Characterization of gender-specific bovine serum

  • Animal cell cultures generally require a nutrient-rich medium supplemented with animal serum. Adult bovine serum contains a variety of nutrients including inorganic minerals, vitamins, salts, proteins and lipids as well as growth factors that promote animal cell growth. To evaluate the potential use of gender-specific bovine serum (GSBS) for cell culture, the biochemical properties of male serum (MS), female serum (FS) and castrated-male serum (CMS) were investigated. Overall, the chemical profile of GSBS was similar to that of bovine references except for glucose, creatine kinase, lactate dehydrogenase and potassium. FS showed elevated total protein and sodium concentrations compared to MS and CMS. Proteins present in MS, FS and CMS but absent in fetal bovine serum (FBS) were selected by two-dimensional gel electrophoresis and identified by peptide mass fingerprinting. Some of the identified proteins are known to be involved in immune responses and the others have unknown physiological roles. Moreover, it was found that some proteins such as alpha-2-macroglobulin appeared to be gender-specific with higher contents in FS. Insulin and testosterone was significantly higher in MS, and $17{\beta}$-estradiol and estrone were higher in FS, as compared to the other sera. Taken together, the results indicate that each GSBS has a different ratio of components. Differences in serum constituents may affect cell cultures in a different manner and could be beneficial, depending on the specific aim of cell cultures.

  • Objectives : Parkinson's disease (PD) is a neurodegenerative disorder threatening the quality of life and highly occurred in over 65 years old. Gastrodia elata Blume (GEB), a traditional medicine used for the treatment of headache and convulsion, has been reported to have neuroprotective effect. This study was designed to investigate the neuroprotective effect of GEB and the proteomic changes in the substantia nigra (SN) of 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP)-treated mice. Methods : Male eleven-week-old C57BL/6 mice were intraperitoneally injected with 30 mg/kg of MPTP at 24-h intervals for 5 days. Two hours after the daily MPTP injection, the mice were orally administered 800 mg/kg of GEB extract, which continued for 7 days beyond the MPTP injections, for a total of 12 consecutive days. Two hours after the final GEB administration, the brain samples were collected, and dopaminergic neuronal death and proteomic changes in the SN were evaluated. Results : GEB prevented the MPTP-induced dopaminergic neuronal death and regulated the expression of 11 proteins including thimet oligopeptidase, T-complex protein 1, glycine tRNA ligase, and pyruvate kinase isozymes M1. Conclusions : GEB prevents MPTP-induced dopaminergic neuronal death by regulating the proteins in the SN.

인체의 폐암과 정상 폐조직에서 Peroxiredoxin 및 Thioredoxin의 발현 양상 (Expression of Peroxiredoxin and Thioredoxin in Human Lung Cancer and Paired Normal Lung)

  • 연구배경 : Peroxiredoxin (Prx) 은 최근에 알려진 항산화제로 세포의 증식과 분화, 세포사멸이나 발암과정에 관여하는 것으로 알려져 있다. 하지만, 현재까지 폐암을 비롯한 각종 질병에서의 이들 Prx단백의 역할은 잘 규명되어 있지 않다. 이에 본 연구는 폐암 조직과 정상폐조직에서 Prx 단백의 발현 양상과 분포를 연구하여 이들의 병태 생리학적인 의미를 찾아보고자 하였다. 방 법 : 아주대학교 병원에서 폐암으로 진단된 환자의 폐암조직과, 동일 환자의 정상 폐조직에서, 1 차원전기영동 (reducing 조건과 non-reducing 조건에서의 SDSPAGE) 혹은 2차원전기영동을 시행한 후 Western blot으로 Prx, Trx 및 TR의 발현 양상을 분석 하였으며, 백서의 정상 폐조직과, 환자의 폐암 조직에서 anti-Prx rabbit polyclonal 항체로 하여 면역조직화학염색법을 통해, Prx 단백의 분포를 관찰하였다. 결 과 : 면역조직화학염색법 결과 백서의 정상 폐조직에서는 Prx I, II, III 및 V 유형이, 주로 기관지상피세포, 폐포상피세포 및 폐포대식세포에서 발현되고 있음을 관찰하였다. 인체 폐암조직에서는, 정상 폐조직 부위에 비해서 Prx I 과 Prx III 유형 및 Trx단백의 발현이 선택적으로 증가되어 있고, 특히, 2차원 전기영동을 통한 프로티옴 분석에서 산화된 형태의 Prx I 과 Prx II가 증가 한 것을 비롯하여, 분자량과 등전점(pI)이 약간 변화된 형태의 Prx III가 폐암조직에 존재함을 알 수 있었다. 한편, 폐암 조직의 non-reducing 전기영동 후 Western blot에서는, monomer와 dimer 사이의 중간 크기에 해당하는 약 40 kDa과 200 kDa 이상 크기의, 항 Prx 항체와 반응하는 단백 띠 (reactive bands)가 관찰되었다. 결 론 : 폐암 조직에서 관찰되는 Prx I 과 Prx III 유형 및 Trx의 과발현 양상은, 종양 세포들이 주변의 미세 환경으로부터 겪는 여러 스트레스에 대하여 단백질을 보호하고 세포의 생명력을 유지하는데 있어 주요한 역할을 하는 것으로 사료된다.

