Effects of Panax ginseng on Alcohol Detoxification (인삼의 알콜해독 효과)

  • To assess the effect of Panax ginseng on the detoxification of ethanol. we examined its effect on blood ethanol clearance in both man and experimental animals and on the rate of ethanol oxidation to carbon dioxide in experimental animals. Fourteen healthy male volunteers were subject to studies. The blood alcohol level in the test group receiving ginseng extract (3g/kg b.w.) along with alcohol (70g/65kg b.w.) was about $35\%$ lower than their control levels at 40 min after ethanol intake. When the blood alcohol level was compared on individual bases. blood alcohol concentrations in 10 subjects ranged from 32 to $51\%$ lower than their control values. The remaining 4 subjects appeared to have a high tolerance level. In experimental animals. the blood alcohol clearance was also much faster in test animals receiving ginseng along with ethanol. The rate of ethanol elimination was determined by the amount of $^{14}CO_2$ in exhaled air following the administration of [$^{14}C$] ethanol. During the first 7 1/4 hr (Phase I) after the ethanol administration. the $CO_2$ output was greater in test animals receving ginseng along with ethanol. whereas from beyond 7 1/4 hr to the near end (Phase II). the $CO_2$ output in control animals was over twice that in test animals. The present studies clearly demonstrate that ginseng promotes the overall metabolism of ethanol. resulting in an enhanced blood alcohol clearance and alcohol elimination.

  • 에탄올에 의한 급성독성은 에탄올, 아세트알데히드 및 에탄을 대사산물의 변형생성물질 등에 기인하므로 알콜 섭취 후 혈중 에탄을 농도 및 아세트알데히드의 농도를 낮추는 것은 음주에 의한 급성 중독상태에 머무는 시간을 단축시키는데 중요하다. 본 실험에서는 Bacillus subtilis natto sp.를 식물 추출액을 배지로 하여 배양한 후 단백 분해효소로 고분자물질을 절단하여 얻은 발효액인 바이오짐(상품명: Biozyme)을 주성분으로 한 '비지니스(조선무약합자회사)' 의 인체 혈중 알콜대사 촉진작용을 검색하였으며, 비지니스 및 바이오짐이 랫드의 에탄을 대사에 미치는 효과를 검토하여 다음과 같은 결과를 얻었다. 인체 혈중 알콜대사 촉진작용을 검색하기 위해 알콜 섭취 전과 후의 혈중 에탄올 농도를 비교하였는데, 대조군에 비해 비지니스 투여군이 30분 후부터 2시간 후까지 혈청 알콜농도가 낮게 나타났다. 대조군의 혈청 알콜농도를 100으로 하였을 때 알콜 투여 30분, 60분, 90분, 120분 경과 후 시험군은 각각 대조군의 84.3%, 89,0%, 85.9%, 75.8%를 나타내어 평균 16% 정도 낮았다. 시험군의 AUC는 대조군의 AUC의 89%로 비지니스 투여군에서 혈액내 알콜의 제거가 빠르게 진전된다는 것을 보여주었다. 또한, 랫드의 에탄을 대사에 미치는 효과를 알아보고자 바이오짐 및 비지니스 투여 후 채혈하여 혈중 에탄을 및 아세트알데히드 농도를 측정하였다. 비즈니스 투여시 혈중 알콜 농도는 알콜 투여 60분 경과후 가장 큰 감소 효과(대조군:83.70$\pm$11.80mg/이, 시험군:45.12$\pm$6.63mg/d1, 47% 감소)를 나타내었으며, 시험군의 AUC는 대조군에 비해 30% 감소하였다. 혈중 아세트알데히드 농도는 투여 60분 후 비지니스 투여군(4.56$\pm$0.51nmol/$m\ell$)이 대조군(6.45$\pm$0.64nmo1/$m\ell$)에 비해 유의성 있는 감소(29%)를 나타내었으며, 시험군의 AUC는 대조군에 비해 35% 감소하였다. 바이오짐 투여시 혈중 에탄을 농도가 알콜 투여 2시간 경과 후 가장 큰 감소 효과(대조군:49.10$\pm$5.20mg/dl, 시험군:25.90$\pm$7.16mg/d1, 47% 감소)를 나타내었으며, 시험군의 AUC는 대조군에 비해 39% 감소하였고, 혈중 아세트알데히드의 농도는 투여 60분후 시험군(3.96$\pm$0.07nmo1/$m\ell$)이 대조군(6.45$\pm$0,64nmo1/$m\ell$)에 비해 유의성 있는 감소(39%)를 나타내었으며, 시험군의 AUC는 대조군에 비해 48% 감소하였다 한편, 시험관내 에탄올 대사 효소에 대한 바이오짐의 효과를 검색해본 결과 바이오짐(2.0 $\mu\textrm{g}$/assay)에 의해 Aldehyde dehydrogenase(1.5unit/assay)의 활성이 14% 증가되었다. 본 연구의 결과로 볼 때, 비지니스 및 바이오짐은 음주 후 상승된 혈중 에탄을 농도 및 아세트알데히드의 농도를 현저히 감소시키는 효과가 있었다.

