stretch vibrational transition, were caused by the deterioration of soybean oil.

Synchronous determination of polycyclic aromatic hydrocarbons(PAHs) in sediment of Ulsan Bay by synchronous 2nd derivative fluorescence spectrophotometry (이차 미분 형광 분광광도법에 의한 울산만 해양 저질토양 중의 다환 방향족 탄화수소(PAHs)의 동시 분석)

  • Determination of some PAHs in sediments at Ulsan bay has been carried out by extraction of the components into n-hexane followed by synchronous spectrofluorimetric technique. 11 PAHs, such as acenaphthene (Ace), anthracene (Anth), benz(a)anthracene (BaA), benzo(b)fluoranthene (BbFt), benzo(k)fluoranthene (BkFt) benzo(a)pyrene (BaP), chrysene (Chry), phenanthrene (Phen), fluoranthene(Ft), perlyrene (Per), and pyrene (Pyr) in sediment samples were able to determine separately by synchronous spectrofluorimetry. Calibration curves for those components were linear for the concentration range of 0.15~166 ppb PAHs with the correlation factor of 0.9985~0.9999. The total amount of PAHs in sediments varied from 68.8 to 324.4 ng/g. The PAHs concentration was shown a tendency to increase from the outer bay to the inner basin as well the predominant contributors to the aromatic ring groups of the PAHs was 4-ring group.

Analysis on Normal Ionospheric Trend and Detection of Ionospheric Disturbance by Earthquake (정상상황 전리층 경향 분석 및 지진에 의한 전리층 교란검출)

  • As the energy generated by earthquake, tsunami, etc. propagates through the air and disturbs the electron density in the ionosphere, the perturbation can be detected by analyzing the ionospheric delay in satellite signal. The electron density in the ionosphere is affected by various factors such as solar activity, latitude, season, and local time. To distinguish from the anomaly, therefore, it is required to inspect the normal trend of the ionosphere. Also, as the perturbation magnitude diminishes by distance it is necessary to develop an appropriate algorithm to detect long-distance disturbances. In this paper, normal condition ionosphere trend is analyzed via IONEX data. We selected monitoring value that has no tendency and developed an algorithm to effectively detect the long-distance ionospheric disturbances by using the lasting characteristics of the disturbances. In the end, we concluded the $2^{nd}$ derivative of ionospheric delay would be proper monitoring value, and the false alarm with the developed algorithm turned out to be 1.4e-6 level. It was applied to 2011 Tohoku earthquake case and the ionospheric disturbance was successfully detected.

Clinical Efficacy of Belotecan (CKD-602), Newly Developed Camptothecin Analog, in the 2nd Line Treatment of Relapsed Small Cell Lung Cancer (재발된 소세포폐암환자에서 이차 약제로 사용되는 Belotecan (CKD-602)의 임상적 효용성)

  • Background: Belotecan (Camtobell, CKD-602, Chongkundang Pharm., Korea), a camptothecin derivative, has anticancer effects by inhibiting topoisomerase I such as topotecan. This study observed the response, survival and toxicity of belotecan monotherapy after the failure of etoposide and platinum (EP). Methods: Forty nine small cell lung cancer (SCLC) patients (M/F=41/8; age, 64.5${\pm}$7.6 (mean${\pm}$SD) years), who failed in their first line chemotherapy were enrolled in this study. Twenty one SCLC patients showed relapsed lung cancer more than 90 days after their priorEP chemotherapy (sensitive relapse group, SR) and 28 patients relapsed within 90 days (refractory relapse group, RR). Results: The response rate was 25%. Eleven patients showed partial responses and 5 patients could not be checked. The response rate of the SR and RR patients was similar. The relative dose intensity was lower in the responders (78${\pm}$15%) than non-responders (83${\pm}$13%, p=0.03). The median survival time (MST) was 10.3 months (290 days). The MST of the non-responders and responders was 186 days (95% CI; 67-305) and 401 days (95% CI; 234-568, p=0.07), respectively. The median progression free survival (MPFS) was similar in the SR (79 days) and RR (67 days) patients. Grade 3-4 neutropenia, anemia, and thrombocytopenia were observed in 59.6%, 12.8% and 23.4% of patients, respectively. Conclusion: The efficacy and survival were demonstrated in the second-line setting. However, a randomized comparative trial with topotecan will be needed.
