고지방식을 섭취한 흰쥐의 체내지질함량에 대한 단백질 가수분해물의 섭취 효과 (Effects of Protein Hydrolysates on Blood and Liver Lipids in Rats fed Fat-enriched Diet)

  • The experiments were performed to investigate the effects of protein and protein hydrolysates on lipid metabolism in the hyperlipidemic/hypercholesterolemic rat model induced by feeding fat-enriched diet. In Except 1 male rats were fed four semi-purified high fat and cholesterol diets that contained different nitrogen source, casein(C), casein hydrolysate(CH), corn gluten(G) and corn gluten hydrolysate(GH), for 6 weeks. In Expt. 2 rats were fed high fat and cholesterol diet for 4 weeks to induce hyperlipidemia and hypercholesterolemia. Then the rats were divided into 4 groups and were fed the four kinds of above experimental diets for 4 weeks consecutively. The contents of total lipid , cholesterol and triglyceride in blood, liver and feces were determined. Serum lipid concentrations of CH, G and GH were significantly lower than that of C. Serum cholesterol concentrations of hydrolysate groups(CH and GH) were significantly lower than those of intact protein groups(C and G). Serum HDL -cholesterol concentration tended to increase by hydrolysate intake. The total lipid, cholesterol contents in liver showed similarity results as above. Fecal lipid excretions of CH, G, and GH groups were significantly higher than that of C group. These results indicate that hypolipidemic and /or hypocholesterolemic effect of corn gluten or protein hydrolysates were detected in the process of inducing hyperlipidemia by high-fat and cholesterol diet or after inducing hyperlipidemia.

콜레스테롤 투여 흰쥐에 있어서 유산균 발효유의 식용결과 (II) (Benefits of Fermented Milk in Rats Fed by Hypercholesterolemic Diet (II))

  • This study was performed to investigate the term effect of fermented milk in drink on rats fed by cholesterol diet. 150 to 5-week-old male rates of Sprague-Dawley strain weer divided into six groups according to diet and content of fermented milk in drink. The first group served as normal control group fed by synthetic pellets of standard diet. The second , cholesterol control group, was treated with standard diet containing 1% of cholesterol and 500000IU/100g of vitamin D2. The other four groups. fermented milk groups, FM-25,FM-50 , FM 75 and FM-100 were treated with the cholesterol and vitamin D2 diet and supplied with 25%, 50%, 75%, 100% of fermented milk in drink , respectively. The animals were sacrificed for analysis in 3, 6, 9 and 12 months. Weight gain, diet intake , and drink consumption were significantly different among groups. Fermented milk reduced serum cholesterol to a significant effect (p<0.05) and significantly increased the ratio of high density lipprotein cholesterol to total cholesterol(p<0.05). Serum triglycerides were reduced to a smaller and less significant effect in fermented milk group. The aorta and liver of fermented milk showed slighter medial calcification and necrosis and milder fat degeneration than did the cholesterol control group. Supplementation of fermented milk may have a helpful effect on hypercholesteremia and atherosclerosis.

방풍통성산이 고지방식이 유도 어린 비만 생쥐의 지방 축적에 미치는 영향 (Effect of Bangpungtongsungsan on Suppressing Fat Accumulation in High-fat Diet-Induced Young Obese Mice)

  • Objectives The aim of this study was to determine the inhibitory effects of Bangpungtongsungsan extract (BTS) on fat accumulation in high-fat diet-induced young obese mice. Methods The extract was administered to 3-week-old C57BL/6 male mice fed with a high-fat diet. The experimental groups were divided into a control group (Ctrl), high-fat diet group (HFDF), and BTS treated group after high fat diet feeding (BTST), with 10 mice assigned to each group. Lipid synthesis was observed to confirm the inhibition of fat synthesis. Changes in body weight, body fat percentage, and total cholesterol in the blood were observed to confirm weight control. Peroxisome proliferator-activated receptor gamma (PPAR-γ) and sterol regulatory element-binding protein (SREBP)-1 positivity was observed to confirm the inhibition of fat accumulation in liver tissue. Results Bangpungtongsungsan significantly inhibited lipid synthesis. Changes in body weight, body fat percentage, and total cholesterol in the blood were significantly lower in BTST rats than in HFDF rats. PPAR-γ and SREBP-1 positivity were significantly lower in BTST rats compared to HFDF rats. Conclusions This study confirms the potential of BTST to inhibit fat accumulation in obesity.