Effects of Angelica gigas Nakai diet on lipid metabolism, alcohol metabolism and liver function of rats administered with chronic ethanol (당귀의 첨가 식이가 흰쥐의 지방대사와 알콜대사 및 간기능에 미치는 영향)

  • To investigate the effects of Angelica gigas Nakai diet on lipid metabolism, alcohol metabolism and liver function of rats administered with chronic ethanol, Sprague-Dawley male rats were fed either AIN-76 diet (control), control diet with ethanol, control plus Angelica gigas Nakai diet, or control plus Angelica gigas Nakai diet with ethanol for 30 days. On the 21st day, all of the rats were given an oral dose of ethanol and blood-ethanol concentration was monitored for the next 5 hours. The results obtained were: 1) Upon ethanol administration, the blood ethanol concentration was decreased from 2 hr significantly in the group of control plus Angelica gigas Nakai diet compared with control diet group; 2) The blood ethanol oxidation rate was increased in the group of control plus Angelica gigas Nakai diet with ethanol compared with control diet group or control plus Angelica gigas Nakai diet group. After 30 days, rats were sacrificed and then lipid and enzyme determinations in blood and liver were carried out. The results obtained were: 1) LDL-cholesterol in the blood of control plus Angelica gigas Nakai diet group was decreased significantly compared with control diet group; 2) Angelica gigas Nakai diet decreased liver triglyceride and total lipid and blood ${\gamma}-GTP$ level increased due to the chronic ethanol administration. These data suggest that Angelica gigas Nakai can have a recovery function on the symptoms of alcohol related diseases.

A Study on Speed and Changes of Physical Reaction due to Alcohol Intake (혈중알콜농도에 따른 신체반응속도 및 변화연구)

  • This study was carried out not only to determine blood alcohol levels by time but also to examine the changes of working ability and reaction speed after ingestion of alcohol. Fifteen healthy students aged from 21 to 27 volunteered as subjects for this study, Liguor (Sojoo) in concentration of 25% ethyl alcohol was administrated with the amount of 1ml of ethyl alcohol per kg of body weight to the subjects. The concentration of alcohol in the blood were determined by the 'Alcohol Sensor 100' at 5, 30, 60 and 90 minutes after the administration of alcohol. Also, the choice reactiontest, the eye-hand coordination test and kraepelin test were examined at the same time after checking of alcohol concentration in the blood. The results of this study can be summarized as follows. 1. Mean blood alcohol level changes resulting from administration of 1ml of ethyl alcohol per kg of body weight were $0.16%(160{\pm}57mg/100ml,\;0.10%(100{\pm}42mg/100ml),\;0.08%(80{\pm}36mg/100ml)\;and\;0.03%(30{\pm}24mg/100ml)$ at the 3, 30, 60 and 90 minutes after the administration respectively The peak in the concentration of blood alcohol was 5 miniutes after the ingestion according to alcohol examination by the respiration. 2. As for choice reaction test, reaction times became prolonged as blood alcohol levels increased. The reaction time showed a significant changes when the blood alcohol concentration reached 0.08% or more after alcohol ingestion. 3. In eye and hand coordination test, the accuracy of the performance became decreased as blood alcohol levels increased. The difference of accuracy of the test was significantly shown when alcohol levels in the blood reached 0.08% or more after alcohol intake. 4. As for kraepelin test, the abilities of calculation also became lowered as blood alcohol levels increased. The abilities of calculation differed signigicantly from control group when alchool levels of 0.08% and more.

