Supplementation of a Fermented Soybean Extract Reduces Body Mass and Prevents Obesity in High Fat Diet-Induced C57BL/6J Obese Mice

  • Obesity is a growing health problem that many countries face, mostly due to the consumption of a Westernized diet. In this present study we observed the effects of a soybean extract fermented by Bacillus subtilis MORI (BTD-1) containing 1-deoxynojirimycin against high fat diet-induced obesity. The results obtained from this study indicated that BTD-1 reduced body weight, regulated hepatic lipid content and adipose tissue, and also affected liver antioxidant enzymes and glucose metabolism. These results suggest that administration of BTD-1 affects obesity by inhibiting hyperglycemia and free radical-mediated stress; it also reduces lipid accumulation. Therefore, BTD-1 may be potentially useful for the prevention of obesity and its related secondary complications.

Effect of ${\alpha}$-Glycosidase Inhibitor in Multidrug Resistant Cell Lines

  • The objective of this study was to evaluate the reversal of multi drug resistance of human cell lines by specific inhibitors of ${\alpha}-glycosidase$ and mannosidases that had been reported to be involved in N-linked oligosaccharide processing of glycoproteins. N-methyldeoxynojirimycin, I-deoxynojirimycin, and castanospermine, which were known to be potent inhibitors of both ${\alpha}-glycosidase$ I and II, showed no activity against the multidrug resistant phenotype of the cell lines of SNU1DOX, KB-V1, and MCF-7/ADR. In contrast, I-deoxymannojirimycin, an inhibitor of mannosidase I, resulted in a slight reversal for the vinblastine resistance of the KB-V1 cell line, but did not show any activity toward the other cell lines. Parallel experiments with tunicamycin, an inhibitor of N-linked glycosylation, also resulted in no significant changes in multidrug resistant (MDR) phenotype of the cell lines tested in this work. These observations suggest that the unglycosylation of P-glycoprotein associated with the inhibitor treatments might not be correlated with the reversal of multidrug resistance of the cell lines tested in this study.

  • The influence of commonly used steeping times and temperatures about mulberry tea on a-glucosidase inhibitory activity were studied. The effects of product and preparation variables on the in-cup chemical composition of mulberry tea extracts is of interest because the appearance and taste characteristics and the possible health effects of a tea liquor arise from the chemical components extracted from the leaf during mulberry tea preparation. A comprehensive study was therefore undertaken to determine the contributions of product and provide a basic data for development of high quality mulberry tea products against diabetes. Specific eight mulberry tea were collected from each region of Korea (A-I) and when it was extracted on 85℃ for 2min, a-glucosidase activity was best for 98-102%. According to various temperature and time extraction methods, this study was carried out to optimize teamaking conditions for maximal DNJ extraction from Korean mulberry tea and and it would be of particular interest for people who drink mulberry tea to control blood glucose levels.

Effect of extraction conditions on chemical composition and antioxidant properties of mulberry fruit

  • Mulberry fruit (Morus alba L.) contains phytochemicals, including 1-deoxynojirimycin, quercetin-glucoside, kaempferol-glucoside, and anthocyanins, which have antioxidant effects. In this study, mulberry fruit extract was prepared at various temperatures (25-100℃) and water/ethanol solvent concentrations (0%-100% ethanol). Fourier-transform infrared spectroscopy (FT-IR) and circular dichroism (CD) data indicated that the content of bioactive compounds such as polyphenols and flavonoids was lower in 100% ethanolic extracts than in 30%-50% ethanolic extracts. Radical scavenging activity determined using 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2'-azino-di-3-ethylbenzthiazoline sulfonic acid (ABTS) assays was highly correlated with polyphenol and flavonoid content. In conclusion, 30%-50% ethanolic extracts contained the highest contents of bioactive compounds and exhibited high levels of radical scavenging activity. These findings may inform the use of mulberry fruit extract as a functional food.

