Histopathological Observations on Hemorrhagic Fatty Liver in Chicken Induced by Thyroidectomy or Thiouracil Administration (갑상선절제(甲狀腺切除) 및 Thiouracil 투여(投與)에 기인(起因)한 닭의 출혈성(出血性) 지방간(脂肪肝)에 관한 병리조직학적관찰(病理組織學的觀察))

  • In the chickens removed the left thyroid gland or administrated with thiouracil, fatty liver (FL) and fatty liver-hemorrhagic syndrome (FLHS) were induced within five weeks after treatment. FL was observed in all of the thyroidectomized layer and broiler hens, and the occurrence of FLHS was higher in rate at the fifth week than that at the third week after removal. FL and FLHS were observed in the thiouracil-administrated broiler hens, but the rate of incidence was appeared to be lower in the thiouracil-administrated hens than in the thyroidectomized chickens. It was suggested that hypothyoidism could be an important factor inducing FLHS.

EFFECTS OF PALMIJIHWANGTANGS ON HYPOTHYROIDISM IN RATS INDUCED BY THIOURACIL (팔미지황탕(八味地黃湯)이 Thiouracil로 유발(誘發)한 흰쥐의 갑상선기능저하증(甲狀腺機能低下症)에 미치는 영향(影響))

  • In order to determine the effects of Palmijihwangtangs on the hypothyroidism, the biochemical and morphologic effects of PALMIJIHWANGTANGS against 2-thiouracil-induced hypothyroidism in rats were examined. 1. Serum $T_3$ value was significantly incresed by the treatment with PALMIJIHWANGTANG-1, 2 and 3(p<0.05). 2. Serum $T_4$ value was significantly higher in PALMIJIHWANGTANG-1 and PALMIJIIIWANGTANG-2 treated groups than that of control group. In PALMIJIHWANGTANG-3 treated group, serum $T_4$ level was significantly higher and that of control group(p<0.01). 3. Scrum free $T_4$ value was significantly incresed in the PALMIJIHWANGTANG-1 and PALMIJIHWANGTANG-2 treated groups(p<0.05) 4. Total cholesterols was significantly decresed by the treatment with PALMIJIHWANGTANG-1(p<0.05). 5. The treament with the PALMIJIHWANGTANG-1, 2 and 3 resulted in the significantly decrease of serum ALT(p<0.05). 6. Thyroid gland weight was significantly increased in PALMIJIHWANGTANG-3 treated group(p<0.01). 7. Treatment with PALMIJIHWANGTANG-1, 2 and 3 effectively recovered the dystropy and degeneration of follicle in thyroid gland caused by 2-thiouracil. 8. There was no histological and functional lesion observed in the liver after treatment with PALMIJIHWANGTANG-1, 2 and 3. These results suggest that PALMIJIHWANGTANG may be effective in the treatment of hypothyroidism.

The Effects of Aconiti Radix on Thyroid Function in Hypothyroidism Rat Model Induced by 6-propyl-2-thiouracil(PTU). (부자(附子)가 6-propyl-2-thiouracil(PTU)로 유발된 rat의 갑상선 기능저하증에 미치는 영향)

  • Objective : Although hypothyroidism is a common disease in the endocrinology system, it is reported that there are a lot of difficulties in treating it effectively. Aconiti Ra얕 traditionally has been used in treatment of coldness, fatigue, and bradycardia. In this study, we investigated the therapeutic effects of Aconiti Radix on hypothyroidism rat model induced by 6-propyl-2-thiouracil (PTU). Methods : Two-month-old rats were used with administration of PTU which induced hypothyroidism in the rats. After 2 weeks, Aconiti Radix and thyroxine were administered, respectively. The body weights were measured every week. After 4 weeks, the blood samples of all rats were taken from their hearts. They were analyzed biochemically and $T_4$ (thyroid hormone) & TSH (thyroid stimulating hormone) was measured by ELISA kits. Results : In comparison with normals, controls showed hypothyroidism with significantly low $T_4$ and high TSH the statistics. In Aconiti Radix administration groups significantly increased $T_4$ was observed in the statistics and its effects were dose-dependent. There was no difference statistically in TSH of Aconiti Radix treatment groups from controls, nor were statistical differences observed significantly in biochemical labs and weight of each group. Conclusions : These findings suggest that Aconiti Radix protects thyroid cells and makes thyroid cells produce thyroid hormones. It is also very safe in the view of liver, kidney function, and other metabolism. It may be a useful agent for treating hypothyroidism.

