백하수오의 식품학적 영양 성분 및 휘발성 향기 성분 분석을 통한 관능적 특성 검토 (Analysis of Nutritional Components, Volatile Properties, and Sensory Attributes of Cynanchi wilfordii Radix: Characterization Study)

  • 본 연구는 백하수오가 가지는 일반 성분과 영양 성분에 대한 탐색을 실시하였다. 무기질에서 높은 칼륨 성분을 보이고 있었으며 라디칼 소거능 역시 우수한 것으로 나타났다. 아미노산에서는 유리 및 구성 아미노산 모두 arginine이 높은 함량을 나타냈으며 생리활성이 우수한 것으로 보고되는 ${\gamma}$-aminobutyric acid(GABA) 역시 높은 함량을 보이고 있다. 지방산 조성을 볼 때 필수지방산으로 알려진 linoleic acid가 가장 주된 지방산으로 나타났으며 ${\omega}$-3 계열인 linolenic acid가 존재하는 것으로 볼 때 우수한 영양적 조성을 가지고 있다고 볼 수 있다. 관능검사를 통해 백하수오차의 경우 녹차에 비해 약간의 쓴맛과 단맛이 부족하다고 측정되었으나 농도 조절이나 부재료 첨가를 통해 개선할 수 있을 것으로 판단된다.

  • Food safety is a term broadly applied to food quality that may adversely affect human health. These include zoonotic diseases and acute and chronic effects of ingesting natural and human-made xenobiotics. There are two major areas of concern over the presence of residues of antibiotics in animal-derived foodstuffs with regard to human health. The first is allergic reactions. Some antibiotics, such as penicillins can evoke allergic reactions even though small amounts of them are ingested or exposed by parenteral routes. The second is development of antibiotic resistance in gut bacteria of human. Recently multi-resistant pneumococcal, glycopeptide-resistant enterococci and gram negative bacteria with extended-spectrum $\beta$-lactamases have spread all over the world, and are now a serious therapeutic problem in human. Although it is evident that drugs are required in the efficient production of meat, milk and eggs, their indiscriminate use should never be substituted for hygienic management of farm. Drug should be used only when they are required. In addition to veterinary drugs, environmental contaminants that were contaminated in feed, water and air can make residues in animal products. Mycotoxins, heavy metals, pesticides, herbicides and other chemicals derived from industries can be harmful both to animal and human health. Most of organic contaminants, such as dioxin, PCBs and DDT, and metals are persistent in environment and biological organisms and can be accumulated in fat and hard tissues. Some of them are suspected to have endocrine disrupting, carcinogenic, teratogenic, immunodepressive and nervous effects. The governmental agencies concerned make efforts to prevent residue problems; approval of drugs including withdrawal times of each preparation of drugs, establishment of tolerances, guidelines regarding drug use and sanitation enforcement of livestock products. National residue program is conducted to audit the status of the chemical residues in foods. Recently HACCP has been introduced to promote food safety from farm to table by reducing hazardous biological, chemical and physical factors. Animal Production Food Safety Program, Quality Assurance Programs, Food Animal Residue Avoidance Databank are para- or non-governmental activities ensuring food safety. This topic will cover classification and usage or sources of chemical residues, their adverse effects, and chemical residue status of some countries. Issues are expanded to residue detection methodologies, toxicological and pharmacokinetic backgrounds of MRL and withdrawal time establishments, and the importance of non-governmental activities with regard to reducing chemical residues in food.

Effects of dietary supplementation of lipid-coated zinc oxide on intestinal mucosal morphology and expression of the genes associated with growth and immune function in weanling pigs

  • Objective: The present study was conducted to investigate the effects of a lipid-coated zinc oxide (ZnO) supplement Shield Zn (SZ) at the sub-pharmacological concentration on intestinal morphology and gene expression in weanling pigs, with an aim to gain insights into the mechanism of actions for SZ. Methods: Forty 22-day-old weanling pigs were fed a nursery diet supplemented with 100 or 2,500 mg Zn/kg with uncoated ZnO (negative control [NC] or positive control [PC], respectively), 100, 200, or 400 mg Zn/kg with SZ for 14 days and their intestinal tissues were taken for histological and molecular biological examinations. The villus height (VH) and crypt depth (CD) of the intestinal mucosa were measured microscopically following preparation of the tissue specimen; expression of the genes associated with growth and immune function was determined using the real-time quantitative polymerase chain reaction. Results: There was no difference in daily gain, gain:feed, and diarrhea score between the SZ group and either of NC and PC. The VH and VH:CD ratio were less for the SZ group vs NC in the jejunum and duodenum, respectively (p<0.05). The jejunal mucosal mRNA levels of insulin-like growth factor (IGF-I) and interleukin (IL)-10 regressed and tended to regress (p = 0.053) on the SZ concentration with a positive coefficient, respectively, whereas the IL-6 mRNA level regressed on the SZ concentration with a negative coefficient. The mRNA levels of IGF-I, zonula occludens protein-1, tumor necrosis $factor-{\alpha}$, IL-6, and IL-10 did not differ between the SZ group and either of NC and PC; the occludin and transforming growth $factor-{\beta}1$ mRNA levels were lower for the SZ group than for PC. Conclusion: The present results are interpreted to suggest that dietary ZnO provided by SZ may play a role in intestinal mucosal growth and immune function by modulating the expression of IGF-I, IL-6, and IL-10 genes.

