Simultaneous Analysis of three Marker Components in Hwangryunhaedok-tang by HPLC-DAD (황련해독탕 중 3종 생리활성 물질의 HPLC-DAD 동시 정량분석법 확립)

  • In this study, a high performance liquid chromatography-diode array detector method was established, for simultaneous determination of three compounds, berberine, palmatine and geniposide in Hwangryunhaedok-tang, To develop and validate method, $C_{18}$ column (5 ${\mu}M$, 4.6 mm${\times}$250 mm) was used with gradient mobile phase, water containing 0.1% trifluoroacetic acid (TFA) and MeOH at the column temperature of $30^{\circ}C$. UV wavelength was set at 230 and 280 nm. Validation of the chromatography method was evaluated by linearity, precision and accuracy test. Calibration curve of standard components showed good linearity ($R^2$ > 0.9999). The limits of detection (LOD) and limits of quantification (LOQ) varied from 0.05 to 0.17 ${\mu}g/ml$ and 0.15 to 0.53 ${\mu}g/ml$, respectively. The relative standard deviations (RSDs) data of intra-day and inter-day test were in less than 2.99% and 1.90%, respectively. The results of the accuracy test were in the range of 98.36 to 102.52% with RSDs values 0.32 to 1.98%. The results of validation indicated that this method was a very accurate and sensitive assay.

Effects of Herb-Mix Supplementation on the Growth Performance and Serum Growth Hormone in Weaned Pigs

  • Two hundred sixteen crossbred ($Landrace{\times}Yorkshire$) castrates with an average weight of $7.4{\pm}0.3kg$ were used in a $3{\times}3$ factorial treatment array. The treatments were three levels of Herb mixture (HM; 0, 0.40 and 0.80 g/kg BW/day) and three levels of dietary nutrient (17.30% CP, Level-1; 17.90% CP, Level-2; and 18.50% CP, Level-3). The influence of HM intake and nutrient level on growth performance and ADG in 0.40- and 0.80-HM pigs increased significantly (p<0.01) as nutritional level was elevated. Although very little enhancement of ADG was observed at Level-1, peak ADG occurred in 0.8-HM treated pigs at Level-3. Feeding of 0.80 g HM/kg/d to pigs consuming Level-1 diet resulted in a 8.7% increase in ADG compared with control pigs, whereas the increase in ADG as a result of 0.80-HM with Level-3 treatment was 39%. ADFI in Level-2 pigs improved linearly (p<0.01) as HM level was increased. Treatment with HM resulted in a 12.0% increase ranging 4.7 to 20% in the ADFI compared with respective controls. ADFI at all nutritional level was significantly higher in 0.80-HM pigs (p<0.02). F/G in Level-2 pigs improved significantly as HM was fed (p<0.01), and in HM-0.80 pigs was also significantly improved as nutritional level was increased (p<0.05). Pigs fed HM had higher bone mineral density (BMD) at Level-1, longer dorsal spine length (DSL) at level-2 (p<0.05) than pigs fed basal diets. Pigs fed HM tended to higher BMD and DSL than those fed basal diets. The level of GH secretion declined with age. There was no difference between treatments (p>0.05) in the serum growth hormone at the same age. The GH was higher in pigs fed HM than those fed basal diets and increased in all pigs after 2wks feeding. A positive effect of added Herb-Mix on growth performance in weaned pigs was demonstrated by measuring the serum growth hormone, bone mineral density and length of dorsal spine.

Simultaneous Determination of Berberine, Cinnamic Acid and Glycyrrhizin in Pharmaceutical Formulations by Capillary Electrophoresis with Diode-Array Detection (모세관 전기이동법에 의한 생약제제중 베르베린, 계피산 및 글리시리진의 동시 정량)

  • A simple, accurate and reproducible capillary electrophoresis(CE) assay has been developed for the determination of berberine, cinnamic acid, and glycyrrhizin which are used in traditional Korean medicinal preparations. Separation of these compounds was performed in 20 mM phosphate buffer(pH 7.5) and acetonitrile(75:25, v/v) using a bare fused silica capillary($57 cm{\times}75 {\mu}m$ i.d.) at 25$^{\circ}C$. With the electric field of 350 V/cm, the time needed for the separation of berberine, cinnamic acid and glycyrrhizin was within 13 min. Calibration curves were linear for 1∼100 ${\mu}g/mL$ berberine, 0.3∼100 ${\mu}g/mL$ cinnamic acid and 2.5∼100 ${\mu}g/mL$ glycyrrhizin. The ranges of relative standard deviations(n=5) for those samples were between 0.96∼2.35%. The limits of detection(S/N=3) for berberine, cinnamic acid and glycyrrhizin were 0.5, 0.1 and 2.0 ${\mu}g/mL$, respectively. The numbers of theoretical plates were 181,000(berberine), 88,000(cinnamic acid) and 169,000(glycyrrhizin), while they were 3,100∼4,800 in HPLC.