면역학적 방법을 이용한 자유생활아메바의 분류학적 접근 (Immunological approach for classification of free-living amoeba in Korea)

  • Acanthnmoeba sp. YM-4의 영양형은 사상의 침상위족을 내어 전형적인 각canthamoeba 속의 특징이 관찰되었으며, 포낭(씨스트)은 외벽이 등근혐이었고 내벽이 단순한 구형이거나 다소 불규칙한 2개의 세포벽을 갖고 있었다. SDS-PAGE 결과, Acanthamoeha sp. Yhf-4는 약 16개의 주요 단백질 분획이 관찰되었는데, 2차원 전기영동 결과와 함께 Acanthamoeba sp. YM-4의 단백질 분획은 A. culbertsoni와 거의 같은 양상이 관찰되었다. Acanthamoeba sp. YM-4로 면역시킨 마우스의 비장세포와 myeloma 세포를 융합한 결과, 항체를 분비하는 17개 clone을 얻을 수 있었다. 생성된 단세포를 항체 중 isotyping을 실시하였는데 실험에 사용된 McAY 6, McAY 7, McAY 8, McAY 13, McAY 16 단세포군은 IgGl 항체를, McAY 10, McAY 11 단세포군은 19M 항체를 분비하였다. 생성된 단세포군 항체는 대부분이 아메바의 세포막에 있는 항원과 반응하였으며, EITB 실험결과, McAY 7 단세포군 항체는 43 kD에서 반응대가 관찰되었고, McAY 10 단세포군 항체는 55 kD과 105 kD에서 반응대가 관찰되었다. ELISA 방법을 이용한 단세포를 항체들과 여러 아메바 간의 교차반응실험 결과, McAY 7 단세포군 항체는 Acanthamoeba sp. YM-4에만 반응하였고, McAY 6과 McAY 10은 A. culbertsoni와도 반응하였다. 또한 McAY 11은 모든 아매바와 교차반응을 보였다.

Proteomic Analysis of the Triglyceride-Rich Lipoprotein-Laden Foam Cells

  • In hypertriglyceridaemic individuals, atherosclerogenesis is associated with the increased concentrations of very low density lipoprotein (VLDL) and VLDL-associated remnant particles. In vitro studies have suggested that VLDL induces foam cells formation. To reveal the changes of the proteins expression in the process of foam cells formation induced by VLDL, we performed a proteomic analysis of the foam cells based on the stimulation of differentiated THP-1 cells with VLDL. Using two-dimensional gel electrophoresis (2-DE) and matrix-assisted laser-desorption ionization time-of-flight mass spectrometry (MALDI-TOF MS) analysis, 14 differentially expressed proteins, containing 8 up-regulated proteins and 6 down-regulated proteins were identified. The proteins are involved in energy metabolism, oxidative stress, cell growth, differentiation and apoptosis, such as adipose differentiation-related protein (ADRP), enolase, S100A11, heat shock protein 27 and so on. In addition, the expression of some selected proteins was confirmed by Western blot and RT-PCR analysis. The results suggest that VLDL not only induces lipid accumulation, but also brings about foam cells diverse characteristics by altering the expression of various proteins.

Comparative analysis of fat and muscle proteins in fenofibratefed type II diabetic OLETF rats: the fenofibrate-dependent expression of PEBP or C11orf59 protein

  • Fenofibrate, an agonist of $PPAR{\alpha}$, plays an important role in activating many proteins catalyzing lipid metabolism, and it also has a considerable effect on improvement of insulin sensitivity in the diabetic condition. To investigate fenofibrate-dependent expression of peripheral tissue proteins in diabetes, we analyzed whole muscle or fat proteins of fenofibrate-fed OLETF rats, an animal model of type II diabetes, using 2-dimensional gel electrophoresis. We found that many proteins were specifically expressed in a fenofibrate-dependent manner in these diabetic rats. In particular, a functionally unknown C11orf59 protein was differentially expressed in the muscle tissues (about 5-fold increase) in fenofibrate-fed OLETF rats as compared to control rats. Additionally, the signal proteins phosphatidylethanolamine binding protein and IkB interacting protein were differentially regulated in the fenofibrate-treated adipose tissues. We suggest here that these proteins might be involved in controlling lipid or carbohydrate metabolism in diabetes via $PPAR{\alpha}$ activation.