요중 AAMA에 의한 한국 성인 아크릴아마이드(AA)의 하루섭취량 추정 및 기여 식습관에 대한 분석 (Estimation of the Daily Human Intake of Acrylamide (AA) Based on Urinary N-acetyl-S-(2-carbamoylethyl)-cysteine (AAMA) and the Contribution of Dietary Habits in South Korean Adults)

  • Objectives: This study estimated the adult Korean daily intake of acrylamide (AA) and investigated its relationship with demographic, lifestyle and dietary habits by using urinary concentrations of N-acetyl-S-(2-carbamoylethyl)-cysteine (AAMA). Methods: Human data (n=1870) was collected in a nationwide cross-sectional biomonitoring program representing the population (18-69 years) residing in South Korea. Urinary AAMA was analyzed with a LC-MS/MS system. Daily intakes of AA were estimated using mass daily AAMA, which was calculated through urinary AAMA concentration and daily creatinine excretion. Statistical analysis was performed with SAS procedures for calculating geometric means, confidence intervals and the exponentiated beta coefficient of multiple linear regressions. Results: Daily intake of AA was estimated at $0.475{\mu}g/kg$ body weight (BW) per day (95% confidence interval (CI): 0.447-0.503). In the case of current smokers, AA intake was $0.957{\mu}g/kg$ BW per day (95% CI: 0.847-1.067), which was significantly higher than that of former smokers and never smoked (p<0.0001). The strong affecting factors were age (95% CI: 0.68-1.14; p=0.0180), education level (95% CI: 1.05-1.42; p=0.0163), body mass index (BMI) (95% CI: 1.00-1.82; p<0.0001), and smoking status (95% CI: 0.97-3.05; p<0.0001). Korean dietary habits increasing AA intake were coffee (p=0.0005), cup noodles (p=0.0010) and canned foods (p=0.0005). Meanwhile, foods decreasing AA intake were fresh fruit (p=0.0076), cooked beef (p=0.0335) and cooked pork (p=0.0147). Conclusion: The Korean daily intake of AA in adults was estimated to be similar with those found in developed countries. The factors increasing daily AA intake were coffee, cup noodles and canned foods, and decreasing factors were fresh fruit, cooked beef and cooked pork.

헬리코박터 파이로리 균의 진단시약 개발을 위한 요소의 체내동태 및 호기 중 배설 (Pharmacokinetics and Excretion into Expired Air of Urea, a Potential Diagnosis Reagent of Helicobacter pylori Infection)

  • Purpose: The purpose of the present study was to investigate the pharmacokinetics of urea, a new potential diagnosis reagent of Helicobacter pylori infection. Methods: Considering the mechanism of urea breath test, we determined the excretion of urea in expired air after its oral administration in rats and beagle dogs at the dose of 2 mg/kg (including 50 mCi/mmol $^{14}C$-urea 50 ${\mu}Ci/kg$ for rats and 13.5 ${\mu}Ci/kg$ for dogs). Results: Urea was rapidly disappeared from the blood circulation by 1 hr after its i.v. bolus injection, followed by a slow disappearance by 24 hr. The half-lives at the distributive phase ($t_{1/2{\alpha}}$) and post-distributive phase ($t_{1/2{\beta}}$) were 2 min and 6 hr, respectively. The bioavailability of urea was 64.3% after its oral administration. The values of the volume of distribution ($V_{dss}$) and the total body clearance ($CL_t$) after the oral administration were compatible with those after i.v. administration. The recovery of urea in the bile was about 0.1% of the dose by 24 hr after its oral administration. Urea was extensively eliminated in the urine by 48 hr. The recovery ratios of urea in the urine and expired air were about 86.8% and 2.99% of the dose by 48 hr, respectively. Moreover, urea was mostly distributed from the blood circulation to the kidney, followed by being eliminated in the urine without metabolism. The concentration of urea in the kidney was 4.0 times higher than that of plasma at 40 min after its oral administration. Conclusions: These findings indicated that oral route appears to be available for the administration of urea. Orally administered urea, thus, was considered to be useful for the diagnosis of Helicobacter pylori infection.