Optimization of Mixing Ratio of Mulberry Leaf, Mulberry Fruit, and Silkworm for Amelioration of Metabolic Syndrome (대사증후군 개선을 위한 뽕잎, 오디, 누에 분말의 혼합 비율 최적화)

  • Objectives: The aim of this study was optimized mixing ratio of mulberry leaf, mulberry fruit, and silkworm for amelioration of the metabolic syndrome by using response surface method (RSM). Methods: Antioxidant, antidiabetic and antihypertensive activities of fifteen mixed powder of mulberry leaf, mulberry fruit, and silkworm by RSM were measured as indicators of metabolic syndrome. Results: The optimal mixing ratio of mulberry leaves, mulberry fruits, and silkworm with the greatest antioxidant, antidiabetic and antihypertensive activities was as follows: 2.5890 of mulberry leaf (A), 0.1222 of mulberry fruit (B), 2.9999 of silkworm (C). At this time, it was predicted that the total polyphenol content was estimated to be 185.51 tannic acid equivalent mg/g, 1, 1-diphenyl-2-picrylhy drazyl radical scavenging activity 84.77%, 1-deoxynojirimycin content 415.66 mg/100 g, ${\alpha}-glucosidase$ inhibitory activity 64.31%, ${\gamma}-aminobutyric$ acid content 267.77 mg/100 g, potassium content 1,899.11 mg/100 g, and angiotensin-converting-enzyme inhibitory activity was estimated to be 73.78%. Conclusions: It was concluded that the significant effect of the mulberry leaf, mulberry fruit, and silkworm on the metabolic syndrome-related biological activity indices (antioxidant activity, antidiabetic activity and antihypertensive activity) was as follows: 2.5890 of mulberry leaf (A), 0.1222 of mulberry fruit (B), 2.9999 of silkworm (C).

Characteristics of the 3rd day of 5th instar silkworm powder: effect of preparation method

  • Silkworm powder was prepared from 3rd day of 5th instar silkworm through various drying technology including freeze drying, hot wind drying, infrared drying, and microwave drying. The shape of silkworm dried was different with the drying methods. Freeze drying and microwave drying silkworm looked its original form, but hot wind drying and infrared drying silkworm looked shriveled and crumpled. The color of silkworm powder freeze-dried changed from yellowish green to hazel with lowering freezing temperature. Heavy metals including Pb, Cd, As, and Hg were lower the food criteria. The results of 1-deoxynojirimycin analysis was shown that BaekOkJam and GoldenSilk silkworm powder was satisfied the criteria of functional food, but YeonNokJam silkworm powder was lower than the criteria. The amino acid composition of silkworm was similar regardless of the frozen temperature in freeze drying process.

A Study on the Analysis of Functional Components and Antioxidative Activity in Mulberry (Morus alba) Silage (뽕나무(Morus alba) 사일리지의 기능성 성분 및 항산화 활성 분석에 관한 연구)

  • This study was conducted to determine application possibility of mulberry (Morus alba) silage as a functional feed in feeding management of Korean native cattle for high quality beef production by analysing active components and antioxidative activity. The chemical analysis of mulberry silage indicates that the content of dry matter, crude protein, ether extract, crude fiber and crude ash was $28.41{\pm}3.12%,\;12.43{\pm}0.28%,\;2.47{\pm}0.18%,\;20.29{\pm}0.75%\;and\;6.98{\pm}0.12%$, respectively. The content of 1-deoxynojirimycin (1-DNJ), which is representative active ingredient of mulberry and blood sugar descending component, was 0.568 mg/g and the content of $\gamma$-Aminobutyric acid (GABA), which is blood pressure descending component, was 5,936.22 pmol. Mulberry silage used in this study did not contain flavonoids but did contain total phenols for 21.69 ${\mu}g/mg$. 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity was increased with increasing the concentration of mulberry silage extracts and there was above 50% of scavenging activity at the concentration of 0.25 mg/ml. Hydroxyl radical scavenging activity was also increased with increasing the concentration of silage extracts. Alkyl radical scavenging activity was high at the low concentration of silage extracts, which was above 50% of scavenging activity at the concentration of 0.125 mg/ml. The result of this study indicated that there was high possibility of mulberry silage as a functional feed for beef cattle.

