Effect of DHU001, a Polyherbal Formula, on Dinitrofluorobenzene-induced Contact Dermatitis (Type I allergy)

  • The effect of DHU001, a mixed herbal formula consisted of 7 types aqueous extracts for various respiratory disorders were evaluated on 2,4-dinitrofluorobenzene (DNFB)-induced contact dermatitis, type I allergic model. Contact dermatitis was induced by sensitization with dinitrophenyl-derivatized ovalbumin (DNP-OVA) and DNFB challenge as antigen. Two different dosages of DHU001 (300 and 150 mg/kg) were orally administered to DNP-OVA sensitization mice once a day for 7 days with reference material, dexamethasone (15 mg/kg, intraperitoneal treatment). End of 7 days oral administration of DHU001 extracts or intraperitoneal treatment of dexamethasone, the changes on the edematous changes and scratching behavior were measured. Immediate after DNFB challenge on ear or paw of DNP-OVA sensitized mice, increases of ear and paw thicknesses and weights were detected with anterior ear skin (dermis to epidermis) thickness and paw scratching behavior increases. However, these contact dermatitis signs induced by DNFB treatment were reduced by treatment of the both different dosages of DHU001 and dexamethasone, respectively. The results obtained in this study suggest that oral treatment of DHU001 extracts also has relatively favorable effects on contact dermatitis.

However, these DNFB-induced increases on ear and paw thicknesses, weights and scratching behaviors were decreased by treatment of all three different dosages of PR extracts and dexamethasone, respectively. In addition, the increases of anterior skin thicknesses were also dramatically inhibited by treatment of all three different dosages of PR extracts and dexamethasone at histopathological observations. The results obtained in this study suggest that oral treatment of PR extracts also has relatively favorable effects on allergic dermatitis.

Regulation of macrophage inflammatory protein-2 gene expression in response to 2,4-dinitrofluorobenzene in RAW 264.7 cells

  • Several skin sensitizers, like 2,4-dinitrofluorobenzene (DNFB), are known to provoke contact hypersensitivity responses after topical application. Here, we show that DNFB can upregulate macrophage inflammatory protein-2 (MIP-2) expression in RAW 264.7 cells via a mechanism that is largely dependent on mitogen-activated protein kinase (MAPK) signaling pathways. ELISA-based transcription factor activation assays and chromatin immunoprecipitation assays revealed that functional interaction between AP-1 and MIP-2 promoter element is necessary for MIP-2 gene expression by DNFB. Interestingly, topical application of DNFB to NC/Nga mice increased MIP-2 expression in dermis, suggesting that MIP-2 contributes to the leukocyte infiltration associated with atopic dermatitis. These results provide additional insight of the mechanism of contact hypersensitivity induced by contact sensitizers.

  • This study aims to evaluate the anti-inflammatory effect of 80% ethanol extracts of Stachys affinis (MQ) on 1-fluoro-2,4-Dinitrofluorobenzene (DNFB) induced atopic dermatitis (AD) in Balb/c mice. AD-like allergic contact dermatitis was induced by challenge of DNFB on the ear after DNFB sensitization on the back sides of mice. MQ alleivated clinical severity in AD-like skin lesions. In addition, ear thickness of epidermis and penetration of inflammatory cells in AD-like skin lesions were decreased by topical application of MQ. The the serum levels of tumor necrosis factor-α (TNF-α) and interleukin-4 (IL-4) were measured in AD mice using ELISA kits. Levels of TNF-α and IL-4 in serum were significantly decreased by topical application of MQ. Therefore, this study could give a clinical basis that MQ could be a agent to prevent AD.

  • A procedure for the high-performance liquid chromatographic determination of Nebramycin factors in fermentation broth of Streptoalloteichus hindustanus ATCC 31218 mutant was investigated using pre-column derivatization and LTV detection. The method is based on pre-column derivatization of Nebramycin factors with 2,4-dinitrofluorobenzene(DNFB) in the presence of Tris (hydroxymethyl)aminoethane. The chromatographic separation of derivatives of Nebramycin factors and unknown impurities is achieved using reversed-phase column (NOVA-PAK $C_{18}$, Waters Co.) and AcCN : H$_{2}$O : AcOH (53.0:46.5:0.5) as a mobile phase. The mixture of these derivatives were separated within 35 minutes and the optimum wavelength($\lambda_{max}$ ) of the UV detector was 353 nm. The linearity of response for derivatives of Nebramycin factors is demonstrated for concentrations up to 500 $\mu\textrm{g}$/ml and the relative standard deviation is less than 0.79%. Detection limit was 1.67 ng for the 10 $\mu\textrm{l}$ sample volume employed.

Effects of nanoparticulate saponin-platinum conjugates on 2,4-dinitrofluorobenzene-induced macrophage inflammatory protein-2 gene expression via reactive oxygen species production in RAW 264.7 cells

  • Nanoparticulate platinum (II) (nano Pt) is a powerful antioxidant that is widely used to scavenge reactive oxygen species (ROS). The antioxidant activity of nano Pt has gained attention as a potentially useful therapeutic for a variety of diseases including cancer and aging. In the present study, we prepared nanoparticulate saponin-Pt (II) (nano saponin-Pt) conjugates using the ethanol reduction method to enhance the permeability and retention effect of Pt. The nano saponin-Pt conjugates were found to restore the viability of approximately 40% of 2,4-dinitrofluorobenzene (DNFB)-treated RAW 264.7 cells. In addition, we found that nano saponin-Pt conjugates acted as a potent antioxidant that reduced the production of ROS and inhibited activation of the MAP kinase pathway and MIP-2 gene expression in response to DNFB. These results provide insight into the potential usefulness of nano saponin-Pt conjugates as a treatment for contact hypersensitivity.

