Phytochemical Constituents of Salsola komarovii and Their Effects on NGF Induction

  • Five lignan glycosides, seven megastigmane glycosides, and seven phenolic compounds were isolated by repeated column chromatography from the MeOH extract of Salsola komarovii. Their structures were determined to be lariciresinol-9-O-${\beta}$-$\small{D}$-glucopyranoside (1), alangilignoside C (2), conicaoside (3), (+)-lyoniresinol 9'-O-${\beta}$-$\small{D}$-glucopyranoside (4), (8S,8'R,7'R)-9'-[(${\beta}$-glucopyranosyl)oxy]lyoniresinol (5), blumenyl B ${\beta}$-$\small{D}$-glucopyranoside (6), blumenyl A ${\beta}$-$\small{D}$-glucopyranoside (7), staphylionoside D (8), icariside $B_2$ (9), (6R,9S)-3-oxo-${\alpha}$-ionol ${\beta}$-$\small{D}$-glucopyranoside (10), 3-oxo-${\alpha}$-ionol 9-O-${\beta}$-$\small{D}$-apiofuranosyl-($1{\rightarrow}6$)-${\beta}$-$\small{D}$-glucopyranoside (11), blumenol B 9-O-${\beta}$-$\small{D}$-apiofuranosyl-($1{\rightarrow}6$)-${\beta}$-$\small{D}$-glucopyranoside (12), benzyl 6-O-${\beta}$-$\small{D}$-apiofuranosyl-${\beta}$-$\small{D}$-glucopyranoside (13), canthoside C (14), tachioside (15), isotachioside (16), biophenol 2 (17), 2-(3,4-dihydroxy)-phenyl-ethyl-${\beta}$-$\small{D}$-glucopyranoside (18), and cuneataside C (19) by spectroscopic methods. All the isolated compounds 1 - 19 were reported from this source for the first time. Compounds 2, 3 and 6 upregulated NGF secretion to $118.8{\pm}3.6%$, $128.2{\pm}9.3%$ and $111.1{\pm}7.1%$ without significant cell toxicity.

Expression of a $\beta$-1,3-Glucanase Gene from Bacillus circulans in B. subtilis and B. megaterium (Bacillus subtilis와 Bacillus megaterium에서의 $\beta$-1,3-glucanase 유전자의 발현)

  • A Bacillus circulans KCTC3004 $\beta$-1,3-glucanase gene contained in a recombinant plasmid pLM460 derived from subcloning the original recombinant plasmid pLM530 was trasferred into a new shuttle vector plasmid pLMS1180 by ligating linearized DNAs of pLM460 and pUB110. B. subtilis RM125 and B. megaterium ATCC14945 transformed with pLMS1180 produced the $\beta$-1,3-glucanase substantially. Most of the enzyme was produced during the exponential growth period. The maxium activities of the $\beta$-1,3-glucanase produced by the Bacillus transformants were compared with that of the B. circulans gene donor strain. The B. subtilis RM125 (pLM1180) enzyme showed the activity 14 times higher than that of the gene donor cells, followed by the B. megaterium ATCC14945 (pLMS 1180) enzyme with activity 5 times higher than that of the gene donor cells. While E. coli secreted about 7% of the produced enzyme, B. subtilis excreted the enzyme into the medium wholly and B. megaterium about 97% of the total product. The SDS-PAGE of this enzyme produced in E. coli (pLMS1180), B subtilis (pLMS1180) or B. megaterium (pLMS1180) indicated a molecular weight of 38,000. The enzymes overproduced in three different host cells hydrolyzed laminarin to produce mainly laminaribiose, laminaritriose, and laminarioligosaccharides. The plasmid pLMS1180 was stable in B. megaterium, E. coli, but was unstable in B. subtilis.

The Clinicopathological and Prognostic Impact of 14-3-3 Protein Isoforms Expression in Human Cholangiocarcinoma by Immunohistochemistry