탄닌산(tannic acid) 첨가가 고지방 식이 흰쥐의 성장과 혈액학치 및 혈액화학치의 변화에 미치는 영향 (The Effect of Supplementary Feeding of Tannic acid on Growth and Hematological Changes in Rats Fed High Fat Diet)

  • This study was performed to investigate the growth rate, hematological and serological changes of the rats when they were fed with the high fat diets supplemented with or without the tannic acid for five weeks. Thirty-two Sprague-Dawley male rats(235.7\pm10.7g\;of\;body\;weight)$ were randomly divided into four groups, control group and three treatment groups(T1, T2 and T3). Rats in control group were fed with the high fat diet containing $15\%\;lard,\;1\%$ cholesterol and $0.5\%$ sodium cholate(wt/wt) which was modified from the formula of American Institute of Nutrition (AIN)-76 diet and rats in treatment groups were fed with above diet supplemented with $0.25\%(T1),\;0.5\%(T2)$ or $0.75\%(T3)$ of tannic acid(wt/wt), respectively. The supplementation of tannic acid(TA) did not affect the final body weight, gain of body weight and feed intake of rats in both control and treatment groups. The numbers of red blood cells, hemoglobin concentrations and hematocrit values in blood of rats showed no significant differences between control group and treatment groups. The glucose concentration and albumin/globulin(A/G) ratio of rats in treatment groups were slightly lower than that of control group without significance. The values of total protein, albumin and globulin showed no significant differences between control group and treatment groups. The values of total cholesterol, low density lipoprotein­cholesterol and atherogenic index in sera of rats in treatment groups were much lower than that of control group without significance. The values of triglycerides in sera of rats in T3 group were significantly lower than that of control group (p<0.05). The values of AST and ALT in sera of rats in T3 group were significantly lower than that of control group (p<0.05). Thus supplementation of tannic acid to high fat diet could be effective to reduce the serum lipid levels such as total cholesterol, high density lipoprotein-cholesterol and triglycerides which were regarded as to cause the cardiovascular diseases.

Onion Supplementation Inhibits Lipid Peroxidation and Leukocyte DNA Damage due to Oxidative Stress in High Fat-cholesterol Fed Male Rats

  • The aim of this study was to investigate effects of onion, red onion, or quercetin on plasma antioxidant vitamin, lipid peroxidation, and leukocyte DNA damage in rats fed a high fat-cholesterol diet. Forty SD male rats were assigned to normal control, high fat-cholesterol diet (HF), or HF+5% onion powder, HF+5% red onion powder, or HF+0.0l% quercetin. The HF diet resulted in significantly higher plasma lipid peroxidation which decreased with onion, red onion, or quercetin supplementation. Leukocyte DNA damage induced by HF diet decreased significantly in rats fed onion and red onion, while quercetin supplementation had no effect on preventing leukocyte DNA damage. $H_2O_2$ induced leukocyte DNA damage exhibited a highly significant negative correlation with plasma retinol and tocopherols. These results suggest that onion or red onion powder exerts a protective effect with regard to DNA damage in rats fed HF diet. However, 0.01% quercetin in pure form might not be effective at preventing DNA damage.