Synthesis of New 2-Thiouracil-5-Sulphonamide Derivatives with Antibacterial and Antifungal Activity

  • 2-Thiouracil-5-sulphonic acid N-(4-acetylphenyl) Amide (1) was reacted with a series of aromatic aldehydes giving chalcones 2 (Claisen-Schemidt reaction), some of these chalcones were reacted with urea and thiourea giving pyrimidine-2-one and pyrimidine-2 thione derivatives respectively of the type 3a,b and 4a,b. In addition many chalcones were reacted with hydroxylamine hydrochloride giving isoxazoline derivatives 5a,b. They could also reacted with phenylhydrazine to give pyrazoline derivatives 5a,b, chalcones also were reacted withethylcyano acetate and/or malononitryl in pyridine giving pyran derivatives 7a,c and 8a,c. In another pathway chalcones were epoxidised by $H_{2}O_{2}$ giving epoxides 9a,c which in turn were reacted with phenylhydrazine giving 4-hydroxypyrazoline derivatives 10a,c. In another reaction chalcones were reacted with ethylcyanoacetate in presence of amm.acetate giving pyridone derivatives 11a,d which could be prepared also in exellent yield from compound 1 by its reaction with certain aromatic aldehydes and ethylcyanoacetate in presence of ammonium acetate. Finally, compound 1 was reacted with semicarbazide giving semicarbazone intermediate 12 which in turn was reacted with thionyl chloride giving thiadiazole derivative 13. The biological effects of some of the new synthesized compounds were also investigated.

  • 2-Thiouracil-5-sulfonylchloride 1 reacted with a series of aromatic and heterocyclic amines to give 2a-j. The same compound 1 was reacted with a series of sulphonamides giving different sulphonamides of type 3a-e. On the other hand compound 1 was allowed to react with p-aminoacetophenone givining compound 4 which in turn was allowed to react with derivatives of alkyl thiosemicarbazides to give thiosemicarbazones of type 5a-e, also compound 4 was monobrominated to give compound 6 which in turn was reacted thiosemicarbazones of some aldehydes to give the corresponding thiazole derivatives 7a-f. In the same time compound 4 was reacted with a series of aromatic and heterocyclic aldehydes givining chalcones 8a-g (Claisen-Schemidt reaction). Also compound 4 was allowed to react with a series of aromatic and heterocyclic aldehydes, ethyl cyano acetate and/or malononitrile, and ammonium acetate giving pyridine derivatives 9a-d and 10a-e respectively. The biological effects of some of the new synthesized compounds was also investigated.

  • 5-Nitrouracil and 5-nitrouridine derivatives which C-2 and C-4 oxo-groups of the pyrimidine base were replaced by thio groups were synthesized. The lipophilicities of thiopyrimidine bases were enhanced significantly as indicated by P-values. Oxygen-sulfur exchange leading to 2-thiouracil (2) and 2,4-dithiouracil (3) were associated with 1.4- and 2.6-fold increase in P-value relative to that of uracil (1). The P-values of 5-nitro-2-thiouracil (5) and 5-nitro-2,4-dithiouracil (6) were increased 13.2- and 79.8-fold relative to that of 5-nitrouracil (4). Most of the 5-nitrothiopyrimidine bases and their nucleosides were found to be moderately active against Staphylococcus aureus and Escherichia coli except 5-nitrouracil (4).

  • A novel fused thiouracil containing a heterocyclic ring system, dipyrimidinopyridine (3), has been prepared through the cyclization of compound 2. Compound 2 was formed by the formylation of 6-amino-2-thiouracil 1, pyrazolopyrimidines 8-10 via the heating of 6-arylhydrazono-2-thiouracils 5-7, compound 11, using Vilsmeier reagent with compound 4, pyrazolylpyrimidine 12, indolodiazinopyrimidine 14 and pyridazinopyrimidine 15. Pyridazino-pyrimidine 15 was formed by the condensation of compound 4 with acetylacetone, isatin and benzyl, respectively.

