Risk assessment on cytotoxicity for benzimidazole fungicides (Benzimidazole계 살균제의 세포독성 평가)

  • To assess potential risk of the benzimidazole fungicides, their cytotoxicities were evaluated. Activities of LDH(Lactic dehydrogenase) in the culture fluid of CHL(chinese hamster lung) fiberoblast cell treated with 4.0, 16.0 or $32.0{\mu}g/mL$ of carbendazim for 24 hours were elevated 2.16, 2.94 and 2.64 folds compared to the control, respectively. DNA synthesis was inhibited by 45% at $2.0{\mu}g/mL$ of carbendazim. Benzimidazole fungicides showed high toxicity to cell and mitochondria of CHL cell by Giemsa and MTT (3-(4,5-dimethylthiazole-2-yl)-2,5-diphenyltetrazolium bromide) assay. $IC_{50}$ by the Giemsa assay of thiophanate-methyl, benomyl, carbendazim and captafol were over 125, 1.2, 30.0 and $0.3{\mu}g/mL$, respectively. $IC_{50}$ by the MTT assay of thiophanate-methyl, benomyl, carbendazim and captafol were over 125, 18.7, 20.4 and $2.6{\mu}g/mL$, respectively. Inhibitory concentration of cell median proliferation by SRB (sulforhodamin B) assay for thiophanate-methyl, carbendazim, benomyl, and captafol were 17.4, 5.3, 1.5 and $0.5{\mu}g/mL$, respectively. Accordingly, benzimidazole fungicides inhibited DNA synthesis, mitochondrial function, cell proliferation and induced cell necrosis.

  • The culture condition of unrecorded species L. aciculospora, L. boryana and L. raphanica was investigated with recorded species of L. edodes as the control group in order to analyze diversity and examed relations of the species belong to Lentinula. The optimal temperature and media for the mycelial growth of L. aciculospora and L. boryana were $22^{\circ}C$ and PDA, MCM medium. L. raphanica was $28^{\circ}C$ and ME1 medium. Each of L. aciculospora, L. boryana, L. raphanica is pH 6, pH 7, pH 5 in the optimal pH respectvely. The optimal carbon and nitrogen source of L. aciculospora were glucose and malt extract. That of L. boryana was glucose and urea that of L. raphanica was sucrose and potassium nitrate. The optimal vitamin of L. aciculospora was Myo-inositol. That of L. boryana and L. raphanica were Riboflabin.

Effect of Heavy Metals (As, Cr, Pb) on the Population Growth Rates of Marine Diatom, Skeletonema castatum (중금속(As, Cr, Pb)이 해산 규조류(Skeletonema costatum)의 개체군 성장률에 미치는 영향)

  • In this study, we evaluated the toxic effects of heavy metals (As, Cr and Pb) on the population growth rate (r) of the marine diatom, Skeletonema costatum. The r of the species was determined after 96 h (4 days) exposure to As (0, 0.31, 0.63, 1.25, 2.50 and 5.00 mg L-1), Cr (0, 0.16, 0.31, 0.63, 1.25 and 2.50 mg L-1) and Pb (0, 1.25, 2.50, 5.00, 10.00 and 20.00 mg L-1). It was observed that r in the control (absence of As, Cr and Pb) were greater than 0.04, however suddenly decreased with increased heavy metal concentrations. As, Cr and Pb reduced r in a dose-dependent manner and a significant reduction were occurred at concentration of greater than 0.31, 0.31 and 5.00 mg L-1, respectively. Based on the toxicity, the heavy metal were ranked as Cr>As>Pb, with EC50 (50% effective concentration) values of 0.80, 2.57 and 13.88 mg L-1, respectively. The lowest-observed-effective-concentration (LOEC) of r in exposed to As, Cr and Pb were 0.63, 0.63 and 10.00 mg L-1, respectively. Precisely, a concentration of greater than 0.63 mg L-1 of As, 0.63 mg L-1 of Cr and 10.00 mg L-1 of Pb in marine ecosystems induced toxic effect on the r of S. costatum. Based on our results, we suggested that the r of S. costatum might be a useful bio-indicator for the toxicity assessment of heavy metals on the producer in marine ecosystems.

  • Recently Ding [4, 5, 8] gives examples of his FC-spaces which are not L-spaces due to Ben-El-Mechaiekh et al. [1]. We show that they are actually L-spaces. We also clarify that all statements in [5] can be stated in corrected and generalized forms for the class of abstract convex spaces beyond FC-spaces.