  • Effects of Bupleuri Radix, Paeonie Radix or Uncariae Ramulus et rncus on experimental yperlipidemia were studied. Hyperlipidemia was induced on male Wistar rats by feeding them on high holesterol diet for one week, as previously described by the authors. Blood lipid profile was verified on these ats by measuring total cholesterol(TC), triglyceride(TG), high density lipoprotein cholesterol(HDL) and low ensity lipoprotein cholesterol(LDL). Then, the rats were followed by feeding normal diet. At the same time, ethanol extracts of the three individual herb medicines were orally administered to the rats for 4 weeks and the parameters above mentioned were monitored. Methanol extract of 5Bupleuri radix reduced the TC value significantly at 2 week point and Paeoneae Radix reduced the TC value at 4 week point in compare to control group, suggesting the antihyperlipidemic effect of the two herbal medicines in vivo. The extract of Uncariae Ramulus et Uncus however did not show antihyperlipidemic effect in our experiment.

The Effect of Supplementation of Persimmon Leaf Extract on Lipid and Antioxidant Metabolism in Rats Fed a High-cholesterol Diet

  • Fruits and vegetables reportedly have a protective effect against hyperlipidemia and oxidative disease. Accordingly, this study aimed to investigate the lipid-lowering effect and antioxidative capacity of persimmon leaf extract (PLE) in rats fed a high-cholesterol diet. Male rats were fed a high-cholesterol (1% wt/wt) or high-cholesterol diet supplemented with Lovastatin (0.02% wt/wt) or PLE (0.2% wt/wt) for 5 weeks. The concentration of plasma total cholesterol was significantly lower in the PLE group than in the lovastatin group. However, the concentration of plasma HDL-cholesterol and the ratio of HDL-cholesterol/total-cholesterol (%) were significantly higher in the PLE group than in the control group. The PLE supplement also significantly lowered the contents of hepatic cholesterol and triglyceride. In comparing fecal sterol contents, the PLE group saw a significant increase of both neutral and acidic sterol compared to the other groups. The PLE supplement significantly lowered plasma GOT and GPT activity, which ave indices of hepatic toxicity. Plasma TBARS concentration was significantly lower in the PLE group than in the control group, while hepatic TBARS level was not significantly different between the groups. In a comparison of hepatic antioxidant parameters, SOD, catalase and GSH-Px activity were significantly higher in the PLE group than in the control group. However, the PLE supplement significantly towered antioxidant enzyme activity in the erythrocyte. Furthermore, these results suggest that supplementation of PLE promoted the excretion of fecal sterols, thereby leading to decreased absorption of dietary cholesterol. In addition, PLE may play an important role in regulating antioxidative capacities by altering SOD and ChT activity.

고콜레스테롤 식이에 있어 키토산 올리고당이 체내 콜레스테롤농도 및 항산화효소 활성에 미치는 영향 (Effect of Chitosan Oligosaccharides on Cholesterol Level and Antioxidant Enzyme Activities in Hypercholesterolemic Rat)

  • 본 연구는 고콜레스테롤을 급여한 흰쥐 에 키토산 올리고당의 첨가 수준을 달리한 식이가 흰쥐의 지방대사에 미치는 영향을 알아보고자 수행하였다. 체중이 174.7$\pm$6.3 g인 Sprague-Dawley종 수컷 흰쥐에게 고콜레스테롤혈증을 유발시키기 위하여 콜레스테롤을 식 이 무게의 0.5%를 급여하고 키토산 올리고당을 식이 무게의 1.0%와 2.0%로 하여 총 3군으로 나누어 4주간 사육하였다. 식이섭취량과 체중증가량은 식이에 의한 영향이 없었다. 혈청 중 총 콜레스테롤, 중성지방은 대조군에 비해 2.0% 키 토산 올리고당을 첨가한 군에서 유의적으로 그 농도가 크게 감소하였으며 HDL-콜레스테롤/총콜레스테롤 비는 키토산 올리고당 첨가에 의해 높아지는 경향을 보였다. 간 조직 중 TBARS 함량은 키토산 올리고당을 첨가하였을 경우 유의적으로 낮게 나타났으며 SOD와 catalase의 활성은 키토산 올리고당을 첨가한 군이 대조군에 비해 유의적으로 낮았다. 이상의 결과를 볼 때 키토산 올리고당은 체내 지방대사에 있어 총콜레스테롤과 LDL-콜레스테롤, 중성지방 및 TBARS 함량을 낮추고 HDL-콜레스테롤/총 콜레스테롤 비는 높이는 효과를 나타냈으며, 또한 생체내 고콜레스테롤에 의한 산화적 스트레스로부터 간을 보호하는 것을 알 수 있었다.

