약산성 다당의 선택적 분해 과정에서 얻어진 특이당 함유 Oligo당의 구조적 분석 (Structural Analysis of the Unusual Sugar-Containing Oligosaccharides Formed by the Selective Cleavage of Weakly Acidic Polysaccharide)

  • 약산성 다당 GL-4IIb2'에 대해 연속적 부분 산가수 분해 과정을 거쳐, 2종의 oligo당 획분 PA-2' 및 PA-1-III를 분리하고 그들의 구조분석을 행하였다. PA-2'는 주요 구성당으로 Rha 와 특이당 Kdo를 동일한 비율로 함유하고 있었으며, permethylated oligosaccharide-alditol로 전환시켜, GC-MS로 분석한 결과, m/z 189$(bA_1)$및 m/z 308 $(aJ_2)$에서 6-deoxyhexose와 Kdo에 기인하는 fragment ion이 관찰되었다. 이 peak는 m/z 162에서 특징적인 ion을 나타낸 반면 m/z 177 ion이 관찰되지 않음으로써 Kdo의 C4위치가 아닌 C5위치가 치환되어 있음을 알 수 있었다. 한 methyl화 분석 결과, PA-2'은 환원말단 Rhap와 5-substituted Kdo가 높은 비율로 검출되었으며 환원 형태의 PA-2'를 대상으로 $^1M-NMR$ 분석을 하였을 때 ${\delta}$ 5.09 ppm에서 ${\alpha}-L-Rha$의 anomeric proton에 기인한 단일 signal이 관찰되었다. 이상의 결과로 부터 PA-2'는 주로 ${\alpha}-L-Rhap-(1{\rightarrow}5)-Kdo$로 구성되었다는 사실을 알 수 있었다 한편. PA-1-III는 소량의 Ara 믹 Dha와, 다량의 Rha 및 Kdo가 주요 구성당으로 나타났으며, PA-1-III 획분의 permethylated oligosaccharide-alditol 유도체는 GC상에서 3개의 peak (1P, 2P 및 3P)로 분리되었다. 이들의 MS 분석 결과, 높은 비율로 검출된 1P는 $Rhap-(1{\rightarrow}5)-Kdo$의 구조로 나타났으며 반면 낮은 비율의 2P및 3P는 $Araf-(1{\rightarrow}5)-Dha$의 동일구조를 갖는 것으로 확인되었는데 이는 Dha의 carbonyl 환원과정 중 생성된 epimer로 판단되었다.

Pattern Recognition of the Herbal Drug, Magnoliae Flos According to their Essential Oil Components

  • This paper describes a pattern recognition method of Magnoliae flos based on a gas chromatographic/mass spectrometric (GC/MS) analysis of the essential oil components. The botanical drug is mainly comprised of the four magnolia species (M. denudata, M. biondii, M. kobus, and M. liliflora) in Korea, although some other species are also being dealt with the drug. The GC/MS separation of the volatile components, which was extracted by the simultaneous distillation and extraction (SDE), was performed on a carbowax column (supelcowax 10; 30 m{\time}0.25 mm{\time}0.25{\mu}m$) using temperature programming. Variance in the retention times for all peaks of interests was within RSD 2% for repeated analyses (n = 9). Of the 74 essential oil components identified from the magnolia species, approximately 10 major components, which is $\alpha$-pinene, $\beta$-pinene, sabinene, myrcene, d-limonene, eucarlyptol (1,8-cineol), $\gamma$-terpinene, p-cymene, linalool, $\alpha$-terpineol, were commonly present in the four species. For statistical analysis, the original dataset was reduced to the 13 variables by Fisher criterion and factor analysis (FA). The essential oil patterns were processed by means of the multivariate statistical analysis including hierarchical cluster analysis (HCA), principal component analysis (PCA) and discriminant analysis (DA). All samples were divided into four groups with three principal components by PCA and according to the plant origins by HCA. Thirty-three samples (23 training sets and 10 test samples to be assessed) were correctly classified into the four groups predicted by PCA. This method would provide a practical strategy for assessing the authenticity or quality of the well-known herbal drug, Magnoliae flos.

Composition and Antimicrobial Activities of Essential Oils in the Peel of Citrus Fruits

