Two New Diphenylethylenes from Arundina graminifolia and Their Cytotoxicity

  • Two new diphenylethylenes, gramniphenols H and I (1 and 2), together with six known diphenylethylenes (3-8), were isolated from Arundina graminifolia. The structures of 1-8 were elucidated by spectroscopic methods including extensive 1D- and 2D-NMR techniques. Compounds 1 and 2 were evaluated for their cytotoxicity against five human tumor cell lines. Compound 1 showed cytotoxicity against PC3 cells with $IC_{50}$ value of 3.5 ${\mu}M$. Compound 2 showed cytotoxicity against NB4 and PC3 cells with $IC_{50}$ values of 3.6 and 3.8 ${\mu}M$, respectively.

초고해상도 미세영상 기법을 이용한 Mouse 뇌의 자기공명영상 연구 (High-Resolution MRI Study on Mouse Brain Using Micro-Imaging)

  • 목적 : 본 연구는 핵자기공명 분광기를 개조한 미세영상 기법을 이용하여, 동물실험에 주류를 이루는 mouse를 대상으로, 0.1 mm 이내의 초고해상도 자기공명영상을 5분 정도 시간 안에 획득할 수 있는 방법을 개발하고자 하였다. 대상 및 방법 : 사용된 mouse는 C57BL/6로서 무게 50 그램 이내의 mouse를 사용하였다. 본 연구에 활용된 초전도 자석은 구경 89 mm, 4.7 T의 자기장 세기를 가진 수직형 자석이며, 사용된 샘플 코일의 직경은 30 mm 이고, 사용된 펄스시퀀스는 fast spin echo (FSE) 및 gradient echo (GE) 기법들이다. 결과 : 최적의 자기공명영상 파라미터를 확보하면서 2차원 영상으로서 수소밀도 및 T2 강조 영상을 획득하였다. 영상으로부터 mouse 뇌의 미세부분까지 상세히 해부학적 구조를 확인할 수 있었고, 또한 입체적인 정보를 획득하기 위하여 3D 영상도 부가적으로 획득하였다. 조영제를 이용한 dynamic contrast 연구에 3D 영상이 매우 유용하였다. 결론 : 본 연구를 통하여 mouse 뇌에 대한 고해상도 자기공명영상 획득을 위한 최적의 파라미터를 확보할 수 있었고, 또한 성공적인 자기공명영상도 획득하였다. 즉, 사람이나 다른 소동물뇌의 경우와 같이 mouse 뇌 조직의 다양한 부위의 미세부분을 확인할 수 있는 충분한 고해상도의 영상을 획득하였다. 최근 국내에서 mouse를 이용한 자기공명영상 연구가 시작되었으나 아직 초기단계라고 평가할 수 있고, mouse는 다른 동물에 비하여 취급/관리하기 쉬우므로 향후 mouse를 이용한 뇌 연구가 활성화 될 것으로 사료된다.

Chemical Constituents from the Stony Coral Alveopora japonica

  • Chemical investigations of the stony coral Alveopora japonica Eguchi (Poritidae) resulted in the isolation of four known compounds (1 - 4). The structures of 1 - 4 were identified as a sterol, ergosta-5,24(28)-dien-$3{\beta}$-ol (1), a mixture of monoacyl glycrols (2), eicosanoic acid and tetracosanoic acid, and two nucleosides, thymine (3) and 2'-deoxythymidine (4), respectively, on the basis of spectroscopic and physicochemical analyses including 1D- and 2D- NMR techniques as well as by comparison of their data with the published values. Compounds 1 - 4 were isolated from this species for the first time. Moreover, these compounds (1 - 4) were found in the genus Alveopora and the family Poritidae for the first time.

