Opuntia ficus-indica is widely used as folk medicine for burned wound, edema and indigestion, etc. After the adaptation of rats for seven days, OF (200mg/kg) was administered into rats intraperitoneally every day for two weeks. On the 14th day, $3.3m\ell$ of $CCl_4$ (50% $CCl_4$ : Olive oil = 1 : 1) was treated to rats and on the 15th day, they were operated. We measured the levels of total cholesterol, HDL-cholesterol, LDL-cholesterol, total lipid and triglyceride (TG) in serum and Malondialdehyde (MDA) in liver tissue. Opuntia ficus-indica and $CCl_4$-treated group (OFC) significantly decreased lipid peroxidation by 32.6% compared to $CCl_4$-treated abnomal group (CTA). Total lipid and TC of OFC administered group were lower than those of CTA group. Cholesterol levels showed significant effect in OFC group. Total cholesterol and LDL-cholesterol in OFC administered group were decreased by 12.7% and 17.4%, respectively, compared to those of CTA group. HDL-cholesterol in OFC administration group was increased by 19.7% compared to CTA group. This finding suggests that lipid peroxidation was inhibited in the significant level when $CCl_4$-induced rats were treated with OF. Based on this analysis, further study to identify the active components in this sample is warranted.

The purpose of this study was to investigate the effects of vitamin C supplementation and yoga practice on total antioxidant status (TEAC), lipid peroxidation, and blood lipid profiles of middle-aged women. Thirty two women ($53.34{\pm}5.08$ years old) in B city volunteered and the signed consents were received before the study. Experimental groups were normal (n=8), vitamin C intake group (n=8), yoga group (n=8), and vitamin C intake with yoga group (n=8). Total experimental period was 12 weeks. Yoga group practiced yoga 3 times a week for 12 weeks with intensity of RPE $13{\sim}15$. Each practice was 60 minutes. Vt. C group was instructed to take 1 g of Vt. C every day after dinner meal. Changes in TEAC after 12 weeks were 28.23% for the normal group, 30% for Vt. C group, 26.58% for yoga group, and 43.66% for Vt. C with yoga group; the increases in TEAC among groups were not significantly different. Serum malondialehyde (MDA) concentration of the normal group increased by 10% during 12 weeks of experiment while that for the Vt. C, yoga, and Vt. C with yoga group were significantly decreased by 25%, 32.14%, and 33.87%, respectively (p<0.05) compared with the normal group. Neither yoga, Vt. C supplementation nor combined program for 12 weeks were effective enough to change the serum lipid profiles compared with the normal group. In conclusion, Vitamin C supplementation or regular yoga practice seems to have health promoting effects of retarding the oxidative stress by decreasing lipid peroxidation in middle-aged woman.
This study was performed to investigate the ameliorating effect of a hangover beverage mixture (SBJ) that contains Dendropanax morbifera Lev. and several medicinal plant extracts, on hepatoprotection and alcohol-metabolizing enzymes in alcohol-induced hangover in both in vitro and in vivo models. In human hepatoma cell line, HepG2, 300 mM of ethanol-induced hepatotoxicity was significantly improved by pretreatment of SBJ by dose-dependent manner. In the in vivo study, administration of alcohol to rats raised to the concentration of blood alcohol and lactate dehydrogenase (LDH). Blood alcohol and LDH levels in SBJ-treated rats significantly decreased at 0.5 h and 8 h after acute ethanol administration (40%, 4.6 g/kg body weight) as compared to alcohol-treated rats. Hepatic alcohol dehydrogenase (ADH) and aldehyde dehydrogenase (ALDH) activity were significantly higher in SBJ-treated rats than in alcohol-treated rats. SBJ supplementation reduced formation of malondialdehyde (MDA), and inhibited reductions of hepatic superoxide dismutase (SOD), hepatic glutathione (GSH), glutathione-S-transferase (GST), glutathione reductase (GR) and glutathione peroxidase (GPx) levels, compared with rats administered alcohol. Plasma catalase (CAT), aspartate aminotransferase (AST) and alanine aminotransferase (ALT) levels showed unaltered resulted in all experimental groups compared with the control group. These results suggest that SBJ exhibit hepatoprotective properties by enhancing ADH, ALDH activity and stimulating the antioxidant defense system in alcohol-induced hangover.