Differential expression of cell surface markers in response to 2,4-dinitrofluorobenzene in RAW 264.7 and primary immune cells

  • We evaluated the expression of the costimulatory molecules CD80 and CD83 and major histocompatibility (MHC) class II induced by 2,4-dinitrofluorobenzene (DNFB) in the RAW 264.7 macrophage cell line. In contrast to the previously reported effect of DNFB on dendritic cells, CD86 expression did not change. Furthermore, we observed that the CD83 expression level transiently increased and then decreased. Induction of CD80 and MHC class II molecule expression and a decrease in CD83 expression by DNFB in vitro were also confirmed in splenocytes of BALB/c and NC/Nga mice. However, DNFB did not influence CD83 expression in peritoneal $CD11b^+$ cells from BALB/c or NC/Nga mice. Detailed in vivo experiments and further studies on the possible contribution of $CD11b^+$ cells to induce atopic dermatitis (AD) would be helpful to attain a better understanding of AD pathogenesis.

Effects of Angelicae Dahuricae Radix on 1-fluoro-2,4-dinitrofluorobenzene (DNFB)-induced contact dermatitis in mice (백지(白芷) 추출물이 DNFB에 의하여 유발된 생쥐의 접촉피부염에 미치는 영향)

  • In the theory of Korean medicine, Angelicae Dahuricae Radix (ADR) can expel wind and relieve exterior syndrome, and eliminate dampness. Recently, ADR has been reported to have possibilities as anti-inflammatory agent and cosmetics. The purpose of this study is to evaluate the efficacy of ADR on contact dermatitis (CD). In order to investigate the anti-inflammatory effects of ADR on CD, we investigated the effects of ADR on ear thickness, ear weight, skin lesion and histopathological changes in mice with CD induced by 1-fluoro-2,4-dinitrofluorobenzene (DNFB). In addition, the effect on spleen weight was also measured. In our results, topical application of ADR lowered ear thickness and weight respectively. ADR treatment also improved skin lesions such as erythema and scale. In the histopathological observation, ADR-treated group showed diminished epidermal hyperplasia and immune cell infiltration in inflammed tissues compared to those of non-treated control group. In conclusion, These data suggest that ADR has anti-inflammatory action in inflammed skin tissue, resulting in improving skin lesion and histopathological abnormalities of CD.

Effect of Lithospermi Radix on Contact Dermatitis Induced by Dinitrofluorobenzene in Mice

  • Objectives: The root of Lithospermum erythrorhizon Sieb. et Zucc. (Lithospermi Radix, LR) is a kind of heat clearing and blood cooling medicinal herbs. It can clear away heat and cool the blood, reduce toxins and disperse maculae. LR has long been used as efficacious therapy for inflammation, burns, frostbite and skin diseases such as eczema and psoriasis. Methods: In the present study, we investigate anti-allergic and anti-inflammatory effects of LR by using the 1-fluoro-2, 4- dinitrofluorobenzene (DNFB)-induced contact dermatitis mouse model. Results: Topical application of 10 mg/mL of LR effectively inhibited skin lesions induced by repeated paintings with DNFB. Topical application of LR also inhibited hyperplasia, edema, spongiosis and infiltrations of mononuclear cells. In addition, production levels of total immunoglobulin and IgG1 in serum were decreased by using LR in vivo. Conclusions: These data suggest that LR acts as an antiinflammatory agent, improving skin lesions in CD mice.

The Ameliorative Effects of Gagam-GongJin-dan (WSY-1075) in Contact Dermatitis-induced Animal Model (가감공진단(加減拱辰丹) (WSY-1075)의 접촉성 피부염 유발 모델동물에 대한 개선효과)

  • Objectives : This study was performed to assess the ameliorative effects of Gagam-GongJin-dan (WSY-1075) composited with Corni Fructus, Angelica gigantis Radix, Lycii Fructus, Ginseng Radix, Cervi parvum Cornu and Cinnamomi Cortex in contact dermatitis animal model. Methods : WSY-1075 was orally administrated the various concentrations (50-400 mg/kg, body weight/day) with one time per day for 10 days from 4 days after DNFB sensitization. We investigated ameliorative effects of WST-1075 on the scratching behavior, skin clinical serverity and inflammatory mediators in 2,4-dinitrofluorobenzene (DNFB)-induced contact dermatitis mice. Results : The orally administration of WSY-1075 (400 mg/kg) inhibited the scratching behavior induced by sensitization and challenge with DNFB. WSY-1075 (200 mg/kg or 400 mg/kg) administration also reduced the skin damage, inflammatory mediators, mast cell infiltration induced by DNFB. Moreover, WSY-1075 (above 200 mg/kg) administration inhibited the serum levels of IgE and IL-4 in DNFB-induced contact dermatitis mice. Conclusions : These results suggest that WSY-1075 administration (200 mg/kg or 400 mg/kg) has the ameliorative effects on the scratching behavior, the clinical signs, mast cell infiltration and inflammatory mediators in DNFB-induced contact dermatitis animal model mice.
