본 연구는 모시잎의 혈청, 간 및 지방조직의 체내 지질대사 개선 및 항비만 효과를 살펴보기 위하여 in vivo에서 5주령 된 흰쥐 수컷 24마리를 1주일간 적응시킨 후 정상 식이군(N), 고지방-고콜레스테롤 식이군(HFC), 고지방-고콜레스테롤 식이와 5% 모시잎 분말 첨가군(HFC-RL) 및 고지방-고콜레스테롤 식이와 10% 모시잎 분말 첨가군(HFC-RH)으로 나누어 4주간 실시하였다. 고지방-고콜레스테롤 식이로 인하여 체중이 유의적으로 증가하였으나, 모시잎 분말의 첨가 수준이 증가할수록 체중이 유의하게 감소하였다. 식이섭취량은 대조군(HFC)이 가장 낮았으며, 모시잎 분말 급여로 감소하는 경향이었다. 식이효율은 10% 모시잎 분말첨가로 유의하게 감소하여 정상 식이군(N)의 식이효율 수준과 유사하였다. 장간막지방조직과 부고환지방조직의 무게는 10% 모시잎 분말 첨가로 유의하게 감소하여 정상 식이군(N)보다 오히려 더 감소하는 경향을 보였다. 고지방-고콜레스테롤 식이로 증가되어진 혈청 중 AST, ALT, ALP 및 LDH 활성은 모시잎 분말 급여로 저하되었다. 혈청 중 중성지방 함량과 총콜레스테롤 함량도 고지방-고콜레스테롤 식이 급여로 증가되었으나, 모시잎 분말 첨가에 따라 농도 의존적으로 감소하였다. 고지방-고콜레스테롤 식이로 감소되어진 혈청 중 HDL-콜레스테롤 함량은 모시잎 분말 급여로 모두 유의하게 증가하였으며, 증가되어진 혈청 중 LDL-콜레스테롤 함량은 모두 유의하게 감소하였다. 동맥경화지수와 심혈관위험지수는 5%와 10%의 모시잎 분말 급여로 모두 유의하게 감소하였으며, 특히 10% 모시잎 분말을 급여하였을 경우 감소효과가 커 정상 식이군(N)과 유사한 경향이었다. 간조직 중 중성지방과 총콜레스테롤 함량은 모시잎 분말 급여로 모두 대조군(HFC)에 비하여 유의하게 감소하였다. 장간막지방조직과 부고환 지방조직의 중성지방 및 총콜레스테롤 함량도 모시잎 분말 급여로 유의하게 감소하였다. 지방조직의 HR-LPL과 TE-LPL 활성은 모시잎 분말의 첨가 급여로 농도 의존적으로 유의하게 감소하였다. 이상의 실험 결과 고지방-고콜레스테롤 식이와 함께 급여한 모시잎 분말은 체중, 간 및 지방조직의 무게 감소와 더불어 혈청 및 지방조직의 지질대사 개선에 긍정적인 영향을 미치는 것으로 나타났다. 이러한 결과는 모시잎 분말에 다량 함유된 식이섬유소와 클로로필 등의 생리활성 물질이 지질대사를 개선시키고 지방축적을 억제하여 고지혈증의 예방 및 치료효과와 비만을 억제하는 효과를 나타내 대사성 합병률과 심혈관계 질환의 발생을 감소시키는데 효과가 있을 것으로 생각되어진다. 그 외에 어떠한 생리활성 성분이 어떻게 작용을 하는지에 대해서는 추후 더 체계적인 연구가 필요할 것으로 사료된다.
Effects of a change in dietary Ca content and an alteration in dietary lipid type on lipid metabolism have been investigated in male Sprague-Dawley rats. The results obtained were summarized as follows ; 1) There was no comparable changes in food consumption and body weight gain among all 9 groups. 2) Serum total lipids and cholesterol were lowest when high Ca-corn all diet was fed, and highest when low Ca-butter diet was fed. 3) The contents of total lipids and cholesterol per g liver and total liver showed no consistent tendency with the dietary treatment. 4) Addition of Ca to the diet caused the significant increase in the dry fecal matter and fecal Ca. And fecal total lipids and fecal cholesterol were much greater if the high-Ca diet was fed than if control and low-Ca diet were fed. It is concluded that Ca and polyunsaturated acids have tendency of hypocholesterolemic effect.
