Antigenotoxic Effect of Paecilomyces tenuipes Cultivated on Soybeans in a Rat Model of 1,2-Dimethylhydrazine-induced Colon Carcinogenesis

  • We evaluated the effect of soybean dongchunghacho [SD, cultivated dongchunghacho fungus (Paecilomyces tenuipes) on soybeans] on dimethylhydrazine (DMH)-induced DNA damage and oxidative stress in male F344 rats. The animals were divided into 3 groups and fed a casein-based high-fat, low fiber diet without (DMH group) or with 13%(w/w) of soybean (DMH+S group), or SD (DMH+SD group). One week after beginning the diets, rats were treated weekly with DMH (30 mg/kg, s.c.) for 6 weeks; dietary treatments were continued for the entire experiment and endpoints measured at 9 weeks after the first DMH injection. SD supplementation reduced DMH-induced DNA damage in colon cells and reduced plasma lipid peroxidation. Thus, SD may have therapeutic potential for early-stage colon carcinogenesis.

  • 본 연구는 로페라마이드로 처리된 흰쥐의 결장점막에 1,2-dimethylhydrazine (DMH)로 유도시 aberrant crypt foci(ACF)에 대한 길항 작용 효과를 갖는 초임계 열처리된 무 복합추출물의 화학적 암 예방 효과를 알아보고자 하였다. 초임계 열처리된 무 복합물추출물을 DMH유발 발암의 개시(12주 동안) 또는 개시 후 단계(9주 동안)에 식수(3.2 mg/ml)에 투여 하였다. 12주 동안 DMH와 함께 초임계 열처리된 무 복합추출물 투여군은 비정상적인 선와(전신경병변)의 총 수와 ACF의 수를 각각 35 %와 30 %로 크게 줄였다. 12주 동안 초임계 열처리된 무 복합추출물을 투여한 흰쥐의 결장에서 4가지 이상의 이상선와소(ACF)가 있는 ACF의 수가 현저히 감소하였다. 9주 동안 개시후 초임계 열처리된 무 복합추출물을 투여한 그룹에서, 4 이상의 이상선와소(ACF)의 수만이 현저히 감소하였다. 12주 동안 단독으로 투여한 초임계 열처리된 무 복합추출물은 ACF의 발생을 유발하지 않았다. 이러한 결과는 초임계 열처리된 무 복합추출물이 화학적으로 유도된 흰쥐의 결장 발암에 화학적 암예방 효과를 가지고 있음을 시사한다.

Chemopreventive Effect of Amorphophallus campanulatus (Roxb.) blume tuber against aberrant crypt foci and cell proliferation in 1, 2-dimethylhydrazine induced colon carcinogenesis

  • Colorectal cancer is one of the leading causes of cancer death, both in men and women. This study investigated the effects of Amorphophallus campanulatus tuber methanolic extract (ACME) on aberrant crypt foci (ACF) formation, colonic cell proliferation, lipid peroxidative damage and the antioxidant status in a long term preclinical model of 1, 2-dimethylhydrazine (DMH) induced colon carcinogenesis in rats. Male Wistar rats were divided into six groups, viz., group I rats served as controls; group II rats treated as drug controls receiving 250 mg/kg body weight of ACME orally; group III rats received DMH (20 mg/kg body weight) subcutaneously once a week for the first 15 weeks; groups IV, V and VI rats received ACME along with DMH during the initiation, post-initiation stages and the entire period of the study, respectively. All the rats were sacrificed at the end of 30 weeks and the intestinal and colonic tissues from different groups were subjected to biochemical and histological studies. Administration of DMH resulted in significant ($p{\leq}0.05$) intestinal and colonic lipid peroxidation (MDA) and reduction of antioxidants such as catalase, glutathione peroxidase, glutathione reductase, glutathione-Stransferase and reduced glutathione. Whereas the supplementation of ACME significantly ($p{\leq}0.05$) improved the intestinal and colonic MDA and reduced glutathione levels and the activities of antioxidant enzymes in DMH intoxicated rats. ACME administration also significantly suppressed the formation and multiplicity of ACF. In addition, the DMH administered rats showed amplified expression of PCNA in the colon and decreased expression of this proliferative marker was clearly noted with initiation, post-initiation and entire period of ACME treatment regimens. These results indicate that ACME could exert a significant chemopreventive effect on colon carcinogenesis induced by DMH.

