Isolation and Characterization of 2-Methyl-4-Chlorophenoxyacetic Acid-Degrading Bacteria from Agricultural Soils

  • Seven numerically dominant 2-methyl-4-chlorophenoxyacetic acid (MCPA)-degrading bacteria were isolated from agricultural soils. The isolates utilized the herbicide MCPA as a sole carbon source, producing significant biomass in MCPA mineral medium. They exhibited diverse herbicide degradation capabilities, but most of them grew very slowly in mineral medium containing herbicide. The chromosomal DNA patterns of the isolates obtained by polymerase chain reaction amplification of repetitive extragenic palindromic sequences were distinct from each other. One isolate, SH3, which was identified as Sphingomonas species by fatty acid methyl ester analysis, was able to degrade 5 different phenoxyacetic acid herbicides within 4 days. This strain contains two plasmids, and the smaller one has a crucial role in herbicide degradation. MCPA treated into agricultural soils without indigenous MCPA-degraders persisted for a long time, but the application of the isolate SH3 resulted in rapid decline of MCPA concentration in the soil.

식물의 Ecdysteroids 함량에 미치는 광과 식물생장조절제의 영향 (Effects of Light and Some Plant Growth Regulators on Ecdysteroids Contents in Polypodium vulgare L. and Achyranthes japonica Nakai)

  • 미역고사리(Polypodium vutgare L.)와 쇠무릎(Achyranthes japonica Nakai)에 있어서 광과 methyl jasmonate(MJ), 6-benzylaminopurine(BA), thidiazuron(TDZ), 2,4-dichlorophenoxyacetic acid(2,4-D)가 식물체 중의 ecdysteroids (${\beta}$-ecdysone+polypodine B) 함량에 미치는 영향을 조사하였다. 수광량을 조절하여 재배한 미역고사리의 잎과 근경 모두 상대광도가 높을수록 ecdysteroids 함량이 감소하였고 쇠무릎 유묘의 배양시 광 조건일 때가 암 조건일 때 보다 ecdysteroids함량이 낮아 식물의 ecdysteroids 함량은 광에 의해 저하되는 것으로 나타났다. 미역고사리 전엽체와 쇠무릎 유묘의 배양계에서 식물체 중의 ecdysteroids 함량은 배지 중의 MJ와 BA 그리고 TDZ의 농도가 높아짐에 따라 증가된 반면 2,4-D의 농도와는 무관하게 일정한 수준으로 유지되어,식물의 ecdysteroids 함량은 MJ와 cytokinin처리에 의해 증가되지만 auxin처리에 의해서는 영향을 받지 않는 것으로 나타났다.

Methyl Jasmonate-mediated Enhancement of Phenylethanoid Glycoside in Callus from Abeliophyllum distichum (cultivar Okhwang1)

  • Abeliophyllum distichum, one of the Korean endemic plant, is a significant pharmaceutical plant resource. A. distichum with phenylethanoid glycoside can use to regulate the development of cancer, DNA damage with radicals, and the generation of inflammatory mediators. In this study, we investigated whether the biomass, content of phenylethanoid glycoside, and growth rate of callus derived from A. distichum (cultivar Okhwang1, CAD) change in the absence or presence of plant hormones (2,4-Dichlorophenoxyacetic acid; 2, 4-D and 1-Naphthaleneacetic acid; NAA). The results showed that the best biomass, the growth rate of callus, and the contents of phenylethanoid glycoside were cultivated on Murashige and Skoog (MS) growth medium fortified with 1 ppm 2,4-D + 2 ppm NAA after 4 weeks. In a further study, CAD was cultivated on MS growth medium fortified with an elicitor (Methyl Jasmonate, MeJA). The results showed that CAD turned to brown color and fragile form with the elicitor. HPLC-PDA analysis revealed that the contents of phenylethanoid glycoside in the elicitor-treated group were higher than in the elicitor-non-treated group. These results are consistent with the findings of Arano-Varela H et al.,'s study which is that acteoside production can increase after the treatment of MeJA. Therefore, this study can be used to develop an effective and sustainable production of useful substances as an alternative to plant cultivation.

Genetic and Phenotypic Diversity of Dichlorprop-Degrading Bacteria Isolated from Soils

  • Nine dichlorprop-degrading bacteria and three pairs of bacteria showing syntrophic metabolism of the herbicide were isolated from soils, and their genetic and phenotypic characteristics were investigated. Analysis of 16S rDNA sequences indicated that the isolates were related to members of the genera, Sphingomonas, Herbaspirillum, and Bradyrhizobium. Twelve different chromosomal DNA patterns were obtained by polymerase-chain-reaction (PCR) amplification of repetitive extragenic palindromic (REP) sequences from the 15 isolates. The isolates were able to utilize the herbicide dichlorprop as a sole source of carbon and energy and their dichlorprop derogative pathways were induced by the presence of dichlorprop. Most of the isolates and syntrophic pairs were able to degrade both (R)- and (S)-dichlorprop, but strain DP522 exhibited enantioselective degradation of (S)-dichlorprop. The isolates degraded 2,4-dichlorophenoxyacetic acid, 2-methyl-4-chlorophenoxyacetic acid , and mecoprop, in addition to dichlorprop. Oxygen uptake experiments indicated that most of the isolates degraded dichlorprop through 2,4-dichlorophenol.