  • Volatile components of the essential oils of Satsuma mandarin (C. unshiu), Dangyuza (C. grandis), Yuza (C. junos), Byungkyul (C. playtymamma), Jinkyul (C. sunki), and Hakyul (C. natsudaidai) grown in Jeju Island were isolated from the fruit peels by hydro distillation and determined by GC-MS. GC-MS analysis identified 58 compounds, with main components being d-limonene $(64.01{\sim}79.34%),\;{\beta}-myrcene\;(3.01{\sim}26.53%),\;{\gamma}-terpinene\;(0.11{\sim}12.88%),\;{\beta}-pinene\;(0.78{\sim}4.74%),\;and\;{\alpha}-pinene\;(1.01{\sim}2.55%)$. Differences in compositions and contents of the essential oils were observed among citrus varieties. Effects of citrus oils on growth inhibitions of Escherchia coli, Staphyllococcus epidermidis, and Candida albicans were investigated using disc diffusion assay and minimal inhibitory concentration (MIC) assay. The essential oils inhibited growths of the test organisms, exhibiting higher levels of activity against Gram-positive S. epidermidis (MIC values $0.04{\sim}0.17mg/mL$), whereas Gram-negative E. coli was moderately resistant (MIC values $1.66{\sim}20.30mg/mL$). MIC of citrus essential oils ranged from $0.82{\sim}23.69mg/mL$ against C. albicans. The essential oils obtained from C. sunki, C. grandis, and C. playtymamma showed the highest antimicrobial activities against S. epidermidis and C. albicans, indicating their potential as natural antimicrobial agents.

Volatile Component of Pine Needles from Pinus densiflora S. using Solid Phase Microextraction-Ges Chromatography-Mass Spectrometry

  • The volatile components of Pinus densiflora needles were studied by gas chromatography-mass spectrometry(GC-MS), using seven kinds of solid phase microextraction (SPME) fibers, seven in SPME fibers: 100 ${\mu}m$ PDMS, 65 ${\mu}m$ PDMS/DVB, 65 ${\mu}m$ SF-PDMS/DVB, 85 ${\mu}m$ PA, 75 ${\mu}m$ CAR/PDMS, 65 ${\mu}m$ CW/DVB and 50/30 ${\mu}m$ DVB/CAR/PDMS fibers. A total of 40 components were identified by using the seven different SPME fibers. The identified components were classified, according to their functionalities, as follows: 26 hydro-carbons, 7 alcohols, 4 carbonyl compounds, and 3 esters. The major volatile components of Pinus densiflora needles identified by these SPME fibers were $\alpha$-pinene ($1.7\~21.7\;{\mu}g/g$), $\beta$-myrcene ($2.0\~20.1\;{\mu}g/g$), $\beta$-phel-landrene ($4.6\~22.8\;{\mu}g/g$), $\beta$-caryophyllene ($6.7\~26.0\;{\mu}g/g$) germacrene D ($1.1\~11.9\;{\mu}g/g$). In the comparison of the seven SPME fibers, PDMS appeared to be the most suitable fiber for the analysis of hydrocarbon compounds and CAR/DPMS, PDMS/DVB, CW/VB and DVB/CAR/PDMS are shown to be optimal for analysis of the alcohols and carbonyl compounds.

천궁의 세포배양에 의한 정유성분의 생산 (The Production of Essential Oils by Tissue Culture of Cnidium officinale)

  • Callus was derived from the shoots of Cnidium officinale. The growth rate of callus and the production of essential oils were studied under different culture conditions. The essential oils in the rhizome of Cnidium officinale and the cultivated callus were analyzed and compared by gas chromatography and mass spectrometry. It appeared that NAA induced higher growth rate and production of essential oils than 2,4-D. The compositions of essential oils were influenced by the illumination. Butyl phthalide, cnidilide, senkyunolide, butylidene phthalide, ligustilide, grandisol, tricosane, 3-methylphenol and 2-pentylthiophene were identified in the cultivated callus.

Antimicrobial Activity of Essential Oil of Pinus koraiensis Seed Against Pathogens Related to Acne

  • The purpose of the present research was to evaluate the antimicrobial activity of the essential oil extracted from Pinus koraiensis seed against pathogens related to acne. The essential oil was extracted by steam distillation method. The chemical compositions of essential oil were analyzed by GC-MS. Alpha-pinene (29.87%), D-limonene (19.26%), betapinene (11.19%), beta-myrcene (3.84%), n-hexadecanoi acid (3.2%), beta-caryphyllene (2.72%), and cyclohexene (2.17%) were main components. This essential oil had antimicrobial activities against Malasseizia furfur, Propionibacterium acnes, and Staphylococcus epidermidis.

삶은 달걀의 부패에 따른 부위별 냄새물질의 발생특성 연구 (Emission Characteristics of Odorous Gases with the Decay of Albumin and Yolk of Boiled Egg)

  • In this study, the concentration of odorants released from albumin (EA) and yolk (EY) portions of boiled egg samples were determined as a function of storage time. The concentrations were measured at storage days of 0, 1, 3, 6, and 9 under room temperature. As such, odorants produced during both fresh and decay conditions were measured through time. A total of 19 compounds were selected as the main target odorants along with 12 reference compounds. GC-MS (for VOC) and GC-PFPD system (for sulfur gases) equipped with thermal desorption (TD) system were employed for odorant analysis in this work. The initial concentrations measured from the chamber system were converted into flux terms ($ng{\cdot}g^{-1}{\cdot}min^{-1}$). The EA showed the highest concentration of $H_2S$ (234 $ng{\cdot}g^{-1}{\cdot}min^{-1}$) at EA-0, and the concentrations of AT (Acetone) was also seen clearly in the range of 11.7 (EA-0) to 58.6 $ng{\cdot}g^{-1}{\cdot}min^{-1}$ (EA-9). The EY showed similar patterns. EtAl (Ethyl alcohol) increased 9.47 (EA-1) to 96.7 $ng{\cdot}g^{-1}{\cdot}min^{-1}$ (EA-9) in EA samples. Ketone, alcohol, sulfur groups generally exhibited high concentrations compared to other odorants. These data were also compared in relation to olfactometry related dilution-to-threshold (D/T) ratio by air dilution sensory (ADS) test and sum of odor intensity (SOI).