Protective Effects of Bifidobacterium spp. on Experimental Colon Carcinogenesis with 1,2-Dimethylhydrazine

  • The protective role of Bifidobacterium spp. (B. breve K-110, B. breve K-111, and B. infantis K-525) isolated from the fecal samples of healthy Koreans was investigated on 1,2-dimethylhydrazine (DMH)-induced aberrant crypt foci(ACF) formation in mouse colon. In mice fed normal diet with DMH treatment, an average of 68.5 ACF/colon was formed, whereas in mice administered with B. breve K-110, B. breve K-111, and B. infantis K-525, the numbers of DMH-induced ACF decreased to 7.2, 10.9, and 6.6 ACF/ colon, respectively. The mean number of crypts/focus was not significantly altered. Fecal harmful enzymes, such as β-glucuronidase, tryptophanase, and urease, were effectively inhibited during the administration of these bifidobacteria to mice. These results suggest that bifidobacteria could prevent colon cancer.

  • Purpose: Protein tyrosine kinase(PTK), protein kinase C(PKC), oxidase, as a mediator, have been known to take a role in signal transduction pathway of angiogenesis. The authors confirmed that PKC is the most noticeable mediator for abnormal proliferation of vascular endothelial cells through in vitro study model using the inhibitors, targeting the formation of three co-enzymes. In this study, we would investigate which isoform of PKC play an important role in abnormal angiogenesis of vascular endothelial cell. Methods: In 96 well plates, $10^4$ HUVECs(human umbilical vein endothelial cells) were evenly distributed. Two groups were established; the control group without administration of DMH(1,2-dimethylhydrazine) and the DMH group with administration of $7.5{\times}10^{-9}M$ DMH. RNA was extracted from vascular endothelial cell of each group and expression of the PKC isoform was analyzed by RT-PCR(reverse transcriptase-polymerase chain reaction) method. Results: RT-PCR analysis showed that $PKC{\alpha}$, $-{\beta}I$, $-{\beta}II$, $-{\eta}$, $-{\mu}$ and $-{\iota}$ were expressed in vascular endothelial cells of each group. DMH incresed the expression of $PKC{\alpha}$ and $PKC{\mu}$, and decreased $PKC{\beta}I$, $PKC{\beta}II$ expression dominantly. Conclusion: Based on the result of this study, it was suggested that $PKC{\alpha}$ and $PKC{\mu}$ may have significant role in abnormal proliferation of vascular endothelial cell.

  • Garlic (Allium sativum L.) with the food supplement material and medicine was used traditionally in Asia and Europe. Epidemiological studies revealed that the intake of garlic reduced incidences of various cancer including digestive system. The present study was designed to investigate the effect of garlic ethanol extract on the development of colonic aberrant crypt foci (ACF) induced by 1,2-dimethylhydrazine (DMH) in male F344 rats. Five-week-old rats were given four times for two weeks to subcutaneous injections by DMH (30 mg/kg body weight) to induce ACF. The animals were divided into groups that fed diet containing garlic ethanol extract at five different doses (0.1, 0.2, 0.5, 2, 5%), respectively, animals were evaluated for the total number of ACF and total aberrant crypts (AC) per colon detected from methylene blue-stained rat colon. ACF were formed in animals in DMH-treated group. The feeding suppressed potently the appearance ACF in the colon of rats. Especially, fed diet containing garlic ethanol extract at intermediate dose (0.5%) significantly reduced the number of ACF and AC per colon (p < 0.05). Garlic ethanol extract inhibited DMH-induced overexpression of Activator Protein-1 (AP-1) and ${\beta}-catenin$ genes related to cell proliferation that also upregulated the expression of p21Waf1/Cip1 mRNA, a cell cycle-regulating gene. These results suggested that garlic ethanol extract may inhibit ACF formation, ${\beta}-catenin$ gene as the early preneoplastic marker of malignant potential in the process of colon carcinogenesis.

Amelioration of 1,2 Dimethylhydrazine (DMH) Induced Colon Oxidative Stress, Inflammation and Tumor Promotion Response by Tannic Acid in Wistar Rats

  • Colon cancer is the third most common malignant neoplasm in the world and it remains an important cause of death, especially in western countries. The toxic environmental pollutant, 1, 2-dimethylhydrazine (DMH), is also a colon-specific carcinogen. Tannic acid (TA) is reported to be effective against various types of chemically induced toxicity and also carcinogenesis. In the present study, we evaluated the chemopreventive efficacy of TA against DMH induced colon toxicity in a rat model. Efficacy of TA against the colon toxicity was evaluated in terms of biochemical estimation of antioxidant enzyme activities, lipid peroxidation, histopathological changes and expression of early molecular markers of inflammation and tumor promotion. DMH treatment induced oxidative stress enzymes (p<0.001) and an early inflammatory and tumor promotion response in the colons of Wistar rats. TA treatment prevented deteriorative effects induced by DMH through a protective mechanism that involved reduction of oxidative stress as well as COX-2, i-NOS, PCNA protein expression levels and TNF-${\alpha}$ (p<0.001) release. It could be concluded from our results that TA markedly protects against chemically induced colon toxicity and acts plausibly by virtue of its antioxidant, anti-inflammatory and antiproliferative activities.

