  • Background: Our previous investigation indicated that the preparation of Panax ginseng Meyer (P. ginseng) inhibited melanogenesis. It comprised salicylic acid (SA), protocatechuic acid (PA), p-coumaric acid (p-CA), vanillic acid (VA), and caffeic acid (CA). In this investigation, the regulatory effects of P. ginseng phenolic acid monomers on melanin production were assessed. Methods: In vitro and in vivo impact of phenolic acid monomers were assessed. Results: SA, PA, p-CA and VA inhibited tyrosinase (TYR) to reduce melanin production, whereas CA had the opposite effects. SA, PA, p-CA and VA significantly downregulated the melanocortin 1 receptor (MC1R), cycle AMP (cAMP), protein kinase A (PKA), cycle AMP-response element-binding protein (CREB), microphthalmia-associated transcription factor (MITF) pathway, reducing mRNA and protein levels of TYR, tyrosinase-related protein 1 (TYRP1), and TYRP2. Moreover, CA treatment enhanced the cAMP, PKA, and CREB pathways to promote MITF mRNA level and phosphorylation. It also alleviated MITF protein level in α-MSH-stimulated B16F10 cells, comparable to untreated B16F10, increasing the expression of phosphorylation glycogen synthase kinase 3β (p-GSK3β), β-catenin, p-ERK/ERK, and p-p38/p38. Furthermore, the GSK3β inhibitor promoted p-GSK3β and p-MITF expression, as observed in CA-treated cells. Moreover, p38 and ERK inhibitors inhibited CA-stimulated p-p38/p38, p-ERK/ERK, and p-MITF increase, which had negative binding energies with MC1R, as depicted by molecular docking. Conclusion: P. ginseng roots' phenolic acid monomers can safely inhibit melanin production by bidirectionally regulating melanin synthase transcription. Furthermore, they reduced MITF expression via MC1R/cAMP/PKA signaling pathway and enhanced MITF post-translational modification via Wnt/mitogen-activated protein kinase signaling pathway.

  • This study aimed to determine the human androgen receptor (AR)-mediated endocrine disrupting potential of parabens and triclosan in food and household products using a cell-based assay in the OECD TG No.458, the 22Rv1/MMTV_GR-KO transcriptional activation assay. Four parabens (methyl-, ethyl-, propyl-, and butyl-) are determined as AR antagonists in OECD TG No.458. However, their AR antagonistic effects were not exhibited in the presence of the S9 hepatic fraction. Triclosan is also classified as an AR antagonist, and the AR antagonistic effect induced by triclosan significantly decreased in the presence of the phase I + II S9 fraction. Regarding the mechanism of AR antagonism induced by parabens and triclosan, the AR-mediated endocrine disrupting effects were exhibited through suppressing the translocation of ligand-bound AR to the nucleus via blocking of AR dimerization in the cytosol. These results indicate that the four parabens and triclosan have AR-mediated endocrine disrupting potential through an AR antagonistic effect via inhibiting AR dimerization; however, their endocrine disrupting effects deceased in the presence of hepatic metabolic enzymes.

  • Objectives: This study was conducted to evaluate and compare the tip and taper compatibility of accessory gutta-percha points (AGPs) with various rotary and reciprocating instruments. Materials and Methods: Using a profile analyzer, tip and taper measurements were taken of 10 AGPs of each of the 14 models available from Odous de Deus and the 4 models available from Dentsply-Maillefer. Diameter measurements were taken at 1-mm intervals, from 3 mm from the tip (D3) to 16 mm. Results: Based on the mean values obtained, 3-dimensional (3D) models of the AGPs were drawn in Autodesk Fusion 360 and superimposed on 3D models of each instrument selected (Mtwo, Reciproc, RaCe, K3, and ProDesign Logic) to determine the compatibility between the instrument and the AGP. Data corresponding to the tips and tapers of the various AGPs, as well as the tip and taper differences between the AGPs and the instruments, were analyzed using descriptive statistics. The tapers of the AGPs were subject to the American National Standards Institute/American Dental Association No. 57 standard. The Odous de Deus extra-long medium and extra-long extra-medium AGPs were shown to be compatible with Mtwo, K3, and ProDesign Logic instruments with taper 0.06 and tip sizes 25 and 30, while the Dentsply fine and fine medium cones were compatible with Mtwo, RaCe, and K3 instruments with conicity of 0.04 and tip sizes 35 and 40. Conclusions: Both the Odous de Deus and Dentsply commercial brands included 2 AGP models with tip (D3) and taper compatibility with Mtwo, RaCe, K3, and/or Prodesign Logic instruments.