  • Effect of combined preparation (DWP715) containing Alaska pollack extract, maltol, ascorbic acid and nicotinamide on decreasing of blood alcohol was evaluated in human blood. Treatment of DWP715 prior to administration of 25% alcohol (100 mL) decreased alcohol concentration in blood and showed significant difference after 2 hours. The pharmacokinetic parameters such as area under the concentration-time curve (AUC), $C_{max},\;T_{max}\;and\;T_{1/2}$ were also decreased and delayed when compared with control values. Effects of DWP715 on anti-fatigue and anti-oxidation activities were also studied in the restraint stress model using various parameters (GOT, GPT, LDH values and organ weights) on mild condition and examined through the content of lipid peroxide induced by 2% $CCl_4$ in mouse livers. While GPT level, thymus and adrenal weight were not influenced by DWP715 dosing, LDH, GOT level and spleen weight used as a parameter against fatigue and stress states were recovered almost to the nomal level. Furthermore, lipid peroxidation due to $CCl_4$ was significantly inhibited by DWP715 treatment. These results suggest that DWP715 seems to metabolize the blood alcohol rapidly and to restore the damaged liver and fatigue conditions which was caused by alcohol metabolism to normal condition.

The Effect of Hangover Drink using Propolis on Ethanol Oxidation (프로폴리스를 이용한 숙취 해소 음료의 알콜 분해 효과)

  • The studies was conducted to investigate the effect of hangover drink using natural products propolis on ethanol oxidation. The hangover drink developed using propolis concentrates and various medicinal plants. The extractions of propolis and acanthopanax senticosus was shown the highest alcohol oxidation among the combination raw. The ethanol oxidation effect were not significantly different between propolis drink and in other company products in acute alcoholic toxic doses. In the case that it was conducted oral administration with acute alcoholic toxic doses 1/3(1.8mg per 1kg body weight of mouse), propolis drink comparative products of other company was shown 0.026% to 270 min, 0.000% to 360 min. In conclusion, propolis drink was shown excellent alcohol oxidation in fixed dose of alcohol, but was not shown in a large quantities of alcohol.

  • 한국의 도로교통법에는 주취상태에서 운전을 하면 안 된다는 규정이 있다. 그런데 주취운전이라는 말보다 음주운전이라는 말이 더 널리 쓰이는 실정이다. 음주운전 역시 말 그대로 술을 마신 상태에서 하는 운전을 말한다. 우리나라를 비롯한 모든 나라에서는 주취운전의 위험성을 고려하여 이에 대한 단속 기준을 제시하고 있다. 한국은 혈중알콜농도를 확인하는데, 0.05를 단속 기준으로 삼고 있으며 현재 단속기준에 대한 논의가 이루어지고 있다. 경찰은 현재의 음주운전 단속 기준을 0.03%로 강화하는 것에 대하여 의견을 수렴 중이다.

선박운항 시뮬레이터 데이터를 이용한 음주운항 패턴 분석

  • 해양사고의 주요 원인으로 언급되고 있는 피로요인은 장시간 소요되는 선박의 운항과정에서 발생할 수 있는 사고발생의 주된 요인이다. 이러한 피로의 유발 요인 중 하나인 음주운항으로, 이로 인한 해양사고는 대형 인명피해 뿐만 아니라 해양환경오염이라는 국가적 대재난으로 이어질 수 있다. 다행히 「해사안전법」 시행령 개정에 따라 주취운항 단속 기준 (혈중알콜농도 기준)이 기존 0.08%에서 2011년 0.05%, 2014년 0.03%로 강화됨에 따라 주취 운항의 위험성에 대한 인식은 개선되고 있으나, 최근 5년 동안 선박 음주 운항 단속 건수는 총 556건으로 나타났으며, 음주운항으로 인한 충돌, 좌초 등의 사고가 지속적으로 발생하고 있다. 본 발표에서는 음주운항이 선박운항에 미치는 영향을 알아보기 위해 선박운항 시뮬레이터 데이터 분석을 통한 음주운항 패턴 분석 결과를 소개하고자 한다.