Regulation of the Korean Radish Cationic Peroxidase Promoter by Phytohormones and Other Reagents

  • The Korean radish cationic peroxidase (KRCP) promoter, comprising nucleotides -471 to +704 relative to the transcriptional initiation site, was fused to the GUS gene and transformed to tobacco BY-2 cells. We examined how auxin (2,4-dichlorophenoxyacetic acid, 2,4-D), cytokinin (6-benzylaminopurine, BAP), gibberellic acid ($GA_3$), abscisic acid (ABA), methyl jasmonate (MeJA), and phosphatidic acid (PA) affect the GUS expression in the presence or absence of 2,4-D in a modified LS medium. Exogenous 2,4-D or BAP greatly decreased the GUS expression regulated by the KRCP promoter in a modified LS medium containing 0.2 mg/l 2,4-D. $GA_3$ increased the GUS expression and ABA completely reduced the inductive effect of $GA_3$. The GUS expression was also increased dose-dependently by plant defense regulators, MeJA and PA. In contrast to the above results, auxin deprivation from the modified LS medium increased the GUS expression after treatment with exogenous 2,4-D whereas BAP still greatly decreased the GUS expression dose-dependently. $GA_3$ or MeJA slightly decreased the GUS expression. The data suggest that auxin deprivation changes the sensitivity of the suspension cells to exogenous chemicals and that the regulation of the KRCP promoter by 2,4-D, $GA_3$, and MeJA is dependent on auxin, whereas the regulation by BAP is not. This study will be valuable for understanding the function and expression mode of the Korean radish cationic peroxidase in Korean radish.

하천수에서 LC-ESI-MS/MS에 의한 극미량 농약류 및 합성원료의 동시분석법 (Simultaneous Determination of Ultra-Trace Pesticides and Synthetic Materials in Surface Water by LC-ESI-MS/MS)

  • 하천수 중에 7가지의 농약류(2,4-dichlorophenoxyacetic acid, 메소밀, 알디캅, MCPA, molinate, 카바릴, 카보퓨란)와 2가지의 합성원료(quinoline, 비스페놀-A)를 액체크로마토그래피 전자분무 이온화 질량분석법(liquid chromatography-electrospray ionization-tandem mass spectrometry, LC-ESI-MS/MS)을 사용하여 분석하였다. 분석물질들은 고체상 추출법(solid-phase extraction, SPE)을 사용하여 추출하였다. 용리액은 질소가스로 농축하였다. 잔여물에 30% 메탄올 수용액 100 μL를 재 용해시켜 0.2 μm PTFE 실린지 필터로 거른 후 그 일부를 LC-ESI-MS/MS에 직접 주입하여 분석하였다. 확립된 조건에서 검정곡선은 0.997 이상의 상관계수를 갖는 직선성을 보였다. 정량한계는 0.002~0.011 μg/L 이었고, 정밀도는 16.4% 이하였다. 또한 정확도는 84~107%의 범위를 보였고, 회수율은 56.2~98.6% 값을 나타내었다. 본 연구에서 개발한 분석법을 실제 하천수 분석에 적용하였다.

  • 2,4-D 제초제(除草劑)의 상호작용(相互作用)을 조사(調査)키 위하여 2,4-D와 제초제(除草劑의 혼합처리(混合處理)가 수도(水稻)의 초기생육(初期生育)에 미치는 영향(影響)을 조사(調査)한 결과(結果)는 다음과 같다. 2,4-D 처리(處理)에 의한 신초생육(新鞘生育)이 더욱 억제(抑制)된 반면에 thiobencarb, butachlor 및 glyphosate 처리(處理)에 의한 IR28의 신초생육(新鞘生育) 억제(抑制)는 Taipei 309 보다 더욱 크게 나타났다. 2,4-D와 thiobencarb 혼합처리시(混合處理時)에 상호작용(相互作用)은 thiobencarb 농(濃度)가 낮은 처리구(處理區)의 신초(新鞘)길이에 대하여 두 품종(品種) 공(共)히 길항작용(拮抗作用)을 나타내었고 thiobencarb 고농도(高濃度)에서는 상승작용(相乘作用)을 보였다. 신초((新鞘)의 생체중(生體重)에 대한 2,4-D와 thiobencarb의 상호작용(相互作用)은 IR28의 경우 모든 처리구(處理區)에서 상승작용(相乘作用)을 나타낸 반면에 Taipei 309의 경우에는 2,4-D의 농도(濃度)가 낮을 때만 상승작용(相乘作用)이 나타났다. 2,4-D와 butachlor 혼합처리(混合處理)에 의해서는 IR28의 신초길이와 생체중(生體重)이 Taiperi 309보다 더욱 높은 억제현상(抑制現象)을 보여 상승작용(相乘作用)을 나타내었다. 2,4-D와 glyphosate 혼합시(混合時)에는 신초(新鞘)의 길이에서는 두 품종(品種) 공히 길항작용(拮抗作用)을 보였으나, 2,4-D와 glyphosate의 최고농도(最高濃度) 혼합시(混合時)에는 신초(新鞘)의 생체중(生體重)에 대해서 IR28은 상승작용(相乘作用)을 보인 반면에 Taipei의 309는 길항작용(拮抗作用)을 나타내었다.