Potential Vaccine Targets against Rabbit Coccidiosis by Immunoproteomic Analysis

  • The aim of this study was to identify antigens for a vaccine or drug target to control rabbit coccidiosis. A combination of 2-dimensional electrophoresis, immunoblotting, and mass spectrometric analysis were used to identify novel antigens from the sporozoites of Eimeria stiedae. Protein spots were recognized by the sera of New Zealand rabbits infected artificially with E. stiedae. The proteins were characterized by matrix-assisted laser desorption ionization time of flight mass spectrometry (MALDI-TOF/TOF-MS) analysis in combination with bioinformatics. Approximately 868 protein spots were detected by silver-staining, and a total of 41 immunoreactive protein spots were recognized by anti-E. stiedae sera. Finally, 23 protein spots were successfully identified. The proteins such as heat shock protein 70 and aspartyl protease may have potential as immunodiagnostic or vaccine antigens. The immunoreactive proteins were found to possess a wide range of biological functions. This study is the first to report the proteins recognized by sera of infected rabbits with E. stiedae, which might be helpful in identifying potential targets for vaccine development to control rabbit coccidiosis.

Biochemical and Genetic Variation of Hordein Subunits in Korean Barley

  • One-dimensional sodium dodecyl sulfate polyacrylamide gel electrophoresis (1D SDS-PAGE) was used to determine whether it would provide improved resolving power of hordein proteins concomitant with improved identification of Korean barley cultivars and germplams. This system gave rapid and reproducible separations of hordein polypeptides. Total fourteen of clear and easily scorable subunits were identified in Korean barley cultivars and germplasms and their polymorphic constitutions could provide biochemical genetic information in progeny analysis and endosperm quality improvement in barley breeding programs. Each hordein polypeptides residing in B, C, and D hordein pattern designations were scored to prepare a cultivar catalogue of protein patterns. On the basis of this character, 7 hordein polypeptide patterns were constructed from 108 barley cultivars and experimental lines. The molecular weight of hordein subunits in Korean barley cultivars and experimental lines varied in the range of 98 to 48 kDa. In contrast, less polymorphic hordein polypeptides were found in the low protein barley lines including malting barleys than those found in Korean barley cultivars and experimental lines.

돼지 품종간 정액 내에서 수정 능력과 단백질 변화와의 관계 분석 (Relative Analysis between Fertility and Protein Changes in Semen of Different Species in Pigs)

  • The objective of this study was to investigate the relationship between fertility and protein pattern change using in vitro fertilization, analysis of sperm characteristics and two-dimensional gel electrophoresis in different pig types. In results, the viability and mitochondria integrity of sperm were higher significantly (p<0.05) but the portions of acrosome reaction was lower significantly (p<0.05) in Duroc and $F_1$ (potbellied ${\times}$ PWG miniature pig) than PWG miniature. On in vitro fertilization to investigate fertility, the fertility of $F_1$ semen war higher significantly (p<0.05) than in Duroc and PWG miniature pig. On the other hand, protein patterns showed similar function among the different boar semen. Especially, the heat shock 70 kDa 1-like and G patch domain-containing protein 4 were significantly (p<0.05) higher expressed in $F_1$ than in Duroc and PWG miniature pig. The proteins associated with mitochondria in Duroc were significantly (p<0.05) higher expressed than in $F_1$ and PWG miniature pig. The developmental rates to blastocyst stage of oocytes fertilized with sperm of $F_1$ pig were significantly (p<0.05) higher than in PWG miniature pig. However, phosphoglycerate kinase 2 and zinc finger protein 431 were significantly (p<0.05) higher expressed in PWG miniature pig than in $F_1$ and Duroc pigs. In conclusion, the results of the present study indicate that different proteins were expressed in different pig types, and were associated with a sperm functions and embryo development.

Identification of Lactoferrin as a Human Dedifferentiation Factor Through the Studies of Reptile Tissue Regeneration Mechanisms

  • In this study, we performed two-dimensional electrophoresis with protein extracts from lizard tails, and analyzed the protein expression profiles during the tissue regeneration to identify the dedifferentiation factor. As a result, we identified 18 protein spots among total of 292 spots, of which proteins were specifically expressed during blastema formation. We selected lactoferrin as a candidate because it is the mammalian homolog of leech-derived tryptase inhibitor, which showed the highest frequency among the 18 proteins. Lactoferrin was specifically expressed in various stem cell lines, and enhanced the efficiency of iPSC generation upto approximately 7-fold relative to the control. Furthermore, lactoferrin increased the efficiency by 2-fold without enforced expression of Klf4. These results suggest that lactoferrin may induce dedifferentiation, at least partly by increasing the expression of Klf4.