  • In this study, dikaryotic strains of Lentinula edodes were generated by mono-mono hybridization using mating type analysis and their fruiting body characteristics were investigated. Approximately 100 monokaryotic strains were isolated from the basidiospores of Sanbaekhyhang, and homokaryotic strains were isolated from Chungheung 1ho using protoplast isolation and regeneration. Their mating types were evaluated and a total of 60 dikaryotic strains were hybridized. Using these strains, fruiting bodies were produced and their characteristics were examined after cultivation on sawdust media. The results indicated that the rate of hybridization was 100% and that 55 of 60 strains formed fruiting bodies. These showed normal pileus and gill structures; however,10 strains also produced fruiting bodies with abnormal pileus and gill structures. The weight and size of the fruiting bodies differed depending on the strains. Overall, further studies are needed for predicting the characteristics of hybridized strains based on their parental strains.

  • Physiological disorders in pear fruit are mainly caused by problems during the growing season, such as lack of calcium in the soil, poor drainage, low porosity, vigorous pruning, and excessive fruiting. In this study, soil physicochemical properties and leaf characteristics were analyzed in pear orchards in four regions of Korea where chlorosis symptoms occurred to determine the causes of chlorosis. The color of chlorotic leaves was diagnosed using the naked eye or SPAD and Hunter values. The soil of the chlorotic orchard had a significantly higher soil pH than that of the regular orchard. Although adequate soil depth was not significantly associated with chlorosis, combined with over-fertilization of the soil with lime, it could potentially impair plant iron uptake. Chlorotic leaves had significantly lower iron and calcium contents and significantly higher magnesium contents than those of regular leaves. Therefore, the intensive occurrence of chlorosis during secondary shoot development around June and July when it is hot and humid may be due to impaired iron and calcium absorption, leading to physiological disorders. To solve this problem, avoiding the over-application of lime and applying foliar fertilizers containing chelated iron is recommended.

  • In the licensing application for the deep geological disposal system of high-level radioactive waste in Finland, the disposal tunnel backfilling has been changed from the block/pellet (for the construction) to the granular type (for the operation). Accordingly, for establishing the design concept for backfilling, it is necessary to examine applicability to the domestic facility through analyzing problems of the existing method and improvements in the alternative design. In this paper, we first reviewed the principal studies conducted for changing the backfill method in the licensing process of the Finnish facility, and identified the expected problems in applying the block/pellet backfill method. In addition, we derived the evaluation factors to be considered in terms of technical and operational aspects for the backfilling solution, and then conducted a comparative analysis for two types of backfill methods. This analysis confirmed the overall superiority of the design change. It is expected that these results could be utilized as the technical basis for deriving the optimum design plan in development process of the Korean-specific deep disposal facility. However, applicability should be reviewed in advance based on the latest technical data for the detailed evaluation factors that must be considered for selecting the backfilling method.

  • This study was performed to investigate the North Korean defectors's depression through the CES-D and a projective test simultaneously. So 40 North Korean defectors and 40 South Koreans participated in this study. The results are the following. First, the North Korean defectors demonstrated more higher CES-D scores than the South Koreans, while there were not significant differences in the Rorschach depression-related indexes, DEPI, CDI, S-CON between two groups. Second, at the analysis of the sex difference effect, women among the North Korean defectors showed the most high CDI index scores. In addition, we analyzed the mean differences between two groups for morbid content, shading response, achromatic response, human movement response, form dimension response, Lamda, and the number of total response. However, there are not significant differences between two groups for those variables, except for form dimension(FD) response. It suggested the probability that the North Korean women defectors could experience more psychological distress underlyingly than the North Korean men defectors. Third, most importantly, the correlation between CES-D and DEPI in the Rorschach test was significantly different in the two groups(the North Korean defectors and the South Koreans). In conclusion, the self-report of the North Korean defectors on depression could be exaggerated. So we should be cautious to interpret the self report results of the North Korean defectors.