Butyrolactones Derivatives from the Fermentation Products of an Endophytic Fungus Aspergillus versicolor

  • Two new butyrolactones, asperphenol A (1) and B (2), together with four known butyrolactones (3-6) were isolated from the fermentation products of an endophytic fungus Aspergillus versicolor. Their structures were elucidated by spectroscopic methods including extensive 1D- and 2D-NMR techniques. Compounds 1-6 were also tested for their anti-tobacco mosaic virus (anti-TMV) activities. The results showed that compound 2 exhibited high anti-TMV activity with inhibition rate of 46.7%. The other compounds also exhibited potential anti-TMV activities with inhibition rates in the range of 21.8-28.4%.

Antioxidant, antinociceptive activity and general toxicity study of Dendrophthoe falcata and isolation of quercitrin as the major component

  • Plants have been used in traditional medicinal system for centuries. Bangladeshi medicinal plants have received considerable attention from the researchers for evaluation of their bioactivity. As a part of our ongoing research of screening the Bangladeshi medicinal plants, the ethanolic extract of Dendrophthoe falcata have been chosen for the present study. The ethanolic extract of the leaves of the plant have been assessed for their antioxidant, antinociceptive, and general toxicity. The extract showed potent antioxidant activity ($IC_{50}5.1{\mu}g/ml$) using DPPH radical scavenging assay, which is comparable to the standard ascorbic acid ($IC_{50}4.6{\mu}g/ml$). The extract significantly and dose dependently inhibited the acetic acid induced writhing in mice (71.2%, P < 0.001 and 28.0%, P < 0.05 for 500 and 250 mg/kg body weight, respectively). A general toxicity was assessed by a simple and low cost assay using brine shrimp lethality as an indicator. The extract showed low level of toxicity ($LC_{50}100{\mu}g/ml$). Using different chromatographic techniques, quercitrin (quercetin 3-O-${\alpha}$-rhamnoside) was separated as the major component from the extract. The structure was elucidated by detailed 1D and 2D NMR and mass spectral analysis.

Minor Phenolic Constituents of the Anemarrhenae Rhizoma

  • A homoisoflavanone, 7,4'-dihydroxyhomoisoflavanone (1) and a flavanone, (2S)-7,4'-dihydroxy-5-methoxyflavanone (2), were isolated from the rhizomes of Anemarrhena asphodeloides, together with 4,4'-dihydroxychalcon (3), 2'-O-methylphlorethin (4), 1,3-bis-di-p-hydroxyphenyl-4-penten-1-one (5), and 2,4'-dihydroxy-4-methoxybenzophenone (6) on the basis of spectroscopic and physicochemical analyses including 1Dand 2D- NMR techniques as well as by comparison of their data with the published values. Compounds 1 - 4 were isolated for the first time from this plant source. Among isolates, compound 2 exhibited moderate inhibitory effect on the differentiation of pre-adipocyte 3T3-L1 cells.

New Polytriazoleimides with High Thermal and Chemical Stabilities

  • A series of novel polytriazoleimides were prepared from various aromatic dianhydrides and a new kind of 1,2,3-triazole-containing aromatic diamine synthesized by the Cu (I)-catalyzed 1,3-dipolar cycloaddition reaction in DMAc, and characterized by FT-IR, $^1H$-NMR, XRD, DSC and TGA techniques. The results show the polytriazoleimides are soluble in most of strong polar solvents and have inherent viscosity values of 0.51-0.62 dL/g(DMAc). The polytriazoleimide films exhibit a tensile strength of 62.3-104.5 MPa and an elongation at breakage of 4.0-8.1%, a glass transition temperature ($T_g$) of $257-275^{\circ}C$, a decomposition temperature (at 5% weight loss) of $350-401^{\circ}C$ in $N_2$ atmosphere, and a dielectric constant of 2.47-3.01 at 10 MHz, which depend on the structure of the polymers. The polytriazoleimides perform good resistance to acid and alkali solution.