Physiochemical Characteristics of Lactobacillus acidophilus KH-l Isolated from the Feces of a Breast-Fed Infant

  • Three lactobacillus strains, two from infant feces, and one from cow's milk, were selected among 172 isolates, from multiple sources, for further study based on the antimicrobial activities against six strains of pathogenic bacteria and identified as Lactobacillus acidophilus. The strains revealed a wide scope of spectrum against pathogenic bacteria. Viable Lactobacillus acidophilus KH-l cell counts at pH 2.0 were slightly decreased to $1.42\times10^7$ CFU/mL from $4.18\times10^7$ CFU/mL, while remaining at $3.42\times10^7$ CFU/mL at pH 4.0 with the survival rate of $33.97\%\;and\;81.82\%$, respectively. At the concentration of $0.1\%$ oxgall, L acidophilus KH-l kept growing up to $3.12\times10^7$ CFU/mL with a mean growth rate constant (k) of 0.25, and cell number was slightly decreased to $1.21\times10^7$ CFU/mL (k=0.19) with $0.3\%$ oxgall, but remained at $7.6\times10^6$ CFU/mL (k=0.17) with $0.5\%$ oxgall. L. acidophilus KH-l had a $D_{60}$ value of 7.14, with viable cell numbers $1.4\times10^5$ CFU/mL after heat treatment at $60^{\circ}C$ for 30 minutes. Stability of L acidophilus KH-l at $-20^{\circ}C$ was significantly higher, when the strain was cultivated under the optimum growth temperature $(54.41\%\;and\;54.35\%)$ than at the temperature $(13.53\%)$.

Regeneration and Maturation Induction for the Free-living Gametophytes of Ecklonia cava Kjellman (Laminariales, Phaeophyta) (대형갈조류 감태(Ecklonia cava Kjellman) 유리배우체의 재생 및 성숙 유도)

  • Induction of regeneration and maturation for the free-living gametophytes of Ecklonia cava Kjellman was conducted under various temperature, irradiance and photoperiod conditions. Culture conditions for female or male gametophyte fragments were 4 temperature (5, 10, 15 and $20^{\circ}C$), 4 irradiance (5, 10, 20 and $40{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$) and 3 photoperiod (14:10, 12:12 and 10:14 h L:D). Female and male gametophyte fragments were maintained in active regeneration without reaching sexual maturity under $5{\sim}10^{\circ}C$, $5{\sim}10{\mu}mol{\cdot}m-2{\cdot}s-1$, 12:12h or 10:14h (L:D) and $15-20^{\circ}C$, $20-40{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$, 14:10h (L:D), respectively. Sexual maturation of female and male gametophytes facilitated under $15^{\circ}C$, $20-40{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$, 14:10h (L:D) and $5-10^{\circ}C$, $5-10{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$, 10:14h (L:D), respectively. These results provide basic informations to control the regeneration or maturation of the free-living gametophytes for artificial seed production of E. cava.

  • Di(10-hemisuccinyloxy)decyl muconate(DDM) and di(6-hemisuccinyloxy)hexyl muconate(DHM) were synthesized, and showed a phase transition in an aqueous solution at 97 and $79^{\circ}C$, respectively. They did not form liposomes by themselves or even in the presence of cholesterol or dioleoylphosphatidylethanolamine(DOPE), but did form liposomes when they were mixed with phosphatidylcholine(PC). DHM molecules in liposome membranes were readily polymerized via 1,2-polymerization process on exposure to 254nm. Liposomes composed of DOPE/dioleoylphosphatidylcholine(DOPC)/DHM(3/3/1) were stable at neutral pH, but leaky at weakly acidic pH. The leakage of entrapped calcein from liposomes in phosphate-buffered saline (PBS, $37^{\circ}C$) of pH 4.8 and 5.8 was complete within 30 and 50 min, respectively. The release of toke entrapped calcein was increased with decreasing pH such that 50% and 100% of the calcein were released at pH near 5.5 and 5.0, respectively.

Pharmacokinetics of antitumer agents, KR 53170 (항암성분 KR 53170 및 관련화합물의 약물동태 연구)

