Antioxidant Activity of Medicinal Plant Extracts Used as Folk Remedies by Diabetic Patients

  • The aim of this study was to investigate the antioxidant effect of medicinal plants used by diabetic patients. Fourteen medicinal plants were selected and antioxidant activity such as 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity, inhibition effect of linoleic acid autoxidation and low density lipoprotein (LDL) oxidation, thiobarbituric acid (TBA) value were measured. The Cornus officinalis had DPPH radical scavenging activity of 84.79%, which was higher than the 78.79% for $\alpha$-tocopherol. Rosa rugosa Thunberg, Pueraria thundergiana Bentham, Artemisia princeps var. orientalis and Sasamorpha purpurascens Nakai also had high values. Extracts with higher DPPH radical scavenging activities had higher total phenol concentrations, and positive correlations between these parameters were found. Inhibitory activities of linoleic acid autoxidation, LDL oxidation and TBA value, used as indices of oxidative stress, were observed in most of the selected medicinal plants. The highest inhibitory activity for TBA value was observed in the extract of Pueraria thundergiana Bentham. Rosa rugosa Thunberg (75.50 %), Sasamorpha purpurascens Nakai (74.00 %), and Cornus officinalis (73.00 %) all had high antioxidant activity against linoleic acid autoxidation, similar to that of $\alpha$-tocopherol. The same 3 plants also had high in inhibitory activity against LDL oxidation. Therefore, we demonstrated that medicinal plants used as folk remedy by diabetic patients had antioxidant activity. Especially, Rosa rugosa Thunberg, Cornus officinalis, Pueraria thundergiana Bentham, Arlemisia princeps var. orientalis, and Sasamorpha purpurascens Nakai had high phenol concentrations which resulted in high values for DPPH radical scavenging. These same plants exhibited high values for inhibitory activities for TBA value, linoleic acid autoxidation and LDL oxidation, indices associated with lipid peroxidation.

연자육의 6-하이드록시도파민으로 유도된 도파민 세포 독성에 대한 보호효과 (Protective Effects of Nelumbinis Semen Against Neurotoxicity fuduced by 6-Hydroxydopamine in Dopaminergic Cells)

  • Objectives : This study was performed to evaluate the neuroprotective effect of water extracts from Nelumbinis semen (NSW) in dopaminergic cells. Methods : We performed 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging assay, 2,2-azinobis3-ethyl-benzothiazoline-6-sulfonic acid (ABTS) cation scavenging assay, and determination of total polyphenolic content to examine the antioxidant effects of NSW. We also evaluated the neuroprotective effects against 6-hydroxydopamine (6-OHDA)-induced toxicity using 3-(4,5-dimethylthiazol-2-yl)- 2,5-diphenyl-tetrazolium bromide assay (MIT) assay, trypan blue cytotoxicity assay, and nitric oxide assay in SH-SY5Y cells and tyrosine hydroxylase (TH) immunohistochemistry in primary rat dopaminergic neurons. Results : NSW showed $IC_{50}$ values of 184.80 and 92.90 ${\mu}$g/mL in DPPH and in ABTS assays, respectively. NSW showed 1.05% of total polyphenol contents. NSW showed protective effect against 6-0HDA-induced neurotoxicity whereas no influence on cell viability at the concentration of 1${\sim}$50 ${\mu}$g/mL. NSW reduced NO generation while 6-OHDA produced it. Moreover, it protected rat dopaminergic neurons against 6-0HDA at a dose of 1 ${\mu}$g/mL. Conclusions : These results indicated that NSW has neuroprotective effect against 6-0HDA-induced neurotoxicity through antioxidant activity in dopaminergic cell culture.

Secondary metabolites (Triterpenes) from Couroupita guianensis

  • The n-hexane and carbon tetrachloride soluble fractions of a methanolic extract of the stem bark of the Couroupita guianensis furnished three compounds, identified as $\beta$-amyrin (1), betulin-$3{\beta}$-caffeate (2) and lupeol-$3{\beta}$-caffeate (3). The structures of the isolated compounds were deduced by extensive spectroscopic analysis as well as by comparison with published values. Compounds 1-3 were subjected to antioxidant screening through free radical scavenging activity by DPPH (1,1-diphenyl-2-picrylhydrazyl), where compound 2 showed moderate antioxidant activity with $IC_{50}$ value $108.0{\mu}g/ml$.