Isolation of Candida albicans Chitin Synthase 1 Inhibitor from Streptomyces sp. A6705 and Its Characterization

  • In the course of searching for potent chitin synthase 1 inhibitors from natural resources, Streptomyces sp. A6705 was found to exhibit potent inhibitory activity against the chitin synthase 1 from C. albicans (CaCHS1p). As a result, the inhibitor was isolated and identified using a series of chromatographies. Through chemical analyses with UV spectrophotometry, MS spectrometry, and various NMR techniques, the inhibitor was identified as N,N-bis(2-phenylethyl)urea. The compound exhibited strong inhibitory activity against the chitin synthase 1 from C. albicans with an $IC_{50}$ of 14 ${\mu}g$/ml, representing a similar inhibitory activity to that of the well-known chitin synthase inhibitor, polyoxin D ($IC_{50}$ 15 ${\mu}g$/ml). However, the compound showed no inhibitory activity against the chitin synthase 2 of Saccharomyces cerevisiae up to 280 ${\mu}g$/ml, which is structurally and functionally analogous to CaCHS 1 p. In addition, the compound exhibited weak antifungal activities against Cryptococcus neoformans and Rhizoctonis solani.

Anti-tumor Constituents from Isodon xerophilus

  • Results of chemistry and biological activity of many years indicate that plants belonging to the Isodon genus are rich in ent-kaurane diterpenoids, which have been revealed to possess biological activities such as antitumor, antibacterial and antiinflammatory effects. In continuation of our research on diterpenoids in medicinal plants of this genus, the acetone extract from the leaves of I. xerophilus, which is a plant native to Yunnan province of China, showed potent antitumor activity against K562. After partition, the most active EtOAc part was studied. Four new diterpenoids named xerophilusin A(l), B(2), C(3), D(4), and eight known compounds including macrocalin B(5) and rabdorosthomin A(6) were isolated, whose structures were elucidated through a series of one- and two-dimensional NMR techniques(DEPT, COSY, HMQC, HMBC and ROESY experiments). Among them, compound 1, 2 and 5 had two unique epoxy units formed by two ether bridges from C-20 to C-7, C-14. Up to now, there are four compounds having such an peculiar structure besides these three compounds. Compound 3 and 4 were two of the few examples possessing $1{\beta}$ substitutes. All the diterpenoid compounds were subjected to the antitumor screening. It is interesting that only xerophilusin A(l), B(2) and macrocalin(5) exhibited significant antitumor activity against K562 by the method of MTT($IC_{50}$ were listed in Table 1.). The results inspired us to infer that the unique ether bridges from C-20 to C-7, C-14 possibly played an important role in the antitumor activity.

New Algicidal Compounds from a Marine Algicidal Bacterium against Cochlodinium polykrikoides

  • In screening of algicidal bacteria, we isolated a marine bacterium which had potent algicidal effects on harmful algal bloom (HAB) species. This organism was identified as a strain very close to Bacillus subtilisby 16S rRNA gene sequencing. This bacterium, Bacillus sp. SY-1, produces very active algicidal compounds against the harmful dinoflagellate Cochlodinium polykrikoides. We isolated three algicidal compounds (MS 1056, 1070, 1084) and identified them by amino acid analyses, fast atom bombardment mass spectrometry (FAB-MS), infrared spectroscopy (IR), $^1H$, $^{13}C$, and extensive two-dimensional nuclear magnetic resonance (2D NMR) techniques including $^1H-^{15}N$ HMBC analysis. One of them, MS 1056, contains a b-amino acid residue with an alkyl side chain of $C_{15}$. MS 1056, 1070, and 1084 showed algicidal activities against C. polykrikoides with an $LC_{50}$ (6 hrs) of 2.3, 0.8, $0.6\;{\mu}g/ml$, respectively. These compounds also showed significant algicidal activities against other harmful dinoflagellates and raphidophytes. In contrast, MS 1084 showed no significant growth inhibition against various organisms coexisting with HAB species in natural environments, including bacteria, eukaryotic microalgae, and cyanobacteria, although it inhibited growth of some fungi and yeasts. These observations imply that algicidal bacterium Bacillus sp. SY-1 and its algicidal compounds could play an important role in regulating the onset and development of HABs in the natural environments.