감귤 주스 착즙박을 이용하여 재배된 버섯균사체의 성분 (Major Components of Mushroom Mycelia Cultivated with Citrus Juice Processing Wastes)

  • 감귤 착즙박을 주 성분으로 하는 버섯배양배지를 만들고 간, 표고, 새송이, 산호침, 참부채 및 영지 버섯 균을 접종하여 균사체 제품을 만들었다. 만들어진 균사체는 착즙박에 비하여 대체적으로 조회분은 증가하였으나, 총산, 환원당, 조단백질 및 조지방은 착즙박과 함량이 비슷하거나 일부 감소하는 경향을 보였다. 대부분의 버섯균사체 중 유기산의 함량은 착즙박의 함량과 비슷하거나 감소하는 경향을 보였으나 새송이 버섯에서는 착즙박의 2.7배 이상 유기산의 함량이 증가하였다. 버섯균사체 중 fructose는 9.20-11.06%로서 착즙박보다 증가하는 경향을 보였으나, 새송이 버섯에서는 2.65%로서 함량이 매우 감소하였으며, glucose의 경우는 모든 버섯 균사체에서 그 함량이 착즙박보다 매우 감소하는 경향을 보였는데, 산호침 버섯 및 참부채 버섯에서 감소하는 현상이 더욱 심하였다. 버섯균사체 중 sucrose는 착즙박의 함량보다 감소하는 경향을 보였는데, 간버섯과 새송이 버섯에서 감소현상이 심하였다. 버섯균사체의 narirutin 함량은 448.67-932.98 mg%로서 대부분 착즙박에 비하여 비슷하거나 약간 감소하는 경향을 보였으나, hesperidin의 함량은 3,019.94-4,980.94 mg%로서 간버섯을 제외하면 착즙박보다 17.81-52.61%증가하여 농축되는 현상을 나타내었다. 버섯균사체 중 TDF 함량은 38.87-51.49%, NDF 함량은 11.94-17.67%, ADF함량은 11.00-14.09%로서 대부분 착즙박의 함량과 비슷하거나 증가하는 현상을 보였다. 버섯균사체의 ${\beta}-glucan$ 함량은 9.43-13.69%로서 착즙박과 비슷하거나 조금 감소하는 경향을 보였다. 총 polyphenol은 모든 버섯 균사체에서 8.01-20.28 mg%로서 그 함량이 비슷하거나 대부분 감소하였다. 버섯균사체의 EDA 및 NSA는 새송이 버섯 균사체를 제외한 모든 버섯 균사체에서 착즙박보다 감소하는 경향을 보였다.

단일 생약재 및 생약복합제 에탄올추출물의 항산화 및 항염증 활성 (Antioxidant and Anti-inflammatory Activities of Ethanol Extracts from Medicinal Herb Mixtures)

  • 본 연구에서는 문헌을 통하여 항산화 혹은 항염증 활성을 가지는 6종의 생약재와 이들 생약재를 혼합하여 제조한 생약복합제의 항산화 및 항염증 활성을 측정하였다. 총 폴리페놀 함량은 어성초, 홍화, MIX-2 에탄올추출물이 각각 58.98, 60.79, 57.74 mg/g의 함량으로 다른 시료들 보다 높게 측정되었다. 또한 플라보노이드 함량도 어성초(HC), 홍화(CT), MIX-2에탄올추출물이 각각 36.86, 19.42, 14.74 mg/g으로 높게 측정되었다. 단일 생약재와 생약복합제 에탄올추출물의 라디칼 소거능을 측정한 결과 DPPH 라디칼의 RC50 값은 어성초, 홍화, MIX-2 에탄올추출물에서 각각 69.64, 50.70, $63.49{\mu}g/mL$로 추출물 가운데 우수한 DPPH 라디칼 소거능을 보였고, ABTS 라디칼의 $RC_{50}$ 값도 어성초, 홍화, MIX-2가 각각 24.06, 18.91, $41.61{\mu}g/mL$로 DPPH 라디칼 소거능과 유사한 경향을 보였다. 시료들의 항염증 효능을 측정하기 위해 5-LO 및 COX-2 효소 저해활성을 측정한 결과 단일 생약재 에탄올추출물보다 생약복합제 에탄올추출물이 5-LO와 COX-2 효소를 동시에 저해하는 효과가 증가되는 것으로 나타났다. 염증유발 효소 저해활성이 우수했던 생약복합제 에탄올추출물들의 NO 소거활성을 비교한 결과, MIX-2 에탄올추출물이 $50{\mu}g/mL$의 농도에서부터 유의적으로 NO의 생성을 저해하는 것을 확인하였다. 따라서 단일 생약재 보다는 생약복합제가 서로 혼합되면서 시너지 효과가 나타나는 것으로 생각되며 앞으로 명확한 기전에 대한 연구들이 지속된다면 기능성 식품 소재로서의 이용이 가능할 것으로 생각된다.