포도 추출물들의 자유 라디칼 소거 작용 마우스 대식세포주의 염증 발현 매개 인자들에 대한 생성 억제 효과 (Effects of Grape Extracts on Free Radical Scavenging Activity and Inhibition of Pro-Inflammatory Mediator Production in Mouse Macrophage Cells)

  • 국내에서 생산된 거봉 및 캠벨 포도의 씨, 줄기 및 껍질 추출물에 대하여 항산화 작용, 염증 관련 인자 생성에 미치는 활성 및 암세포 성장에 대한 영향 등을 resveratrol과 비교하여 평가하였다. 그 결과 포도 추출물 중 거봉줄기, 캠벨줄기, 캠벨씨 및 거봉씨 추출물들이 항산화 능력을 나타내었고 그 중 거봉씨 추출물은 vitamin C와 효력이 유사하게 나타나 항산화 효능이 우수함을 알 수 있었다. 또한 마우스 대식세포주인 RAW 264.7 cell을 이용하여 포도 추출물들의 LPS처리에 의한 $PGE_2$ 및 NO 생성을 저해 여부를 확인한 결과, 거봉줄기, 거봉씨, 및 캠벨씨 추출물이 $50\;{\mu}g/mL$에서 $PGE_2$ 및 NO 생성을 50% 가량 저해하는 효능을 나타내었다. 또한 사람 폐암 및 대장암 세포주를 이용하여 포도 추출물들이 암세포 성장 저해 효과를 나타내는지를 확인하였는데 거봉줄기 및 씨 추출물 $50\;{\mu}g/mL$에서 30% 정도의 암세포 성장 저해 작용을 나타내었다.

The Isolation and Antioxidative Effects of Vitexin from Acer palmatum

  • Free radicals and reactive oxygen species (ROS) caused by UV exposure or other environmental factors are critical players in cellular damage and aging. In order to develop a new antiphotoaging agent, this work focused on the antioxidant effects of the extract of tinged autumnal leaves of Acer palmatum. One compound was isolated from an ethyl acetate soluble fraction of the A. palmatum extract using silica gel column chromatography. The chemical structure was identified as apigenin-8-C-beta-D-glucopyranoside, more commonly known as vitexin, by spectral analysis including LC-MS, FT-IR, UV, $^{1}H-$, and $^{13}C-NMR$. The biological activities of vitexin were investigated for the potential application of its anti-aging effects in the cosmetic field. Vitexin inhibited superoxide radicals by about $70\%$ at a concentration of $100\;{\mu}g/mL$ and 1,1-diphenyl-2-picrylhydrazyl (DPPH) radicals by about $60\%$ at a concentration of $100\;{\mu}g/mL$. Intracellular ROS scavenging activity was indicated by increases in dichlorofluorescein (DCF) fluorescence upon exposure to UVB $20\;mJ/cm^2$ in cultured human dermal fibroblasts (HDFs) after the treatment of vitexin. The results show that oxidation of 5-(6-)chloromethyl-2',7'-dichlo-rodihydrofluorescein diacetate ($CM-H_{2}DCFDA$) is inhibited by vitexin effectively and that vitexin has a potent free radical scavenging activity in UVB-irradiated HDFs. In ROS imaging using a confocal microscope we visualized DCF fluorescence in HDFs directly. In conclusion, our findings suggest that vitexin can be effectively used for the prevention of UV-induced adverse skin reactions such as free radical production and skin cell damage.

Antioxidant Activities of Various Solvent Extracts from Ginseng (Panax ginseng C.A. Meyer) Leaves

  • Water, methanol and ethanol extracts of ginseng leaves were assayed for total phenolics and flavonoids, ascorbic acid, cupric and ferrous ion chelating activities, 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity, ferric reducing antioxidant power (FRAP) assay and ABTS radical cation decolourization (TEAC) assay for their antioxidant properties. The ethanol extract of ginseng leaves contained significantly (p<0.05) higher amounts of total phenolics and flavonoids (600.57 and 1701 mg/100 g) than methanol (374.43 and 1512.64 mg/100 g) and water extracts (248.30 and 680.05 mg/100 g). Among solvent extracts of ginseng leaves, the ethanol extract showed the most powerful antioxidant activities. However, the ferrous ion chelating activity of ginseng leaf extracts were lower than the cupric ion chelating ability. These differences in concentrations of key antioxidants among various solvent extracts seemed to be responsible for their differences in antioxidant activities. These results suggest that ethanol extract of ginseng leaves has the most effective antioxidant capacity compared to the methanol and water extracts tested in the present study. Thus, it can be applied for the effective extraction of functional material from ginseng leaves for the usage of pharmaceutical and/or food industries.