Effects of Ginseng Radix on the Rat Hypothyroidism Induced by PTU (6-n-propyl-2-thiouracil) (인삼이 PTU (6-n-propyl-2-thiouracil)로 유발된 Rat 갑상선 기능저하증에 미치는 영향)

  • Object : This study was to evaluate the effect of Ginseng Radix, aqueous extracts of the root part of Panax ginseng on the 6-n-propyl-2-thiouracil(PTU)-induced rat hypothyroidism. Methods : Aqueous extracts of Ginseng Radix(GR; yield = 11.70%) were administered, once day for 42 days from 2 weeks before start of PTU treatment as an oral dose of 300 and 150 mg/kg(body weight), and hypothyroidism was induced by daily subcutaneous treatment of PTU 10mg/kg for 28 days. The changes in the body weight, thyroid gland weights, serum levels of thyroid hormone-thyroid stimulating hormone(TSH), tri-iodothyronine(T3) and thyroxine(T4), total cholesterol, low density lipoprotein(LDL), high density lipoprotein(HDL) and triglyceride, liver antioxidant defense system-lipid peroxidation, $H_2O_2$, superoxide dismutase(SOD) and catalase(CAT) were observed with histopathology of thyroid glands. Results : Results were compared with LevoT4 0.5mg/kg treated rats. GR extracts suppressed the decreases in the body weight, thyroid gland weights, T3 and T4, TG, liver $H_2O_2$ and SOD activities as results of PTU treatment. And GR extracts suppressed the increases of HDL contents, liver CAT activities, thyroid gland weight as results of PTU treatment. In addition, marked hyperplasia of follicular cells with decreases of follicular colloid contentsand sizes were demonstrated at histopathological inspections. However, these PTU-induced histopathological changes related to hypothyroidism were dramatically decreased by treatment of both different dosages of GR extract, respectively Conclusions : This study suggest that GR extracts have favorable effects on the thyroid hormone productions with beneficial effects on the hypothyroidism mediated by the modulatory effects on the antioxidant defense system.

Effects of Bupleuri Radix on the Rat Hypothyroidism Induced by PTU (6-n-propyl-2-thiouracil) (시호가 PTU (6-n-propyl-2-thiouracil)로 유발된 Rat 갑상샘 기능저하증에 미치는 영향)

The changes on the body weight, thyroid gland weights, serum thyroid hormone - thyroid stimulating hormone (TSH), triiodothyronine ($T_3$) and thyroxine ($T_4$), serum lipid profiles - total cholesterol, low density lipoprotein (LDL), high density lipoprotein (HDL) and triglyceride, liver antioxidant defense system - lipid peroxidation, $H_2O_2$, superoxide dismutase (SOD) and catalase (CAT), serum aspartate aminotransferase (AST) and alanine aminotransferase (ALT) were observed with histopathology of thyroid glands. Results were compared with $LevoT_4$ 0.5 mg/kg treated rats. As results of PTU treatment, marked decreases of body weights, triglyceride contents, liver CAT activities and changes of serum thyroid hormone levels were observed with increases of serum AST, HDL contents, liver $H_2O_2$ and SOD activities and thyroid gland weight. In addition, marked hyperplasia of follicular cells with decreases of follicular colloid contents and sizes were demonstrated at histopathological inspections. However, these PTU induced hypothyroidism were dose-dependently inhibited by treatment of BR extracts, and BR extracts effectively regulated the hypothyroidism related changes on the antioxidant defense system. The results obtained in this study suggest that BR extracts have favorable effects on the thyroid hormone productions with beneficial effects on the hypothyroidism mediated by the modulatory effects on the antioxidant defense system.

Aqueous extracts of Lonicerae Flos (LF yield = 23.80%) were administered, once day for 42 days from 2 weeks before start of PTU treatment as an oral dose of 500 and 250 mg/kg (body weight), and hypothyroidism was induced by daily subcutaneous treatment of PTU 10 mg/kg for 28 days. The changes on the body weight, thyroid gland weights, serum thyroid hormone - thyroid stimulating hormone (TSH), tri-iodothyronine ($T_3$) and thyroxine ($T_4$), serum lipid profiles - total cholesterol, low density lipoprotein (LDL), high density lipoprotein (HDL) and triglyceride were observed with liver antioxidant defense system - lipid peroxidation, $H_2O_2$, superoxide dismutase (SOD) and catalase (CAT) and serum asparte aminotransferase (AST) and alanine aminotransferase (ALT) analysis. Results were compared with Levothyroxine ($LT_4$) 0.5 mg/kg treated rats. As results of PTU treatment, marked decreases of body weights, serum thyroid hormone levels and triglyceride contents, liver $H_2O_2_3$ and SOD activities were observed with increases of serum AST and HDL contents, liver CAT activities, thyroid gland weight. However, these PTU induced hypothyroidism were dose-dependently inhibited by treatment of LF extracts, and LF extracts effectively regulated the hypothyroidism related changes on the antioxidant defense system.
