Skin Whitening and Anti-Wrinkle Effects of Extract from Jubak of Oriental Herbal Liquor (한방 발효주 주박 추출물의 미백 및 피부 주름 개선 효과)

  • Oriental herbal liquor (Yakju) is a type of Korean traditional alcoholic beverage that uses Nuruk and oriental herbs for fermentation. The purpose of this study was to develop cosmetic ingredients using Jubak, which is a by-product of alcoholic fermentation of oriental herbal liquor. To investigate antioxidant, whitening, and anti-aging effects of Jubak, we prepared extract of Jubak and its solvent fractions. Ethyl acetate fraction (KSD E4-3) showed the most prominent free radical [1,1-diphenyl-2-picrylhydrazyl (DPPH)] scavenging activity ($SC_{50}$: 0.75 mg/mL). KSD E4-3 significantly inhibited in vitro mushroom tyrosinase activity ($IC_{50}$: 0.82 mg/mL) and reduced the melanin contents in mouse melanoma melanocyte, B16F10 cells. KSD E4-3 down-regulated protein expression of tyrosinase related proteins (TRP)-1, -2, which play key roles in melanogenesis. For anti-aging effects, inhibition of matrix metalloproteinase (MMPs) expression was evaluated using human keratinocyte, HaCaT cells. Treatment of HaCaT cells with KSD E4-3 reduced expression of MMP-1, -2, -9 and inhibited proteolytic activities of MMP-2, -9. These results suggest that KSD E4-3 induces down-regulation of cellular melanogenesis and protects against photoaging induced by UVB-induced damage. Thus KSD E4-3 could potentially be a valuable cosmetic ingredient.

Antioxidant activity and neuroprotective effects of ethanol extracts from the core of Diospyros kaki (감 심지 에탄올 추출물의 항산화 활성 및 신경세포 보호 효과)

  • This study examined the antioxidant activity and neuroprotective effects of ethanol extracts obtained from Diospyros kaki core (DCE). The total polyphenol and flavonoid contents in DCE was 786.47±15.27 and 31.14±0.82 mg/g, respectively. In addition, DCE exhibited a dose-dependent induction of radical scavenging activity, determined by 1,1-diphenyl-picrylhydrazyl (DPPH), 2,2'-azino-bis-(3-ethylbenzothiazoline-6-sulfonicacid) (ABTS), ferric reducing antioxidant power (FRAP), and reducing power assays. The viability of HT22 hippocampal cells was examined to investigate the neuroprotective effect of DCE. DCE treatment did not induce cytotoxicity at concentrations below 1,000 ㎍/mL. Additionally, DCE treatment in the background of H2O2 induce oxidative stress revealed a significant increase in the survival rat, indicated by increased SOD activity and decreased levels of MDA, a lipid peroxidation product. Therefore, the results suggest that DCE can be used as a source of natural antioxidants source and a therapeutic agent for the treatment of brain disorders induced by oxidative stress and neuronal damage.

Antioxidant activities and quality characteristics of Matcha (powdered green tea) spreads containing coconut milk (말차 첨가 코코넛밀크 스프레드의 항산화 활성 및 품질특성)

  • The antioxidant activities and quality characteristics of Matcha (powdered green tea) spreads containing coconut milk were evaluated. Matcha spreads containing coconut milk was prepared with different amounts (0, 4, 8, 12, and 16 g) of Matcha, and indicated as S1, S2, S3, S4 and S5, respectively. The moisture content, viscosity, color, pH and sugar content were influenced by the concentration of Matcha. Total phenolic content and total flavonoid content of Matcha were found to be 226 mg GAE/g and 809.42 mg QE/g respectively. Also, 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging activity and reducing power of Matcha was found to be $5.76{\mu}g/mL$ ($IC_{50}$) and 2.26 O.D., respectively. The antioxidant activities of the Matcha spreads containing coconut milk increased with increasing concentration of Matcha. Sensory evaluation indicated that S3 (8 g) was the most preferred in terms of color, taste, texture, and overall acceptance. Based on these results, it is suggested that S3 (8 g) was appropriate for achieving good quality coconut milk based spread containing Matcha.

