Effects of Chaperones on mRNA Stability and Gene Expression in Escherichia coli

  • Effects of chaperones on mRNA stability and gene expression were studied in order to develop an efficient Escherichia coli expression system that can maximize gene expression. The stability of mRNA was modulated by introducing various secondary structures at the 5'-end of mRNA. Four vector systems providing different 5'-end structures were constructed, and genes encoding GFPuv and endoxylanase were cloned into the four vector systems. Primer extension assay revealed different mRNA half-lives depending on the 5'-end secondary structures of mRNA. In addition to the stem-loop structure at the 5'-end of mRNA, coexpression of dnaK-dnaJ-grpE or groEL-groES, representative heat-shock genes in E. coli, increased the mRNA stability and the level of gene expression further, even though the degree of stabilization was varied. Our work suggests that some of the heat-shock proteins can function as mRNA stabilizers as well s protein chaperones.

  • MI(Magneto-Impedance) sensor which is made by thin films has significantly high detecting sensitivity in weak magnetic field. It also has a merit to be able to build in low power system. Its structure is simple, which makes it easier to prepare a miniature. In this study, its magnetic permeability and anisotropy field(H$\sub$k/) as a function of a thickness of sputtered amorphous CoZrNb thin film with high saturation magnetostriction and excellent soft magnetic property are investigated. In order to make a uniaxial anisotropy, thin film was subjected to post annealing with a static magnetic field with 1KOe intensity at 250, 300, and 320$^{\circ}C$ for 2 hour. Anisotropy field(H$\sub$k/)of thin film is measured by using MH loop tracer. Its magnetic permeability of thin film is measured over the frequency range from 1 MHz to 750MHz. It has shown that the magnetic permeability of amorphous CoZrNb thin film is decreased due to the skin effect with increasing a thickness of CoZrNb thin film, and hence its driving frequency is lowered.

  • Ferroelectric lean-titanate thin films were fabricated by sol-gel processing. Sol-gel derived $PbTiO_3$ thin films crystallized into the expected tetragonal perovskite structure when heated to $600^{\circ}C$ and above. The effects of annealing temperature on grain size made with SEM observation are reported. The films heated at $650^{\circ}C$ for 30min showed a square-type hysteresis loop with $P_r$ and $E_c$ of $11.5{\mu}C/cm^2$, 115kV/cm, respectively.

A Study on the Antenna for Mobile Communication (이동체 통신용 안테나에 관한 연구)

  • In this paper, the total electric field antenna in the small loop and aperture plane structure to load a capacitance on transformed QMSA(Quarter-wavelength Microstrip Antenna) from MSA(Microstrip Antenna) is proposed, and the total electric field is shown that the calculated value is agreed resonably with experimental value. Itis a smalled antenna of the radiation element and ground plane with equal width. The designed and manufactured antenna can receive borth vertically and horizontally polarised waves to operating frequency of 1.5GHz; therefore this is available as a smalled antenna for wireless telephones and will be quit useful for indoor communication. Also, antenna's equivalent circuit with transmission line model is designed to find more accurate resonant frequency and is calculated return loss value. The calculated value is agreed resonably with experimental value.

  • This paper proposes a high speed frequency synthesizer with dual phase-locked loop(PLL) structure to improve phase noise level and shape in a wideband receiver. To reduce phase noise and fractional spur, a output frequency of $1^{st}$ PLL used as reference frequency of $2^{nd}$ PLL is changed. The frequency synthesizer has been designed with 1 Hz frequency resolution using digital NCO in 6.5~8.5 GHz wide spectrum. The measured results of the fabricated frequency synthesizer show that the output power is about -3 dBm, the maximum lock-in time and phase noise are within 60 us and -95 dBc/Hz at 10 kHz offset, respectively.

A Rotational Decision-Directed Joint Algorithm of Blind Equalization Coupled with Carrier Recovery for 32-QAM Demodulation (회전결정 경계를 이용한 32-QAM 목조용 반송파 복구와 채널등화의 Joint 알고리즘)

  • We introduce a rotational decision-directed joint algorithm of blind equalization coupled with carrier recovery for 32-QAM demodulation with high symbol rate. The proposed carrier recovery, which we call a rotational decision-directed carrier recovery(RDDCR), removes the residual phase difference by rotating the decision boundary for the kth received symbol by the frequency detector output of the (k-1)th received symbol. Since the RDDCR includes the function of PLL loop filter by rotating the decision boundary, it gives a simpler demodulator structure. The rotational decision-directed blind equalization(RDDBE) with the rotated decision boundary based on the Stop-and-Go Algorithm(SGA) operated during tracking the frequency offset by the RDDCR and removes intersymbol interference due to multipaths and channel noise. Test results show that symbol error rate of $10^{-3}$ is obtained before the forward error correction when SNR equals 15dB with 150KHz of carrier frequency offset and two multipaths, which is the channel condition for 32-QAM receiver.

A Research on Shifting Controller Development of a Automatic Transmission far Construction Vehicles (건설차량용 자동변속기의 변속제어기 개발연구)

  • In this research, a transmission control unit was developed for a wheel loader on the basis of 16-bit microcontroller system. The TCU has the basic functions which include shifting control algorithm, actuation of six proportional solenoid valves, signal conditioning for four speed sensors, remote data monitoring capability with RF-module and duty cycle identification system which could identify the duty cycles from PWM signals. In order to design the control system, the overall transmission structure of the wheel loader was investigated and its characteristics of shifting were analyzed in advance. For the purpose of identifying the existing control algorithm and acquire some information about the shifting performance, the shifting experiments were performed for various shifting conditions with the conventional TCU. From the previous work on the conventional TCU, the shifting scheme was designed with the open-loop control which is based on the experimental data only to verify the feasibility of the developing TCU's shilling capability. The experimental results show comparable shifting characteristics to that of conventional TCU though the tests were performed at restricted shilling conditions. Hence, we could have the confidence for the development of the wheel loader automatic transmission TCU and its shifting quality could be improved with the adoption of appropriate feedback control scheme.

A Design of Wide band Dual Folded Microstrip Antennas (광대역 이중 폴디드 마이크로스트립 안테나 설계)

  • In this paper, a single plane wide band microstrip antenna for integrated circuit as MMIC and LTCC is designed and fabricated. A new configuration for a wide band microstrip antenna with omni-directional pattern is proposed. This antenna consists of two rectangular folded dipoles, which are fed by a coplanar waveguide(CPW). It was effected stabilization ground that a signal plane of CPW feed have been limited ground plane. Therefore, a ground plane of folded structure should be extended outside folded antenna in this research. The characteristics of the proposed antenna were analyzed by using an FDTD methods. The return loss and radiation patterns were simulated and measured. The proposed antenna is get 120MHz bandwidth of PCS band and 250MHz bandwidth of IMT2000 band, used ISM band.