Effect of a Functional Food Containing Bacillus polyfermenticus on Dimethylhydrazine-Induced Colon Aberrant Crypt Formation and the Antioxidant System in Fisher 344 Male Rats

  • The goal of this study was to investigate the effects of a newly developed functional food containing Bacillus polyfermenticus (BP) and other physiologically active materials on the antioxidant system and the process of colon carcinogenesis in male F344 rats. Following a one-week adaptation period, the rats were divided into 3 groups and fed either a high-fat, low-fiber diet (control and DMH groups), or a high-fat, low-fiber diet supplemented with B. polyfermenticus ($3.1{\times}10^8\;CFU/day$) and other physiologically active materials (chitosan, chicory, ${\alpha}$-tocopherol, and flavonoids) (DMH+BP group). One week after the initiation of the diets, 2 groups of rats were subjected to six weeks of treatment with 1,2-dimethylhydrazine (DMH, 180 mg/kg BW, s.c.). The dietary treatments remained consistent throughout the entire experimental period. Nine weeks after the initial DMH injection, the rats supplemented with B. polyfermenticus had significantly lower numbers of aberrant crypt foci than those in the DMH group. Injections with DMH resulted in significantly higher leukocytic DNA damage and plasma lipid peroxidation levels, as well as in a lower plasma total antioxidant potential. These effects were reversed following supplementation with B. polyfermenticus and other physiological materials. Our results indicate that a functional food containing B. polyfermenticus exerts a protective effect on the antioxidant system and on the process of colon carcinogenesis, thereby suppressing the development of preneoplastic lesions.

Protective Effect of Antler in Experimental Colon Carcinogenesis

  • There are epidemiological evidences that the population with high fecal ${\beta}-glucuronidase$ activity has greater risk of colon cancer than the population with low fecal ${\beta}-glucuronidase$. This relationship was investigated by using the mouse-dimethylhydrazine colon carcinogenesis model and the extract of antler which was a ${\beta}-glucuronidase$ inhibitor. Mice with low fecal ${\beta}-glucuronidase$ activity induced by administration of water and Folch's fraction of antler had significantly fewer aberrant crypts after injections of 1,2-dimethylhydrazine (DMH) than mice treated with DMH alone. The result supports the hypothesis that the inhibitor of ${\beta}-glucuronidase$ such as antler extract can protect an animal against the induction of colon cancer.

  • The study was designed to compare the anti-carcinogenic effect of different level of conjugated linoleic acid (CLA) in 1,2-dimethylhydrazine (DMH)-treated rats by determining biomarkers (apoptosis, cell proliferation, eicosanoids, 1,2-diacylglycerol) and phospholipid fatty acid profile in colonic mucosa. Eighty male Sprague Dawley rats weighing 180-220g were randomly divided into 4 groups depending on the content of CLA, i.e. 0.0% CLA, 0.5% CLA, 1.0% CLA, 1.5% CLA. The experimental diet contained protein 21.6%, carbohydrate 54.6%, and fat 14.5% including CLA mixture at different level by weight. The experimental diet was fed for 14 weeks with the initiation of intramuscular injection of DMH, which was injected twice a week for 6 weeks to give total amount of 180 mg/kg body weight. Regardless of the amount of CLA supplemented to diet, CLA significantly increased the apoptotic index but did not have significant effect on cell proliferation in colonic mucosa. CLA was undetected in colonic mucosal phospholipid of rats fed the 0% CLA diet and increased to 5.9mg/g phospholipid in rats fed the 0.5% diet. The apoptotic index was increased by 251% and the 1,2-DAG content was decreased by 57% in rats fed 0.5% CLA. No further changes in these variables were observed when CLA in the diet was raised to 1.0% or 1.5%. However, dietary CLA decreased mucosal levels of prostaglandin (PG)E$_2$, thromboxane (TX)B$_2$, and arachidonic acid in dose-dependent manner. The present data indicate that dietary CLA can inhibit DMH-induced colon carcinogenesis by mechanism probably involving increased apoptosis.