  • The 14-3-3 proteins are highly conserved, ubiquitous molecules involved in a variety of biologic phenomena, such as cell cycle control, and apoptosis. However, their expression in cholangiocarcinoma has not been previously characterized. In this paper, immunohistochemistry using specific anti-14-3-3 monoclonal antibodies was performed on formalin-fixed;, paraffin embedded archival tissue from 86 patients of cholangiocarcinoma. We also examined the correlation between expression and survival rate and clinicopathologic factors such as tumor location, tumor size, pathologic differentiation, lymphatic permeation, lymph node metastasis, and tumor stage. Positive 14-3-3 proteins expression was observed for 6 isoforms (${\beta}$, ${\sigma}$, ${\gamma}$, ${\theta}$, ${\delta}$, ${\eta}$) of these proteins in 86 patients of cholangiocarcinoma. ${\beta}$ and ${\sigma}$ isoform immunoreactivity was correlated with lymph node metastasis, tumor stage and patients' survival rate. In addition, ${\delta}$ isoform immunoreactivity showed trends with tumor location, tumor size, pathologic differentiation and tumor stage, while the ${\theta}$ isoform was correlated with pathologic differentiation. These results indicated that upregulated expression of some isoforms of 14-3-3 may be a common mechanism for evading apoptosis in cholangiocarcinoma, so that targeting 14-3-3 may be a novel promising strategy for the treatment of this tumor.

Essential Oil Composition of Chrysanthemum boreale and Chrysanthemum indicum (산국과 감국의 정유성분 조성비교)

  • The compositions of essential oils isolated from the aerial parts of Chrysanthemum boreale and C. indicum by steam distillation were analyzed by GC and GC-MS. Ninty-four components were identified in the essential oil from C. boreale, with camphor (15.40% as GC peak area), cir-chrysanthenol (14.11%), ${\alpha}-thujone$ (13.27%), 1,8-cineole (4.16%), ${\alpha}-pinene$ (3.80%), ${\beta}-caryophyllene$ (3.58%), gremacrene D (2.69%), camphene (2.40%), umbellulone (2.24%) and ${\beta}-pinene$ (2.00%) as the major constituents. In the oil from C. indicum, the major constituents among 80 components identified were germacrene D (16.50%), camphor (10.04%), ${\alpha}-thujone$ (6.40%), ${\alpha}-pinene$ (4.50%), ${\alpha}-cadinol$ (3.97%), camphene (3.82%), ${\beta}-pinene$ (3.67%), zingiberene (3.64%), cis-chrysanthenol (3.45%), piperitone (3.09%), 1,8-cineole (2.61%) and chrysanthenone (2.42%). The oil from C. boreale, although similar to that from C. indicum in many respects, contained proportionately higher levels of camphor, cis-chrysanthenol, ${\alpha}-thujone$, 1,8-cineole and umbellulone, while that from C. indicum contained higher levels of germacrene D, ${\beta}-caryophyllene$, ${\alpha}-cadinol$, zingiberene, cis-chrysanthenol and piperitone.

Aging Behavior and Effect of Heat Treatment on High Temperature Mechanical Properties in Ti-15V-3AI-3Cr-3Sn (Ti-15V-3Al합금의 시효거동과 열처리에 따른 고온 기계적 특성)

  • Titanium alloys are the one of promising candidate materials for medium high temperature parts in the aircraft, automobile, petrochemistry and electrochemistry because of their high strength with low density in medium high temperature. In this study, the effects of aging and heat treatments on the mechanical properties of Ti-15-3 alloy in medium high temperature, which was $400^{\circ}C$, were studied. Solid solution treatment was performed at $8000^{\circ}C$ of $\beta$ phase region for 1 h and the alloy was quenched in water. The alloy was aged at $5000^{\circ}C$ of $\alpha$ and $\beta$ two-phase region for 1, 2, 4, 8, ... and 100 h to increase the mechanical property. The $\beta$ single phase was observed at all parts of specimens in Ti-15-3 alloy after ST. As the aging at $500^{\circ}C$, fine precipitates of a phase was generated from matrix of $\beta$ phase and the microstructure was consisted of weaving structure such as Widmanstiitten a phase. The most suitable aging time is 24h in$ 400^{\circ}C$. At this time, strength is 1164 MPa and elongation is about 12%. In room temperature, elongation of Ti-15-3 alloy aged at $500^{\circ}C$ for 16 h is poor (=3%) in spite of high tensile strength (1458 MPa).