Simultaneous Determination of B Group Vitamins in Supplemented Food Products by High Performance Liquid Chromatography-Diode Array Detection

  • A simple HPLC-DAD method was developed and validated to determine B group vitamin content (thiamine, riboflavin, nicotinamide, pantothenic acid, pyridoxine and folic acid) in supplemented food samples, i.e., infant formula, cereal, low-calorie food, a multi-vitamin pill and a vitamin drink. In this study, the most significant advantages were simultaneous determination of the six B group vitamins in various food matrices and a small number of sample treatment steps that required only an organic solvent, acetonitrile. Moreover, this method prevents reduction of column durability, because the mobile phase does not contain ion-pairing reagents. Analytes were separated on a Develosil RPAQUEOUS $C_{30}$ (4.6 mm ${\times}$ 250 mm, 5 ${\mu}M$ particle size) column with a gradient elution of acetonitrile and 20 mM phosphate buffer (pH 3.0) at a flow rate between 0.8 and 1.0 mL/min. Detection was performed at 275 nm, except for that of pantothenic acid (205 nm). The calibration curves for all six vitamins showed good linearity with correlation coefficients ($r^2$) higher than 0.995. The developed method was validated with respect to linearity, intra- and inter-day accuracy and precision, limit of quantification (LOQ), recovery and stability. The method showed good precision and accuracy, with intra- and inter-assay coefficients of variation less than 15% at all concentrations. The recovery was carried out according to the standard addition procedure, with yields ranging from 89.8 to 104.4%. This method was successfully applied to the determination of vitamin B groups in supplemented food products.

  • A DDFS(Direct Digital Frequency Synthesizer) used in spread spectrum communication systems must need fast switching speed, high resolution(the step size of the synthesizer), small size and low power. The chip has been designed with four parallel sine look-up table to achieve four times throughput of a single DDFS. To achieve a high processing speed DDFS chip, a 24-bit pipelined CMOS technique has been applied to the phase accumulator design. To reduce the size of the ROM, each sine ROM of the DDFS is stored 0-.pi./2 sine wave data by taking advantage of the fact that only one quadrant of the sine needs to be stored, since the sine the sine has symmetric property. And the 8 bit of phase accumulator's output are used as ROM addresses, and the 2 MSBs control the quadrants to synthesis the sine wave. To compensate the spectrum purity ty phase truncation, the DDFS use a noise shaper that structure like a phase accumlator. The system input clock is divided clock, 1/2*clock, and 1/4*clock. and the system use a low frequency(1/4*clock) except MUX block, so reduce the power consumption. A 107MHz DDFS(Direct Digital Frequency Synthesizer) implemented using 0.8.mu.m CMOS gate array technologies is presented. The synthesizer covers a bandwidth from DC to 26.5MHz in steps of 1.48Hz with a switching speed of 0.5.mu.s and a turing latency of 55 clock cycles. The DDFS synthesizes 10 bit sine waveforms with a spectral purity of -65dBc. Power consumption is 276.5mW at 40MHz and 5V.