In order to investigate the effect of riboflavin on cholesterol and MLHP (methyl linoleate hydroperoxide) -induced fatty liver electron microscopically, riboflavin was given to rate receiving high cholestero and MLHP diet for 4 weeks, 8 week old male Sprague-Dawley rats were administrated diets daily, as following group ; usual diet (control ), riboflavin 910 ug/20gm BW), cholesterol (50mg/gm BW), MLHP(0.1ml/20gm BW), cholesterol plus MLHP, cholesterol plus riboflavin, cholesterol plus MLHP plus riboflavin, by means of intubation into the stomach for 4 weeks. Each group consisted of 10 rats, The liver of the animals were examined ultrstructurally by transmission electronmicroscope. The results wee as follows ; 1) The three group including cholesterol, MLHP, cholesterol plus MLHP feeding resulted in fatty liver. 2) The main finding of the fatty liver were swelling and vacuolation of mitochondria, a lot of lipid droplets, disarrangements and loss of rough endoplamic reticulums. 3) the most striking features in the group of cholesterol plus riboflavin, cholesterol plus MLHP plus riboflavin feeding were decrease of size and number of lipid droplets, mitochondrial swelling and vesiculation, with restortion of rough endoplasmic retriculums. It is postulated with above findings that riboflavin was effective in prevention of cholesterol and MLHP -induced fatty liver.
In order to independently examine the effects of long-chain saturated fatty acids and dietary cholesterol levels on plasma and hepatic cholesterol concentrations, six different diets were fed to male Mongolian gerbils (14 gerbils pes grcup) for an 8-week period. Purified diets contained 36% energy as fat (each saturated fatty acid tested comprised about 20% of the total fat energy) and 0.06% (w/w) cholesterol, corresponding to typical human consumption patterns in Western diets. Fat blends were formulated with natural fat sources. To determine the effects of different saturated fatty acids on plasma and liver cholesterol levels, four of the six diets contained constant levels of all nutrients except for the amounts of lauric acid (12 0), myristic acid (14 : 0), palmitic acid (16 0), and stearic acid (18 : 0). Dietary cholesterol effects were tested using 16 : 0-enriched diets containing 0, 0.006, and 0.06% (w/w) cholesterol. None of the plasma lipids were influenced by fatty acid treatment, including triglycerides, plasma total-, VLDL+LDL-, and HDL-cholesterol. However, hepatic esterified cholesterol concentrations were increased in the palmitic and stearic arid diet groups compared to the lauric and myristic acid diet group. The molar ratios of hepatic EC/FC were the highest in the palmitic acid diet (12.2 $\pm$0.6) and the lowest in the myristic acid diet (6.4$\pm$0.2). Dietary cholesterol significantly (p<0.001) increased the plasma total cholesterol which was due to the increase of both HDL-and VLDL+LDL-cholesterol. In the absence of dietary cholesterol and compared to other species, the gerbil exhibited a high molar ratio of hepatic EC/FC, which was funker elevated by dietary cholesterol feeding (0.06%). The results from this study indicate that hepatic cholesterol concentrations are sensitive to both low levels of dietary cholesterol and saturated fatty acid chain length and also, that plasma cholesterol concentrations are sensitive to low levels of dietary cholesterol .
The purpose of this study was to investigate the effects of Crataegii Fructus on the diet-induced hyperlipidemia in rats. Rats were divided into 4 groups, normal group(supplied enough water and feeds only), high fat diet administered group(supplied high fat diet for 4 weeks, Control group) and Crataegii Fructus administered group(supplied high fat diet and Crataegii Fructus lyophilization extract for 4 weeks, 397.3 mg/kg(rat) in sample A, 662.5 mg/kg(rat) in sample B). Body weight, liver weight and serum lipid levels were evaluated. The water extract of Crataegii Fructus decreased liver weight and triglyceride in high fat diet induced hyperlipidemia in rats, and increased HDL-cholesterol.