Antiobesity Effect of Major Korean Spices (Red Pepper Powder, Garlic and Ginger) in Rats Fed High Fat Diet

  • Commonly used spices in Korean cooking (red pepper powder, garlic, ginger) were evaluated for anti-obesity properties and effects on triglyceride (TG) and cholesterol in blood and adipose tissues in rats fed a high fat (20%) diet. SD rats were raised for four weeks on either a normal diet (ND, based on the AIN-93M diet), high fat diet (HFD, supplemented with 16% lard oil in the ND), or diets containing 5% red pepper powder (RPP), garlic or ginger. These spices significantly decreased weight gain compared to HFD, but garlic and ginger showed a greater effect on reducing weight gain than RPP. The weights of liver and epididymal and perirenal fat pads in garlic and ginger diet groups were lower than those of the HFD groups (p < 0.05). The garlic and ginger also decreased triglyceride and cholesterol contents in liver and epididymal and perirenal fat pad, reversing the higher levels seen in HFD. RPP, garlic and ginger supplemented diets were effective in lowering serum triglyceride and cholesterol levels (p< 0.05). These results indicated that garlic and ginger more effectively suppressed the effects of HFD on body fat gain and lipid values of adipose tissues and serum than RPP.

나이가 다른 흰쥐에서 식이내 지방수준과 식이횟수가 체내 지방대사에 미치는 영향 (Effect of Age on Liquid Metabolism in Rats Fed Diets with Different Fat Lieval and in Meal Fed Rats)

  • This study was designed to investigate the effect of age on the lipid metabolism in the rats fed different diets. In experiment A male Wistar rats of 5 weeks of age and of 32 weeks of age were divided into low fat diet groups and high fat-cholesterol groups. The rats were sacrificed 1 week, 2 weeks and 4 weeks after experiment begins. Also after 4 weeks. the rats in low and high fat diet groups were tube-fed 500mg of choelsterol and were sacrified 3 days later. In experiment b, male Wistar rats of 4 weeks of age and of 6 months of age were divided into 2 groups of butter and cron oil groups. And then eachgroup were divided into 2 subgroups ; meal feeding and nibbling groups . Each diet was fed for 4 weeks. In experiment A, age of the rats and experimental diets did not affect the serum cholesterol and triglycerides concentrations were higher in rats fed high fat cholesterol diet than in those fed low-fat or high -fat diets, but age of the animals had no effect on liver lipid content. The weight and cholesterol content of epidymal fat pad, however were higher in adult rats than in young ones regardless of the diets fed. When the rats were challenged with 500mg cholesterol, the rates of increase in serum and hepatic cholesterol level were higher in adult rats compared to young rats regardless of the diets . On the other hand, the rate of increase of small intestinal cholesterol content was lower in adult rats than in young rats. In experiment B, serum cholesterol and triglyceride content were relatively higher in young rats than adult ones. Stored body lipid was higher in adult rats, as judged by epididymal fat pad weight and total carcass lipid. Meal frequency and the kinds of fat in the diet did not affect the serum choelstero concentration . The serum triglyceride levels. however, was higher in butter fed rats thancron oil fed ones. The cholesterol content of live rand epididymal fat pad was lower in butter fed groups than corn oil groups for both young and adult rats, but there was no difference in liver triglycerides livel.