Combination of air stripping and biological processes for landfill leachate treatment

  • Landfill waste decomposition generates a dark effluent named, leachate which is characterized by high organic matter content. To minimize these polluting effects, it becomes necessary to develop an effective landfill leachate treatment process. The objective of this study was to evaluate the performance of an innovative approach based on air stripping, anaerobic digestion (AD) and aerobic activated sludge treatment. A reduction of 80% of ammonia and an increase of carbon to nitrogen ratio to 25 were obtained, which is a suitable ratio for AD. This latter AD was performed in fixed bed reactor with progressive loading rate that reached 2 and 3.2 g COD/L/d for the raw and diluted leachate (1:2), respectively. The anaerobic treatment led to significant removal of chemical oxygen demand (COD) and biogas production, especially for the diluted leachate. The COD removal was of 78% for the raw leachate and a biogas production of 4 L/d with 70% methane content. The use of the diluted leachate led to 81% of COD removal and 7 L/d biogas with 75% methane content. It allowed a removal of 77% COD and more than 97% of the organic compounds present in the initial leachate sample.

Structure and Foaming Properties of Viscous Exopolysaccharides from a Wild Grape-Associated Basidiomycetous Yeast Papiliotrema flavescens Formerly Known as Cryptococcus flavescens

  • Exopolysaccharide produced by the yeast Papiliotrema flavescens, isolated from wine grape berries of Champagne vineyard, was investigated for both chemical and functional characterization. SECMALLS and colorimetric assay analyses showed that the EPS is a high MW heteropolymer (2.37 × 106 g/mol) majorily consisting of mannose, glucose, xylose and glucuronic acid as monosaccharide constituents, with two substituents (sulphate and phosphate groups), and a minor protein moiety. Structural enchainment of these carbohydrates based on methylation, GC-MS and NMR analyses revealed a linear main backbone built up of α-(1 → 3)-D-mannopyranosyl residues on which are branched side chains consisting of a single β-D-glucopyranosyluronic acid residue and β-(1 → 2)-xylopyranoses (2-5 residues). Suggestion of some xylopyranose side chains containing a mannose residue at the nonreducing terminal end was also proposed. This is first report on EPSs from the grape P. flavescens yeast with such structural characteristics. Furthermore, investigations for valuating the application performance of these EPS in relation with their structural features were carried out in 8% alcohol experiment solutions. Very exceptional viscosifying and foaming properties were reported by comparison with commercial biopolymers such as Arabic, gellan and xanthan gums. The intrinsic properties of the natural biopolymer from this wild grape-associated P. flavescens yeast make it a potential candidate for use in various biotechnology applications.

4-Isopropyl, 5-isopropyl-3-methylphenol 유도체들의 합성과 식물 병원균에 대한 항균 활성 (Antifungal activities for derivatives of 4-isopropyl-3-methylphenol and 5-isopropyl-3-methylphenol against plant pathogenic fungi)

  • 살균활성이 있는 4-isopropyl-3-methylphenol(I)과 5-isopropyl-3-methylphenol(II)을 출발물질로 하여 ester, sulfonyl ester, carbamate, ether 및 phosphoyl ester계열의 50개 화합물들을 합성하였으며, IR, GC/MS와 $^1H$-NMR spectrum을 이용하여 합성을 확인하였다. 이 유도체들에 대한 in vitro 살균활성 실험을 10종의 식물병원균에 대하여 실시한 결과 몇 가지 화합물이 우수한 항균활성을 나타내었다. 이들 화합물을 5가지 식물병(벼도열병, 토마토역병, 오이 잿빛곰팡이병, 벼 잎집무늬 마름병, 오이탄저병)에 대하여 in vivo 항균활성을 조사하였다. 그 결과, 4-isopropyl-3-methylphenyl (2-aminothiazole-4-yl)methoxyiminoacetate(I-7a)는 벼 도열병(Pycularia oryzae)에 methyl (4-isopropyl-3-methylphenoxy)acetate(I-4d)와 methyl (5-isopropyl-3-methylphenoxy) acetate (II-4d)는 오이 잿빛곰팡이병(Botrytis cinerea)에 높은 방제활성을 나타내었다.