Gender differences in the effects of gonadectomy and high cholesterol diet on body weight and body cholesterol were investigated by using Sprague Dawley rats. Body weight, plasma and liver levels of cholesterol and triglyceride and platelet aggregation were examined in ovariectomized(OVX) or orchidectomized(ODX) rats with their intacts after feeding diet with or without 0.5% cholesterol. Body weight was significantly increased(p < 0.01) in OVX rats and significantly decreased(p < 0.01) in ODX rats compared to their respective intact rats, and cholesterol diet significantly(p < 0.05) decreased body weight in gonadectomized rats. Liver lobes from rats fed cholesterol diet were opaque and larger than those from rats find control dict, resulting in a significant increase(p < 0.01) in LW/BW ratio. Plasma and liver levels of total cholesterol were significantly increased (p < 0.01) in female rats regardless ovariectomy when find 0.5% cholesterol diet, but those levels in male rats were increased only when they were orchidectomized(p < 0.0l). Plasma HDL-cholesterol was significantly decreased(p < 0.05) in both sexes when find cholesterol diet. HDL-cholesterol were higher in female than male rats regardless treatments(p < 0.05). Liver triglyceride was significantly increased(p < 0.05) in both sexes when find cholesterol dict. Plasma level of triglyceride was not different among groups except significant decrease(p < 0.05) in cholesterol find ODX rats. Maximum platelet aggregation in female rats was significantly lower(p < 0.05) than male, but ovariectomy and cholesterol diet caused an increase te the level of male rats. Microscopic examination showed cholesterol diet caused a lipid accumulation in liver. Results indicate that intact female rats have higher response to hypercholestcrolemic diet than intact male rats and orchidectomy causes male rats more responsive to hypercholesterolemic diet. However, ovariectomy causes an increase female food efficiency ratio to the level of male rats, significantly increasing body weight.
Objectives : In order to investigate the anti-obesity effects of Wolbi-tang(here in after referred to WBT) on the obese gene and obese inhibitory, C57BL/6 mice were induced by high-fat diet. Methods : C57BL/6 mice were divided into 5 groups(normal, only high-fat diet, high-fat diet with Reductil, high-fat diet with WBT 400, 200 mg/kg extract) and fed for 5 weeks. And observed body weight change, total cholesterol, low density lipoprotein cholesterol(LDL-cholesterol), high density lipoprotein cholesterol (HDL-cholesterol), triglyceride, glucose, leptin change, alanine transaminase(ALT), aspartate transaminase(AST), serum creatinine, the expression of ${\beta}3$-adrenergic receptor(${\beta}3AR$), leptin, uncoupling protein(UCP2) gene in 3T3-L1 adipocyte, 3T3-L1 adipocyte proliferation, histological analysis of adipose tissue and liver tissue. Results : 1. Refer to cell cytotoxicity, viability of human fibroblast cells(hFCs) showed not significant changes. 2. The amount of ALT, AST was decreased significantly in WBT 400 mg/kg, 200 mg/kg groups. The amount of creatinine showed not significant changes. 3. Body weight was decreased significantly in WBT 400 mg/kg, 200 mg/kg groups. 4. The amount of total cholesterol and triglyceride was decreased significantly in WBT 400 mg/kg, 200 mg/kg groups. LDL-cholesterol was decreased and HDL-cholesterol was increased significantly in WBT 400 mg/kg groups. 5. The amount of glucose was decreased significantly in WBT 400 mg/kg groups. 6. The amount of serum leptin was decreased significantly in WBT 400 mg/kg, 200 mg/kg groups. 7. The revelation of ${\beta}3AR$ in 3T3-L1 adipocyte was increased significantly in WBT $100{\mu}g/ml$, $50{\mu}g/ml$ groups. The revelation of leptin was decreased significantly in WBT $100{\mu}g/ml$, $50{\mu}g/ml$ groups. The revelation of UCP2 was decreased significantly in WBT $100{\mu}g/ml$ group. 8. 3T3-L1 adipocyte proliferation was decreased significantly in WBT $100{\mu}g/ml$, $50{\mu}g/ml$ groups. The size of adipocyte was decreased relative to the control group in WBT 400 mg/kg group. 9. The adipose vacuoles in liver tissue was decreased relative to the control group. Conclusions : These results suggested that WBT has inhibitory effects of obesity. WBT might be applicated on treatment of obesity and metabolic syndrome. Further studies analysing its effects were needed.
