Effect of Ethyl Alcohol Fraction of Cervus nippon on Mouse T-Lymphocyte (녹용 에탄올 분획이 생쥐의 T-Lymphocyte에 미치는 영향)

  • In this study, the effect of 70% ethyl alcohol fraction of Cervus nippon(CN-E) on mouse T-lymphocyte was investigated in vivo. The administration of CN-E(100 mg/kg) enhanced the proliferation of thymocytes, the population of $CD4^+CD8^-$ single-positive cells and the production of $interferon-{\gamma}$ in thymocytes and splenocytes. The administration of CN-E did not induce DNA fragmentation and reduce mitochondrial transmembrane potential in thymocytes. These results indicate that the CN-E contams a stimulative component on the proliferation of thymocytes, the population of $T_H$ cells and the production of $interferon-{\gamma}$ in T-lymphocytes.

Effects of Sipyukmiyukieum on DNA Synthesis, cAMP Synthesis and MHC-class II Expression of FRTL-5 Thyroid Cells (십육미류기음(十六味流氣飮)이 FRTL-5 갑상선 세포의 DNA와 cAMP의 합성 및 MHC-class II의 발현에 미치는 영향)

  • Objective: Graves' disease encompasses hyperthyroidism and diffused goiter associated with auto-antibodies to the thyroid stimulating hormone(TSH) receptors. In clinical environment, treatments of Graves' disease have many side effects such as recurrence and hypothyroidism. We've studied the effects of Sipyukmiyukieum on DNA synthesis, cAMP synthesis, and MHC-class II expression of FRTL-5 thyroid cells were studied. Methods: DNA synthesis was investigated by using BrdU staining and cAMP synthesis by ELISA kit, and expression of $interferon-{\gamma}$ activated MHC class II by Flow cytometer. Results: After introduction of Sipyukmiyukieum, significant inhibition of DNA synthesis. cAMP synthesis, and expression of $interferon-{\gamma}$ activated MHC class II of FRTL-5 thyroid cells was observed. Conclusions: Judging from these results, Sipyukmiyukieum has potential as a potent herbal treatment for inhibiting the enlargement of goiter, synthesis of abnormal thyroidal hormones, and autoimmuine responses of Graves' disease.

  • A rapid assay for interferon based on reduction of cytopathic effect was developed with HEp-2 and Vesicular Stomatitis Virus. The number of manipulations and the lengths of the various incubation steps were reduced to minimum. The assay is simple to perform and can be completed with 22-24 hr. Moreover, it was precise method than CPE-reading method.

The Effect of Lipopolysaccharide on Noxa Expression Is Mediated through IRF1, 3, and 7

  • Lipopolysaccharide (LPS), a component of the cell wall of gram-negative bacteria, elicits the secretion of cytokines, such as interferons, that stimulate the host defense system. Previously, we demonstrated that interferons induce interferon regulatory factors (IRFs) 1, 3, and 7, which regulate the transcription of Noxa and alter the expression profiles of Bcl-2 family proteins in tumors. However, the immediate consequences of LPS stimulation on Noxa and BH3 expression in tumor cells remain uncharacterized. In this study, we determined that LPS induced Noxa expression in CT26 cells. Furthermore, studies in HCT116 parental and HCT116 p53-deficient cells revealed that LPS-mediated Noxa was independent of p53. Meanwhile, IRF1, 3, and 7 in CT26, HCT116 parental, and HT116 p53-deficient cells were upregulated by LPS stimulation, suggesting that LPS induces the expression of these IRFs in a p53-independent manner. The responsiveness of IRF1, 3, 4, and 7 binding to the Noxa promoter region to LPS indicated that IRF1, 3, and 7 activated Noxa expression, whereas IRF4 repressed Noxa expression. Together, these results suggest that LPS directly affects Noxa expression in tumor cells through IRFs, implicating that it may contribute to LPS-induced tumor regression.

  • To search for immunoactive natural products exerting anti-inflammatory activity, we have evaluated the effects of the ethanol extracts of Rubus coreanus Miq. (ERC) on lipopolysaccharide-induced nitric oxide (NO), tumor necrosis $factor-{\alpha}\;(TNF-{\alpha})$, and $Interferon-{\gamma}\;(IFN-{\gamma})$ production by RAW 264.7 macrophage cell line. Our data indicate that this extract is a potent inhibitor of NO production and it also significantly decreased $IFN-{\gamma}\;and\;TNF-{\alpha}$ production. Consistent with these results, the protein level of inducible Nitric Oxide Synthase (iNOS) and cyclooxygenase-2 (COX-2) was inhibited by ethanol extracts of ERC in a dose-dependent manner. These results suggest that ERC may exert anti-inflammatory and analgesic effects possibly by suppressing the inducible NO synthase and COX-2 expressions.