The present study was conducted to investigate the effect of whole wheat bread with added Lindera obtusiloba (LO) and Capsosiphon fulvescens (CF) ethanol extracts on serum glucose and lipid levels in Sprague Dawley rats. Rats were divided into five groups depending on the diet administered: normal bread (NC), whole wheat bread (W), whole wheat bread with LO leaves extract (WL), whole wheat bread with CF extract (WC), and whole wheat bread with freeze-drying CF (WDC). After 4 weeks of consuming the experimental diet, the blood glucose level and hemoglobin A1c contents were found to be significantly lower in the W, WL, WC, and WDC groups than in the NC group. The high-density lipoprotein-cholesterol levels increased in the WL group when compared to those in the NC group and triglycerides levels decreased in all wheat groups compared to those in the NC group. These results suggest that wheat breads containing LO and CF extracts are effective for preventing hypercholesterolemia and obesity.
This study was to investigate the effect of long-term high intensity endurance training on the activation of antioxidation enzyme activity, lipid peroxidation and lipoprotein metabolism. 15 subjects were divided into, endurance exercise + antioxidation Vitamin supplement(n=5), endurance exercise(n=5), and the control(n=5) groups. The endurance exercise groups(endurance exercise + antioxidation Vitamin supplement and endurance exercise) had 12 week of endurance exercise program. The antioxidation Vitamin supplement group was taken a Vitamin C tablet with 1000mg/day and Vitamin E tablet with 671.14mg/day right after lunch. The results obtained from this study were as follows; 1. Looking at the changes of SOD, Endurance exercise+antioxidation Vitamin supplement group and endurance exercise groups showed the significantly greater decrease in the activation of SOD after 12 weeks of all-out exercise. 2. Looking at the changes of CAT, Endurance exercise+antioxidation Vitamin supplement group revealed subjects tended to increase CAT after all-out exercise although statistically non-significant. Endurance exercise+antioxidation Vitamin supplement group showed the significantly greater increase in the activation of CAT after 12 weeks treatment for all-out exercise. 3. Looking at the changes of GPX, Endurance exercise+antioxidation Vitamin supplement group revealed subjects tended to increase GPX for the rest and after all-out exercise although statistically non-significant. Endurance exercise+antioxidation Vitamin supplement group showed the significantly greater increase in the activation of GPX after 12 weeks treatment for all-out exercise. 4. The MDA change showed the significant decrease after 6 weeks, after 12 weeks for the all-out exercise of Endurance exercise + antioxidation Vitamin supplement group. 5. There was non-significant change in lipoprotein metabolism for the rest and after all-out exercise.

Effect of Korean red ginseng (KRG) on the level of serum and liver lipids and lipid peroxidation was investigated in the rats fed high fat diet. Content of serum total cholesterol was significantly decreased (P<0.05) in KRG I group and KRG II group. Content of HDL-cholesterol was significantly increased by 69.75% and 39.15% in KRG I and KRG II group compared to control group, respectively. Atherogenic index (hi) was also significantly decreased by 74.76% and 37.38% in KRG I and KRG II groups compared to control group, respectively. Serum triglyceride content was significantly decreased (p<0.05) in only KRG II group. Antioxidative activity of KRG on the lipid peroxidation of serum and tissues in rats was also studied in vivo by measuring the formation of thiobarbituric acid reactive substances (TBARS). Contents of TBARS in the serum of both KRG groups were significantly decreased (p<0.05) and that of nonheme iron in serum was significantly increased (p<0.05) in a dose-dependent manner, which suggested that lipid peroxidation contents are inversely correlated with serum nonheme iron content. Content of TBARS in liver was significantly decreased (p<0.05) in KRG I and KRG II groups, without any influence in other tissues. Content of TBIARS in liver microsomal fractions stimulated by Fe$^{2+}$/ascorbate was significantly decreased (p<0.05) in KRG I and KRG II groups, whereas this observation did not occur in liver mitochondrial fractions. When the effect of KRG on TBARS content in the liver fractions of homogenates, microsomes, and mitochodria stimulated by Fe$^{2+}$/ascorbate was tested in vitro experimental model, TBARS of liver three fractions was significantly decreased at 6 mg/mL KRG compared with those of control. These results suggested that KRG powder have hypocholesterolemic effect as well as antioxidative effect in the serum and liver of the rats fed high fat diet.