The effect of conjugated linoleic acid (CLA) isomers esterified in diacylglycerol (DAG)-rich oil on lipid metabolism was investigated. Since dietary DAG has been known to induce the regression of atherosclerosis, CLA-DAG and olive-DAG oils containing similar levels of DAG (51.4~54.2%) were synthesized from olive oil. Hyperlipidemic C57BL/6J mice were then fed high-fat high-cholesterol diets supplemented with these oils (5% each) for 7 wk. The CLA-DAG diet containing 2.1% CLA isomers (0.78% c9,t11-CLA; 1.18% t10,c12-CLA) remarkably increased the levels of total plasma cholesterol and glutamic oxaloacetic transaminase (GOT) along with hepatic cholesterol and triacylglycerol (TAG) contents. Furthermore, the CLA-DAG diet inhibited fat uptake into adipose tissue whereas fat deposition (especially in the liver) was increased, resulting in the development of fatty livers. Hepatic fatty acid composition in the CLA-DAG mice was different from that of the olive-DAG mice, showing higher ratios of C16:1/C16:0 and C18:1/C18:0 in the liver. The activity of hepatic acyl-CoA:cholesterol acyltransferase (ACAT) was higher in CLA-DAG mice while plasma lecithin:cholesterol acyltransferase (LCAT) activity and the ferric reducing ability of plasma (FRAP) were lower in CLA-DAG mice compared to the olive-DAG animals. Results of the present study suggest that CLA incorporation into DAG oil could induce atherosclerosis in mice.
Recently, Korean people are consuming seaweeds almost 3.5 times more now than three decades ago. It is well known that seaweeds contain lots of soluble dietary fiber in addition to micronutrients such as ${\beta}$-carotene, iodine and some bioactive components. Seaweeds are considered to be effective for preventing chronic diseases including obesity, diabetes mellitus, atherosclerosis, cancer or constipation. This study was conducted to investigate the effect of seamustard intake on body weight gain, blood glucose level and lipid profiles in rats fed diets with different energy nutrient composition. Male Sprague-Dawley rats (average initial weight 103.7g) were divided into groups for two experiments as follows; Control, M2.5 & M5 groups (Exp. I) and M5, M10, HCM5, HCM10, HFM5 & HFM10 groups (Exp. II). The rats were fed diet and water ad libitum for 4 weeks. In general, there was no significant difference in blood glucose and triglyceride concentration among groups. In Exp. I, serum LDL-cholesterol level of rats fed diet with 5% seamustard powder (M5) was significantly lower than that of control group, while HDL-cholesterol level, TC/LDL ratio and weight of adrenal gland were higher. In Exp. II, food intake, body weight gain and EER of high fat diet with 10% seamustard group (HFM10) were the lowest among groups. Except gastrocnemius muscle, all organ weights of HFM10 group were the lowest. Fecal cholesterol excretion and serum LDL-cholesterol concentration of HFM10 group were the highest, while serum HDL-cholesterol level was the lowest among groups. Interestingly, HDL-cholesterol concentration was the highest in HCM5 group among groups. From these results, it was suggested that seamustard intake might be more effective for body weight control, but not for improving blood lipid profiles in high fat diet than in high carbohydrate diet.
Accumulation of cholesterol may occur due to liver injury like hepatic steatosis and fibrosis as well as athrosclerosis and coronary heart disease. The aim of this study was search for protective effect of active ceramic water on experimental hepatic injury induced by high cholesterol diet(containing 1.0 % cholesterol and 0.3 % sodium cholate) in rats.