  • Falun Gong (FG) is an advanced system of cultivation and practice, which is beneficial for both mind and body. In this study we investigated the effects of FG on the production of cytokines in FG practitioner (FGP). To study whether plasma cytokines levels were affected by FG, their levels were analyzed. The amount of $interferon-{\gamma}$ $(IFN-{\gamma})$, interleukin (IL)-2, IL-4 and IL-6 (2.5-fold for $IFN-{\gamma}$, 1.2-fold for IL-2, 2.1-fold for IL-4 and 2.5-fold for IL-6, respectively) were significantly higher in the FGP group than normal group (P<0.05). Peripheral blood mononuclear cells obtained from normal healthy control and FGP were cultured for 24 h in the presence or absence of lipopolysaccharide. The amount of $IFN-{\gamma}$, IL-2, IL-4 and IL-6 in culture supernatant was quantified. However, there were no significant differences in the level of the same cytokines between the normal and FGP group. These data suggest that FG cultivation may contribute to immune-enhancement in vivo.

Inhibitive Effect of KH against Toxicity Induced by 5-Flurouracil(FU) (화학요법제 5-flurouracil의 조혈 및 면역독성 억제를 위한 조성물 개발 연구)

  • The objective on this study was to investigate effect of KH, which was composed of 9 kinds of oriental herbs tonifing the blood, against toxicity of 5-flurouracil(FU) through animal study. Reduction of WBC and platelet after treating S-FU was significantly recovered by KH. KH also rehabilitated immune gene expression of interleukin-2(IL2) and $Interferon-{\gamma}\;(IFN-{\gamma})$ by RT-PCR. In addition, in situ hybridization of spleen showed that KH treatment increased mRNA expression of IL2. In addition to the immune action, antitumor activity on 5-FU was not affected by KH treatment. In conclusion, our study demonstrated that KH alleviated damage of hematopoiesis system induced by f-FU without toss of antitumor activity.

  • Drug-induced toxic hepatitis is a relatively common hepatic disease in children, and it is usually self-limiting upon cessation of the offending drugs. Antituberculous drugs are well known for inducing hepatitis. Some cases of drug-induced hepatitis with autoimmune features have been reported; in these cases, the offending drugs were usually methyldopa, nitrofurantoin, minocycline, and interferon. The authors report the first case in Korea of drug-induced autoimmune hepatitis associated with thyroiditis and multiple autoantibodies that was induced by the antituberculous drugs isoniazid and rifampin.

Gamma Irradiation-reduced IFN-γ Expression, STAT1 Signals, and Cell-mediated Immunity

  • The signal transducer and activator of transcription (STAT)1 is a cytoplasmic-transcription factor that is phosphorylated by Janus kinases (Jak) in response to interferon $\gamma$ (IFN-$\gamma$). The phosphorylated STAT1 translocates to the nucleus, where it turns on specific sets of IFN-$\gamma$-inducible genes, such as the interferon regulatory factor (IRF)-1. We show here that gamma irradiation reduces the IFN-$\gamma$ mRNA expression. The inhibition of the STAT1 phosphorylation and the IRF-1 expression by gamma irradiation was also observed. In contrast, the mRNA levels of IL-5 and transcription factor GATA-3 were slightly induced by gamma irradiation when compared to the non-irradiated sample. Furthermore, we detected the inhibition of cell-mediated immunity by gamma irradiation in the allogenic-mixed lymphocytes' reaction (MLR). These results postulate that gamma irradiation induces the polarized-Th2 response and interferes with STAT1 signals, thereby causing the immunosuppression of the Th1 response.

A Case Report of HBeAg Seroconversion in Chronic Viral Hepatitis B (만성 바이러스성 B형 간염 환자에서 HBeAg이 혈청전환된 1례)

  • Objectives: The purpose of this case study is to report the efficacy and safety of treatment with Korean medicine of a patient with HBeAg-positive chronic viral hepatitis B.Methods: The patient took Korean medicine (mainly Injinchunggan-tang-gamibang) from July 20th, 2010, to March 14th, 2016, without any antiviral or interferon therapy. Changes to laboratory records, abdomen ultrasonography, and clinical symptoms were reviewed.Results: The laboratory records showed that AST, ALT, and HBV DNA had decreased to normal ranges, and HBeAg showed seroconversion. Clinical symptoms also improved after taking Korean medicine.Conclusion: The results suggest that treatment with Korean medicine and without antiviral or interferon therapy could be effective for HBeAg-positive chronic hepatitis B.