Effects of Dietary Supplementation of Castor Aralia (Kalopanax pictus Nakai) on Performance and Fatty Acid Composition of Chicken Meat (사료 내 엄나무 첨가 급여가 육계의 생산성 및 계육의 지방산 조성에 미치는 영향)

  • The effects of dietary supplementation with Kalopanax pictus branch, leaves, and extracts on performance, proximate composition, pH, total phenol and 2,2-diphenyl-1-picrylhydrazyl (DPPH) scavenging activity, cholesterol and fatty acid composition of chicken meat. Broiler chicks were fed for 5 weeks with a diet containing 5% Kalopanax pictus branch (T1), 2% Kalopanax pictus leaves (T2), and 5% Kalopanax pictus extracts (60% leaves + 40% branch) (T3). T1 and T3 resulted in much better feed conversion than T2. In proximate composition, crude fat of chicken meat was increased, but not signi-ficantly (p>0.05) in Kalopanax pictus-supplemented diets than control diet. Total phenol contents and DPPH radical scavenging activity were significantly increased (p<0.05) by the dietary-supplementation with Kalopanax pictus branch and leaves compared to the control diet. Total and high density lipoprotein cholesterols of treatment groups were higher than the control group, and control group low density lipoprotein cholesterolwas higher than treatment groups (p<0.05). Especially, T3 was significantly (p<0.05) more effective in improving cholesterol compared to other treatment groups. The treatment groups (especially T3) showed significantly higher total fatty acid and unsaturated fatty acid compositions compare to the control group.

Antioxidation and Antigenotoxic Effects of Buckwheat Sprout Extracts (메밀싹 추출물의 항산화 효과 및 유전독성억제 효과)

  • This study was carried out to determine the antioxidative and antigenotoxic effects of buckwheat (Fagopyrum esculentum Moench) sprout using 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical donating method and micronucleus test. Buckwheat sprout were extracted with 70% ethanol and then further fractionated to n-hexane, chloroform, ethyl acetate (EtOAc), butanol and water. Among the five fractions, the EtOAc fraction showed the highest electron donating activity ($RC_{50}$ 26.1 ${\mu}g/mL$). The effects of buckwheat sprout extracts on the frequencies of micronucleated polychromatic erythrocytes (MNPCEs) induced by MNNG (N-methyl-N'-nitro-N-nitrosoguanidine) were investigated in the bone marrow. 10, 20, 40 and 80 mg/kg of each extract were administered to animals immediately after injection of MNNG and the exposure time was 36 hrs. Inhibition effects of buckwheat sprout ethanol extract were 23.4%, 40.6%, 56.3% and 73.4%, respectively. When the fraction of hexane, chloroform, ethyl acetate, butanol and water from 70% ethanol extract were treated with concentration of 80 mg/kg, the suppression rates of the MNPCE were 64.1, 67.9, 75.8, 74.2 and 63.3%, respectively.

Quality Characteristics and Antioxidant Activities of Yanggaeng Supplemented with Licorice Powder (감초 분말을 첨가한 양갱의 품질 특성과 항산화 활성)

  • Feasibility of incorporation of licorice powder (LP) as a value-added food ingredient into convenient food products was investigated using yanggaeng as a model system. LP was incorporated into yanggaeng at amounts of 0, 2, 4, 6, and 8% (w/w) based on total weight of cooked white beans and LP. pH level decreased while soluble solid content significantly increased with increasing levels of LP (P<0.05). In terms of color, lightness decreased while redness and yellowness increased significantly (P<0.05) with increasing levels of LP. Hardness also decreased significantly with a higher amount of LP in the formulation (P<0.05). 2,2'-Azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) and 2,2'-diphenyl-1-picrylhydrazyl radical scavenging activities were significantly elevated by LP addition, and they increased significantly as LP concentration increased in the formulation (P<0.05). Finally, consumer acceptance test indicated that the highest levels of LP incorporation (8%) had an adverse effect on general consumer preferences. In contrast, yanggaeng with moderate levels of LP (2%) is recommended based on its overall scores to take advantage of the antioxidant properties of LP without sacrificing consumer acceptability.

Nutritional Characteristics and Screening of Biological Activity of Agarum cribrosum (구멍쇠미역의 식품영양학적 접근 및 생리활성 분석)

  • The purpose of this study is to determine the possibility of using Agarum cribrosum as a natural health food source. To accomplish this purpose, the contents of the general and antioxidant activities of Agarum cribrosum were measured. The contents of carbohydrate, crude protein, crude lipid and ash were 45.4%, 15.0%, 2.3% and 33.1%. The calories of Agarum cribrosum were 262.3 kcal and total dietary fiber of Agarum cribrosum was 34.0%. The protein contained a total of 18 different kinds of amino acids. The content of amino acids was 12,402.42 mg/100 g. The K was the largest mineral followed by Ca, Na and Mg, implying that Agarum cribrosum is an alkali material. The antioxidant activity of Agarum cribrosum was assessed by various radical scavenging assays using DPPH (2,2-diphenyl-1-picrylhydrazyl), FRAP (ferric ion reducing antioxidant power), reducing power, and ABTS (2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid)). All antioxidant activity of Agarum cribrosum extract increased the concentration of the dependents. Total phenolic contents of Agarum cribrosum extract were $34.1{\pm}2.56mg/g$, and total flavonoids contents were estimated at $4.9{\pm}0.26mg/g$. General nutrients and other antioxidant bioactive materials in Agarum cribrosum were also potential materials for good health food. It is expected that a follow-up study of Agarum cribrosum through developing processed food and evaluation of their functional properties would provide useful information or sources of functional foods.