Clostridium 종을 이용한 미역으로부터 아세톤, 부탄올, 에탄올 (ABE) 생산 (Acetone, Butanol, Ethanol Production from Undaria pinnatifida Using Clostridium sp.)

  • 본 연구에서는 미역을 이용하여 초고온 열산 가수분해, 효소 당화, 발효과정을 거쳐 아세톤, 부탄올, 에탄올을 생성하는 실험에 대해 진행하였다. 초고온 열산 가수분해에서의 최적 조건은 10%의 slurry, 270 mM의 황산, $160^{\circ}C$에서의 7.5분이었다. 초고온 열산 가수분해는 열처리 시간을 줄이고 적은 농도의 황산을 사용해도 더 많은 당과 적은 저해물질을 생성해 낸다는 장점이 있다. 효소 당화에서는 Viscozyme L (${\beta}-glucanase$, Novozymes)을 12 unit/ml으로 처리하는 것이 25.1 g/l로 가장 많은 단당을 생성했다. 발효에서는 C. acetobutylicum KCTC 1724이 비교적 낮은 pH 5.0에서 많은 아세톤, 부탄올, 에탄올을 생성하는 장점이 있었지만 mannitol을 모두 소비하지 못하는 단점이 있어 고농도의 mannitol 배지에 순치한 C. acetobutylicum KCTC 1724을 사용하여 발효를 진행하였다. 그 결과, 아세톤, 부탄올, 에탄올이 각각 0.99 g/l, 5.62 g/l, 2.44 g/l로 순치하지 않은 C. acetobutylicum KCTC 1724를 이용해 발효했을 때 보다 부탄올은 2.45 g/l, 에탄올은 1.10 g/l 증가했으며 수율($Y_{ABE}$)은 0.24에서 0.37로 증가했다.

CalTOX 모델을 이용한 벤젠 종합위해성평가의 불확실성 분석과 민감도 분석 (Uncertainty and Sensitivity Analyses of Human Aggregate Risk Assessment of Benzene using the CalTOX Model)

  • Objectives: The purpose of this study was to perform an aggregate human risk assessment for benzene in an industrial complex using the CalTOX model and to improve the reliability and predictability of the model by analyzing the uncertainty and sensitivity of the predicted assessment results. Methods: The CalTOXTM 4.0 beta model was used to evaluate a selected region, and @Risk 7.6 software was used to analyze uncertainty and sensitivity. Results: As a result of performing the aggregate risk assessment on the assumption that 6.45E+04 g/d of benzene would be emitted into the atmosphere over two decades, 3% of the daily source term to air remained in the selected region, and 97% (6.26E+04 g/d) moved out of the region. As for exposure by breathing, the predicted LADDinhalation was 2.14E-04 mg/kg-d, and that was assessed as making a 99.99% contribution to the LADDtotal. Regarding human Riskcancer assessment, the predicted human cancer risk was 5.19E-06 (95% CI; 4.07E-06-6.81E-06) (in the 95th percentile corresponding to the highest exposure level, a confidence interval of 90%). As a result of analyzing sensitivity, 'source term to air' was identified as the most influential variable, followed by 'exposure time, active indoors (h/day)', and 'exposure duration (years)'. Conclusions: As for the results of the human cancer risk assessment for the selected region, the predicted human cancer risk was 5.19E-06 (95% CI; 4.07E-06-6.81E-06) (in the 95th percentile, corresponding to the highest exposure level, a confidence interval of 90%). As a result of analyzing sensitivity, 'source term to air' was found to be most influential.

차량하중에 의한 사장교의 동적거동에 관한 연구 (A Study on Dynamic Behaviour of Cable-Stayed Bridge by Vehicle Load)

  • 본 연구는 사장교의 차량하중에 의한 동적거동을 파악하고자 수치해석상 비교적 간단한 일본의 풍리(豊里)(Toyosato)대교(大橋)의 자료를 근거로하여 수치해석 대상모델을 구조형식별로, 여러가지 설계변수-즉, (1) 경간비, (2) 중앙경간장과 주탑높이와의 비, (3) 거어더의 강성, (4) 주탑의 강성, (5) 케이블의 강성-을 변화시켜 수치해석을 수행하여 동적거동을 파악하고, 그 결과를 가지고 설계변수의 영향 및 충격계수의 변화에 대하여 비교 분석하였다. 이때 변위 및 단면력의 영향선을 구하기 위한 해석은 전달행렬법을 이용하였으며, 동적해석에 있어서는 평면구조계의 집중질량계로 모델을 가정하여 차량과 교량의 운동방정식을 유도한 후 모드중첩법을 사용하여 각 질점에 대한 변위 및 단면력의 동적시간이력을 구하였다.
