Antioxidant activity of black rice and grains (흑미 및 곡류 추출물의 항산화 효과)

  • In this study, the comparison of antioxidative effect of the methanol (MeOH) extracts from grains were investigated in vitro radical scavenging system. Ten grains (black rice, rice, barley, wheat, millet, sorghum, glutinous millet, buckwheat, phellines linteus rice and brown rice) were extracted with MeOH. Among the MeOH extracts of grains, sorghum and black rice showed effective scavenging activities of 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical. The $IC_{50}$ values of sorghum and black rice were $47.4{\mu}g/mL$ and $50.6{\mu}g/mL$, respectively. In addition, black rice also exerted the strongest activities on hydroxyl radical (${\cdot}OH$) scavenging activity. Furthermore, the MeOH extracts of black rice showed effective and dose dependant scavenging activities of DPPH radical and ${\cdot}OH$. These results indicated that black rice showed strong free radical scavenging activity. It suggested that black rice could be a promising natural antioxidant against free radical-induced oxidative damage.

  • As part of an ongoing search for natural plants with antioxidant compounds by measuring the radical scavenging effect on 1,1-diphenyl- 2-picrylhydrazyl (DPPH), a total extract of the twigs of Galium spurium L. (Rubiaceae) was found to show potent antioxidant activity. Subsequent activity-guided fractionation of the methanolic extract led to the isolation of nine compounds, asperulosidic acid methyl ester (1), asperuloside (2), caffeic acid (3), kaempferol-3-O-L-rhamnopyranoside (4), quercetin-3-O-[${\alpha}$-Lrhamnopyranosyl($1{\rightarrow}6$)-${\beta}$-D-glucopyranoside] (5), isorhamnetin-3-O-glucopyranoside (6), quercetin-3-O-${\alpha}$-L-rhamnopyranoside (7), kaempferol-3-O-[${\alpha}$-L-rhamnopyranosyl($1{\rightarrow}6$)-${\beta}$-D-glucopyranoside] (8), and quercetin (9). Their structures were elucidated by spectroscopic studies. Compounds 1, 3-8 were isolated for the first time from this plant. Among them, compounds 3 and 9 showed the significant radical scavenging effects on DPPH, and compounds 3 and 7 showed the potent riboflavin originated superoxide quenching activities.

Effects of Vitex rotundifolia on radical scavenging and nitric oxide production

  • Vitex rotundifolia (V. rotundifolia) has been used for treating headache, dizziness, toothache and removal of fever as a traditional medicine in Korea. In the present study, we examined the antioxidant and anti-inflammatory activities of 85% methanol extract of V. rotundifolia. In various radical scavenging assays, V. rotundifolia exhibited strong scavenging effect on 1, 1-diphenyl-2-picrylhydrazyl free radical, superoxide radical, nitric oxide. To elucidate the anti-inflammatory properties of V. rotundifolia, we investigated the inhibition effects of nitric oxide production in IFN-gamma and LPS-stimulated mouse peritoneal macrophages. V. rotundifolia suppressed nitric oxide production, iNOS and COX-2 expression dose-dependently through suppression of NF-$\hat{e}B$ activation without notable cytotoxicity. These findings mean that V. rotundifolia may be beneficial in oxidative stress-mediated inflammatory disorders.

Antioxidant Activity and Characterization of Exiguobacterium sp. SC2-1 Isolated from Sea Water (해양에서 분리한 Exiguobacterium sp. SC2-1의 항산화 활성 및 특성)

  • For the reseach of the natural marine antioxidant, an antioxidant-producing bacterium was isolated from seawater in Jeju costal area. The isolated strain SC2-1 was Gram-positive, catalase positive, oxidase negative, motile and small rods. The strain utilized sucrose, dextrose, fructose, mannitol and maltose as a sole carbon and energy source and NaCl required for growth. The radical scavenging activity of the culture supernatant was detemined by DPPH method. This bacterium was identified based on morphological, biochemical characteristics and 16S rDNA sequence analysis, and then was named Exiguobacterium sp. SC2-1. The optimum conditions of culture for production antioxidnat were $25^{\circ}C$, pH 6-8 and $4\%$ NaCl. The stain showed the highest activity and growth cultured in medium which added $1\%$ maltose. Hydroxyl radical activity of the supernatant of Exiguobacterium sp. SC2-1 was $73\%$. The SOD activity of the culture supernatant was estimated about $35\%$.

Isolation and Identification of Antioxidative Compounds 3,4-Dihydroxybenzoic acid from Black Onion (흑양파로부터 항산화 활성 물질인 3,4-Dihydroxybenzoic acid의 분리 및 동정)

  • The antioxidant substance in black onion was identified. The assays that used 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azino-bis(3-ethylbenzthiazo line-6-sulphonic acid) ($ABTS^+$) radicals showed that the ethyl acetate fraction of black onion methanol extract had a higher level of radical-scavenging activity than the other fractions. Two antioxidative compounds were purified and isolated from the ethyl acetate fraction via column chromatographies of silica gel and Sephadex LH-20 using the guided DPPH radical-scavenging assay. The isolated compounds were identified as 3,4-dihydroxybenzoic acid (1) and quercetin (2) based on mass spectrometry and nuclear magnetic resonance. The isolated compounds showed a high level of DPPH and ABTS+ radical-scavenging activity. Compound 2 had a higher level of radical-scavenging activity than 1.