Physicochemical characteristics and storage stabilities of soy sauce and soybean paste sauce added with extracts of pear and sugar (배당침액을 첨가한 간장소스와 된장소스의 이화학적 특성 및 저장성)

  • This study was conducted to investigate the physicochemical characteristics and storage stabilities of soy sauce and soybean paste sauce added with extracts of pear and sugar. The commercial possibility of these sauces was investigated by sensory and preference evaluations. First, the sensory evaluation of two different particle sizes of soybean paste sauce (Type I, chopped; Type II, ground) was performed. The result of sensory and preference evaluations of soybean paste sauce Type I was higher than those of soybean paste sauce Type II, which the former was used for further analysis. The total polyphenol contents and the 2,2'-azino-bis (3-ethylb enzothiazoline-6-sulfonic acid) (ABTS${\cdot}$+) radical scavenging activities of two sauces were not significantly different (p<0.05). The 2,2-Diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity of soy sauce was higher than that of soybean paste sauce I(p<0.05). The pH, total bacterial count, saltiness, and hunter's color values of both sauces were not changed during the whole storage period (60 days). Viscosity of the soybean paste sauce I was increased while the viscosity of soy sauce was significantly decreased during the storage periods (p<0.05). From these results, it was suggested that the soy sauce and soybean paste sauce I added with pear extract of pear and sugar would be a suitable sauce for smoked duck and meat foods.

Quality Characteristics of Muffins Containing Wheat Sprout Powder (밀싹 분말 첨가 머핀의 품질 특성)

  • This study investigated the quality of muffins prepared with various concentrations of wheat sprout powder. Muffins were prepared by adding 0, 1, 3, 5, and 7% wheat sprout powder to flour of the basic formulation. The weight of muffins increased upon addition of 3~5% wheat sprout powder. Height and volume index of the muffins containing 1~5% wheat sprout powder were higher than those of the control. The pH, lightness, and redness of muffins decreased with 3~5% wheat sprout powder, whereas yellowness and moisture contents increased. The hardness of muffins increased upon addition of wheat sprout powder, but there was no significant difference among the sample groups. The total polyphenol contents and antioxidative activity as measured by 2,2-diphenyl-1-picrylhydrazyl radical scavenging activity of muffins increased as the concentration of wheat sprout powder increased. Muffins containing 1~5% wheat sprout powder showed acceptable sensory properties, including color, flavor, taste, and overall acceptability. Results indicate that the optimal concentration of wheat sprout powder in the muffin formula was 3~5% (w/w).

Component Analysis and Digestive Enzyme Activities of Fermented Crataegi Fructus Extracts (산사 발효액의 함유 성분 분석 및 소화 활성)

  • Currently many studies aimed at enhancing efficacy of medicinal food on biological activity using bioconversion technology including fermentation process. In this study, the quality characteristics and antioxidative activity of fermented Crataegi fructus was investigated. The antioxidant activity of fermented Crataegi Fructus was assessed by various radical scavenging assays using DPPH (2,2-Diphenyl-1-picrylhydrazyl), FRAP (Ferric ion reducing antioxidant power), Reducing power and ABTS (2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid)). Moisture content of fermented Crataegi Fructus was $39.3{\pm}0.06%$. Contents of crude ash, crude protein, and crude fat were $0.20{\pm}0.01$, $1.77{\pm}0.04$, and $1.40{\pm}0.59%$, respectively. Moreover, the hunter's color values of fermented Crataegi Fructus were 79.24 (lightnees), 1.58 (redness), and 31.25 (yellowness), respectively. Total phenolic contents of fermented Crataegi Fructus were $3,015{\pm}250$ GAE ${\mu}g/g$. The antioxidative activities of fermented Crataegi Fructus significantly increased in a dose dependent manner. In addition, fermented Crataegi Fructus slightly (10.4%) inhibited ${\alpha}$-glucosidase activity; however, there was no inhibitory activity against ${\alpha}$-amylase. In terms of proteolytic activity, fermented Crataegi Fructus showed a strong activity than pancreatin (used as a positive control). These results indicate that fermented Crataegi Fructus can be used as a natural resource for material aiding digestion.