Production of Lignocellulytic Enzymes from Spent Mushroom Compost of Pleurotus eryngii (큰느타리버섯 수확 후 배지로부터 리그닌섬유소분해효소 생산)

  • The lignocellulytic enzymes including a-amylase (EC 3.2.1.1), lignin peroxidase (EC 1.11.1.14), laccase (EC 1.10.3.2), xylanase (EC 3.2.1.8), ${\beta}$-xylosidase (EC 3.2.1.37), ${\beta}$-glucosidase (EC 3.2.1.21) and cellulase (EC 3.2.1.4) were extracted from spent mushroom compost (SMC) of Pleurotus eryngii. Different extraction buffers and conditions were tested for optimal recovery of the enzymes. The optimum extraction was shaking incubation (200 rpm) for 2 h at $4^{\circ}C$. ${\alpha}$-Amylase was extracted with the productivity range from 1.20 to 1.6 Unit/SMC g. Cellulase was recovered with the productivity range from 2.10 to 2.80 U/gf. ${\beta}$-glucosidase and ${\beta}$-xylosidase productivities showed lowest recovery producing 0.1 U/g and 0.02 U/g, respectively. The P. eryngii SMCs collected from three different mushroom farms showed different recovery on laccase and xylanse, cellulase. Furthermore, the water extracted SMC was compared to commercial enzymes for its industrial application in decolorization and cellulase activity.

  • Carotenoid pigments from muscle of mussel, Mytilus coruscus, and blue mussel edulis, were separated by thin layer and column chromatography. The isolated carotenoids were identified by comparative test with reference carotenoids, reduction with sodium borohydride, isomerization with iodine and absorption spectrophotometry. The carotenoid content in the muscle of mussel were 0.4mg% in male and 2.7mg% in female, and the carotenoids were composed of 23.4%, 33.4% mytiloxanthin, 26.3%, 22.5% 3, 4, 3'-trihydroxy-7', $8'-didehydro-{\beta}-carotene$, 24.8%, 22.8% pectenoxanthin, 14.0%, 9.9% pectenolone and 5.1%, 6.1% diatoxanthin in male and female, respectively. While, the carotenoid contents in the muscle of blue mussel were 1.1mg% in male and 3.2mg% in female, and the carotenoids were composed of 33.8%, 35.6% mytiloxanthin, 28.4%, 44.7% pectenoxanthin, 18.1%, 5.0% diatoxanthin, 9.7%, 8.7% pectenolone and 5.5%, 3.1%, 3, 4, 3'-trihydroxy-7', $8'-didehydro-{\beta}-carotene$ in male and female, respectively.

Lipids of Fresh-Water Cockle, Corbicula elatior (재첩의 지질에 관한 연구)

  • Present investigation was carried out to clarify chemical characteristics, lipid composition, fatty acid components and the sterol composition of the lipids from the fresh-water cockle, Corbicula slatier. The results obtained are as follows :1) The oil content, iodine value and unsaponifiable matter of the total .lipids were 2.17, 119 and $16.6\%$ respectively. 2) The main components of the total lipids were phospholipids (ahout $43\%$), triglyceride and sterol. 3) The main fatty acids of the total lipids were $C_{16:0}\;(48.4\%),\;C_{14:0}\;(10.0\%),\;C_{18:3}\;(8.3\%),\;C_14:1\;(5.4\%)\;and\;C_{18:1}\;(5.3\%)$. 4) Sterols found were 24-methylenecholesterol ($14.1\%$), $\beta-sitosterol(22.5\%)$, fucosterol ($14.9\%$), isofucosterol (?) ($14.0\%$), $\Delta^7-avenasterol (?)(5.2\%)$ and cholesterol ($2.3\%$).

Taxoids from the Heartwood of Taxus baccata L. Growing in Turkey

  • The ethanolic extract of the heartwood of Taxus baccata L. afforded six taxoids, belonging to the skeletally three different groups, $2{\alpha},\;5{\alpha},\;10{\beta}-triacetoxy-14{\beta}-(2-methyl)-butryloxy-4(20)$,11-taxadiene (1), taxusin (2), baccatin VI (3), baccatin III (4), $1{\beta}-hydroxybaccatin$ I (5) and taxol (6). The isolation and structure elucidation of three taxoids (3-5) have been presented in this study. This is the first report for the presence of these taxoids in Taxus baccata L. growing in Turkey.

  • Five electrophoretic bands of crude enzyme extracted from rice leaves were found to possess both chitinase and ${\beta}-1,3-glucanase$ activities. These $chitinase/{\beta}-1,3-glucanase$ were resolved into acidic and basic fractions of protein by DEAE-cellulose and chitin affinity column chromatography. The optimal pH and temperature for ${\beta}-1,3-glucanase$ activity of two fractions were in the same extent as pH 5 and $60^{\circ}C$, whereas those for chitinase activity differed from one another; pH 3 and $60^{\circ}C$ for the acidic and pH 4 and $50^{\circ}C$ for the basic fraction, respectively. In addition, lysozyme activity was found in both fractions.