In Vitro Antioxidant Activity of Alnus firma Extracts (사방오리(Alnus firma) 추출물의 in vitro 항산화 활성)

  • This study evaluated the antioxidant activity of the extract and fractions of Alnus firma. Alnus firma had the highest total phenolic content ($452.80{\pm}7.01{\mu}g$ gallic acid equivalents/mg) in a methanol (MeOH) fraction and the highest total flavonoid content ($112.29{\pm}11.14{\mu}g$ rutin equivalents/mg) and antioxidant capacity ($936.23{\pm}0.07{\mu}g$ ${\alpha}$-tocopherol equivalents/mg) in an ethylacetate (EA) fraction. The antioxidant activities of various solvent extract fractions of Alnus firma were evaluated using various antioxidant assays, including ${\beta}$-carotene-linoleate assay, reducing power assay, 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging assay, metal chelating activity assay, superoxide anion radical scavenging assay, and lipid peroxidation inhibition assay using the ferric thiocyanate method. These activities were compared with those of ascorbic acid, butylated hydroxyanisole (BHA), gallic acid (GA), butylated hydroxytoluene (BHT), and ${\alpha}$-tocopherol. First, at a $250{\mu}g/ml$ concentration, the EA and MeOH fractions of A. firma showed 92.43% and 89.20% DPPH radical scavenging activity, respectively. Second, $50{\mu}g/ml$ of the EA fraction exhibited 72.49% superoxide anion radical scavenging activity, a little greater than the same dose of GA (60.88%). Finally, 0.5 and 1 mg/ml of the EA fraction showed 73.45% and 73.29% inhibition of peroxidation in the ${\beta}$-carotene-linoleic acid system, respectively. The decreasing order of reducing power was EA fraction > n-butanol (BuOH) fraction > dichloromethane (DCM) fraction > n-hexane (HX) fraction. The results obtained in the present study indicated that Alnus firma can be used as an easily accessible potential source of natural antioxidants.

Free radical scavenging activity and protective effect of three glycyrrhiza varieties against hydrogen peroxide-induced oxidative stress in C6 glial cells (종류별 감초의 라디칼 소거능 및 H2O2에 의한 C6 glial 세포의 산화적 스트레스 개선 효과)

  • Oxidative stress is common cause of neurodegenerative diseases. The purpose of this study is to investigate the in vitro free radical scavenging activity and protective effect of three Glycyrrhiza species including Glycyrrhiza uralensis, G. glabra, and a new variety of Glycyrrihza (Shinwongam, SW) against hydrogen peroxide-induced oxidative stress in C6 glial cells. In vitro assays, radical scavenging activities of G. uralensis, G. glabra, and SW against 2,2-diphenyl-1-picrylhydrazyl, ·OH, and O2- increased as concentration-dependent manner. In addition, the SW was found to contain the highest polyphenol and flavonoid contents. The treatment of H2O2 to C6 glial cell induced oxidative stress, whereas G. uralensis, G. glabra, and SW significantly increased the cell viability as dose-dependent manner. In particular, SW exerted stronger protective effect on H2O2-induced cytotoxicity, than G. uralensis and G. glabra. Furthermore, reactive oxygen species (ROS) formation was significantly elevated by H2O2 in C6 glial cells. However, treatments of G. uralensis, G. glabra, and SW decreased ROS formation. In addition, SW decreased pro-inflammatory related protein expression levels such as inducible nitric oxide synthase and cyclooxygenase-2, compared to H2O2-treated control group. These results indicated that G. uralensis and G. glavra, especially SW, may be useful for preventing from oxidative stress-induced neuronal damage by regulating inflammatory reaction.

Antioxidative Activity of Carotenoids in Mideodeok Styela clava

  • Carotenoids were found in high levels in both muscle and tunic samples, with the highest and lowest values observed in March and January, respectively. The average values in muscle (GM) and tunic (GT) harvested in Geoje were 49.1 mg/100g and 56.7 mg/100g, respectively, whereas those in muscle (TM) and tunic (TT) harvested in Tongyeong were 42.0 mg/100g and 50.2 mg/100g, respectively. The total phenol contents of the tunic were not significantly different (P < 0.05) between sampling area and month. We investigated the antioxidative activities of the carotenoids against linoleic acid peroxidation [1,1-diphenyl-2-picrylhydrazyl (DPPH)] and hydroxyl radicals as well as their reducing power. The DPPH radical scavenging activity was 7.6-13.5% in GM, which is relatively weak, whereas it was 21.1-29.9% in GT, 9.6-12.4% in TM and 19.3-24.1% in TT. In comparison to ${\alpha}$-tocopherol, the carotenoids were found to have strong inhibitory effects against linoleic acid peroxidation, and exhibited strong hydroxyl radical scavenging activities and reducing power at 120 ${\mu}g/mL$ of each sample.

Antioxidant activity and total polyphenolics of Shiitake Mushroom extracts(Lentinus edodes)

  • Methanol and water crude extracts, crude polysaccharides and crude polysaccharides-free of water crude extracts from Shiitake mush-room (Lentinus edodes) were investigated for their antioxidant capacity in total polyphenolics and 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity. Total polyphenolics in the crude polysaccharides-free of water crude extracts were higher than that of the other ones. And crude polysaccharides-free of water crude extracts and water crude extracts show high on DPPH free radical scavenging activity. Positive correlations were found between total polyphenolics and their antioxidant activity.

The Effects of Bamboo Extract on Human Melanocytes and B16 Melanoma Cells in vitro

  • To identify inhibitory effect of Bamboo extract on melanogenesis, the effect was compared with arbutin, ascorbic acid, hydroquinone, and kojic acid on the melanin biosynthesis in B16 mouse melanoma cells and cultured human melanocytes. The cell viability of the agent was tested on cultured human melanocytes. We also examined its. free radical scavenging activity on 1,1-diphenyl-2-picrylhydrazyl(DPPH). Bamboo extract showed considerable effect against melanin production and did not reduce cell viability at the concentration tested. It also showed potent free radical scavenging activity.