  • The conclusions which was acquired one renal cell carcinoma and renal disease 132 cases that was tested renal echogram among 4,499 cases for recent 16 months at medical department, Dae Han Kyoyuk Insurance company from August, 1984 to November, 1985 are as follows: 1. On bur ultrasonography, the echo of tumor was demonstrated with echogenicity as compared with renal parenchyme. 2. The case was stage I by Robson's modification method for pathologic histology. 3. There is no fever, typical triad of renal cell carcinoma and the result of serum biochemical test was within normal limit. 4. The frequency with disease was renal cell carcinoma(0.76%), ureteral stone(1.5%), multicystic kidney(2.27%), hydroureter(2.27%), Bilateral poly cystic kidney(4.55%), hydronephrosis(4.55%), renal agenesis(6.06%) renal calculi(18.18%), simple cyst(60.61%). 5. The frequency with age was 55/1200 case(4.58%) in $41{\sim}50$ years, 13/296 cases(4.39%) in $51{\sim}60$ years, 43/2144 cases(2.01%) in $31{\sim}40$ years, 14/791 cases(1.77%) in $21{\sim}30$ years, 7/53 cases(1.32%) more than 61 years and 0/15(0%) under 20 years. 6. The affected site of renal agenesis 8 cases was right side all. 7. In total renal disease 132 cases, the affected site of 126 cases except bilateral polycystic kidney 6 cases was right kidney 72 cases, left kidney the proportion of right to left 1.6:1 8. In total renal disease 132 cases except bilateral polycystic kidneys 6 cases, the patients affected with both side kidneys were 14 cases. 9. The affection rate with sex in total renal disease 132 cases was 98/2860 cases in male, 34/1819 cases in female and the former was about 2 times than the latter. 10. Classifying the stone with part, nephrolithiasis 24 cases were appeared high frequency, on the contray, ureteral stone 2 cases. 11. 2 cases of ureteral stone developed complication, hydronephrosis and hydroureter. 12. The linear array type transducer was not helpful for the diagnosis of lower ureteral calculi but for the lower ureteral calculi, we could see the stone with high echo in accompanying with acoustic shadowing. 13. In 24 cases of renal calculi, both side nephrolithiasis was 3 cases(12.5%). 14. In renal calculi, solitary stone could be seen extremely much and the number of stone was so much variable from 2 to 10. 15. In 26 cases with renal calculi and ureteral stone, the common clinical manifestation was a intermittent and slight pain. 16. In 80 cases of renal cyst, as one's get older, the affection rate of cyst extremely rised. 17. In bilateral polycystic kidney, large cyst had septum on the whole. 18. The patients with complication were 14 cases(10.6%) of total patients.

Patient-Specific Quality Assurance in a Multileaf Collimator-Based CyberKnife System Using the Planar Ion Chamber Array

  • This paper describes the clinical use of the dose verification of multileaf collimator (MLC)-based CyberKnife plans by combining the Octavius 1000SRS detector and water-equivalent RW3 slab phantom. The slab phantom consists of 14 plates, each with a thickness of 10 mm. One plate was modified to support tracking by inserting 14 custom-made fiducials on surface holes positioned at the outer region of $10{\times}10cm^2$. The fiducial-inserted plate was placed on the 1000SRS detector and three plates were additionally stacked up to build the reference depth. Below the detector, 10 plates were placed to avoid longer delivery times caused by proximity detection program alerts. The cross-calibration factor prior to phantom delivery was obtained by performing with 200 monitor units (MU) on the field size of $95{\times}92.5mm^2$. After irradiation, the measured dose distribution of the coronal plane was compared with the dose distribution calculated by the MultiPlan treatment planning system. The results were assessed by comparing the absolute dose at the center point of 1000SRS and the 3-D Gamma (${\gamma}$) index using 220 patient-specific quality assurance (QA). The discrepancy between measured and calculated doses at the center point of 1000SRS detector ranged from -3.9% to 8.2%. In the dosimetric comparison using 3-D ${\gamma}$-function (3%/3 mm criteria), the mean passing rates with ${\gamma}$-parameter ${\leq}1$ were $97.4%{\pm}2.4%$. The combination of the 1000SRS detector and RW3 slab phantom can be utilized for dosimetry validation of patient-specific QA in the CyberKnife MLC system, which made it possible to measure absolute dose distributions regardless of tracking mode.

Detection Method for Bean Cotyledon Locations under Vinyl Mulch Using Multiple Infrared Sensors