Purpose: Sprouts of evening primrose (Oenothera laciniata, OL) were reported to have high contents of flavonoids and potent antioxidant activity. This study examined the antioxidant and antiobesity activities of OL sprouts to determine if they could be a natural health-beneficial resource preventing obesity and oxidative stress. Methods: OL sprouts were extracted with 50% ethanol, evaporated, and lyophilized (OLE). The in vitro antioxidant activity of OLE was examined using four different tests. The antiobesity activity and in vivo antioxidant activity from OLE consumption were examined using high fat diet-induced obese (DIO) C57BL/6 mice. Results: The IC50 for the 2,2-diphenyl-1-picryl-hydrazyl (DPPH) radical scavenging and superoxide dismutase (SOD)-like activities of OLE were 26.2 ㎍/mL and 327.6 ㎍/mL, respectively. OLE exhibited the ferric reducing antioxidant power (FRAP) activity of 56.7 ㎍ ascorbic acid eq./mL at 100 ㎍/mL, and an increased glutathione level by 65.1% at 200 ㎍/mL compared to the control in the hUC-MSC stem cells. In an animal study, oral treatment with 50 mg or 100 mg of OLE/kg body weight for 14 weeks reduced the body weight gain, visceral fat content, fat cell size, blood leptin, and triglyceride levels, as well as the atherogenic index compared to the high fat diet control group (HFC) (p < 0.05). The blood malondialdehyde (MDA) level and the catalase and SOD-1 activities in adipose tissue were reduced significantly by the OLE treatment compared to HFC as well (p < 0.05). In epididymal adipose tissue, the OLE treatment reduced the mRNA expression of leptin, PPAR-γ and FAS significantly (p < 0.05) compared to HFC while it increased adiponectin expression (p < 0.05). Conclusion: OLE consumption has potent antioxidant and antiobesity activities via the suppression of oxidative stress and lipogenesis in DIO mice. Therefore, OLE could be a good candidate as a natural resource to develop functional food products that prevent obesity and oxidative stress.

We performed a randomized placebo-controlled trial to determine whether or not Ishige okamurae extract supplements modulate blood glucose and antioxidant systems in type 2 diabetic patients. A total of 46 patients were randomized to either an Ishige okamurae extract group or a placebo group. The patients consumed either 1,600 mg of Ishige okamurae extract or cornstarch supplement per day for 10 weeks. The lifestyle factors and dietary intake of patients were not altered during the 10 week trial period. After 10 weeks, the fasting blood glucose level was slightly decreased in the Ishige okamurae extract group, but a significant decrease was not observed. Also, glycosylated hemoglobin was significantly (p<0.01) decreased. Especially, low-glycosylated hemoglobin ($7.12{\pm}0.38%$ to $6.56{\pm}0.53%$) was significantly decreased compared to high-glycosylated hemoglobin ($8.65{\pm}0.92%$ to $8.60{\pm}0.85%$) in that group. The superoxide dismutase, catalase, and glutathione peroxidase levels were increased in the Ishige okamurae extract group compared to the placebo group. The increase of these enzymes was associated with the decrease of MDA concentration in the Ishige okamurae extract group, but a significant decrease was not observed. The Ishige okamurae extract supplement showed no adverse effects on liver and kidney functions. Findings from this study suggest that an Ishige okamurae extract supplement can help blood glucose status in type 2 diabetic patients without adverse effects.
Hepatoprotective effects of corn gluten hydrolysates (CGH) were investigated in rats orally treated with ethanol (30%(v/v), 3 g/kg body weight/day) for 4 weeks. Six-week old Sprague-Dawley male rats were divided into four dietary groups: normal diet (N), alcohol diet (E), E+CGH 1% diet (CGH-1%), and E+CGH 3% diet (CGH-3%). Body weights and liver indices were not significantly different among the four groups. However, food intakes were lower in the CGH groups than in the normal group (p<0.05). The administration of CGH significantly reduced serum alkaline phosphatase activity by 30% compared to the alcohol diet group. Among the antioxidative enzymes assessed, catalase activity was significantly decreased by 79% in the CGH diet groups compared to the alcohol diet group. In comparison to the alcohol-treated group, aldehyde dehydrogenase activity was increased by 20%, while microsomal ethanol oxidizing system activity was decreased by 20% in the CGH-treated groups. Furthermore, the area under the curve of the blood acetaldehyde concentration versus time profile after the administration of ethanol was significantly lower for the CGH rats than for the ethanol or asparaginic acid treated groups. Thus, CGH seems to offer beneficial effects by protecting against ethanol-induced hepatotoxicity by improving the acetaldehyde-related metabolizing system.