뽕잎의 기능성 효과구명

  • 뽕잎의 지금까지 박ㄹ혀진 생리활성은 중금속 흡착과 해독효과, 황산화 효과, 혈중지질 억제효과 및 혈당강하 효과를 들 수 있는데, 요약하면 다음과 같다. 1) 뽕잎이 Cd과 Pb같은 중금속에 대한 흡착 및 해독작용에 미치는 영향을 조사해 본 결과, 납중독으로 인하여 감소된 Hb와 Haematocritcl가 뽕잎 투여로 증가되었고 특히 gpa합성에 관여하는 DALAD효소 활성을 증가시켰다. 또한 뇨로의 Pb의 배설 효과가 뽕잎 투여로 증진되었으며 각 조직으로의 Pb축적을 감소시켰다. Cdwndehr으로 중독시킨 흰쥐에서도 감소된 Hd와 Haematocritcl가 뽕잎투여로 증가되었고, 특히 간조직 중의 Cd축적을 61%로 감소시켜 유의성있게 감소시켰다. 또한 뽕잎투여로 인하여 변으로의 Cd의 배설효과는 38%였고 이는 녹차투여 효솨보다는 낮은 수준이었다. 2)뽕잎의 향산화 효과는 녹차와 거의 비슷하였고 특히 혐기 처리함으로써 그 활성이 증가하였다. 또한 뽕잎중의 황산화 활성 성분은 10여종 flavonoidfb 이었고 그 중 quercetin이 가장 높은 황산화 활성을 나타냈다. 특히 dlavonidfb 화합물 중 kaempferol계열의 화합뭉이 황산화 효과가 높음을 알수 있었다. 3) 뽕잎의 고지혈 억제작용에 미치는 영향을 알아본 결과 뽕잎은 혈중 총 콜레스테롤이 차지하는 비율을 보였고 특히 혈중 중성지질 함량을 감소시킴으로써 정상의 80%수준까지 회복시켰다. 또한 콜레스테롤 생합성에 관여하는 효소활성을 뽕잎의 메탄올 추출물 중 물분획뭉이 59.9%수준으로 억제시켰고 지방분해효소 활성은 유의적인 수준은 아니었지만 16% 증가시켰다. 4) 뽕잎의 혈당강하 효과는 뽕잎을 질소가스를 이용하여 혐기처리하는 경우 유의적으로 그 효과가 증강되었다. 또한 뽕잎은 대조약물인 acarbose의 10배 용량에서 대조약물보다 23% 이상의 혈당강하 효과를 나타냈다. 이들의 혈당강하 효과는 같은 농도에서 인슐린 함량을 27%회복시켰으며 특히 surcrose와 maltose와 같은 이당류를 부하시켰을 때 혈당 강하 효과는 유의적이었다. 이는 뽕잎의 혈당강화 효과는 인슐린생합성 증가에 의한 것보다는 glucosidase의 활성을 억제시키는 기전이 더 많이 관여하는 것으로 판단할수 있었다. 또한 뽕잎 중의 혈당강하 성분으로 알려진 1-deoxynojirimycin 함량은 혐기처리로 인하여 그 함량이 5%정도 증가됨을 확인할 수 있었다. 이러한 뽕잎의 생리활성 결과에 기초할 때, 뽕잎은 고지혈증을 포함한 성인병 질환의 예방과 회복에 관련되는 조절기능을 생체에 충분히 발휘할수 있는 기능성 식품의 중요한 자원으로 이용될 수 있으리라 판단된다.