Hydrocolloids Decrease the Digestibility of Corn Starch, Soy Protein, and Skim Milk and the Antioxidant Capacity of Grape Juice

  • Hydrocolloids have many applications in foods including their use in dysphagia diets. We aimed to evaluate whether hydrocolloids in foods affect the digestibility of starch and protein, and their effects on antioxidant capacity. The thickening hydrocolloids: locust bean gum and carboxymethyl cellulose, and the gel-forming agents: agar agar, konjacglucomannan, and Hot & Soft Plus were blended with corn starch and soy protein, skim milk, or grape juice and were examined for their in vitro-digestability by comparing the reducing sugar and trichloroacetic acid (TCA)-soluble peptide, for antioxidant capacity by total polyphenol contents and the 2,2-diphenyl-1-picrylhydrazyl radical scavenging activity. The hydrocolloids resulted in a decrease in starch digestibility with the gel-forming agents. Hydrocolloids diminished TCA-soluble peptides in skim milk compared to soy protein with the exception of locust bean gum and decreased free radical scavenging capacities and total phenolic contents in grape juice. Our findings may provide evidence for the use of hydrocolloids for people at risk of nutritional deficiencies such as dysphagia patients.

Multi-response Optimization for Unfertilized Corn Silk Extraction Against Phytochemical Contents and Bio-activities

  • This study was designed to optimize ethanol extraction process of unfertilized corn silk (UCS) to maximize phytochemical contents and bioactivities. The response surface methodology (RSM) with central composite design (CCD) was employed to obtain the optimal extraction conditions. The influence of ethanol concentration, extraction temperature and extraction time on total polyphenol contents, total flavonoid contents, maysin contents, 2,2-diphenyl-1-picrylhydrazyl(DPPH) radical scavenging activities and tyrosinase inhibition were analyzed. For all dependable variables, the most significant factor was ethanol concentration followed by extraction temperature and extraction time. The following optimum conditions were determined by simultaneous optimization of several responses with the Derringer's desirability function using the numerical optimization function of the Design-Expert program: ethanol concentration 80.45%, extraction temperature $53.49^{\circ}C$, and extraction time 4.95 h. Under these conditions, the predicted values of total polyphenol contents, total flavonoid contents, maysin contents, DPPH radical scavenging activity and tyrosinase inhibition were $2758.74{\mu}g\;GAE/g$ dried sample, $1520.81{\mu}g\;QUE/g$ dried sample, 810.26 mg/100g dried sample, 56.86% and 43.49%, respectively, and the overall desirability (D) was 0.74.

Phenolic compounds and antioxidant activity of berries produced in South Korea

  • Berries are rich sources of phenolic compounds, which are known to have health-promoting effects. In this study, phenolic compounds of seven popularly consumed berries were analyzed by HPLC-ESI-MS/MS. In addition, 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical and nitrite scavenging activities were investigated for comparison of the biological activity of the berry extracts. Chlorogenic acid was the major phenolic acid, which had the highest content in aronia, followed by blueberry. The rutin and isoquercetin contents were the highest in mulberry, followed by aronia, black raspberry, and blueberry. The anthocyanin content was the highest in black raspberry. Aronia showed the highest DPPH free radical and nitrite scavenging activities with the highest contents of total phenolics and proanthocyanidins.

창상을 유발한 흰쥐에서 금은화(金銀花) 추출물의 치료 효과 (The Effect of Lonicera japonica Extract in Wound-induced Rats)

  • Objectives This study is carried out to investigate the effects of Lonicera japonica in wound-induced rats. Methods Rats were divided into 5 groups; normal (Nor), control (Veh), positive comparison (PC), Lonicera japonica 100 mg/kg (LL), Lonicera japonica 200 mg/kg (LH), each n=8. Total polyphenol and flavonoid were quantified. 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2'-azino-bis(3 ethylbenzothiazoline-6-sulfonic acid) (ABTS) free radical scavenging activation were measured. Reactive oxygen species (ROS) was measured in serum. Antioxidant factors and inflammatory factors were measured in skin tissue, and also hydroxyproline content. Skin tissue was analyzed by Hematoxylin & Eosin and Masson's trichrome staining method. Results Total polyphenol and flavonoid were 32.86±0.14 mg/g and 67.17±0.57 mg/g. The IC50 values of DPPH and ABTS free radical scavenging activation were 26.69±1.50 ㎍/mL and 49.33±4.52 ㎍/mL. ROS was significantly lower in LL and LH groups. Nuclear factor-erythroid 2-related factor 2 (Nrf2) was significantly higher in LH group and higher in LL group but not significant. Superoxide dismutase 1 (SOD-1), catalase, and heme oxygenase 1 (HO-1) were significantly higher in LL and LH groups. Nuclear factor kappa-B p65 (NF-κBp65), phosphorylated iκBα (p-iκBα), cyclooxygenase 2 (COX-2), and tumor necrosis factor alpha (TNF-α) were significantly lower in LL and LH groups. Hydroxyproline was significantly higher in LL and LH groups. The histopathologic analysis showed that skin tissue had recovered further more in LL and LH groups than in Veh group. Conclusions These results suggest that Lonicera japonica has the anti-oxidant, anti-inflammatory and healing effects in wound-induced rats.