  • Purpose: Pulse crop damage due to wild birds is a serious problem, to the extent that the rate of damage during the period of time between seeding and the stage of cotyledon reaches 45.4% on average. This study investigated a method of fundamentally blocking birds from eating crops by conducting vinyl mulching after seeding and identifying the growing locations for beans to perform punching. Methods: Infrared (IR) sensors that could measure the temperature without contact were used to recognize the locations of soybean cotyledons below vinyl mulch. To expand the measurable range, 10 IR sensors were arranged in a linear array. A sliding mechanical device was used to reconstruct the two-dimensional spatial variance information of targets. Spatial interpolation was applied to the two-dimensional temperature distribution information measured in real time to improve the resolution of the bean coleoptile locations. The temperature distributions above the vinyl mulch for five species of soybeans over a period of six days from the appearance of the cotyledon stage were analyzed. Results: During the experimental period, cases where bean cotyledons did and did not come into contact with the bottom of the vinyl mulch were both observed, and depended on the degree of growth of the bean cotyledons. Although the locations of bean cotyledons could be estimated through temperature distribution analyses in cases where they came into contact with the bottom of the vinyl mulch, this estimation showed somewhat large errors according to the time that had passed after the cotyledon stage. The detection results were similar for similar types of crops. Thus, this method could be applied to crops with similar growth patterns. According to the results of 360 experiments that were conducted (five species of bean ${\times}$ six days ${\times}$ four speed levels ${\times}$ three repetitions), the location detection performance had an accuracy of 36.9%, and the range of location errors was 0-4.9 cm (RMSE = 3.1 cm). During a period of 3-5 days after the cotyledon stage, the location detection performance had an accuracy of 59% (RMSE = 3.9 cm). Conclusions: In the present study, to fundamentally solve the problem of damage to beans from birds in the early stage after seeding, a working method was proposed in which punching is carried out after seeding, thereby breaking away from the existing method in which seeding is carried out after punching. Methods for the accurate detection of soybean growing locations were studied to allow punching to promote the continuous growth of soybeans that had reached the cotyledon stage. Through experiments using multiple IR sensors and a sliding mechanical device, it was found that the locations of the crop could be partially identified 3-5 days after reaching the cotyledon stage regardless of the kind of pulse crop. It can be concluded that additional studies of robust detection methods considering environmental factors and factors for crop growth are necessary.

Quantitative Analysis of Glycyrrhizic Acid in Fermented Glycyrrhizae Radix by HPLC (HPLC를 이용한 발효 감초의 지표 성분 분석)

  • The aim of this study was to study the quantitative analysis of glycyrrhizic acid in Glycyrrhizae Radix extract fermented with Paecilomyces japonica, Ganoderma lucidum, honey or Nuruk. The amounts of dry on loss were measured and the quantitative analysis of glycyrrhizic acid was performed by high performance liquid chromatographic (HPLC). HPLC method was performed on C18 column ($250\;mm\;{\times}\;4.6\;mm$, $5\;{\mu}m$, RS tech) using gradient solvent mixtures of water-acetonitrile with photodiode array detector (254 nm). The flow rate was $1.0\;m{\ell}/min$. Retention time of glycyrrhizic acid was about 23.96 min and linearity of calibration was $R^2$=0.9998. Contents of glycyrrhizic acid in Glycyrrhizae Radix extract (control) was $5.048\;{\pm}\;0.14$; Contents of glycyrrhizic acid in Glycyrrhizae Radix extract fermented with Paecilomyces japonica (SDT) was $1.975\;{\pm}\;0.07$; Contents of glycyrrhizic acid in Glycyrrhizae Radix extract fermented with Ganoderma lucidum (SYT) was $2.676 \;{\pm}\;0.07$; Contents of glycyrrhizic acid in Glycyrrhizae Radix extract fermented with honey (SST) was $5.191\;{\pm}\;0.06$; Contents of glycyrrhizic acid in Glycyrrhizae Radix extract fermented with Nuruk (SNT) was $5.305\;{\pm}\;0.34$, respectively. Contents of glycyrrhizic acid in SDT and SYT were decreased but that in SST and SNT was increased when compared to control.

Simultaneous Determination of Six Components in the Traditional Herbal Medicine 'Oryeongsan' by HPLC-DAD and LC-MS/MS

  • A simple high performance liquid chromatography - diode array detector (HPLC-DAD) method has been developed and validated for simultaneous determination of the six components (cinnamic acid, cinnamaldehyde, eugenol, atractylenolide I, atractylenolide III, and ergosterol) in Oryeongsan. In addition, identification of six marker compounds was conducted by a LC-MS/MS analysis. The six compounds in Oryeongsan were separated on Shishedo $C_{18}$ column (5 ${\mu}m$, $4.6{\times}250$ mm) at a column temperature of $30^{\circ}C$. The mobile phase was a mixture of 0.1% trifluoroacetic acid (TFA) water and acetonitrile employing gradient elution at a flow rate of 1.0 mL/min. The detection wavelength was set at 205, 250, 280, and 330 nm. The developed method had good linearity ($R^2$ > 0.9997) and the limit of detection (LOD) and limit of quantification (LOQ) were observed within the ranges 0.01~0.15 and 0.05~0.45 ${\mu}g/mL$, respectively. The relative standard deviation (RSD) values of intra- and inter-day testing were indicated that less than 3% and 90.31~103.31% of accuracy. The results of recovery test were 90.56~106.72% and RSD range was measured from 0.84 to 2.95%. In conclusion, this HPLC-DAD method has been successfully applied to the simultaneous determination of six compounds in Oryeongsan samples.
