The Effects of Endocrine Disrupters on Reproduction and Development of Wild Animals (내분비 장애물질, 환경호르몬이 야생동물의 생식과 발생에 미치는 영향)

  • Recently public concerns about the endocrine disrupting chemicals (EDC) have resulted in the increased legistrative and regulatory attentions in many countries. Endocrine disrupter is an exogeneous chemical interfering the synthesis, transport binding, action or elimination of natural hormones in the body, which are responsible for the maintenance of homeostasis, reproduction, development and/or behavior. Reported possible harmful effects on human and animal lives, possible developmental anomalities, reproductive malfunctions and behaviors, and the ways of EDC accumulation in animal kingdom are reviewed. The current scientific papers and knowledges on the global contamination of EDCs demonstrates the conclusive data that EDCs have the potentials to cause the calamitous extermination of human and animal species by the global contamination.

Isolation and Cultural Characteristics of Styrene Dimer [Endocrine Disrupter] Biodegrading Microorganism (Styrene dimer [환경호르몬 물질] 분해균주의 분리 및 배양특성)

  • We examined the culture conditions and degrading characteristics of styrene dimer (endocrine disrupter) using microorganism. The isolated microbe were consisted of 3 kinds of strain. The strains were identified to Pseudomonas sp. and Klebsiella pneumoniae by API 20E kit, but one was not identified. Single strain was not grown on the C-medium containing styrene dimer. However the complex strain YH3 could grow and we confirmed it by the broth color and O.D$_{660nm}$ (optical density 660 nm). The optimal culture conditions of complex strain YH3 were 35$^{\circ}C$, 1,000 ppm (v/v) of styrene dimer and pH 7.0, respectively. In tolerance test against the organic solvents, the complex strain YH3 could grow above log P=3.1, and could degrade ethyl benzene and 2,4-D, one kind of herbicide. As a result of TLC (Thin Layer Chromatography) analysis, we confirmed that the metabolite of styrene dimer was created by YH3 after 5th day, but not at control samples.

Responses in Hepatic Xenobiotic Metabolizing Enzymes and Sex Hormones of Yellowfin Goby Acanthogobius flavimanus in Nakdong Estuary (낙동강 하구에서 채집한 문절망둑 Acanthogobius flavimanus의 간장 약물대사효소계와 성호르몬 농도)

  • To assess effects of contaminants on fish in Nakdong river, feral yellowfin goby Acanthogobius flavimanus were caugt in two different sites and its hepatic monooxygenase enzyme, including cytochrome P450 (CYP), NADPH-cytochrome P450 reductase (P450R), NADH-cytochrome b5 reductase (b5R), ethoxyresorufin deethylase (EROD), glutathione S-transferase (GST) were quantitatively determined. Gonadosomatic index (GSI), hepatosomatic index (HSI) and three sex steroid hormone (17$\beta$-estradiol, E2; testosterone, T; 11-ketotestosterone, 11-KT) levels of the fish were also investigated. HSI of fish from polluted site (site 1) were significantly higher than that of unpolluted site (site 2), but GSI levels were significantly lower in polluted site. No significant differences in plasma 11-KT and T levels were observed in two sites surveyed. E2 level was, however, significantly (p<0.05) higher in female fish from site 1 than site 2. In addition, hepatic EROD activity and CYP level of site 1 fish were lower than those of site 2 fish, whereas relatively high levels of P450R, b5R and GST activities were found in site 1. The results imply that yellowfin goby, especially female fish in Nakdong river estuary are affected from contaminants disrupting sex steroid hormone system.

  • A crude crystal of phytoecdysone was obtained 355 mg per Kg dry radix of A. japonica, and it was identified to $\beta$-ecdysone by TLC. Feeding silkworms on mulberry leaves sprayed with the extract, mounting time shortened and pupation ratio improved. The feeding effects of the extract was differed according to the rearing seasons, spring and summer. In spring season, only mounting time shortened without any difference in pupation ratio from the control. In summer season, not only mounting time shortened, but also pupation ratio remarkably advanced.

  • Stress has been linked to the pathophysiology and pathogenesis of various psychiatric illnesses. Over the past few years, our understanding of the brain and neuroendocrine systems that are linked to stress responses has increased enormously. This article reviews a series of animal and human studies to understand what are the central pathways by which stress is perceived, processed, and transduced into a neuroendocrine response. We focus on the limbic-hypothalamic-pituitary-adrenal(LHPA) axis and several neurotransmitter systems such as norepinephrine, CRF, serotonin, acetylcholine, and dopamine. LHPA stress circuit is a complex system with multiple control mechanisms which are altered in pathological states. CRF and related peptides in the central nervous system appear to enhance behavioral responses to stressors. Norepinephrine systems are also activated by stressors and cause the release of catecholamines from the autonomic nervous system. CRF-norepinephrine interaction makes a feed-forward system which may be important for an organism to mobilize not only the pituitary system but also the central nervous system, in response to environmental challenges. The interactions among several neurotransmitters and endocrine systems appear to play key roles in mediating various behavioral and psychological stress responses involving abnormal responses to stressors such as anxiety and affective disorders.