  • 실험목적: KR 53170 및 KR 53234 는 천연물에서 분리 추출한 화합물로 terpene lacton ring을 가진 약산성 물질이다. in vitro 항종양 실험에서 종양억제효과가 있었고 in vivo 항종양 실험에서도 유의성 있는 효과가 있었으며 급성 경구독성시험에서도 독성이 낮은 것으로 평가 되었다. 이에 따라 KR 53170 등의 약물동태학적인 성질을 연구하여 독성실험과 임상실험을 위한 용법용량을 결정하고 나아가 본약물의 제제개발에 도움이 되도록 본 연구를 시행하였다. 방법: KR 53170 및 53234은 경구시 20mg/kg, 정맥투여시는 10mg/kg로 투여하였다. Rat의 혈장 sample 채취는 ether로 흡입마취 시킨 후 heart puncture 하였다. 뇨시료는 대사 cage를 이용하여 채취하였으며 얻어진 모든 시료는 HPLC로 분석 하였다. 분석결과는 computer program 'Multi-free'를 이용하여 주요 pqarameter를 산출하였다. 인체혈액에 대한 혈청단백결합율 측정은 ultrafiltration법을 이용하였다. 즉 YMT membrane을 이용하여주요 parameter를 산출하였다. 인체혈액에 대한 혈청단백결합율 측정은 ultrafiltration법을 이용하였다. 즉 YMT membrane을 이용하여 유리약물을 분리하여 HPLC로 정량하였다. 결과 및 고찰: 1. KR 53170 10mg/kg 정맥투여시 최고혈중농도는 0.55ug/ml, 반감기는 0.51hr, 분포용적은 4.5L이었다. 20mg/kg를 경구 투여시 최고 혈중농도는 0.18ug/ml, 반감기는 3.5기산이고 AUC는 0.91ug.ml, 분포용적 28Lm, Ka 3.49$hr^{-1}$ 그리고 Cl는 5.5L/hr/kg이었다. 이는 투여용량에 비해 매우 적은량이 흡수되고 배설 된 것으로 약물이 혈액에 대한 용해도 문제에 기인하는 것으로 간주된다. 2. KR 53234 10mg/kg 정맥투여후의 최고혈중농도는 1.14ug/ml, 반감기는 0.50hr, 분포용적은 2.2L이었다. 20mg/kg 경구 투여시의 최소 혈중 농도는 0.33 ug/ml, 소실반감기는 1.5시간, AUC는 0.942ug.hr/ml, 분포용적 11L, Ka는 3.05 $hr^{-1}$ 그리고 Cl는 5.3L/hr/kg이었다. 이는 KR 53170에서와 같이 매우 적은량이 흡수되고 배설되었다. 3. KR 53170의 혈청단백 결합율은 5-500 ug/ml 범위에서 78.7-86.2%이었고 KR 53234의 혈청단백결합율은 5-100 ug/ml 범위에서 79.6-71.2%이었다.이었다.

Pollutant Loading and Changes of Water Quality at the Namdae-cheon Watershed in Keum river Districts (금강수계 남대천유역의 수질변화와 오염부하량)

  • This study was carried out to investigate the changes of water quality, the pollutant loading by population, livestock, and land use at the Namdae-cheon in Keum river districts. Water samples were taken at the four sites of watershed, the nutrients of which were analyzed periodically from July, 1995 to April, 1998. Content of nutrients in water samples ranged from 1.8 to 5.8 mg/L for total nitrogen, from 0.01 to 0.08 mg/L for total phosphorus and from 0 to 0.007 mg/L for ortho-phosphate respectively. Nitrogen loading by pollutant unit was investigated 550.4 ton/yr as 358.1 ton/yr by livestock, 129.3 ton/yr by land use and 63.0 ton/yr by population. Phosphorus loading by pollutant unit was investigated 79.1 ton/yr as 60.6 ton/yr by livestock, 5.2 ton/yr by land use and 13.3 ton/yr by population. The loading of measured pollutant was investigated 452.5 ton/yr of nitrogen and 5.4 ton/yr of phosphorus in Namdae-cheon watershed.

Effects of 2,4-D and NAA on Nicotine Production during Callus Culture of Nicotiana gluauca Graham (담배(Nicotiana glauca Graham) Callus 배양시 Nicotine 생성에 미치는 2,4-D 및 NAA의 영향)

  • Effects of two auxins, 2,4-dichlorophenoxyacetic acid (2,4-D) and a-napthaleneacetic acid (NAA) on nicotine production during callus culture of a wild tobacco (Nicotiana glauca) were investigated using a high performance liquid chromatography (HPLC). The high concentration ($11.5\;\mu\textrm{M}$)of 2,4-D and NAA had peaks of nicotine contents at 4th and 2nd week, respectively. Thereafter, the concents decreased and the nicotine was metabolized to other alkaloids. The low concentration ($1.5\;\mu\textrm{M}$) of 2,4-D on the medium supplemented with 0.1 mM of L-aspartic acid or L-arginine inhibited nicotine production. However, the low NAA promoted it only when the medium was supplemented with L-aspartic acid. From these results, it could be concluded that both auxins exhibit different action mechanisms on nicotine production pathway and the low NAA promotes the activities for the pathway with L-aspartic acid as a precursor.cursor.