  • The recombinant fluorescent chinese hamster ovary (CHO) cell line was developed and optimized through this study for biomonitoring system. This cell line, called KFC-A10, contains recombinant plasmid(pKCFG) constructed in this study for detecting toxic conditions (Mitomicyn C, EDCs, ${\gamma}-ray$, etc.). It is known that c-Fos is involved in proliferation and differentiation of the signal transduction and overexpression of this gene can lead cell to death under the toxic conditions including apoptosis status. Therefore, pKCFG which has the c-fosSRE::GFP is induced by toxic chemicals, especially DNA damage agents and apoptotic chemicals, and produces green fluorescence protein(GFP) under these toxic conditions. Through the characterization of KFC-A10 using fluorescent assays of GFP, it was shown that KFC-A10 cell line had a manifest GFP expression pattern due to various toxicants especially mitomycin C, ${\gamma}-ray$ and bisphenol A. Therefore this study proved the possibility of using GFP as a reporter for detecting various toxicants

  • This study was carried out effect of red ginseng extract(1.0g/kg) on organotin compounds (10, 20 and 40 mg/kg) which poisons against some organs like thyroid gland, liver, kindey, testis, ovary, serum immuinty and sex hormone activity of rats were examinned by gastric tubing for 3 weeks. The weight of each organ in TBTO treated group were significantly increased other organs which excepted kinedy in males and only liver in females.(p<0.05, p<0.01). In case of Immunity activity of each sex, IgM level was small change comparsion with that of control group in all sex. but IgG level was significantly decreased females rather than males comparsion with that of control group.(p<0.05, p<0.01) In case of sex hormone activity, the testosterone activity of males and the estradiol activity of females were significantly decreased rather than the control group. on the other hand, red singsong treated group was only significantly increased estradiol activity.( p<0.05, p<0.01)

  • 17β-estradiol (E2) is a natural hormone secreted by ovary, and continuously discharged from household and livestock wastewater into aquatic environment. Due to its strong estrogenic activity, it has adverse effects on development and reproduction in crustacean as an endocrine disrupting chemical. Although ecdysteroid signaling pathway play a key role in development in crustacean, little information on transcriptional modulation of ecdysteroid-related genes in response to E2 is available in small crustacean. Here, we investigated the acute toxicity of E2 to obtain 24-h LCx values in the brackish water flea Diaphanosoma celebensis. Time-dependent expression patterns of seven ecdysteroid pathway - related genes (CYP314a1, EcRA, EcRB, USP, ERR, Vtg, VtgR) were further examined using quantitative real time reverse transcriptase polymerase chain reaction (qRT-PCR). As results, 24-h LC50 and LC10 values were 9.581 mg/l and 4.842 mg/l, respectively. The mRNA expression of CYP314a1, EcRA, USP, VtgR was significantly up-regulated at 12 or 24 h after exposure to E2. These findings indicate that E2 can affect their molting and reproduction by modulating the expression of ecdysteroid pathway - related in D. celebensis. This study will be useful for better understanding of molecular mode of action of endocrine disrupting chemicals on molting process in small crustacean.

  • Melatonin Is a multifunctional hormone secreted from the pineal gland in the middle of cerebrum and cerebellum. Its synthesis and release reflect photopedod;Photopedod is a yearly predictable ambient factor that most animals utilize as an environmental cue for maximum survival. Hamsters maintaln reproductive activity in summer during which day length exceeds night time. Upon the advent of autumnal equinox they undergo gonadal regression. The photoperiodic effects are prevented by removal of the pineal gland and restored by the timed repiacument of melatonin. The results suggest that melatonin constitutes part of control mechanism whereby environmental information is transduced to neuroendocrine signal responsIble for the functional integrity of the reproductive system. From the studies for the action site of melatonin following the treatment of photopedod or melatonin in the lesion of a spedflc portion of hypothalamus, suprachiasmatic nuclei and pars tuberalis are shown to be a consensus site for melatonIn. The action of melatonin. In the regulation of reproduction is largely unknown. It is mainly due to the lack of acute effect of melatonin on gonadotropin secretion. However, reduction of the gonadotropln release and augmentation of the hypothalamic gonadotropin-releasing hormone (GnRH) content by long-term treatment of melatonln Indicate that constant presence of melatonln may partidpate in the regulation of sexual activity via the GnRH neuronal system. The action mechanism by which melatonin exerts Its effect on GnRH neuron needs to be eluddated. The inability of opiold analogues to affect the reproductive hormones in sexually regressed animals by inhibftory photopedod and melatonin suggests that the opioldergic neuron may be a prime intervening mediator. Recent cloning of melatonin receptor will contribute to investigate its anatomical Identification and the action mechanism of melatonin on target tissues at the molecular level.

Changes of Phytohormones in Rice and Tobacco Protoplasts under Different Cultural Conditions (벼와 담배의 원형질체 배양조건에 따른 식물호르몬의 변화)

  • This study was carried out to establish simple and easy methods to judge the survival, senescence and death of the protoplasts in culture system by identifying the marker substance related to metabolic status of the cells. When rice and tobacco protoplasts were cultured in MS and KM-8P media containing 2,4-D or coconut milk ABA decreased especially in the media containing coconut milk, but GA$_3$, IAA and zeatin increased as the cultures progressed. The decrease of ABA and increase of zeatin was especially remarkable. When the supraoptimal amount of osmoticum (mannitol) was added to the culture media ABA decreased after a momentary increase, but other growth hormones slowly increased as the concentration of the osmoticum increased. Contents of individual hormones were contrasted when protoplasts rice and tobacco were cultured on the same medium containing 10mM super mine or NaCl. Tobacco protoplasts were more sensitive to NaCl stress and stopped protoplast division at the late stage of culture. Protoplast viability decreased greatly in 48 hours when the protoplast were at 32$^{\circ}C$ on a medium lacking several components. ABA content increased up to 10 days from incubation in negative proportion to the protoplast viability. On the other hand contents of other growth hormones, especially zeatin, decreased. The present results clearly showed that the contents of individual growth hormones in the plant protoplasts in culture varied sensitively in response to environmental factors that they are faced with. This indicates that the physiological states of the protoplast, such as survival, senescence or death can be simply judged based on the quantitative analysis of those hormones by ELISA.
